The impact of agriculture on wood ants of the Formica rufa group was investigated in a small-scale agricultural landscape with many woodland fragments in the east of the Netherlands. An inventory of nests was carried out in 1986, and repeated in 2014. The number of nests of F. rufa and F. polyctena had drastically decreased during the intervening 28 years, especially in small patches of woodland bordering a maize field. The species F. rufa, which mainly disperses by flight of the queen, seems to be better adapted to habitat fragmentation than F. polyctena, where dispersal is mainly by colony splitting. However, the flying strategy can only be successful in a landscape where most woodland fragments have a good quality habitat; in the study area, the habitat quality had deteriorated. This is ascribed to the development of a tall, nitrophilous vegetation, a result of considerable nitrogen deposition from the atmosphere and the spraying of manure. The regional survival of wood ants in the east of the Netherlands is at risk in the agricultural landscapes we studied. This is due to the increasing probability of colonies becoming extinct and the decreased probability of a queen colonizing habitat patches. However, the negative influence of agriculture on the edge vegetation of patches of woodland can be decreased by leaving a wide strip between the arable land and the woodland that receives no manure.  相似文献

Bixin is an apocarotenoid obtained from the seed aril of Bixa orellana L., a tropical plant known as achiote in Mexico. This compound is the second most commonly used natural colouring for food and pharmaceutical industries. B. orellana is an outcrossing species that displays high genetic variability. Recently, the colour traits of sexual organs were associated with the biosynthesis and accumulation of bixin in mature seeds. Herein, we describe a new approach for genotype–phenotype association by surveying lycopene beta-cyclase (Boβ-LCY1) gene variation in sixteen achiote accessions divided into three groups according to contrasting traits, such as flower colour, fruit colour and bixin production. Using a combination of single-strand conformational polymorphism techniques and the sequencing of polymorphic bands, we identified several single-nucleotide polymorphisms that divided the accessions into three haplotypes. Surprisingly, we observed that these three haplotypes were consistent with the same three groups previously characterized by phenotypic traits. We derived a putative sequence for the Boβ-LCY1 gene and surveyed the variations in this sequence. The heterozygosity of Boβ-LCY1 alleles resulted in a higher bixin content, likely associated with heterosis for this metabolite. These findings augment the toolbox available for the selection and genetic improvement of B. orellana and provide a reliable phenotype–genotype association method for commercial varietal selection, contributing to the development of laboratory techniques to identify desirable traits of commercial plant species.  相似文献

A mechanical grooming test was performed on large scale steel test panels coated with a fouling-release (FR) coating (International Intersleek 900), at four different frequencies, during the high fouling season in Port Canaveral, Florida. Grooming at frequencies of three or two times per week was effective at removing heavy biofilm growth and significantly reduced macrofouling settlement. Mechanical grooming at lower frequencies of weekly or bi-weekly removed heavy biofilm growth but was much less effective at reducing macrofouling settlement. The results indicated that frequent mechanical grooming could reduce the fouling rating of ships coated with FR coatings. The reduction in the fouling rating of ships’ hulls by frequent grooming could offer significant reductions in drag, fuel consumption, and the emission of exhaust gases. Frequent grooming could also eliminate the need for hull cleaning and increase the time between dry docking which would reduce the operational costs for many vessel operators.  相似文献

As an initial contribution to understanding the adaptive value of behavioral and life-history strategies, the life cycle and mating behavior of an unstudied species of tephritid fruit fly in the genus Rhagoletis are characterized for the first time. Over a 9-month fruiting period, a small proportion of Solanum appendiculatum Dunal (< 10 %) was found to be infested with a single larva of Rhagoletis solanophaga (Hernández & Frías). The average duration of R. solanophaga lifecycle (c.a. 140 days from egg laying to death of adults) exceeded the three month fruitless period. Additionally, R. solanophaga is capable of exploiting Solanaceous plants in at least two genera. These features could have selected for a non-diapausing species of Rhagoletis, a genus where most species are univoltine. Nevertheless, some individuals in the population became dormant. As other members of the genus, R. solanophaga exhibited a resource defense mating system with forced copulations and multiple mating. Both males and females could be highly promiscuous and individual mating success exhibited a wide range of outcomes. Regardless of mating success, mated females stored similar amounts of sperm in two spherical spermathecae. Long copulations were observed, perhaps functioning as a form of mate guarding with probable disadvantages for females. We outline hypotheses and opportunities for future comparative studies examining sperm competition and mate guarding.  相似文献

The current study describes the electrografting of 2,4-diamino-1,3,5-triazine (AT) groups at the surfaces of glassy carbon electrode (GCE) and indium tin oxide (ITO) through in situ diazotization of melamine. The presence of AT groups at the surface of the electrode was confirmed by X-ray photoelectron spectroscopy (XPS), scanning electron microscopy (SEM), electrochemical impedance spectroscopy (EIS), and cyclic voltammetry (CV). Furthermore, graphene oxide (GO) was self-assembled on AT grafted GCE. The oxygen functional groups present on the surface of GO were electrochemically reduced to form an electrochemically reduced graphene oxide (ERGO) on AT grafted electrode surface. Raman spectra show the characteristic D and G bands at 1340 and 1605 cm−1, respectively, which confirms the successful attachment of GO on AT grafted surface, and the ratio of D and G bands was increased after the electrochemical reduction of GO. EIS shows that the electron transfer reaction of [Fe(CN)6]3−/4− was higher at the ERGO modified electrode than at bare, AT grafted, and GO modified GCEs. The electrocatalytic activity of ERGO was investigated toward the oxidation of methylxanthines. It shows excellent electrocatalytic activity toward these methylxanthines by not only shifting their oxidation potentials toward less positive potentials but also enhancing their oxidation currents.  相似文献

An epigenetic profile defining the DNA methylation age (DNAm age) of an individual has been suggested to be a biomarker of aging, and thus possibly providing a tool for assessment of health and mortality. In this study, we estimated the DNAm age of 378 Danish twins, age 30–82 years, and furthermore included a 10‐year longitudinal study of the 86 oldest‐old twins (mean age of 86.1 at follow‐up), which subsequently were followed for mortality for 8 years. We found that the DNAm age is highly correlated with chronological age across all age groups (r = 0.97), but that the rate of change of DNAm age decreases with age. The results may in part be explained by selective mortality of those with a high DNAm age. This hypothesis was supported by a classical survival analysis showing a 35% (4–77%) increased mortality risk for each 5‐year increase in the DNAm age vs. chronological age. Furthermore, the intrapair twin analysis revealed a more‐than‐double mortality risk for the DNAm oldest twin compared to the co‐twin and a ‘dose–response pattern’ with the odds of dying first increasing 3.2 (1.05–10.1) times per 5‐year DNAm age difference within twin pairs, thus showing a stronger association of DNAm age with mortality in the oldest‐old when controlling for familial factors. In conclusion, our results support that DNAm age qualifies as a biomarker of aging.  相似文献

Dietary calorie restriction is a broadly acting intervention that extends the lifespan of various organisms from yeast to mammals. On another front, magnesium (Mg2+) is an essential biological metal critical to fundamental cellular processes and is commonly used as both a dietary supplement and treatment for some clinical conditions. If connections exist between calorie restriction and Mg2+ is unknown. Here, we show that Mg2+, acting alone or in response to dietary calorie restriction, allows eukaryotic cells to combat genome-destabilizing and lifespan-shortening accumulations of RNA–DNA hybrids, or R-loops. In an R-loop accumulation model of Pbp1-deficient Saccharomyces cerevisiae, magnesium ions guided by cell membrane Mg2+ transporters Alr1/2 act via Mg2+-sensitive R-loop suppressors Rnh1/201 and Pif1 to restore R-loop suppression, ribosomal DNA stability and cellular lifespan. Similarly, human cells deficient in ATXN2, the human ortholog of Pbp1, exhibit nuclear R-loop accumulations repressible by Mg2+ in a process that is dependent on the TRPM7 Mg2+ transporter and the RNaseH1 R-loop suppressor. Thus, we identify Mg2+ as a biochemical signal of beneficial calorie restriction, reveal an R-loop suppressing function for human ATXN2 and propose that practical magnesium supplementation regimens can be used to combat R-loop accumulation linked to the dysfunction of disease-linked human genes.  相似文献

Emerging data on cancer suggesting that target-based therapy is promising strategy in cancer treatment. PI3K-AKT pathway is extensively studied in many cancers; several inhibitors target this pathway in different levels. Recent finding on this pathway uncovered the therapeutic applications of PI3K-specific inhibitors; PI3K, AKT, and mTORC broad spectrum inhibitors. Noticeably, class I PI3K isoforms, p110γ and p110δ catalytic subunits have rational therapeutic application than other isoforms. Therefore, three classes of inhibitors: isoform-specific, dual-specific and broad spectrum were selected for molecular docking and dynamics. First, p110δ structure was modelled; active site was analyzed. Then, molecular docking of each class of inhibitors were studied; the docked complexes were further used in 1.2?ns molecular dynamics simulation to report the potency of each class of inhibitor. Remarkably, both the studies retained the similar kind of protein ligand interactions. GDC-0941, XL-147 (broad spectrum); TG100-115 (dual-specific); and AS-252424, PIK-294 (isoform-specific) were found to be potential inhibitors of p110γ and p110δ, respectively. In addition to that pharmacokinetic properties are within recommended ranges. Finally, molecular phylogeny revealed that p110γ and p110δ are evolutionarily divergent; they probably need separate strategies for drug development.  相似文献

We aimed to identify quantitative trait loci (QTL) for secondary traits related to grain yield (GY) in two BC1F2:3 backcross populations (LPSpop and DTPpop) under well-watered (4 environments; WW) and drought stressed (6; DS) conditions to facilitate breeding efforts towards drought tolerant maize. GY reached 5.6 and 5.8 t/ha under WW in the LPSpop and the DTPpop, respectively. Under DS, grain yield was reduced by 65% (LPSpop) to 59% (DTPpop) relative to WW. GY was strongly associated with the normalized vegetative index (NDVI; r ranging from 0.61 to 0.96) across environmental conditions and with an early flowering under drought stressed conditions (r ranging from -0.18 to -0.25) indicative of the importance of early vigor and drought escape for GY. Out of the 105 detected QTL, 53 were overdominant indicative of strong heterosis. For 14 out of 18 detected vigor QTL, as well as for eight flowering time QTL the trait increasing allele was derived from CML491. Collocations of early vigor QTL with QTL for stay green (bin 2.02, WW, LPSpop; 2.07, DS, DTPpop), the number of ears per plant (bins 2.02, 2.05, WW, LPSpop; 5.02, DS, LPSpop) and GY (bin 2.07, WW, DTPpop; 5.04, WW, LPSpop), reinforce the importance of the observed correlations. LOD scores for early vigor QTL in these bins ranged from 2.2 to 11.25 explaining 4.6 (additivity: +0.28) to 19.9% (additivity: +0.49) of the observed phenotypic variance. A strong flowering QTL was detected in bin 2.06 across populations and environmental conditions explaining 26–31.3% of the observed phenotypic variation (LOD: 13–17; additivity: 0.1–0.6d). Improving drought tolerance while at the same time maintaining yield potential could be achieved by combining alleles conferring early vigor from the recurrent parent with alleles advancing flowering from the donor. Additionally bin 8.06 (DTPpop) harbored a QTL for GY under WW (additivity: 0.27 t/ha) and DS (additivity: 0.58 t/ha). R2 ranged from 0 (DTPpop, WW) to 26.54% (LPSpop, DS) for NDVI, 18.6 (LPSpop, WW) to 42.45% (LPSpop, DS) for anthesis and from 0 (DTPpop, DS) to 24.83% (LPSpop, WW) for GY. Lines out-yielding the best check by 32.5% (DTPpop, WW) to 60% (DTPpop, DS) for all population-by-irrigation treatment combination (except LPSpop, WW) identified are immediately available for the use by breeders.  相似文献

Techniques to expand human hematopoietic stem cells ex-vivo could be beneficial to the fields of clinical hematopoietic stem cell transplantation and gene therapy targeted at hematopoietic stem cells. NUP98-HOXA10HD is a relatively newly discovered fusion gene that in mouse transplant experiments has been shown to increase numbers of hematopoietic stem cells. We evaluated whether this fusion gene could be used to expand engrafting human primitive CD34+ cells in an immunodeficient mouse model. Gene transfer was achieved using a lentiviral based vector. The engraftment of mobilized peripheral blood human CD34+ cells grown in culture for one week after gene transfer was evaluated 3–4 months after transplant and found to be 2–3 fold higher in the NUP98-HOXA10HD groups as compared to controls. These data suggest an expansive effect at least at the short term human repopulating cell level. Further evaluation in long term repopulating models and investment in a NUP98-HOXA10HD protein seems worthy of consideration. Additionally, the results here provide strong impetus to utilize NUP98-HOXA10HD as a tool to search for underlying genes and pathways involved in hematopoietic stem cell expansion that can be enhanced and have an even more potent expansive effect.  相似文献
