Introduction
Osteogenesis imperfecta (OI) is a clinically and genetically heterogeneous disease with skeletal fragility and variable extra-skeletal manifestations. To date several point mutations in 18 different genes causing different types of OI have been identified. Mutations in WNT1 compromise activity of the osteoblasts leading to disturbed bone mass accrual, fragility fractures and progressive skeletal abnormalities. The present study was conducted to determine the underlying genetic cause of an autosomal recessive skeletal dysplasia in a large consanguineous family from Chinute, Pakistan.Materials and methods
Blood was collected from 24 individuals of affected family along with clinical data. Homozygosity mapping was performed to confirm consanguinity. SNPs were identified, followed by whole exome and Sanger sequencing. In silico characterization of WNT1 mutation was performed using multiple platforms.Results
Nine affected family members exhibited severe bone deformities, recurrent fractures, short stature and low bone mineral density. SNP array data revealed homozygous segments >?1 Mb in length accounting for 2.1–12.7% of the genome in affected individuals and their siblings and a single 6,344,821 bp homozygous region in all affected individuals on chromosome 12q12-q13. This region includes two potential OI candidate genes WNT1 and VDR. We did whole-exome sequencing for both genes in two patients and identified a novel damaging missense mutation in exon 4 of WNT1: c.1168G?>?T (NM_005430) resulting in p.G324C. Sanger sequencing confirmed segregation of mutation with the disease in family.Conclusion
We report a novel mutation responsible for OI and our investigation expands the spectrum of disease-causing WNT1 mutations and the resulting OI phenotypes.72.

Background
Newcastle disease (ND) outbreaks are global challenges to the poultry industry. Effective management requires rapid identification and virulence prediction of the circulating Newcastle disease viruses (NDV), the causative agent of ND. However, these diagnostics are hindered by the genetic diversity and rapid evolution of NDVs.Methods
An amplicon sequencing (AmpSeq) workflow for virulence and genotype prediction of NDV samples using a third-generation, real-time DNA sequencing platform is described here. 1D MinION sequencing of barcoded NDV amplicons was performed using 33 egg-grown isolates, (15 NDV genotypes), and 15 clinical swab samples collected from field outbreaks. Assembly-based data analysis was performed in a customized, Galaxy-based AmpSeq workflow. MinION-based results were compared to previously published sequences and to sequences obtained using a previously published Illumina MiSeq workflow.Results
For all egg-grown isolates, NDV was detected and virulence and genotype were accurately predicted. For clinical samples, NDV was detected in ten of eleven NDV samples. Six of the clinical samples contained two mixed genotypes as determined by MiSeq, of which the MinION method detected both genotypes in four samples. Additionally, testing a dilution series of one NDV isolate resulted in NDV detection in a dilution as low as 101 50% egg infectious dose per milliliter. This was accomplished in as little as 7 min of sequencing time, with a 98.37% sequence identity compared to the expected consensus obtained by MiSeq.Conclusion
The depth of sequencing, fast sequencing capabilities, accuracy of the consensus sequences, and the low cost of multiplexing allowed for effective virulence prediction and genotype identification of NDVs currently circulating worldwide. The sensitivity of this protocol was preliminary tested using only one genotype. After more extensive evaluation of the sensitivity and specificity, this protocol will likely be applicable to the detection and characterization of NDV.74.

A study to determine the variation of ovipositioning behavior of stingless bees, Heterotrigona itama (Cockerell, 1918) was conducted on three colonies on June 2015. A digital single-lens reflex (DSLR) camera with a macro lens attached was used to record every movement of H. itama in its colonies for 20 min hour between 0800 h and 2000 h for seven days and seven month. The daily egg laying rate and time for laying eggs in colony-B and colony-C were significantly (P ? 0.05) higher than the colony-A. However, time to close the brood was not significantly different among colonies. The fastest egg oviposition time was 4 s by the colony-B and the slowest was 6 s by the colony-A. In addition there are no significant trends on brood produced per day, laying time of eggs, and the closing time of the brood after the oviposition process from June to December 2016. This result is useful for understanding the behavior of egg laying process by the queen bees and necessary to deal with problems of management and reproduction in the near future. 相似文献

Middle East Respiratory Syndrome Coronavirus (MERS-CoV) is a zoonotic pathogen capable of causing severe respiratory disease in humans. Although dromedary camels are considered as a major reservoir host, the MERS-CoV infection dynamics in camels are not fully understood. Through surveillance in Pakistan, nasal (n = 776) and serum (n = 1050)samples were collected from camels between November 2015 and February 2018. Samples were collected from animal markets, free-roaming herds and abattoirs. An in-house ELISA was developed to detect IgG against MERS-CoV. A total of 794 camels were found seropositive for MERS-CoV. Prevalence increased with the age and the highest seroprevalence was recorded in camels aged [ 10 years (81.37%) followed by those aged 3.1–10 years (78.65%) and B 3 years (58.19%).Higher prevalence was observed in female (78.13%) as compared to male (70.70%). Of the camel nasal swabs, 22 were found to be positive by RT-qPCR though with high Ct values. Moreover, 2,409 human serum samples were also collected from four provinces of Pakistan during 2016–2017. Among the sampled population, 840 humans were camel herders.Although we found a high rate of MERS-CoV antibody positive dromedaries (75.62%) in Pakistan, no neutralizing antibodies were detected in humans with and without contact to camels. 相似文献

Abstract The biology of the Australian phytoseiid mite Typhlodromips montdorensis is described from material collected in Queensland and South Australia in 1994−1996. At 25°C, when fed on cumbungi ( Typha sp.) pollen, the life cycle was completed in approximately 7 days, with an intrinsic rate of natural increase ( r m ) of 0.32. Female−male pairs produced a mean total of 52.7 eggs within 28 days of oviposition. Females that were deprived of males after first mating stopped laying eggs after 7−19 days; however, if another male was added, they resumed egg laying and produced, on average, a total of 49.4 eggs. The sex ratio was 2.24 females to one male. At 25°C, fecundity on a diet of thrips larvae (first-instar Frankliniella schultzei Trybom) was high, ranging from 2.72 to 3.58 eggs per day on the third day, depending on previous diet. Consumption rate of thrips was also high, with an average of 7.23−14.44 first-instar larvae eaten per day on the third day, depending again on previous diet and also on number of thrips larvae made available. The species was also observed to feed on: (i) broad mite, Polyphagotarsonemus latus (Banks); (ii) tomato russet mite, Aculops lycopersici (Massee); and (iii) two-spotted mite, Tetranychus urticae Koch. No diapause was observed under conditions of 25°C, 8 h light and 10°C, 16 h dark. Eggs were sensitive to low humidity, with 50% failing to hatch below 70.8% relative humidity. Summary Diurnal variations in water chemistry and plankton in this eutrophic tropical freshwater ponds were quite apparent. Large fluctuations were noted in dissolved oxygen, pH, carbonate and bicarbonate concentrations.
