Avian eggs contain a variety of maternally-derived substances that can influence the development and performance of offspring. The levels of these egg compounds vary in relation to environmental and genetic factors, but little is known about whether there are correlative links between maternal substances in the egg underlying common and different pathways of maternal effects. In the present study, we investigated genetically determined variability and mutually adjusted deposition of sex hormones (testosterone-T, androstenedione-A4 and progesterone-P4), antibodies (IgY) and antimicrobial proteins (lysozyme) in eggs of Japanese quail (Coturnix japonica). We used different genetic lines that were independently selected for yolk T concentrations, duration of tonic immobility and social reinstatement behaviour, since both selections for behavioural traits (fearfulness and social motivation, respectively) produced considerable correlative responses in yolk androgen levels. A higher selection potential was found for increased rather than decreased yolk T concentrations, suggesting that there is a physiological minimum in egg T levels. Line differences in yolk IgY concentrations were manifested within each selection experiment, but no consistent inter-line pattern between yolk IgY and T was revealed. On the other hand, a consistent inverse inter-line pattern was recorded between yolk IgY and P4 in both selections for behavioural traits. In addition, selections for contrasting fearfulness and social motivation were associated with changes in albumen lysozyme concentrations and an inverse inter-line pattern between the deposition of yolk IgY and albumen lysozyme was found in lines selected for the level of social motivation. Thus, our results demonstrate genetically-driven changes in deposition of yolk T, P4, antibodies and albumen lysozyme in the egg. This genetic variability can partially explain mutually adjusted maternal deposition of sex hormones and immune-competent molecules but the inconsistent pattern of inter-line differences across all selections indicates that there are other underlying mechanisms, which require further studies.  相似文献

Filamentous marine cyanobacteria produce bioactive natural products with both potential therapeutic value and capacity to be harmful to human health. Genome sequencing has revealed that cyanobacteria have the capacity to produce many more secondary metabolites than have been characterized. The biosynthetic pathways that encode cyanobacterial natural products are mostly uncharacterized, and lack of cyanobacterial genetic tools has largely prevented their heterologous expression. Hence, a combination of cutting edge and traditional techniques has been required to elucidate their secondary metabolite biosynthetic pathways. Here, we review the discovery and refined biochemical understanding of the olefin synthase and fatty acid ACP reductase/aldehyde deformylating oxygenase pathways to hydrocarbons, and the curacin A, jamaicamide A, lyngbyabellin, columbamide, and a trans-acyltransferase macrolactone pathway encoding phormidolide. We integrate into this discussion the use of genomics, mass spectrometric networking, biochemical characterization, and isolation and structure elucidation techniques.  相似文献

The effect of high concentrations of arsenic has been investigated in Caenibacter arsenoxydans, a beta-proteobacterium isolated from an arsenic contaminated environment and able to oxidize arsenite to arsenate. As the genome of this bacterium has not yet been sequenced, the use of a specific proteomic approach based on nano-high performance liquid chromatography tandem mass spectrometry (nanoLC-MS/MS) studies and de novo sequencing to perform cross-species protein identifications was necessary. In addition, a random mutational analysis was performed. Twenty-two proteins and 16 genes were shown to be differentially accumulated and expressed, respectively, in cells grown in the presence of arsenite. Two genes involved in arsenite oxidation and one in arsenite efflux as well as two proteins responsible for arsenate reduction were identified. Moreover, numerous genes and proteins belonging to various functional classes including information and regulation pathways, intermediary metabolism, cell envelope and cellular processes were also up- or down-regulated, which demonstrates that bacterial response to arsenic is pleiotropic.  相似文献

Simulation models can be used to perform virtual profiling in order to analyse eco‐physiological processes controlling plant phenotype. To illustrate this, an eco‐physiological model has been used to compare and contrast the status of a virtual fruit system under two situations of carbon supply. The model simulates fruit growth, accumulation of sugar, citric acid and water, transpiration, respiration and ethylene emission, and was successfully tested on peach (Prunus persica L. Batsch) for two leaf‐to‐fruit ratios (6 and 18 leaves per fruit). The development stage and the variation in leaf number had large effects of the fruit model variables dealing with growth, metabolism and fruit quality. A sensitivity analysis showed that changing a single parameter value, which could correspond to a genotypic change induced by a mutation, either strongly affects most of the processes, or affects a specific process or none. Correlation analysis showed that, in a complex system such as fruit, the intensity of many physiological processes and quality traits co‐varies. It also showed unexpected co‐variations resulting from emergent properties of the system. This virtual profiling approach opens a new route to explore the impact of mutations, or naturally occurring genetic variations, under differing environmental conditions.  相似文献

In a previous paper we reported that beta-D-glucans isolated from Saccharomyces cerevisiae could adsorb zearalenone, reduce its bioavailability in the digestive tract, and protect animals against its adverse effects. We have now investigated, in vitro, the kinetics of the interaction between other mycotoxins and beta-D-glucans from several sources at three pH values found along the digestive tract (3.0, 6.0, and 8.0). Acid and neutral conditions gave the highest affinity rates for aflatoxins B1 > deoxynivalenol > ochratoxin A and involved both the (1 --> 3)-beta-D-glucans and the (1 --> 6)-beta-D-glucans. Alkaline conditions, owing to their destructuring action on glucans, were favorable only for the adsorption of patulin. Using molecular mechanics, we found that hydroxyl, ketone, and lactone groups are involved in the formation of both hydrogen bonds and van der Waals interactions between aflatoxins B1, deoxynivalenol and patulin, and beta-D-glucans. Differences in the binding capacity of the mycotoxins are due to their specific physical and chemical characteristics.  相似文献

In several mammalian species, prenatal exposure to odours can elicit later positive consummatory behaviour in response to substrates bearing that odorant. In birds, the sense of smell has been considerably underestimated, and very little is known about the effects of early sensory experience on the regulation of feeding behaviour. We tested the hypothesis that the feeding behaviour of the domestic chicken could be regulated by olfactory learning during the embryonic life. To that end, chicken embryos were exposed to an olfactory stimulus (blend of essential oil of orange and nature identical vanillin) from embryonic day (ED) 12 to ED20, and chicks were tested between 4 and 9 d of age. In short‐term choice tests, at day 4 and 5, chickens previously exposed to a low concentration (LC) of the olfactory stimulus spent a higher proportion of time eating a familiar or an unfamiliar food bearing the olfactory stimulus compared to non‐exposed control chickens. Conversely, chickens previously exposed to a high concentration (HC) of the olfactory stimulus were found to avoid all foods bearing the olfactory stimulus. On a 24‐ h time scale at day 7–8, LC and HC birds, but not controls, ingest significantly less familiar food containing the olfactory stimulus. This result indicated a long‐term effect of the early olfactory experience on feeding preferences. We demonstrated that chickens can utilize information from their pre‐hatch chemosensory environment to guide their later feeding behaviour. A pre‐hatch effect of the intensity of odour signals in the regulation of feeding behaviour is reported here for the first time.  相似文献

Iron toxicity occurs under flooded conditions such as those prevailing in lowland rice fields and is due to an excess of ferrous ions. Ferritin is a multimeric protein responsible for Fe sequestration and storage, playing a key role in Fe homeostasis. Our aim was to study the modalities of overall ferritin synthesis in different organs of young seedlings from the African rice species (Oryza glaberrima) in relation to the putative involvement of abscisic acid (ABA) and oxidative stress in signalling processes. Seedlings from a moderately resistant to iron toxicity cultivar were grown in hydroponic culture for 2 weeks and treated with 500 mg l?1 Fe2+ in the presence or in the absence of 200?µM ABA, 50?µM methylviologen or 50?µM fluridone. Iron treatment increased iron and malondialdehyde concentration in all organs as well as ABA in roots and laminae. Although ferritin protein was detected in controls plants, iron treatment strongly reinforced its accumulation in sheaths and laminae after 24 h and 72 h. Ferritin mRNA was induced as early as 24 h after the beginning of the Fe-treatment in sheaths and, to a higher extent, in laminae. In the absence of iron treatment, exogenous ABA increased ferritin mRNA in laminae only but did not lead to further ferritin accumulation. Unexpectedly, it decreased ferritin mRNA levels in the sheaths of iron-treated plants and may thus have a dual influence depending on the considered organ. The inhibitor of ABA synthesis fluridone reduced endogenous ABA but did not compromise ferritin gene expression or ferritin synthesis, whatever the iron dose. Methyviologen application induced obvious oxidative damages but reduced ferritin synthesis. It is suggested that the signalling pathway leading to ferritin synthesis in the semi-aquatic African rice species may involve other components than those reported for typical terrestrial plants.  相似文献
