Aims: Salmonella spp. has the capability to form biofilm on various surfaces. Biofilm‐associated protein (bapA), a large surface protein has been shown to play a leading role in the development of biofilm in Salmonella. Objective of this study was to investigate the presence of bapA gene in different serotypes of Salmonella spp. and to characterize DNA fragment encoding bapA protein of Salmonella Enteritidis. Methods and Results: Sixty‐seven Salmonella strains belonging to 34 serovars isolated from diverse sources in India were screened for the presence of bapA gene employing a primer designed for the purpose. All the strains yielded a positive amplification indicating that the bapA gene is well conserved in Salmonella spp. The amplified gene fragment of bapA was cloned in Escherichia coli (DH5 α) cells by using pGEM‐T easy cloning vector. On partial sequence analysis, the product exhibited 667 base pairs, corresponding to 218 amino acids. Conclusions BapA gene was found to be highly conserved in Salmonella. Partial sequence analysis of this gene from a strain of Salm. Enteritidis revealed close association with serotypes of poultry origin and also with some other animal/zoonotic serotypes. Significance and Impact of the Study: BapA gene can be targeted for the genus‐specific detection of this organism from different sources. Antigenic index of bapA protein indicates its protective and diagnostic potentials.  相似文献

A class of organic polymers, known as conducting polymers (CPs), has become increasingly popular due to its unique electrical and optical properties. Material characteristics of CPs are similar to those of some metals and inorganic semiconductors, while retaining polymer properties such as flexibility, and ease of processing and synthesis, generally associated with conventional polymers. Owing to these characteristics, research efforts in CPs have gained significant traction to produce several types of CPs since its discovery four decades ago. CPs are often categorised into different types based on the type of electric charges (e.g., delocalized pi electrons, ions, or conductive nanomaterials) responsible for conduction. Several CPs are known to interact with biological samples while maintaining good biocompatibility and hence, they qualify as interesting candidates for use in a numerous biological and medical applications. In this paper, we focus on CP-based sensor elements and the state-of-art of CP-based sensing devices that have potential applications as tools in clinical diagnosis and surgical interventions. Representative applications of CP-based sensors (electrochemical biosensor, tactile sensing 'skins', and thermal sensors) are briefly discussed. Finally, some of the key issues related to CP-based sensors are highlighted.  相似文献

The development of devices for the precise and controlled delivery of therapeutics has grown rapidly over the last few decades. Drug delivery materials must provide a depot with delivery profiles that satisfy pharmacodynamic and pharmacokinetic requirements resulting in clinical benefit. Therapeutic efficacy can be limited due to short half-life and poor stability. Thus, to compensate for this, frequent administration and high doses are often required to achieve therapeutic effect, which in turn increases potential side effects and systemic toxicity. This can potentially be mitigated by using materials that can deliver drugs at controlled rates, and material design principles that allow this are continuously evolving. Affinity-based release strategies incorporate a myriad of reversible interactions into a gel network, which have affinities for the therapeutic of interest. Reversible binding to the gel network impacts the release profile of the drug. Such affinity-based interactions can be modulated to control the release profile to meet pharmacokinetic benchmarks. Much work has been done developing affinity-based control in the context of polymer-based materials. However, this strategy has not been widely implemented in peptide-based hydrogels. Herein, we present recent advances in the use of affinity-controlled peptide gel release systems and their associated mechanisms for applications in drug delivery.  相似文献

As the battle against the deadly Covid-19 pandemic is still continuing worldwide, several complications are being reported in patients who have recovered post-covid. One such lethal complication being reported in patients in India in recent times, who have tested positive for Covid-19 and are gradually recovering, is a fungal disease called Mucormycosis or the black fungus. With several hundreds of cases being reported all over the country, it has triggered an additional wave of panic among the general public. Post-Covid-19 patients who are more vulnerable to Mucormycosis are those with a history of poorly controlled diabetes mellitus and also those who are immuno-compromised and have been treated with steroids and other drugs for Covid-19. The aim of this short review is to briefly cover the epidemiology of mucormycosis, its possible pathophysiology in Post Covid scenario, the clinical presentation and its diagnosis and management.  相似文献

The backwater system of Kerala is well known. In the present investigation, we have tried to explore the marine fungal diversity of selected backwater and brackish water habitats of the North Malabar region of Kerala, India. A total of 30 marine fungi were isolated, which include 19 Ascomycetes, 1 Basidiomycete, and 10 Mitosporic fungi. Periconia prolifica emerged as the most dominant one in terms of percent frequency of occurrence and percent relative abundance.  相似文献

We have studied the effect of several structurally related mansonones on the cytotoxicity of plant and bacterial toxins in Vero and BER-40, a brefeldin A-resistant mutant of Vero cells. Mansonone-D (MD), a sesquiterpenoid ortho-naphthoquinone, inhibited the cytotoxicity of ricin, modeccin, Pseudomonas toxin, and diphtheria toxin in Vero cells to different extents. The inhibition of ricin cytotoxicity was dose dependent and reversed upon removal of the drug. Protection of ricin cytotoxicity was also observed in the presence of cycloheximide, indicating that de novo protein synthesis is not required for the protective effect. Although MD inhibited the degradation and excretion of ricin, the binding and internalization of ricin was not affected. In contrast, MD strongly reduced the specific binding of diphtheria toxin in Vero cells. Fluorescence microscopic studies show that MD treatment dramatically alters the morphology of the Golgi apparatus in Vero cells. The kinetic studies reveal that the protection of ricin cytotoxicity is the consequence of decreased toxin translocation to the cytosol in MD-treated cells. The reactive ortho-quinone moiety of MD is important for the protective effect as thespesone, a para-naphthoquinone with a heterocyclic ring structure identical to that of MD, did not inhibit the cytotoxicity of toxins. Thespone, a dehydromansonone-D, lacking two hydrogens from the heterocyclic dihydrofuran ring of MD, inhibited the cytotoxicity of ricin, but was albeit less potent than MD. Neither mansonone-E nor mansonone-H with reactive ortho-quinone moiety, but with a different heterocyclic structure, had any effect on the cytotoxicity of ricin indicating that the protective effect of MD is specifically related to the overall structure of the metabolite.     The effect of nitrogen fertilisation on growth, foliar nutrients and water relations of four families of radiata pine (Pinus radiata, D. Don) currently in the Australian breeding program was examined from age six to 11 years. At this stage, the stand was ready for commercial thinning. The annual rainfall at the site varied from 563 to 733 mm.Application of nitrogen fertiliser resulted in stem wood volume at age nine years of 178 m3 ha-1 in the controls, compared with 228 m3 ha-1 in plots treated with 600 kg N ha-1. Pre-dawn needle water potential () measured in three consecutive summers (when rainfall ranged from 53 to 106 mm) were consistently higher (less water stress) in nitrogen fertilised than in control trees. Similarly, the water stress integral (S) decreased consistently with increasing levels of nitrogen, although total water use in fertilised trees would have been substantially higher because fertiliser application increased the leaf area index. The relationship between S and basal area was strong and paralleled that of foliar nitrogen concentration and basal area growth. Therefore, nitrogen application increased growth rates of trees by improving the nutrient status of trees and lowering the water stress on trees in summer.Families showed markedly different responses of basal area growth to nitrogen, ranging from an increase of 9.4% over three years for the least responsive family to 99.0% for the most responsive. There was no nitrogen × family interaction on or S suggesting that the large genetic variation in the growth response to nitrogen is mediated by factors other than water relations. These results have implications for managing highly productive plantations grown in an environment where rainfall is low compared to potential evapotranspiration.  相似文献

    Prosimulium multidentatum (Twinn) has three metacentric pairs in its chromosome complement. All six arms are individually identifiably in polytene nuclei. XY1 males are heterozygous for a small basal non-conformity in section 59 of the non-pairing sex differential segments which extends from sextion 58 to section 62 of the IIL arm. XY2 males carry an additional large heterozygous inversion in the center of this same arm. Meiosis is chiasmate in both kinds of male. In XY2 individuals 14.2% of the pachytene nuclei show reverse loop pairing and 12.5% of the anaphase I cells form bridge-fragment configurations. A majority of these bridges persist into second division and 7.1% double sized spermatids are formed. No pachytene loops or anaphase bridges were found in XY1 males. It is concluded therefore that the bridges and fragments of XY2 males result from chiasma formation within the Y2 inversion.  相似文献

    Summary Absorption of Zn65 by plant roots from a layer of dry top soil (suction > 15 bar) was measured using a technique that obviated water stress in the plant.Roots growing through the dry layer absorbed significant amounts of Zn65 provided they absorbed water from lower depths. Mucilage accumulated around the roots in the dry soil and may have facilitated ion transfer in the immediate vicinity of the root. The ability of roots to grow in dry soil and to absorb Zn65 varied between plant species.  相似文献

    Background  Seizures were observed in a 16-year old male Guyanese squirrel monkey with a history of inappetence and weakness.
    Methods and results  Complete blood count, biochemical profile, and urinalysis indicated systemic disease. Nematode larvae were detected in the feces. Cerebrospinal fluid (CSF) analysis revealed leukocytes and gram-positive cocci. Staphylococcus aureus was isolated from the CSF. Histopathological evaluation revealed systemic lesions with inflammation and nematodes in the small and large intestine.
    Conclusion  This is the first report describing spontaneous staphylococcal CNS infection in a squirrel monkey.  相似文献
