Thermal denaturation of four oligonucleotides, viz. 3'-d(AT)5pO(CH2)6Opd(AT)5-3'(par(AT], 3'-d(AT)5pO(CH2)6Opd(AT)5-5'(anti(AT],3'-d(A)10pO(CH2) 6Op(T)10-3'(par(A-T], and 3'-d(A)10pO(CH2)6Opd(T)10-5' (anti(A-T], was studied in 0.01 M phosphate buffer, pH 7, in the presence of 0.1, 0.25, 0.5 and 1.0 M NaCl. All the oligomers were found to exist at a lower temperature (0 to 20 degrees C) as complexes composed either of two oligomer molecules (a canonical duplex) or of more oligomer molecules whereas, at a higher temperature (30 to 70 degrees C), they formed hairpins with a parallel (par(AT) and par(A-T] or antiparallel (anti(AT) and anti(A-T) orientation of the chains. Melting curves (A260(T] were used to calculate thermodynamic parameters for the formation of hairpins and "low-temperature" duplexes. Experiments on ethidium bromide binding to the oligonucleotides have shown that the oligomer anti(A-T) exists, at a low ionic strength, as a four stranded complex ("quadruplex") contains two antiparallel helices, d(A).d(T), which have a parallel orientation and are bound to one another owing to the formation of additional hydrogen bonds between nucleic acid bases. Antibacterial activity of 14 drugs against clinical strains of asporogenic anaerobes causing wound infections in the soft tissues i. e. Bacteroides fragilis and Bacteroides melaninogenicus as well as anaerobic gram-positive++ cocci was assayed with the method of serial dilutions in agar. It was shown that among the investigated species B. fragilis had the most marked resistance since out of the 14 drugs only 8 were sufficiently active against it i.e. carbenicillin, levomycetin, lincomycin, dioxidine, metronidazole, thinidazole, nitrazole and erythromycin. The choice of drugs for treating infections caused by B. melaninogenicus and anaerobic grampositive cocci unlike those caused by B. fragilis offered no difficulty since practically++ all the investigated drugs were highly active against the causative agents. There was observed relationship between the frequency of asporogenic anaerobes and the wound genesis. The characteristic features of the species composition connected with localization of the suppurative foci were indicated. The detected specific antimicrobial profiles of the asporogenic anaerobes causing wound infections and the peculiarity of their participation in development of purulent infections of the soft tissues provided a differential approach to empirical antibacterial therapy prior to the pathogen bacteriological investigation and availability of the antibioticograms. 相似文献

We examined the uptake of five heavy metals (Cu, Fe, Ni, Zn, and Mn) in Ceratophyllum demersum L. (hornwort) and Potamogeton alpinus Balb. (pondweed) from Iset’ river, Ural region, Russia. This study was conducted in a territory that is highly urbanized where the surface waters are contaminated by a wide spectrum of pollutants. The environmental situation in this territory drastically deteriorated due to anthropogenic activity. The water quality in most of the water bodies in the Ural region is rather poor. In a comparative study ofC. demersum and P. alpinus, differential accumulation pattern was noted for heavy metals (HMs). Higher amounts of HMs accumulated in C. demersum compared to P. alpinus. Also it was shown that in leaves of C. demersum there were high amount of total phosphorus, nitrogen, organics acids and ash; high activity of guaiacol peroxidase; high content of non-enzymatic antioxidants viz., flavonoids, ascorbate, glutathione and proline; high amount of thiols (soluble and membrane bound) compared to P. alpinus. 相似文献

The effect of the cholesterol-depleting agent methyl-β-cyclodextrin (MβCD) on exocytotic, transporter-mediated, tonic release,
the ambient level and uptake of l-[14C]glutamate was assessed in rat brain synaptosomes using different methodological approaches of MβCD application. The addition
of 15 mM MβCD to synaptosomes (the acute treatment, AT) immediately resulted in the extraction of cholesterol and in a two
times increase in the extracellular l-[14C]glutamate level. When 15 mM MβCD was applied to synaptosomes for 35 min followed by washing of the acceptor (the long-term
pretreatment, LP), this level was only one-third higher than in the control. The opposite effects of MβCD on tonic l-[14C]glutamate release and glutamate transporter reversal were found in AT and LP. Tonic release was dramatically enlarged in
AT, but decreased after LP. Transporter-mediated release was increased several times in AT, but attenuated in LP. Depolarization-evoked
exocytotic release of l-[14C]glutamate was completely lost in AT, whereas after LP, it was decreased by half in comparison with the control. Na+-dependent l-[14C]glutamate uptake was decreased by ~60% in AT, whereas in LP, it was lowered by ~40% only. The presence of MβCD in the incubation
media during AT caused dramatic dissipation of the proton gradient of synaptic vesicles that was shown with the pH-sensitive
dye acridine orange, whereas after LP, no statistically significant changes were registered in synaptic vesicle acidification.
It was concluded that the diverse changes in glutamate transport in AT and LP were associated with the difference in the functional
