Saprotrophy of Conidiobolus and Basidiobolus in leaf litter   总被引:1,自引:0,他引:1  
This study of the putative saprotrophs of Conidiobolus and Basidiobolus aids the understanding of their ecological roles in litter, and their relationship with the entomogenous fungi of the Entomophthorales. A total of 47 isolates (ten spp.) were screened for their ability to utilise pure compounds, arthropod cadavers, and plant leaf fragments as substrates. Isolates co-occurred in a larch plantation (Larix sp.) or were from adjacent habitats. Of the 21 isolates (nine spp.) tested on potential prime carbon sources, none could utilise common plant structural polymers. Conidiobolus adiaeretus, C. iuxtagenitus, and B. ranarum from litter and some soil isolates of C. heterosporus, C. pumilus, and C. firmipilleus could use starches and glycogen. In marked contrast, all could utilise animal chitin, gelatine, casein, N-acetyl glucosamine, and trehalose. The lipids tributyrin and sunflower oil also supported growth. Conidia on cadavers usually led to high levels of colonisation as was the case for 30 isolates (ten species). Collembola were more frequently and rapidly colonised than mites. Cadavers of many other arthropods were also internally colonised. The ability to utilise cadavers of diverse arthropods indicates that trophic competition between co-occurring test species may be minimal. Niche differentiation may depend more on non-trophic features of their life history. Negative correlation of performance with the presence of naturally occurring, non-test fungi suggests competition with (or antibiosis from) at least some of the other fungi. In washed or unwashed plant fragments of larch litter (F-layer) only occasional local growth and resting spore formation occurred. Extra nutrients did not facilitate colonisation. Alternative forms of repetitional conidia showed a strong association with plant fragments but not with cadavers.  相似文献

Mesocoelium lanfrediaesp. nov. (Digenea: Mesocoeliidae) inhabits the small intestine of Rhinella marina (Amphibia: Bufonidae) and is described here, with illustrations provided by light, scanning electron microscopy and molecular approachs. M. lanfrediae sp. nov. presents the typical characteristics of the genus, but is morphometrically and morphologically different from the species described previously. The main diagnostic characteristics of M. lanfrediae sp. nov. are (i) seven pairs of regularly-distributed spherical papillae on the oral sucker, (ii) ventral sucker outlined by four pairs of papillae distributed in a uniform pattern and interspersed with numerous spines, which are larger at the posterior margin and (iii) small, rounded tegumentary papillae around the opening of the oral sucker, which are morphologically different from those of the oral sucker itself, some of which are randomly disposed in the ventrolateral tegumentary region of the anterior third of the body. Addionally, based on SSU rDNA, a phylogenetic analysis including Brachycoeliidae and Mesocoeliidae taxa available on GenBank established the close relationship between M. lanfrediae sp. nov. and Mesocoelium sp.  相似文献

Cornish Path-moss is one of the world’s rarest bryophytes, presently known from just three sites, former copper mines in the UK and Ireland. Covering the two UK sites, for the first time this study provides a set of location coordinates (x-, y- and z-axis) that is comprehensive, accurate and precise, in addition to investigating aspects of its ecology and assessing the success of trial habitat management plots. The species is shown to be rare, with a total population occupying 490 Ordnance Survey (OS) 10?cm grid cells, 77% at Phoenix United Mine and the remainder at South Caradon Mine, confined to bare patches of metalliferous soil that are especially high in the phytotoxins arsenic, cobalt and copper. Whilst the hostile chemical environment limits competition, various tolerant and larger bryophytes, lichens and vascular plants are shown to occupy the same niche, so that competitive exclusion is a constant threat. Turf-stripping of trial habitat management plots using a mechanical excavator at Phoenix United Mine, re-exposing bare metalliferous ground, is shown to be remarkably successful, providing a 20-fold increase in the population, presently making it the most important site for the plant in the world. Priorities for future survey and research are considered.  相似文献

Alloantibody is an important effector mechanism for allograft rejection. In this study, we tested the hypothesis that regulatory T cells with indirect allospecificity can prevent humoral rejection by using a rat transplant model in which acute rejection of MHC class I-disparate PVG.R8 heart grafts by PVG.RT1(u) recipients is mediated by alloantibody and is dependent upon help from CD4 T cells that can recognize the disparate MHC alloantigen only via the indirect pathway. Pretransplant treatment of PVG.RT1(u) recipients with anti-CD4 mAb plus donor-specific transfusion abrogated alloantibody production and prolonged PVG.R8 graft survival indefinitely. Naive syngeneic splenocytes injected into tolerant animals did not effect heart graft rejection, suggesting the presence of regulatory mechanisms. Adoptive transfer experiments into CD4 T cell-reconstituted, congenitally athymic recipients confirmed that regulation was mediated by CD4 T cells and was alloantigen-specific. CD4 T cell regulation could be broken in tolerant animals either by immunizing with an immunodominant linear allopeptide or by depleting tolerant CD4 T cells, but surprisingly this resulted in neither alloantibody generation nor graft rejection. These findings demonstrate that anti-CD4 plus donor-specific transfusion treatment results in the development of CD4 regulatory T cells that recognize alloantigens via the indirect pathway and act in an Ag-specific manner to prevent alloantibody-mediated rejection. Their development is associated with intrinsic tolerance within the alloantigen-specific B cell compartment that persists after T cell help is made available.  相似文献

Serum amyloid P (SAP) is a common component of human amyloid deposits and has been identified in atherosclerotic lesions. We investigated the extent of the colocalization of SAP with apolipoprotein A-I (apoA-I), apoB, apoC-II, and apoE in human coronary arteries and explored potential roles for SAP in these regions, specifically the effect of SAP on the rate of formation and macrophage recognition of amyloid fibrils composed of apoC-II. Analysis of 42 human arterial sections by immunohistochemistry and double label fluorescence microscopy demonstrated that SAP and apoA-I, apoB, apoC-II, and apoE were increased significantly in atherosclerotic lesions compared with nonatherosclerotic segments. SAP colocalized with all four apolipoproteins to a similar extent, whereas plaque macrophages were found to correlate most strongly with apoC-II and apoB. In vitro studies showed that SAP accelerated the formation of amyloid fibrils by purified apoC-II. Furthermore, SAP strongly inhibited the phagocytosis of apoC-II amyloid fibrils by primary macrophages and macrophage cell lines and blocked the resultant production of reactive oxygen species. The ability of SAP to accelerate apoC-II amyloid fibril formation and inhibit macrophage recognition of apoC-II fibrils suggests that SAP may modulate the inflammatory response to amyloid fibrils in atherosclerosis.  相似文献

Background

Telomeric 3′ overhangs can fold into a four-stranded DNA structure termed G-quadruplex (G4), a formation which inhibits telomerase. As telomerase activation is crucial for telomere maintenance in most cancer cells, several classes of G4 ligands have been designed to directly disrupt telomeric structure.

Methods

We exposed brain tumor cells to the G4 ligand 3,11-difluoro-6,8,13-trimethyl-8H-quino[4,3,2-kl]acridinium methosulfate (RHPS4) and investigated proliferation, cell cycle dynamics, telomere length, telomerase activity and activated c-Myc levels.

Results

Although all cell lines tested were sensitive to RHPS4, PFSK-1 central nervous system primitive neuroectodermal cells, DAOY medulloblastoma cells and U87 glioblastoma cells exhibited up to 30-fold increased sensitivity compared to KNS42 glioblastoma, C6 glioma and Res196 ependymoma cells. An increased proportion of S-phase cells were observed in medulloblastoma and high grade glioma cells whilst CNS PNET cells showed an increased proportion of G1-phase cells. RHPS4-induced phenotypes were concomitant with telomerase inhibition, manifested in a telomere length-independent manner and not associated with activated c-Myc levels. However, anti-proliferative effects were also observed in normal neural/endothelial cells in vitro and ex vivo.

Conclusion

This study warrants in vivo validation of RHPS4 and alternative G4 ligands as potential anti-cancer agents for brain tumors but highlights the consideration of dose-limiting tissue toxicities.  相似文献

Phytosterols and their oxidation products have become increasingly investigated in recent years with respect to their roles in diet and nutrition. We present a comprehensive review of recent literature on Phytosterol Oxidation Products (POP) identifying critical areas for future investigation. It is evident that POP are formed on food storage/preparation; are absorbed and found in human serum; do not directly affect cholesterol absorption; have evidence of atherogenicity and inflammation; have distinct levels of cytotoxicity; are implicated with high levels of oxidative stress, glutathione depletion, mitochondrial dysfunction and elevated caspase activity.  相似文献
