The human immunodeficiency virus type 1 (HIV-1) transactivator (tat) protein produced in one cell activated HIV-1 promoter-directed gene expression in a second cell, provided the cells were in direct contact with one another. Short muscle fibers (less than 1.5 mm) from the m. lumbricalis IV digiti of Rana pipiens were voltage-clamped at -100 mV with a two-microelectrode technique, in normal Ringer's solution containing 10(-6) g/ml tetrodotoxin. The activation curve relating peak tension to membrane potential could be shifted toward more negative or less negative potential values by hyperpolarizing or depolarizing the fiber membrane to -130, -120, or -70 mV, respectively, which indicates that contractile threshold depends on the fiber membrane potential. Long (greater than 5 s) depolarizing (90 mV) pulses induce prolonged contractile responses showing a plateau and a rapid relaxation phase similar to K contractures. Conditioning hyperpolarizations prolong the time course of these responses, while conditioning depolarizations shorten it. The shortening of the response time course, which results in a decrease of the area under the response, is dependent on the amplitude and duration of the conditioning depolarization. Depending on the magnitude and duration, a conditioning depolarization may also reduce peak tension. When the area under the response is reduced by 50%, the level of membrane potential also affects the repriming rate. During repriming, peak tension is restored before the contracture area. Thus, when peak tension is reprimed to 80%, the area is reprimed by 50% of its normal value. Repriming has a marked temperature dependency with a Q10 higher than 4. These results are compatible with the idea that an inactivation process, voltage and time dependent, regulates the release of calcium from the sarcoplasmic reticulum during these responses.  相似文献

Celiac disease (CD) is characterized by abnormally high concentrations of certain peptides in the small bowel. These peptides can be grouped in ‘toxic’ and ‘immunogenic’ classes, which elicit an innate immune response and an HLA‐mediated adaptive response, respectively. It is not clear on which molecular mechanisms responses to these different classes are based, but the 31–43 (P31–43) and the 56–68 (P56–68) A‐gliadin fragments are usually adopted as sequence representatives of toxic and immunogenic peptides, respectively. Here we report fluorescence experiments aiming to mimic the interaction of these peptides with the cell membrane surface by using sodium dodecyl sulphate (SDS) as a membrane‐mimetic medium. We show that P31–43 is able to bind SDS micelles in a way that resembles mixed micelle formation. On the other hand, no binding at all could be detected for P56–68. This different behaviour could be related to the paracellular or transcellular route through which gluten peptides may cross the intestinal epithelium, and open new insights into the pathogenetic mechanisms of CD. Seven freshwater Anabaena sp. strains have been examined for esterase, malate dehydrogenase, acid phosphatase and aldolase isozyme patterns. The morphological differences among them did not allow a sure understanding of their relationships, whereas a phylogenetic analysis of the esterase and malate dehydrogenase patterns led to a single fully resolved tree. A phenetic analysis on the same data set resulted in the same pattern of relationships. Successive cultures of the same strains did not show identical band phenotypes, but nevertheless the phenetic and phylogenetic relationships among the various strains did not change.  相似文献

The 2016 World Health Organisation revised classification of lymphoma has sub‐classified well‐defined entities and added a number of provisional entities on the basis of new knowledge on genetic, epigenetics and phenotypical data; prognostic and predictive features are also part of this classification. New knowledge on well‐defined entities further enlightens the mechanisms of lymphomagenesis, which are more complex and multifactorial than once believed. Therapies are also more complex because traditional clinical trials have been integrated with new drugs and compounds with unique mechanisms of actions against distinct molecular targets. As lymphoma acquires additional genetic and phenotypic features over the time, pathological assessment is also necessary. Histological evaluation and tissue collection by surgical biopsies are necessary for phenotypical and molecular purposes; however, these are demanding procedures for both the patient and the health care system. At the same time, the choice of the best treatment for a specific entity, in different phases and different patients requires information that may not be available when the biopsy is performed. Fine needle aspiration cytology (FNAC) is successfully used in lymph nodes (LNs) in combination with different ancillary techniques and might be used to assess the phenotypic and genetic profile of specific targets and to get key information for therapy, in different phases and stages of the disease, with the option to re‐check the same target over time, without surgical excision. This brief review describes LN‐FNAC diagnostic criteria, current therapies for lymphomas and the potential role of LN‐FNAC in selecting non‐Hodgkin lymphomas patients for specific targeted treatments.  相似文献

TLR3 recognizes dsRNA and activates antiviral immune responses through the production of inflammatory cytokines and type I IFNs. Genetic association studies have provided evidence concerning the role of a polymorphism in TLR3 (rs3775291, Leu412Phe) in viral infection susceptibility. We genotyped rs3775291 in a population of Spanish HIV-1-exposed seronegative (HESN) individuals who remain HIV seronegative despite repeated exposure through i.v. injection drug use (IDU-HESN individuals) as witnessed by their hepatitis C virus seropositivity. The frequency of individuals carrying at least one 412Phe allele was significantly higher in IDU-HESN individuals compared with that of a matched control sample (odds ratio for a dominant model = 1.87; 95% confidence interval, 1.06-3.34; p = 0.023). To replicate this finding, we analyzed a cohort of Italian, sexually HESN individuals. Similar results were obtained: the frequency of individuals carrying at least one 412Phe allele was significantly higher compared with that of a matched control sample (odds ratio, 1.79; 95% confidence interval, 1.05-3.08; p = 0.029). In vitro infection assays showed that in PBMCs carrying the 412Phe allele, HIV-1(Ba-L) replication was significantly reduced (p = 0.025) compared with that of Leu/Leu homozygous samples and was associated with a higher expression of factors suggestive of a state of immune activation (IL-6, CCL3, CD69). Similarly, stimulation of PBMCs with a TLR3 agonist indicated that the presence of the 412Phe allele results in a significantly increased expression of CD69 and higher production of proinflammatory cytokines including IL-6 and CCL3. The data of this study indicate that a common TLR3 allele confers immunologically mediated protection from HIV-1 and suggest the potential use of TLR3 triggering in HIV-1 immunotherapy.  相似文献
