Human peripheral lymphocytes in G(0) phase were irradiated with 1-5 Gy of gamma rays. The biochemical and morphological changes characteristic of apoptosis were examined for 72 h after irradiation. In parallel, changes in chromatin conformation were studied by the method of anomalous viscosity time dependence (AVTD) and by measurements of nuclear halo size. An immediate and dose-dependent relaxation of chromatin, which became saturated at doses above 2-3 Gy, was revealed by the AVTD method. The state of relaxed chromatin lasted up to 12-24 h after irradiation, a response considerably longer than the time attributable to repair of radiation-induced DNA breaks. Measurements of nuclear halo size also indicated the initial relaxation of chromatin in the irradiated cells and its subsequent condensation. This condensation of chromatin as revealed with AVTD correlated well with nuclear condensation, as measured with dual fluorescence staining, and with DNA fragmentation, as measured by conventional and pulsed-field gel electrophoresis (PFGE). Late apoptotic cells did not contribute significantly to the AVTD signal, showing that the chromatin of these cells was completely condensed and fragmented.  相似文献

Propagation and breeding of red clover ( Trifolium pratense L.) in Russia was initiated about 200 years ago but the origin of present-day cultivars is disputed. Some authors argue that most Russian cultivars were derived from western European ones, whereas others support a Russian origin of the cultivars from local wild populations. In the present study we assessed the genetic variation at 17 allozyme loci in seven Russian cultivars, bearing the names of localities of the Urals, two American ones that have been used in Russia for scientific experiments and seven wild populations of the Urals and Western Siberia. Variation at the 17 protein loci supports the western European origin of the cultivars and also indicates that gene flow between cultivars and wild populations was limited or has not acted sufficiently long to affect the genetic composition of the red clover wild populations of the Urals.  相似文献

The cDNAs encoding lactate dehydrogenase isozymes LDH-A (muscle) and LDH-B (heart) from alligator and turtle and LDH-A, LDH-B, and LDH-C (testis) from pigeon were cloned and sequenced. The evolutionary relationships among vertebrate LDH isozymes were analyzed. Contrary to the traditional belief that the turtle lineage branched off before the divergence between the lizard/alligator and bird lineages, the turtle lineage was found to be clustered with either the alligator lineage or the alligator-bird clade, while the lizard lineage was found to have branched off before the divergence between the alligator/turtle and bird lineages. The pigeon testicular LDH-C isozyme was evidently duplicated from LDH-B (heart), so it is not orthologous to the mammalian testicular LDH-C isozymes.   相似文献

Movement-deficient potato virus X (PVX) mutants tagged with the green fluorescent protein were used to investigate the role of the coat protein (CP) and triple gene block (TGB) proteins in virus movement. Mutants lacking either a functional CP or TGB were restricted to single epidermal cells. Microinjection of dextran probes into cells infected with the mutants showed that an increase in the plasmodesmal size exclusion limit was dependent on one or more of the TGB proteins and was independent of CP. Fluorescently labeled CP that was injected into epidermal cells was confined to the injected cells, showing that the CP lacks an intrinsic transport function. In additional experiments, transgenic plants expressing the PVX CP were used as rootstocks and grafted with nontransformed scions. Inoculation of the PVX CP mutants to the transgenic rootstocks resulted in cell-to-cell and systemic movement within the transgenic tissue. Translocation of the CP mutants into sink leaves of the nontransgenic scions was also observed, but infection was restricted to cells close to major veins. These results indicate that the PVX CP is transported through the phloem, unloads into the vascular tissue, and subsequently is transported between cells during the course of infection. Evidence is presented that PVX uses a novel strategy for cell-to-cell movement involving the transport of filamentous virions through plasmodesmata.  相似文献

The location of the 3a movement protein (MP) of cucumber mosaic virus (CMV) was studied by quantitative immunogold labeling of the wild-type 3a MP in leaves of Nicotiana clevelandii infected by CMV as well as by using a 3a-green fluorescent protein (GFP) fusion expressed from a potato virus X (PVX) vector. Whether expressed from CMV or PVX, the 3a MP targeted plasmodesmata and accumulated in the central cavity of the pore. Within minor veins, the most extensively labeled plasmodesmata were those connecting sieve elements and companion cells. In addition to targeting plasmodesmata, the 3a MP accumulated in the parietal layer of mature sieve elements. Confocal imaging of cells expressing the 3a-GFP fusion protein showed that the 3a MP assembled into elaborate fibrillar formations in the sieve element parietal layer. The ability of 3a-GFP, expressed from PVX rather than CMV, to enter sieve elements demonstrates that neither the CMV RNA nor the CMV coat protein is required for trafficking of the 3a MP into sieve elements. CMV virions were not detected in plasmodesmata from CMV-infected tissue, although large CMV aggregates were often found in the parietal layer of sieve elements and were usually surrounded by 3a MP. These data suggest that CMV traffics into minor vein sieve elements as a ribonucleoprotein complex that contains the viral RNA, coat protein, and 3a MP, with subsequent viral assembly occurring in the sieve element parietal layer.  相似文献

The connection between the efficiency of phenolic lipids and their hydrophobic property (solubility) and hydrophobic property of microorganisms’ cell structure is shown. The mixture of amphiphilic di(oxiphenil)-phenil-methanes, which act bacteriostatically under 15 mg/l, possesses maximal efficiency against Staphylococcus aureus. Against Mycobacterium smegmatis with hydrophobic cell wall, hydrophobic 2,4-dialkylocibenzol 70 mg/l was the most effective. Hexylresorcin (HR) stops the development of gram-positive bacteria in concentrations 20–50 mg/l, that of gram-negative bacteria in concentration 65 mg/l, that of M. smegmatis at 70 mg/l, and that of yeast and fungi at 300 mg/l. HR prevails bacteria spores germination in the concentration 25–100 mg/l. The dependence of antibacterial action of isomers and homologues of alkylresorcins on their structure-number, position, and length of alkyl substituents-is studied.  相似文献

The liquid phase of different units of an underground gas storage (UGS) in the period of gas injection was studied with respect to its hydrochemical composition and characterized microbiologically. The presence of viable aerobic and anaerobic bacteria was revealed in the UGS stratal and associated waters. An important source of microorganisms and biogenic elements in the ecosystem studied is water and various technogenic admixtures contained in trace amounts in the gas entering from the gas main in the period of gas injection into the storage. Owing to this fact, the bacterial functional diversity, number, and activity are maximal in the system of gas treatment and purification and considerably lower in the observation well zone. At the terminal stages, the anaerobic transformation of organic matter in the UGS aqueous media occurs via sulfate reduction and methanogenesis; exceptionally high rates of these processes (up to 4.9 × 105 ng S2? l?1 day?1 and 2.8 × 106 nl CH4 l?1 day?1, respectively) were recorded for above-ground technological equipment.  相似文献

Two cultivars of potato (Solanum tuberosum L.) were transformed with a barley antiporter gene HvNHX2 driven by the CaMV 35S promoter. The expressed transgene conferred a higher NaCl tolerance to one of the cultivars. Under salt stress, the more salt-tolerant transgenic plants had longer roots, higher dry weight, and suppressed cell expansion as compared to wild-type plants. The salt tolerance of the plants grown in vitro was not accompanied by elevated total sodium in any plant organs tested. Instead, higher potassium was found in roots of transgenic plants. Possible mechanisms of plant salt tolerance are discussed.  相似文献
