Crosslinking of proteins by tissue transglutaminase (tTG) is enhanced in amyloid (Abeta) deposits characteristic of Alzheimer's disease and sporadic inclusion body myositis. Small heat shock proteins (sHsps) also occur in amyloid deposits. We here report the substrate characteristics for tTG of six sHsps. Hsp27, Hsp20 and HspB8 are both lysine- and glutamine-donors, alphaB-crystallin only is a lysine-donor, HspB2 a glutamine-donor, and HspB3 no substrate at all. Close interaction of proteins stimulates crosslinking efficiency as crosslinking between different sHsps only takes place within the same heteromeric complex. We also observed that alphaB-crystallin, Hsp27 and Hsp20 associate with Abeta in vitro, and can be readily crosslinked by tTG.  相似文献

The anteroposterior and dorsoventral axes of the future embryo are specified within Drosophila oocytes by localizing gurken mRNA, which targets the secreted Gurken transforming growth factor-alpha synthesis and transport to the same site. A key question is whether gurken mRNA is targeted to a specialized exocytic pathway to achieve the polar deposition of the protein. Here, we show, by (immuno)electron microscopy that the exocytic pathway in stage 9-10 Drosophila oocytes comprises a thousand evenly distributed transitional endoplasmic reticulum (tER)-Golgi units. Using Drosophila mutants, we show that it is the localization of gurken mRNA coupled to efficient sorting of Gurken out of the ER that determines which of the numerous equivalent tER-Golgi units are used for the protein transport and processing. The choice of tER-Golgi units by mRNA localization makes them independent of each other and represents a nonconventional way, by which the oocyte implements polarized deposition of transmembrane/secreted proteins. We propose that this pretranslational mechanism could be a general way for targeted secretion in polarized cells, such as neurons.  相似文献

Statistical predictions of the impact of climate change on biodiversity assume that the environmental and spatial characteristics of contemporary species’ distributions reflect the conditions needed for their continued and prolonged existence. Here we explore this assumption by testing whether a species’ threatened status is associated with the amount of variation in its distribution range attributable to environmental and spatial patterns. Using a variation partitioning approach, we decomposed variation in the distribution ranges of 4423 vertebrate species in sub-Saharan Africa into components attributable exclusively to environmental variables (E|S), exclusively to spatial variables (S|E) or to the collinearity between environmental and spatial variables (E∩S). We found that species’ threatened status was unrelated to E|S, S|E or E∩S variation components, but that unexplained variation was higher for species threatened with extinction. This suggests that spatio-environmental patterns in species’ ranges likely underestimate the overall extinction threat caused by climate change. We also found clear geographic patterns in the strength of E|S, S|E or E∩S that differed amongst biogeographical regions, but no component was over- or underrepresented in the present-day protected area network. While there may be benefits to tailoring protected area expansion to differences between biogeographical regions, this should aim to incorporate species-specific information wherever possible.  相似文献

Both constitutive and inducible antipredator strategies are ubiquitous in nature and serve to maximize fitness under a predation threat. Inducible strategies may be favored over constitutive defenses depending on their relative cost and benefit and temporal variability in predator presence. In African temporary ponds, annual killifish of the genus Nothobranchius are variably exposed to predators, depending on whether larger fish invade their habitat from nearby rivers during floods. Nonetheless, potential plastic responses to predation risk are poorly known. Here, we studied whether Nothobranchius furzeri individuals adjust their life history in response to a predation threat. For this, we monitored key life history traits in response to cues that signal the presence of predatory pumpkinseed sunfish (Lepomis gibbosus). While growth rate, adult body size, age at maturation, and initial fecundity were not affected, peak and total fecundity were higher in the predation risk treatment. This contrasts with known life history strategies of killifish from permanent waters, which tend to reduce reproduction in the presence of predators. Although our results show that N. furzeri individuals are able to detect predators and respond to their presence by modulating their reproductive output, these responses only become evident after a few clutches have been deposited. Overall our findings suggest that, in the presence of a predation risk, it can be beneficial to increase the production of life stages that can persist until the predation risk has faded.  相似文献

S‐adenosyl‐l ‐methionine (SAM) is the major methyl donor in cells and it is also used for the biosynthesis of polyamines and the plant hormone ethylene. During climacteric ripening of tomato (Solanum lycopersicum ‘Bonaparte’), ethylene production rises considerably which makes it an ideal object to study SAM involvement. We examined in ripening fruit how a 1‐MCP treatment affects SAM usage by the three major SAM‐associated pathways. The 1‐MCP treatment inhibited autocatalytic ethylene production but did not affect SAM levels. We also observed that 1‐(malonylamino)cyclopropane‐1‐carboxylic acid formation during ripening is ethylene dependent. SAM decarboxylase expression was also found to be upregulated by ethylene. Nonetheless polyamine content was higher in 1‐MCP‐treated fruit. This leads to the conclusion that the ethylene and polyamine pathway can operate simultaneously. We also observed a higher methylation capacity in 1‐MCP‐treated fruit. During fruit ripening substantial methylation reactions occur which are gradually inhibited by the methylation product S‐adenosyl‐l ‐homocysteine (SAH). SAH accumulation is caused by a drop in adenosine kinase expression, which is not observed in 1‐MCP‐treated fruit. We can conclude that tomato fruit possesses the capability to simultaneously consume SAM during ripening to ensure a high rate of ethylene and polyamine production and transmethylation reactions. SAM usage during ripening requires a complex cellular regulation mechanism in order to control SAM levels.  相似文献

Nitrite-oxidizing bacteria are key members of the global nitrogen cycle but their study is hampered by their limited availability in culture, mostly due to laborious cultivation procedures and the lack of stable preservation methods. In this study, it was demonstrated that long-term cryopreservation of nitrite-oxidizing bacteria assigned to the genera Nitrobacter, Nitrospina, Nitrococcus, Nitrotoga and Nitrospira was possible using a simple and rapid protocol. Their survival was tested with different cryoprotecting agents, DMSO and Hatefi, and in various carbon-rich preservation media, ten-fold diluted TSB, and ten-fold diluted TSB supplemented with 1% trehalose, and 1% sucrose. Optimal preservation conditions were strain-dependent and marine strains appeared to be more sensitive to freezing than non-marine strains. Nevertheless, a general cryopreservation protocol using 10% dimethyl sulfoxide with or without ten-fold diluted trypticase soy broth as a preservation medium allowed successful preservation of all tested strains.  相似文献

Explaining the variance of local communities in a spatial‐environmental matrix is one of the core interests of ecology today. Recent progress in metacommunity theory has made a substantial contribution to this field, however good empirical data in support of available theories are still relatively scarce. In this study we sampled a cluster of 36 temporary rock pools four times during one season to assess invertebrate metacommunity structure and dynamics and to search for steering processes and variables. Both Mantel tests and redundancy models indicate that local abiotic factors were dominant over spatial factors in explaining community structure and both were acting independently. Spatial variables were only important for passive dispersers and significantly explained 11% of variation in this community component. Pools connected by temporary overflows hosted more similar communities of passive dispersers than unconnected ones while community dissimilarity significantly increased with inter‐pool distance. A negative curvilinear relation was discovered between taxon richness and isolation in passive dispersers, providing some support for existing theoretical models of Mouquet and Loreau. Of different metacommunity perspectives, a combination of species sorting and mass effects best explains the observed patterns. Additionally, priority effects and monopolization may buffer against the homogenising effects of dispersal and contribute to the distinctness of isolated communities. This is one of the first studies to present evidence for spatial patterns in aquatic communities on such a small spatial scale (a rock ledge of ±9000 m2). Bridging the gap between theory and observed patterns in natural systems is one of the main challenges for future metacommunity research. Small aquatic habitats such as pitcher plants and freshwater rock pools may well have an important role to play as model systems to study ecological processes in a natural spatially explicit environment.  相似文献
