The apex of a 3-leaf pea plant was chilled in cold chambers maintained at 5–7°C. The lateral shoots 1 through 5 grew, and shoot 5 eventually dominated other lateral shoots. The apex when returned to the ambient temperature did not reimpose apical dominance. The growing lateral shoots competed with the stem apex. The apices of 2- and 3-leaf plants were chilled and P-32 distribution in these plants was studied in the entire plant, at various intervals of time. Phosphorus-32 accumulation followed the growth pattern of the plant. The lateral shoots accumulated P-32 activity and very little activity was accumulated by the apex. The dominating shoots 2 and 5 accumulated the maximum amount of activity in 2- and 3-leaf plants, respectively. Labeled-IAA moved basipetally through the stem when applied to the cut stump simulating the apex. By cold treatment the translocation of IAA was influenced more than its absorption. The plant seems to metabolize this compound in the later periods of application. The plant now becomes “insensitive” to auxin and the lateral shoots grow.  相似文献

Xanthomonas campestris pv. glycines , (Xcg), the causative agent of the bacterial pustule disease of soybean was isolated and characterized. On susceptible soybean the pathogenic isolates displayed characteristic chlorotic lesions around the site of infection within 48 h of inoculation. The pathogenic isolates were found to contain two cryptic plasmids. A smaller plasmid of 1.5 kb and a larger one of size about 25 kb. SDS-PAGE profile of the soluble proteins of the pathogenic isolatess, howed a different pattern compared to that of the non-pathogenic isolates.  相似文献

Employing a photoaffinity labeling procedure with 8-azido-S-adenosyl-l-[methyl-3H]methionine (8-N3-Ado[methyl-3H]Met), the binding sites for S-adenosyl-l-methionine (AdoMet) of three protein N-methyltransferases [AdoMet:myelin basic protein-arginine N-methyltransferase (EC2.1.1.23); AdoMet:histone-arginin N-methyltransferase (EC2.1.1.23); and AdoMet:cytochromec-lysine N-methyltransferase (EC2.1.1.59)] have been investigated. The incorporation of the photoaffinity label into the enzymes upon UV irradiation was highly specific. In order to define the AdoMet binding sites, the photolabeled enzymes were sequentially digested with trypsin, chymotrypsin, and endoproteinase Glu-C. After each proteolytic digestion, radiolabeled peptide from each enzyme was resolved on HPLC first by gradient elution and further purified by an isocratic elution. Retention times of the purified radiolabeled peptides from the three enzymes from the corresponding proteolysis were significantly different, indicating that their sizes and compositions were different. Amino acid composition analysis of these peptides confirmed further that the AdoMet binding sites of these protein N-methyltransferases are quite different.  相似文献

Membrane vesicles of Escherichia coli can be produced by 2 different methods: lysis of intact cells by passage through a French pressure cell or by osmotic rupturing of spheroplasts. The membrane of vesicles produced by the former method is everted relative to the orientation of the inner membrane in vivo. Using NADH, D-lactate, reduced phenazine methosulfate, or ATP these vesicles produce protonmotive forces, acid and positive inside, as determined using flow dialysis to measured the distribution of the weak base methylamine and the lipophilic anion thiocyanate. The vesicles accumulate calcium using the same energy sources, most likely by a calcium/proton antiport. Calcium accumulation, therefore, is presumably indicative of a proton gradient, acid inside. The latter type of vesicle, on the other hand, exhibits D-lactate-dependent proline transport but does not accumulate calcium with D-lactate as an energy source. NADH oxidation or ATP hydrolysis, however, will drive the transport of calcium but not proline in these vesicles. Oxidation of NADH or hydrolysis of ATP simultaneous with oxidation of D-lactate does not result in either calcium or proline transport. These results suggest that the vesicles are a patchwork or mosiac, in which certain enzyme complexes have an orientation opposite to that found in vivo, resulting in the formation of electrochemical proton gradients with an orientation opposite to that found in the intact cell. Other complexes retain their original orientation, making it possible to set up simultaneous proton fluxes in both directions, causing an apparent uncoupling of energy-linked processes. That the vesicles are capable of generating protonmotive forces of the opposite polarity was demonstrated by measurements of the distribution of acetate and methylamine (to measure the ΔpH) and thiocyanate (to measure the Δψ).  相似文献

We have earlier identified and purified two protein-lysine N-methyltransferases (Protein methylase III) fromEuglena gracilis [J. Biol. Chem.,260, 7114 (1985)]. The enzymes were highly specific toward histone H1 (lysine-rich), and the enzymatic products were identified as ε-N-mono-, di- and trimethyllysines. These earlier studies, however, were carried out with rat liver histone H1 as thein vitro substrate. Presently, histone H1 has been purified fromEuglena gracilis through Bio-Rex 70 and Bio-Gel P-100 column chromatography. TheEuglena histone H1 showed a single band on SDS-polyacrylamide gel electrophoresis and behaved like other histone H1 of higher animals, whereas it had a much higherR f value than the other histones H1 in acid/urea gel electrophoresis. When theEuglena histone H1 was [methyl-3H]-labeledin vitro by a homologous enzyme (one of the twoEuglena protein methylase III) and analyzed on two-dimensional gel electrophoresis, three distinctive subtypes of histone H1 were shown to be radiolabeled, whereas five subtypes of rat liver histone H1 were found to be labeled. Finally, by the combined use of a strong cation exchange and reversed-phase Resolve C18 columns on HPLC, we demonstrated thatEuglena histone H1 contains approximately 9 mol% of ε-N-methyllysines (1.40, 1.66, and 5.62 mol% for ε-N-mono-, di- and trimethyllysines, respectively). This is the first demonstration of the natural occurrence of ε-N-methyllysines in histone H1.  相似文献
