Background

E-communities, social groups interacting online, have recently become an object of interdisciplinary research. As with face-to-face meetings, Internet exchanges may not only include factual information but also emotional information – how participants feel about the subject discussed or other group members. Emotions in turn are known to be important in affecting interaction partners in offline communication in many ways. Could emotions in Internet exchanges affect others and systematically influence quantitative and qualitative aspects of the trajectory of e-communities? The development of automatic sentiment analysis has made large scale emotion detection and analysis possible using text messages collected from the web. However, it is not clear if emotions in e-communities primarily derive from individual group members'' personalities or if they result from intra-group interactions, and whether they influence group activities.

Methodology/Principal Findings

Here, for the first time, we show the collective character of affective phenomena on a large scale as observed in four million posts downloaded from Blogs, Digg and BBC forums. To test whether the emotions of a community member may influence the emotions of others, posts were grouped into clusters of messages with similar emotional valences. The frequency of long clusters was much higher than it would be if emotions occurred at random. Distributions for cluster lengths can be explained by preferential processes because conditional probabilities for consecutive messages grow as a power law with cluster length. For BBC forum threads, average discussion lengths were higher for larger values of absolute average emotional valence in the first ten comments and the average amount of emotion in messages fell during discussions.

Conclusions/Significance

Overall, our results prove that collective emotional states can be created and modulated via Internet communication and that emotional expressiveness is the fuel that sustains some e-communities.  相似文献

The increased spread of insect outbreaks is among the most severe impacts of climate warming predicted for northern boreal forest ecosystems. Compound disturbances by insect herbivores can cause sharp transitions between vegetation states with implications for ecosystem productivity and climate feedbacks. By analysing vegetation plots prior to and immediately after a severe and widespread outbreak by geometrid moths in the birch forest-tundra ecotone, we document a shift in forest understorey community composition in response to the moth outbreak. Prior to the moth outbreak, the plots divided into two oligotrophic and one eutrophic plant community. The moth outbreak caused a vegetation state shift in the two oligotrophic communities, but only minor changes in the eutrophic community. In the spatially most widespread communities, oligotrophic dwarf shrub birch forest, dominance by the allelopathic dwarf shrub Empetrum nigrum ssp. hermaphroditum, was effectively broken and replaced by a community dominated by the graminoid Avenella flexuosa, in a manner qualitatively similar to the effect of wild fires in E. nigrum communities in coniferous boreal forest further south. As dominance by E. nigrum is associated with retrogressive succession the observed vegetation state shift has widespread implications for ecosystem productivity on a regional scale. Our findings reveal that the impact of moth outbreaks on the northern boreal birch forest system is highly initial-state dependent, and that the widespread oligotrophic communities have a low resistance to such disturbances. This provides a case for the notion that climate impacts on arctic and northern boreal vegetation may take place most abruptly when conveyed by changed dynamics of irruptive herbivores.  相似文献

Binding of class I MHC molecules (MHCI) to an inhibitory receptor, PIR-B, expressed on B cells and myeloid cells provides constitutive cellular inhibition, thus ensuring peripheral tolerance. Recent unexpected findings pointed to a novel inhibitory role of PIR-B in neurite regeneration through binding to three axonal outgrowth inhibitors of myelin, including Nogo. Thus, it becomes interesting to determine whether the actions of the inhibitory myelin proteins and MHCI could coexist independently or be mutually exclusive as to the PIR-B-mediated immune and neural cell inhibition. Here, we present data supporting the competition of Nogo- and MHCI-mediated inhibition where they coexist. Kinetic analyses of Nogo and MHCI binding to the whole or a part of the recombinant PIR-B ectodomain revealed that PIR-B binds with higher affinity to Nogo than MHCI and that the MHCI binding only occurred with the N-terminal domains of PIR-B, whereas Nogo binding occurred with either the N- or C-terminal ectodomains. Importantly, kinetic tests indicated that the binding to PIR-B of Nogo and MHCI was competitive. Both endogenous and exogenous Nogo intensified the PIR-B-mediated suppression of interleukin-6 release from lipopolysaccharide-stimulated wild-type, but not PIR-B-deficient, cultured mast cells, indicating that PIR-B mediates Nogo-induced inhibition. Thus, we propose a novel mechanism by which PIR-B-mediated regulation is achieved differentially but competitively via MHCI and Nogo in cells of the immune system.  相似文献

Proteomic analysis of Inner Medullary Collecting Duct (IMCD3) cells adapted to increasing levels of tonicity (300, 600, and 900 mosmol/kg H(2)O) by two-dimensional difference gel electrophoresis and mass spectrometry revealed several proteins as yet unknown to be up-regulated in response to hypertonic stress. Of these proteins, one of the most robustly up-regulated (22-fold) was S100A4. The identity of the protein was verified by high pressure liquid chromatography-mass spectrometry. Western blot analysis confirmed increased expression with increased tonicity, both acute and chronic. S100A4 protein expression was further confirmed by immunocytochemical analysis. Cells grown in isotonic conditions showed complete absence of immunostaining, whereas chronically adapted IMCD3 cells had uniform cytoplasmic localization. The protein is also regulated in vivo as in mouse kidney tissues S100A4 expression was many -fold greater in the papilla as compared with the cortex and increased further in the papilla upon 36 h of thirsting. Increased expression of S100A4 was also observed in the medulla and papilla, but not the cortex of a human kidney. Data from Affymetrix gene chip analysis and quantitative PCR also revealed increased S100A4 message in IMCD3 cells adapted to hypertonicity. The initial expression of message increased at 8-10 h following exposure to acute sublethal hypertonic stress (550 mosmol/kg H(2)O). Protein and message half-life in IMCD3 cells were 85.5 and 6.8 h, respectively. Increasing medium tonicity with NaCl, sucrose, mannitol, and choline chloride stimulated S100A4 expression, whereas urea did not. Silencing of S100A4 expression using a stable siRNA vector (pSM2; Open Biosystems) resulted in a 48-h delay in adaptation of IMCD3 cells under sublethal osmotic stress, suggesting S100A4 is involved in the osmoadaptive response. In summary, we describe the heretofore unrecognized up-regulation of a small calcium-binding protein, both in vitro and in vivo, whose absence profoundly delays osmoadaptation and slows cellular growth under hypertonic conditions.  相似文献

BACKGROUND AND AIMS: White clover (Trifolium repens) is, due to nitrogen (N) fixation, important to the N dynamics of several northern temperate agroecosystems. This study aimed at monitoring growth and death of major white clover plant organs to assess their potential contribution to within-season N input and risk of off-season N losses. METHODS: White clover ('Snowy') was studied in a plot and root window experiment in southeast Norway (60 degrees 42'N, 10 degrees 51'E). Leaves, stolons and roots were tagged for lifespan measurement in harvested and unharvested stands during two experimental years. The availability of soil inorganic N was measured by plant root simulator (PRS) probes. KEY RESULTS: The longevity of leaves and petioles ranged from 21 to 86 d (mean = 59 d), of main stolon sections from 111 to over 677 d (mean = 411 d) and of roots from 27 to 621 d (mean = 290 d). About 60 % of the leaves produced had turned over by the end of the growing season and another 30 % had died or disappeared by the subsequent spring. Harvesting reduced the longevity of stolons and increased plant fragmentation, but did not decrease leaf or root lifespan or increase soil N availability. From the plant organ turnover data, it was estimated that the gross N input to the soil-plant system from white clover in pure stand during two growing seasons corresponded to a 2.5-fold increase over the total N in harvestable shoots. CONCLUSIONS: The short lifespan and poor over-wintering of leaves showed their potential importance as a nitrogen source in the soil-plant ecosystem but also their potential contribution to the risk of off-season N losses.  相似文献

Switchgrass (Panicum virgatum L.) is considered to be a highly promising bioenergy crop. However, little is known about insect pests that impact its utilization for this purpose. The switchgrass gall midge [Chilophaga virgati Gagné (Diptera: Cecidomyiidae)], which was first discovered in 2008 at Brookings, SD, USA, is shown to have a negative impact on biomass and seed yields of switchgrass. Our objectives were to increase knowledge of the biology of the midge by describing its life stages and any parasitoids that have biological control potential. Data collections were made during May to December in 2011 and April to late autumn in 2012. The gall midge adult is active from early June to late July. This insect overwinters as a late instar larva, usually in large aggregations, enclosed in the sheath of the flag leaf of dry tillers. The mean number of larvae was 31, with a range of 6 to 85 per tiller. Infested tillers m?2 varied among three phenologically distinct cultivars. The late flowering cultivar ‘Cave-In-Rock’ was more heavily infested (>2×) than the early flowering ‘Dacotah’. A newly discovered parasitoid, Platygaster chilophagae Buhl (Hymenoptera: Platygastridae) and a species of Quadrastichus sp. (Hymenoptera: Eulophidae) were reared from gall midge larvae. These results will be valuable to entomologists, switchgrass breeders, and agronomists as a guide to the occurrence and activities of the gall midge.  相似文献

Prairie cordgrass has been reported as a multi-polyploidy species having three cytotypes: tetra- (2n?=?4x?=?40), hexa- (2n?=?6x?=?60), and octoploid (2n?=?8x?=?80). A mixed-ploidy population comprising tetraploids and hexaploids was recently found at a single location in Illinois. However, adaptation and morphological differences between tetra- and hexaploids occurring in natural conditions as well as the contact zones of these cytotypes have yet to be determined. In this study, the cytotypes of 147 individuals of prairie cordgrass collected across the contact zone (4x?+?6x) were determined by flow cytometry using somatic G1 nuclei, and the results were confirmed by chromosome counts. Nineteen morphological characteristics were compared between the cytotypes. Tetra- and hexaploid plants have 2C genome sizes of 1.57 and 2.36?pg with chromosome counts of 40 and 60, respectively. This increase in polyploidy resulted in a greater variability of morphological expression in Illinois prairie cordgrass. Substantial differences in the flowering time, stomatal size, and plant morphological characteristics were observed between tetra- and hexaploids. The results indicate that the increasing of ploidy level in prairie cordgrass resulted in increased plant size in ploidy mixtures. The recent event of ploidy mixtures in prairie cordgrass natural populations offers unique opportunities for studying the formation and establishment of neopolyploidy.  相似文献

The synaptic vesicle protein 2A (SV2A), the brain-binding site of the anti-epileptic drug levetiracetam (LEV), has been characterized by Protein Tomography™. We identified two major conformations of SV2A in mouse brain tissue: first, a compact, funnel-structure with a pore-like opening towards the cytoplasm; second, a more open, V-shaped structure with a cleft-like opening towards the intravesicular space. The large differences between these conformations suggest a high degree of flexibility and support a valve-like mechanism consistent with the postulated transporter role of SV2A. These two conformations are represented both in samples treated with LEV, and in saline-treated samples, which indicates that LEV binding does not cause a large-scale conformational change of SV2A, or lock a specific conformational state of the protein. This study provides the first direct structural data on SV2A, and supports a transporter function suggested by sequence homology to MFS class of transporter proteins.  相似文献
