Arginase is a manganese metalloenzyme that catalyzes the hydrolysis of L-arginine to form L-ornithine and urea. The structure and stability of the binuclear manganese cluster are critical for catalytic activity as it activates the catalytic nucleophile, metal-bridging hydroxide ion, and stabilizes the tetrahedral intermediate and its flanking states. Here, we report X-ray structures of a series of inhibitors bound to the active site of arginase, and each inhibitor exploits a different mode of coordination with the Mn(2+)(2) cluster. Specifically, we have studied the binding of fluoride ion (F(-); an uncompetitive inhibitor) and L-arginine, L-valine, dinor-N(omega)-hydroxy-L-arginine, descarboxy-nor-N(omega)-hydroxy-L-arginine, and dehydro-2(S)-amino-6-boronohexanoic acid. Some inhibitors, such as fluoride ion, dinor-N(omega)-hydroxy-L-arginine, and dehydro-2(S)-amino-6-boronohexanoic acid, cause the net addition of one ligand to the Mn(2+)(2) cluster. Other inhibitors, such as descarboxy-nor-N(omega)-hydroxy-L-arginine, simply displace the metal-bridging hydroxide ion of the native enzyme and do not cause any net change in the metal coordination polyhedra. The highest affinity inhibitors displace the metal-bridging hydroxide ion (and sometimes occupy a Mn(2+)(A) site found vacant in the native enzyme) and maintain a conserved array of hydrogen bonds with their alpha-amino and -carboxylate groups.  相似文献

Computer simulations are rapidly becoming a standard tool to study the structure and dynamics of lipids and membrane proteins. Increasing computer capacity allows unbiased simulations of lipid and membrane-active peptides. With the increasing number of high-resolution structures of membrane proteins, which also enables homology modelling of more structures, a wide range of membrane proteins can now be simulated over time spans that capture essential biological processes. Longer time scales are accessible by special computational methods. We review recent progress in simulations of membrane proteins.  相似文献

Cdc42p is a member of the RAS superfamily of GTPases and plays an essential role in polarized growth in many eukaryotic cells. We cloned the Candida albicans CaCDC42 by functional complementation in Saccharomyces cerevisiae and analyzed its function in C. albicans. A double deletion of CaCDC42 was made in a C. albicans strain containing CaCDC42 under the control of the PCK1 promoter. When expression of the heterologous copy of CaCDC42 was repressed in this strain, the cells ceased proliferation. These arrested cells were large, round, and unbudded and contained predominantly two nuclei. The PCK1-mediated overexpression of wild-type CaCdc42p had no effect on cells. However, in cells overexpressing CaCdc42p containing the dominant-negative D118A substitution, proliferation was blocked and the arrested cells were large, round, unbudded, and multinucleated, similar to the phenotype of the cdc42 double-deletion strain. Cells overexpressing CaCdc42p containing the hyperactive G12V substitution also ceased proliferation in yeast growth medium; in this case the arrested cells were multinucleated and multibudded. An intact CAAX box is essential for the phenotypes associated with either CaCdc42pG12V or CaCdc42pD118A ectopic expression, suggesting that membrane attachment is involved in CaCdc42p function. In addition, the lethality caused by ectopic expression of CaCdc42pG12V was suppressed by deletion of CST20 but not by deletion of CaCLA4. CaCdc42p function was also examined under hypha-inducing conditions. Cdc42p depletion prior to hyphal induction trapped cells in a round, unbudded state, while depletion triggered at the same time as hyphal induction permitted the initiation of germ tubes that failed to be extended. Ectopic expression of either the G12V or D118A substitution protein modified hyphal formation in a CAAX box-dependent manner. Thus, CaCdc42p function appears important for polarized growth of both the yeast and hyphal forms of C. albicans.  相似文献

Abstract Two oligonucleotide primers were used in a polymerase chain reaction-protocol to amplify a region (approx. 850 bp) of the 16S rRNA gene of Aeromonas schubertii and Aeromonas jandaei . Hybridization of the polymerase chain reaction products to specific internal probes provided a highly specific method for the identification of these two species.  相似文献

The interaction between physiological stress and arthropod herbivory was studied using two perennial species of Hypericum. Seedlings of H. perforatum, a herb introduced to Australia and weedy in places, and H. gramineum, an indigenous species, were subjected to water stress and/or herbivory by a mite, Aculus hyperici and an aphid, Aphis chloris. Both arthropods have recently been released in Australia for biological control of H. perforatum. Individually, stresses reduced measures of plant growth. Combinations of the three stresses decreased plant growth by slightly more than the product of their separate effects, suggesting that there is a weak positive interaction exacerbating the damage caused by each stress. Seedlings of the target weed and the non-target indigenous species were equally affected by the arthropods. The implications for weed biological control are discussed.  相似文献

The anterior alimentary tract of infective-stage Wuchereria bancrofti is divided into the following segments: stoma or buccal capsule, muscular esophagus, glandular esophagus, esophageal-intestinal valve, and intestine. Invaginated external cuticle lines only the anterior stoma. External cuticle and esophageal lining are not continuous and are ultrastructurally distinct; the latter is compared morphologically to the amorphous component of elastin. The glandular esophagus is a composite structure of a stellate contractile epithelial core, surrounded by a sleeve of secretory epithelium. The glandular cytoplasm shows evidence of formation and release of dense secretory granules. At least 2 nerve cell bodies lie within the esophagus approximately 15 micrometer anterior to the esophageal-intestinal valve and their associated processes pass forward and backward through the contractile epithelium. Materials interpreted as ingested flight muscle mitochondria of the mosquito vector appear in various stages of degeneration within the intestinal lumen. It is suggested that, although simple by comparison to some other nematodes, the anterior alimentary tract of infective-stage W. bancrofti functions in the ingestion and breakdown of nutrient materials. The ultrastructure of the excretory cell likewise suggests a functional capability.  相似文献

The enhancement of the longitudinal proton relaxation rate of solvent water protons which occurs when Mn(II) is bound to the "tight" metal ion site of unadenylylated glutamine synthetase (GS) was used to determine the binding constant of L-methionine (SR)-sulfoximine to GS-Mn(II) complexes. The binary enhancement for GS-Mn(II) is 22 at 24 MHz, 25 degrees C. The enhancement is lowered in the presence of the sulfoximine and the computed dissociation constant is 30 muM with epsilont, the enhancement for the ternary complex, equal to 3.0. Titration curves for the sulfoximine were also obtained in the presence of Mg-ADP, Mg-ADP plus Pi, and Mg-ATP. The dissociation constants were 9, 5, and 0.8 muM, respectively. The progressive tightening of the dissociation constants is symptomatic of conformational changes at the active site as the total subsite occupied by ATP is filled. The number of rapidly exchanging water molecules drops from 2 to approximately 0.1 when saturating concentrations of L-methionine (SR)-sulfoximine and nucleotide are present. The kinetically determined KI value of approximately 4 muM for the sulfoximine is about three orders of magnitude tighter than thee Km' value of approximately 3 mM for L-glutamate. The previously mentioned dissociation constants obtained by enhancement titrations are also orders of magnitude tighter than Km'. These data suggest that L-methionine (SR)-sulfoximine is a "transition-state" analogue for the glutamine synthetase reaction. ...  相似文献
