Islands played a key role in Charles Darwin's observations and experiments on plant dispersal. By means of these experiments, he expunged the old idea that a given species could originate at multiple times and in multiple places. More importantly, by seeing the capabilities for dispersal of plant seeds, fruits and branches, he was able to develop ideas of how plants reach islands and thus he is one of the founders of plant biogeography. For facts regarding floristic distribution of plants, Darwin relied on other workers, most notably Sir Joseph Dalton Hooker. Among his insights were the differences between oceanic and continental islands on a floristic basis, ideas on how age of island and distance from mainland areas influenced composition of island floras, the nature of endemism on islands and the role islands and archipelagos served as stepping stones in dispersal. Ingenious at proposing hypotheses, but always respectful of facts, Darwin sought explanations for plant adaptations on islands at a time when knowledge of island botany was little more than floristic in nature. N-Linked glycosylation of a baculovirus-expressed recombinant glycoprotein in insect larvae and tissue culture cells 总被引:5,自引:3,他引:2
The potential of insect cell cultures and larvae infected with recombinant
baculoviruses to produce authentic recombinant glycoproteins cloned from
mammalian sources was investigated. A comparison was made of the N-linked
glycans attached to secreted alkaline phosphatase (SEAP) produced in four
species of insect larvae and their derived cell lines plus one additional
insect cell line and larvae of one additional species. These data survey
N-linked oligosaccharides produced in four families and six genera of the
order Lepidoptera. Recombinant SEAP expressed by recombinant isolates of
Autographa californica and Bombyx mori nucleopolyhedroviruses was purified
from cell culture medium, larval hemolymph or larval homogenates by
phosphate affinity chromatography. The N-linked oligosaccharides were
released with PNGase-F, labeled with 8- aminonaphthalene-1-3-6-trisulfonic
acid, fractionated by polyacrylamide gel electrophoresis, and analyzed by
fluorescence imaging. The oligosaccharide structures were confirmed with
exoglycosidase digestions. Recombinant SEAP produced in cell lines of
Lymantria dispar (IPLB-LdEIta), Heliothis virescens (IPLB-HvT1), and Bombyx
mori (BmN) and larvae of Spodoptera frugiperda, Trichoplusia ni ,
H.virescens , B.mori , and Danaus plexippus contained oligosaccharides that
were structurally identical to the 10 oligosaccharides attached to SEAP
produced in T.ni cell lines. The oligosaccharide structures were all
mannose-terminated. Structures containing two or three mannose residues,
with and without core fucosylation, constituted more than 75% of the
oligosaccharides from the cell culture and larval samples.

Quantitative and qualitative data are presented for seven collections representing two varieties (unlike in habit) of Gnetum gnemon. Tracheids are present, but abundant and intermixed with them are septate fibre-tracheids rich in starch. Axial parenchyma has been reported only once previously for the species. Axial parenchyma is in strands of 4–10 cells, is rich in starch, is primarily vasicentric (paratracheal) in distribution, less commonly diffuse. About equally common are simple and compound perforation plates; the latter are composed of from two to about ten bordered foraminate perforations, the shape of which may be altered by crowding or coalescence, but is clearly still foraminate. Lateral walls of vessels bear pits that are vestured around pit cavities, not facing the pit membrane. Rays are composed mostly of procumbent cells; the tangential walls bear bordered pits. Crystals, present in ray cells and (rarely) axial parenchyma vary widely in size. Crystalliferous sclereids with layered walls, starch-rich parenchyma, and gelatinous secondary phloem fibres are the main components of bark. Early stages in origin of successive vascular cambia in bark are newly described. When representative conditions are derived from study of large numbers of slides, the classical view that Gnetum vessels are unlike those of angiosperms is supported. Features of Gnetum gnemon wood are discussed in the light of ecology and conductive physiology. 相似文献

Wood samples of stems, lignotubers, and roots of the majority of species of Penaeaceae were analyzed with respect to qualitative and quantitative features. Virtually no data have hitherto been presented on xylem features of this family, restricted to Cape Province, South Africa. Presence of vestured pits in vessels, septate crystalliferous parenchyma in wood, intraxylary phloem, predominantly erect ray cells in the typically narrow, multiseriate rays and in the uniseriate rays, and amorphous deposits in ray cells place Penaeaceae securely in Myrtales and help to define that order. By comparison of ecological preferences of the species, as observed during field work, with quantitative analysis of conductive tissue, close correspondence of the wood structure to habit and habitat is demonstrated. 相似文献

In situ hybridization has become a standard method for localizing DNA or RNA sequences in cytological preparations. We developed two methods to extend this technique to the transmission electron microscope level using mouse satellite DNA hybridization to whole mount metaphase chromosomes as the test system. The first method devised is a direct extension of standard light microscope level using mouse satellite DNA hybridization to whole mount metaphase chromosomes as the test system. Radioactively labeled complementary RNA (cRNA) is hybridized to metaphase chromosomes deposited on electron microscope grids and fixed in 70 percent ethanol vapor; hybridixation site are detected by autoradiography. Specific and intense labeling of chromosomal centromeric regions is observed even after relatively short exposure times. Inerphase nuclei present in some of the metaphase chromosome preparations also show defined paatterms of satellite DNA labeling which suggests that satellite-containing regions are associate with each other during interphase. The sensitivity of this method is estimated to at least as good as that at the light microscope level while the resolution is improved at least threefold. The second method, which circumvents the use of autoradiogrphic detection, uses biotin-labeled polynucleotide probes. After hybridization of these probes, either DNA or RNA, to fixed chromosomes on grids, hybrids are detected via reaction is improved at least threefold. Wood and stem anatomy is studied for seven species of six genera (root anatomy also reported for one species) of Amaranthaceae s.s. Quantitative data on vessels correlate closely with relative xeromorphy of respective species, agreeing with values reported for dicotyledons without successive cambia in comparable habitats. Libriform fibre abundance increases and vessel diameter decreases as stems and roots of the annual Amaranthus caudatus mature. Long, thick-walled fibres in Bosea yervamora may be related to the upright nature of elongate semi-climbing stems. Non-bordered or minutely bordered perforation plates characterize Amaranthaceae, as they do most other Caryophyllales. Amaranthaceae have idioblastic cells containing druses, rhomboidal crystals or crystal sand: these forms intergrade and seem closely related. Rays are present in secondary xylem of the Amaranthaceae studied. Cells intermediate between ray cells and libriform fibres occur in Charpentiera elliptica . Degrees of diversity in rays and reports of raylessness in Amaranthaceae induce discussion of definition and identification of rays in dicotyledons; some sources recognize both rays and radial plates of conjunctive tissue in Amaranthaceae. The action of successive cambia is described: lateral meristem periclinal divisions produce secondary cortex externally, conjunctive tissue internally and yield vascular cambia as well. Vascular cambia produce secondary phloem and secondary xylem, in both ray and fascicular zones, as in a dicotyledon with a single cambium. For the single species of Austmbaileya (Austrobaileyaceae), quantitative and qualitative data are offered on the basis of a mature stem and a root of moderate diameter. Data available hitherto have been based on stems of small to moderate diameter, and roots have not previously been studied. Scanning electron microscope (SEM) photographs are utilized for roots, and show compound starch grains. Roots lack sclerenchyma but have relatively narrow vessels and abundant ray tissue. Recent phylogenies group Austrobaileyaceae with the woody families Illiciaceae, Schisandraceae, and Trimeniaceae (these four may be considered Illiciales), and somewhat less closely with the vesselless families Amborellaceae and Winteraceae and the aquatic families Cambombaceae and Nymphaeaceae. The vessel-bearing woody families above share vessels with scalariform perforation plates; bordered bars on plates; pit membrane remnants present in perforations; lateral wall pitting of vessels mostly alternate and opposite; tracheids and/or septate fibre-tracheids present; axial parenchyma vasicentric (sometimes abaxial); rays Heterogeneous Type I; ethereal oil cells present; stomata paracytic or variants of paracytic. Although comparisons between vessel-bearing and vesselless families must depend on fewer features, Amborellaceae and Winteraceae have no features incompatible with their inclusion in an expanded Illiciales. 相似文献
