Ribosome biogenesis requires, in addition to rRNA molecules and ribosomal proteins, a multitude of trans-acting factors. Recently it has become clear that in the yeast Saccharomyces cerevisiae many RNA helicases of the DEAD-box and related families are involved in ribosome biogenesis. Here we show that the previously uncharacterised open reading frame YDL031w (renamed DBP10 for DEAD-box protein 10) encodes an essential putative RNA helicase that is required for accurate ribosome biogenesis. Genetic depletion of Dbp10p results in a deficit in 60S ribosomal subunits and an accumulation of half-mer polysomes. Furthermore, pulse-chase analyses of pre-rRNA processing reveal a strong delay in the maturation of 27SB pre-rRNA intermediates into 25S rRNA and 7S pre-rRNA. Northern blot analyses indicate that this delay leads to higher steady-state levels of 27SB species and reduced steady-state levels of 7S pre-rRNA and 25S/5.8S mature rRNAs, thus explaining the final deficit in 60S subunit and the formation of half-mer polysomes. Consistent with a direct role in ribosome biogenesis, Dbp10p was found to be located predominantly in the nucleolus.  相似文献

Because previous studies have shown that a high molecular mass constituent of cranberry juice inhibited adhesion of Escherichia coli to epithelial cells and coaggregation of oral bacteria, we have examined its effect on the adhesion of Helicobacter pylori to immobilized human mucus and to erythrocytes. We employed three strains of H. pylori all of which bound to the mucus and agglutinated human erythrocytes via a sialic acid-specific adhesin. The results showed that a high molecular mass constituent derived from cranberry juice inhibits the sialic acid-specific adhesion of H. pylori to human gastric mucus and to human erythrocytes.  相似文献

A neural network model with incremental Hebbian learning of afferent and lateral synaptic couplings is proposed,which simulates the activity-dependent self-organization of grating cells in upper layers of striate cortex. These cells, found in areas V1 and V2 of the visual cortex of monkeys, respond vigorously and exclusively to bar gratings of a preferred orientation and periodicity. Response behavior to varying contrast and to an increasing number of bars in the grating show threshold and saturation effects. Their location with respect to the underlying orientation map and their nonlinear response behavior are investigated. The number of emerging grating cells is controlled in the model by the range and strength of the lateral coupling structure.  相似文献

Damselfishes are well known for their aggressive, territorial behaviour during which the use of vocalization behaviour has been well documented. However, agonistic acoustic signalling has been understudied in particular when the vocalizations are interspecific. In this study, we characterize and compare the previously undescribed vocalization behaviour of longfin damselfish (Stegastes diencaeus), in an agonistic context, with the closely related and sympatric dusky damselfish (Stegastes adustus). Next, we examined if these congeneric species modulate their vocalizations in a similar pattern to previously described aggressive behaviour patterns. Audio field recordings of territorial males were obtained in response to three separate stimuli: (1) conspecific male damselfish, (2) heterospecific male damselfish and (3) a common intruder, the slippery dick wrasse (Halichoeres bivittatus). The vocal repertoires of both longfin and dusky damselfish comprised the same three distinct call types: chirps, pops and pulse trains. However, temporal measures of the calls showed significant differences between species. Additionally, dusky damselfish were more vocal overall, producing more calls and spending more time calling than longfin damselfish. These responses were stimulus and species dependent, as the two species modulated acoustic response by modulating pulse number based on intruder species. These results suggest that these closely related species of damselfish use vocalization behaviours that are both unique and context dependent.  相似文献

Specialist and generalist bees use olfactory and visual cues to find and recognise flowering plants. Specialised (oligolectic) bees rely on few host plants for pollen collection. These bee species are suggested to use specific volatiles, but it is unknown whether they have dedicated adaptations for these particular compounds compared to bees not specialised on the same plants. In the present study, we investigated the perception of host odorants and its neuronal substrate with regard to host-plant finding behaviour in oligolectic bees. We reconstructed the antennal lobes (AL) in the Salix specialist, Andrena vaga, and counted about 135 glomeruli and thereby less than the approximately 160 in honeybees. Using calcium imaging experiments to measure neural activity in the bee brain, we recorded odorant-evoked activity patterns in the AL of A. vaga and, for comparison, in the generalist honeybee, Apis mellifera. Our physiological experiments demonstrated that A. vaga bees were particularly sensitive to 1,4-dimethoxybenzene, a behaviour-mediating odorant of Salix host flowers. We found more sensitive glomeruli in the specialised bees as compared to generalist honeybees. This neural adaptation might allow oligolectic A. vaga bees to effectively locate host plants from distances.  相似文献

Butterflies are important components of biodiversity in grassland ecosystems and provide ecosystem services such as pollination. Although agricultural intensification has led to a scarcity of native grassland habitats within most agricultural landscapes of North America, fragmented remnants and semi-natural habitats may support diverse communities, including butterflies, as long as vital resources such as host plants are available. The United States Department of Agriculture’s (USDA) Conservation Reserve Program practice CP33 Habitat Buffers for Upland Birds (USDA 2004) provides semi-natural grassland habitat in agricultural landscapes, but a knowledge gap exists about impacts of prescribed disturbance (e.g. burning or disking) on butterflies. We monitored butterfly habitat and butterfly communities on experimentally manipulated CP33 grassland buffers in Clay County, Mississippi from 2007 to 2009. Disturbance guild butterfly species richness did not differ among treatments. However, disturbance guild abundance was positively affected by disking in both the first and second growing seasons following disking, and the magnitude of this response varied between years. Effects of burning on disturbance guild abundance did not differ from the control treatment. There were no treatment differences for grassland guild butterfly abundance and species richness suggesting that periodic disturbance does not unduly impact grassland-associated butterflies in the southeastern US. Our results support current USDA practice standards that require periodic disturbance during the 10-year contract, but restrict the disturbance to 1/3 or 1/4 of grassland buffer area in a given year.  相似文献

The ubiquitin-proteolytic system controls the stability of proteins in space and time. In this study, using a temperature-sensitive mutant allele of the cul-2 gene, we show that CRL2LRR-1 (CUL-2 RING E3 ubiquitin-ligase and the Leucine Rich Repeat 1 substrate recognition subunit) acts at multiple levels to control germline development. CRL2LRR-1 promotes germ cell proliferation by counteracting the DNA replication ATL-1 checkpoint pathway. CRL2LRR-1 also participates in the mitotic proliferation/meiotic entry decision, presumably controlling the stability of meiotic promoting factors in the mitotic zone of the germline. Finally, CRL2LRR-1 inhibits the first steps of meiotic prophase by targeting in mitotic germ cells degradation of the HORMA domain-containing protein HTP-3, required for loading synaptonemal complex components onto meiotic chromosomes. Given its widespread evolutionary conservation, CUL-2 may similarly regulate germline development in other organisms as well.  相似文献

Chordomas are rare primary bone tumors that occur along the neuraxis. Primary treatment is surgery, often followed by radiotherapy. Treatment options for patients with recurrence are limited and, notably, there are no FDA approved therapeutic agents. Development of therapeutic options has been limited by the paucity of preclinical model systems. We have established and previously reported the initial characterization of the first patient-derived chordoma xenograft model. In this study, we further characterize this model and demonstrate that it continues to resemble the original patient tumor histologically and immunohistochemically, maintains nuclear expression of brachyury, and is highly concordant with the original patient tumor by whole genome genotyping. Pathway analysis of this xenograft demonstrates activation of epidermal growth factor receptor (EGFR). In vitro studies demonstrate that two small molecule inhibitors of EGFR, erlotinib and gefitinib, inhibit proliferation of the chordoma cell line U-CH 1. We further demonstrate that erlotinib significantly inhibits chordoma growth in vivo. Evaluation of tumors post-treatment reveals that erlotinib reduces phosphorylation of EGFR. This is the first demonstration of antitumor activity in a patient-derived chordoma xenograft model and these findings support further evaluation of EGFR inhibitors in this disease.  相似文献
