Mice with uniparental disomies (uniparental duplications) for defined regions of certain chromosomes, or certain disomies, show a range of developmental abnormalities most of which affect growth. These defects can be attributed to incorrect dosages of maternal or paternal copies of imprinted genes lying within the regions involved. Combinations of certain partial disomies result in interactions between the imprinting effects that seemingly independently affect foetal and/or placental growth in different ways or modify neonatal and postnatal development. The findings are generally in accord with the 'conflict hypothesis' for the evolution of genomic imprinting but do not demonstrate common growth axes within which imprinted genes may interact. Instead, it would seem that any gene that favours embryonic/foetal development, at consequent cost to the mother, will have been subject to evolutionary selection for only paternal allele expression. Reciprocally, any gene that reduces embryonic/foetal growth to limit disadvantage to the mother will have been selected for only maternal allele expression. It is concluded that survival of the placenta is core to the evolution of imprinting.  相似文献

Uncoupler resistance in Escherichia coli: the role of cellular respiration   总被引:2,自引:0,他引:2  
Bioenergetic properties of a mutant strain of Escherichia coli K12 designated TUV, which is resistant to the protonophoric uncoupling agent 4,5,6,7-tetrachloro-2-trifluoromethylbenzimidiazole (TTFB) have been compared with those of its non-resistant parent, E. coli K12 Doc-S. Strain TUV grew and respired some 20-30% faster than strain Doc-S, and was cross-resistant to carbonylcyanide p-(trifluoromethoxy)phenylhydrazone and triphenyltin, but not to 2,4-dinitrophenol. Phosphorus nuclear magnetic resonance demonstrated the TTFB-mediated collapse of the transmembrane pH gradient at identical rates in starved cells of both strains, indicating that uncoupler access and function were unimpaired in the mutant under these conditions. Strain TUV displayed enhanced uncoupler resistance and maintained intracellular pH and ATP levels only when respiring. On the other hand, strain TUV also showed increased resistance to novobiocin, implying that its outer wall permeability had been lowered. We suggest that the active resistance of strain TUV results from the exclusion of uncoupler by the interaction of inner and outer membrane components in a manner modulated by the degree of cellular energization.  相似文献

In the mouse, gametes with gross chromosome duplications and deficiencies can complement each other to give viable zygotes (with some notable exceptions involving particular chromosomes). These complementation-type offspring can be recognised in intercrosses between translocation heterozygotes in which one parent is homozygous for a recessive genetic marker not carried by the other. This system has beeN used by Lyon and colleagues (1976) to study non-disjunction in heterozygotes for tobacco mouse and laboratory-derived Robertsonian translocations. Although non-disjunction is frequent in the former group, still higher frequencies are needed for a workable test system in which wild type mice are treated and mated to a tester stock generating many aneuploid gametes. Possible approaches include (1) use of semidominant markers, (2) marking both arms, (3) combining two or three independent Robertsonians in the tester stock, (4) use of compounds of Robertsonians wih monobrachial homology, since these give very high frequencies of non-disjunction, (5) generation of a compound of three Robertsonians with tribrachial homology, which should produce aneuploid gametes only. This last seems the most promising approach, if the compound proves fertile, and would be analogous to the isochromosome system of Drosophila.  相似文献

Growth of winter cereals at low, nonfreezing temperatures is essential for the establishment of a cold-hardy state and subsequent survival of the overwintering plant. In this paper, I describe the consequences of growth and development of Secale cereale L. cv. Puma at cold-hardening temperatures with respect to leaf morphology, anatomy, and biochemistry, and with particular emphasis on their relationship to photosynthetic acclimation. Low temperature-induced structural and functional alterations at the level of ribulose bisphosphate carboxylase-oxygenase and the chloroplast thylakoid membrane are described and related to overall photosynthetic efficiency and capacity for CO2 utilization. Growth and development at cold-hardening temperatures appear to result in changes in protein conformation and membrane organization, but not in basic composition. It is proposed that developmental temperature imparts a significant effect on the assembly of these multimeric, photosynthetic components, which leads to distinct structural and functional changes.  相似文献
