A purification procedure is described for the components of Bacillus polymyxa nitrogenase. The procedure requires the removal of interfering mucopolysaccharides before the two nitrogenase proteins can be purified by the methods used with other nitrogenase components. The highest specific activities obtained were 2750 nmol C2H4 formed . min-1 . mg-1 MoFe protein and 2521 nmol C2H4 formed . min-1 . mg-1 Fe protein. The MoFe protein has a molecular weight of 215 000 and contains 2 molybdenum atoms, 33 iron atoms and 21 atoms of acid-labile sulfur per protein molecule. The Fe protein contains 3.2 iron atoms and 3.6 acid-labile sulfur atoms per molecule of 55 500 molecular weight. Each Fe protein binds two ATP molecules. The EPR spectra are similar to those of other nitrogenase proteins. MgATP changes the EPR of the Fe protein from a rhombic to an axial-type signal.  相似文献

Most species of pycnogonids are sexually dimorphic and all have exclusive male care of the offspring, characteristics that make them essential for studies on sex-roles, sexual selection, and parental investment. However, sea spiders have been understudied because of their small size, cryptic coloration, and often patchy distribution, and little is known about their courtship and mating behaviors. The mating habits of both male and female Achelia simplissima (Hilton 1939) were studied experimentally and observationally. This species mates year-round and females commonly initiate courtship by actively ‘pumping’ their bodies near a male. When both nonparental and parental males were present, females mated significantly more often with males that were not carrying egg masses. Each mating event resulted in a single egg mass. Most often, males placed the first egg mass on their right oviger and then alternated between ovigers with subsequent matings. The newest (least developed) egg mass was always placed at the tip of the oviger. Both males and females routinely mated multiple times and had multiple mates; in the field, males were found carrying up to twelve egg masses simultaneously. This is the first experimental study to describe both the male and female mating system of a pycnogonid, and the first observations of courtship and mating for any species with the ammotheid type of ovigers.  相似文献

Selenium (Se) content of feedstuffs is dependent on the Se level of the soil. Even though Se in grass and forage crops is primarily present in organic forms, Se is commonly supplemented in cattle diets in an inorganic (sodium selenite) form in geographic regions where Se soil concentrations are low. The purpose of this study was to answer two important questions about inorganic (ISe) vs organic (OSe) forms of dietary supplementation of Se (3?mg/day) to growing beef heifers (0.5?kg/day): (1) what would the effect of supplementing Se with an equal blend of ISe:OSe (Mix) have on Se tissue concentrations and (2) how long does it take for the greater assimilation with OSE to occur and stabilize? A long-term (224?day) Se dietary supplementation trial was conducted with serial sampling performed (days?28, 56, 112, and 224) to determine the length of time required to achieve Se supplement (OSE, Mix, and ISe)-dependent changes in Se assimilation in blood fractions and liver tissue. Forty maturing Angus heifers were fed a corn silage-based diet for 98?days with no Se supplementation, and then a cracked corn/cottonseed hull-based diet (basal diet) without Se supplementation for 74?days. Liver biopsies were taken for Se analysis, and heifers were fed the same diet for another 14?days. Heifers were assigned (n?=?10) to one of four Se treatment groups such that basal liver Se contents were stratified among groups, and then fed enough of the basal diet (0.08?mg Se per day) and a mineral-vitamin mix that provided 0.16 (control) or 3.0?mg Se per day in ISe (sodium selenite), OSe (Sel-Plex(?)), or Mix (1:1 ISe:OSe) form to support 0.5?kg/day growth for 224?days. More Se was found in whole blood, red blood cells, serum, and liver of Mix and OSe heifers than ISe heifers, and all were greater than control. Se content either increased until day?56 then was stable (liver and plasma), or was stable until day?56 (whole blood) or day?112 (red blood cells) and then increased steadily through day?224, for all supplemental Se treatments. These data indicate that a 1:1 mix (1.5?mg Se:1.5?mg Se) of supplemental ISe and OSe is equal to 3?mg/day OSe supplementation and greater than 3?mg/day ISe supplementation. The data also indicate that Se levels stabilized in liver and plasma by 56 to 112?days whereas whole blood and red blood cell concentrations were still increasing through 224?days of supplementation, regardless of the form of supplemental Se.  相似文献

Though the presence of antioxidized low-density lipoprotein IgG is well documented in clinical and animal studies, the role for FcγRs to the progression of atherosclerosis has not been studied in detail. In the current study, we investigated the role for activating FcγR in the progression of atherosclerosis using apolipoprotein E (apoE)-Fcγ-chain double-knockout (DKO) mice. Relative to apoE knockout (KO) mice, arterial lesion formation was significantly decreased in apoE-Fcγ-chain DKO mice. Bone marrow chimera studies showed reduced lesions in apoE KO mice receiving the bone marrow of apoE-Fcγ-chain DKO mice. Compared to apoE KO mice, antioxidized low-density lipoprotein IgG1 (Th2) and IgG2a (Th1), IL-10, and IFN-γ secretion by activated T cells was increased in apoE-Fcγ-chain DKO mice. These findings suggest that reduced atherosclerotic lesion in apoE-Fcγ-chain DKO mice is not due to a Th1/Th2 imbalance. Interestingly, the number of Th17 cells and the secretion of IL-17 by activated CD4(+) cells were decreased in apoE-Fcγ-chain DKO mice. Notably, the number of regulatory T cells, expression of mRNA, and secretion of TGF-β and IL-10 were increased in apoE-Fcγ-chain DKO mice. Furthermore, secretions of IL-6 and STAT-3 phosphorylation essential for Th17 cell genesis were reduced in apoE-Fcγ-chain DKO mice. Importantly, decrease in Th17 cells in apoE-Fcγ-chain DKO mice was due to reduced IL-6 release by APC of apoE-Fcγ-chain DKO mice. Collectively, our data suggest that activating FcγR promotes atherosclerosis by inducing a Th17 response in the hyperlipidemic apoE KO mouse model.  相似文献

The goal of this research was to generate a breeding population of horseweed segregating for glyphosate resistance. In order to generate a marker to select between hybrids of glyphosate resistant (GR) and glyphosate susceptible (GS) horseweed, a GR horseweed accession from western Tennessee was transformed with a green fluorescent protein (GFP) transgene. The GFP marker allowed for the simple and accurate determination of GR hybrid plants by visual observation. GR plants were shown to be transgenic via the green fluorescence under UV light, and resistant to glyphosate when sprayed with the field-use-rate 0.84 kg acid equivalent ha−1 of glyphosate (i.e. RoundupTM) herbicide. An in vitro screen for glyphosate resistance in seedlings was developed, and a 5 μM glyphosate concentration was found to reduce dry weight in GS seedlings but not in GR seedlings. The GR plants containing GFP were then hand-crossed with GS plants from eastern Tennessee under greenhouse conditions, with GS plants acting as the pollen acceptor. Resulting seed was collected and germinated for GFP fluorescence screening. Seedlings that exhibited the transgenic GFP phenotype were selected as F1 hybrids between GR and GS horseweed. Thirty GS×GR hybrids were produced on the basis of a green-fluorescent GFP phenotype of GR plants. GS×GFP/GR F1 hybrids produced F2 seeds, and F2 plants were shown to segregate for GFP fluorescence and glyphosate resistance independently. Both traits segregated at a Mendelian 3:1 ratio, indicating a single gene is responsible for each phenotype.  相似文献

Hutchinson‐Gilford Progeria Syndrome (HGPS) is a devastating premature aging disease. Mouse models have been instrumental for understanding HGPS mechanisms and for testing therapies, which to date have had only marginal benefits in mice and patients. Barriers to developing effective therapies include the unknown etiology of progeria mice early death, seemingly unrelated to the reported atherosclerosis contributing to HGPS patient mortality, and mice not recapitulating the severity of human disease. Here, we show that progeria mice die from starvation and cachexia. Switching progeria mice approaching death from regular diet to high‐fat diet (HFD) rescues early lethality and ameliorates morbidity. Critically, feeding the mice only HFD delays aging and nearly doubles lifespan, which is the greatest lifespan extension recorded in progeria mice. The extended lifespan allows for progeria mice to develop degenerative aging pathologies of a severity that emulates the human disease. We propose that starvation and cachexia greatly influence progeria phenotypes and that nutritional/nutraceutical strategies might help modulate disease progression. Importantly, progeria mice on HFD provide a more clinically relevant animal model to study mechanisms of HGPS pathology and to test therapies.  相似文献

For nearly a century, articular cartilage has been known for its exceptional tribological properties. For nearly as long, there have been research efforts to elucidate the responsible mechanisms for application toward biomimetic bearing applications. It is now widely accepted that interstitial fluid pressurization is the primary mechanism responsible for the unusual lubrication and load bearing properties of cartilage. Although the biomechanics community has developed elegant mathematical theories describing the coupling of solid and fluid (biphasic) mechanics and its role in interstitial lubrication, quantitative gaps in our understanding of cartilage tribology have inhibited our ability to predict how tribological conditions and material properties impact tissue function. This paper presents an analytical model of the interstitial lubrication of biphasic materials under migrating contact conditions. Although finite element and other numerical models of cartilage mechanics exist, they typically neglect the important role of the collagen network and are limited to a specific set of input conditions, which limits general applicability. The simplified approach taken in this work aims to capture the broader underlying physics as a starting point for further model development. In agreement with existing literature, the model indicates that a large Peclet number, Pe, is necessary for effective interstitial lubrication. It also predicts that the tensile modulus must be large relative to the compressive modulus. This explains why hydrogels and other biphasic materials do not provide significant interstitial pressure under high Pe conditions. The model quantitatively agrees with in-situ measurements of interstitial load support and the results have interesting implications for tissue engineering and osteoarthritis problems. This paper suggests that a low tensile modulus (from chondromalacia or local collagen rupture after impact, for example) may disrupt interstitial pressurization, increase shear stresses, and activate a condition of progressive surface damage as a potential precursor of osteoarthritis.  相似文献
