A deletion mutation in GDF9 in sisters with spontaneous DZ twins.   总被引:1,自引:0,他引:1  
A loss of function mutation in growth differentiation factor 9 (GDF9) in sheep causes increased ovulation rate and infertility in a dosage-sensitive manner. Spontaneous dizygotic (DZ) twinning in the human is under genetic control and women with a history of DZ twinning have an increased incidence of multiple follicle growth and multiple ovulation. We sequenced the GDF9 coding region in DNA samples from 20 women with DZ twins and identified a four-base pair deletion in GDF9 in two sisters with twins from one family. We screened a further 429 families and did not find the loss of function mutation in any other families. We genotyped eight single nucleotide polymorphisms across the GDF9 locus in 379 families with two sisters who have both given birth to spontaneous DZ twins (1527 individuals) and 226 triad families with mothers of twins and their parents (723 individuals). Using case control analysis and the transmission disequilibrium test we found no evidence for association between common variants in GDF9 and twinning in the families. We conclude that rare mutations in GDF9 may influence twinning, but twinning frequency is not associated with common variation in GDF9.  相似文献

The rat genome sequencing and mapping consortium found evidence for an association between the catenin‐δ2 gene (CTNND2) and anxious behaviour. We replicated these results in humans by carrying out a genetic association test in patients with panic disorder, social phobia, generalized anxiety disorder and/or agoraphobia (N = 1714) and controls (N = 4125). We further explored the association between CTNND2 and other psychiatric disorders based on publicly available genome‐wide association results. A gene‐based test showed that single nucleotide polymorphisms (SNPs) in CTNND2 have a significantly increased signal (P < 1e?5) and decreased P‐values. Single nucleotide polymorphism rs1012176 showed the strongest association with any anxiety disorder (odds ratio: 0.8128, SE = 0.063, P = 0.00099), but this effect was not significant after correction for multiple testing. In available genome‐wide association results from the Psychiatric Genomics Consortium we found that SNPs in CTNND2 collectively showed an increased signal for schizophrenia (P < 1e?5) and major depressive disorder (P < 1e?5), but not for bipolar disorder. These signals remained significant after correction for potential confounders. The association between CTNND2 and anxiety was not strong enough to be picked up in the current generation of human genome‐wide analyses, indicating the usefulness of and need for animal genetic studies to identify candidate genes for further study in human samples .  相似文献

Eusocial insects offer special opportunities for the comparativestudy of sperm traits because sperm competition is absent (inspecies with obligatory monandry) or constrained (in lineageswhere queens mate multiply but never remate later in life).We measured sperm length in 19 species of fungus-growing ants,representing 9 of the 12 recognized genera, and mapped theseonto the ant phylogeny. We show that average sperm length acrossspecies is highly variable and decreases with mature colonysize in basal genera with singly mated queens, suggesting thatsperm production or storage constraints affect the evolutionof sperm length. Sperm length does not decrease further in multiplymating leaf-cutting ants, despite substantial further increasesin colony size. In a combined analysis, sexual dimorphism explained63.1% of the variance in sperm length between species. As colonysize was not a significant predictor in this analysis, we concludethat sperm production trade-offs in males have been the majorselective force affecting sperm length across the fungus-growingants, rather than storage constraints in females. The relationshipbetween sperm length and sexual dimorphism remained robust inphylogenetically independent contrasts. Some of the remainingvariation was explained by the relative size of the sperm-storageorgan, but only in the multiply mating leaf-cutting ants, suggestingthat sperm-storage constraints become important for the evolutionof sperm length in this derived group. Mate number affectedsperm length to a minor extent, and only in interaction withother predictor variables, suggesting that sperm competitionhas not been a major selective force for sperm length evolutionin these ants.  相似文献

High-performance liquid chromatography (HPLC) and liquid chromatography-tandem mass spectrometry (LC-MS/MS) are generally accepted as the preferred techniques for detecting and quantitating analytes of interest in biological matrices on the basis of the rule that one chemical compound yields one LC-peak with reliable retention time (Rt.). However, in the current study, we have found that under the same LC-MS conditions, the Rt. and shape of LC-peaks of bile acids in urine samples from animals fed dissimilar diets differed significantly among each other. To verify this matrix effect, 17 authentic bile acid standards were dissolved in pure methanol or in methanol containing extracts of urine from pigs consuming either breast milk or infant formula and analyzed by LC-MS/MS. The matrix components in urine from piglets fed formula significantly reduced the LC-peak Rt. and areas of bile acids. This is the first characterization of this matrix effect on Rt. in the literature. Moreover, the matrix effect resulted in an unexpected LC behavior: one single compound yielded two LC-peaks, which broke the rule of one LC-peak for one compound. The three bile acid standards which exhibited this unconventional LC behavior were chenodeoxycholic acid, deoxycholic acid, and glycocholic acid. One possible explanation for this effect is that some matrix components may have loosely bonded to analytes, which changed the time analytes were retained on a chromatography column and interfered with the ionization of analytes in the MS ion source to alter the peak area. This study indicates that a comprehensive understanding of matrix effects is needed towards improving the use of HPLC and LC-MS/MS techniques for qualitative and quantitative analyses of analytes in pharmacokinetics, proteomics/metabolomics, drug development, and sports drug testing, especially when LC-MS/MS data are analyzed by automation software where identification of an analyte is based on its exact molecular weight and Rt.  相似文献

We have investigated transmitter release from small and large dense-core vesicles in nerve terminals isolated from guinea pig hippocampus. Small vesicles are found in clusters near the active zone, and large dense-core vesicles are located at ectopic sites. The abilities of Ca2+ channel activation and uniform elevation of Ca2+ concentration (with ionophores) to evoke secretion of representative amino acids, catecholamines, and neuropeptides were compared. For a given increase in Ca2+ concentration, ionophore was less effective than Ca2+ channel activation in releasing amino acids, but not in releasing cholecystokinin-8. Titration of the average Ca2+ concentration showed that the Ca2+ affinity for cholecystokinin-8 secretion was higher than that for amino acids. Catecholamine release showed intermediate behavior. It is concluded that neuropeptide release is triggered by small elevations in the Ca2+ concentration in the bulk cytoplasm, whereas secretion of amino acids requires higher elevations, as produced in the vicinity of Ca2+ channels.  相似文献

Summary In 1977, 1978, and 1979 nuptial flights of Lasius niger L., Lasius flavus F., Myrmica rubra L., and Myrmica scabrinodis Nyl. were observed on the island of Schiermonnikoog and in the area around Amsterdam. Weather conditions during these flights were determined using data from meteorological stations at Schiermonnikoog and Schiphol Airport. Significant differences were found concerning daytime, global radiation and relative humidity at the beginning of flights of Lasius niger, Lasius flavus, and Myrmica rubra; Myrmica scabrinodis had no defined preferences for these parameters. Wind velocity at 2 m of height was less than 1.7 m.s–1 during all flights before 20 August. After that date all species tended to fly at higher wind velocities as well.The calculated ranges for daytime, temperature, global radiation, relative humidity, and wind velocity appeared to be sufficient to characterize all nuptial flight occasions at Schiermonnikoog.Micrometeorological measurements in typical habitats of different ant species revealed that during flights the air temperature at 20 cm above ground and the soil temperature at 5 to 7 cm below ground were about equal in the habitat of the flying species, but unequal in the neighbouring habitats of coexisting ants.  相似文献

Queens of eusocial Hymenoptera are inseminated only during a brief period before they start to lay eggs. This has probably been kin-selected because repeated insemination of old queens would normally be against the inclusive fitness interest of their daughter workers. Army ants have been considered to be the only possible exception to this rule due to their idiosyncratic life-history. We studied two distantly related species of army ants, the African Dorylus (Anomma) molestus and the Neotropical Eciton burchellii and present data from microsatellite genotyping, behavioural observations and sperm counts.We also describe the copulation behaviour of African army ants for the first time. Our results strongly suggest that, contradictory to earlier contentions, army ant queens do not mate repeatedly throughout their life and thus do not constitute an exception among the eusocial Hymenoptera in this respect. Sperm counts for males and queens of both species show that army ant queens have to mate with several males to become fully inseminated. However, sperm limitation by queens is unlikely to have been the prime reason for the evolution of high queen-mating frequencies in this group. Birth weight is in large extent influenced by gestational age. In addition genetic and environmental factors determine intrauterine growth and birth weight. The contributions of these factors may be influenced by maternal smoking during pregnancy. We examined birth weight and maternal smoking in a sample of 2930 twin pairs from the Netherlands Twin Register using structural equation modelling. Gestational age accounted for 27-44% of the variance in birth weight. A lower variability of birth weight and a lower association of birth weight with gestational age was found in twins whose mothers smoked during pregnancy. The variance not associated with gestational age was independent of maternal smoking during pregnancy. A systematic smaller part of the variability in birth weight was associated with variability in gestational age in second born twins compared to first born twins. The heritability of interindividual differences in birth weight was modest (10% for twins with non-smoking mothers and 11% for twins with smoking mothers). Common environmental influences other than gestational age accounted for a slightly larger part of the variance not associated with gestational age (17-20%).  相似文献
