Wildlife species damaging crops can cause substantial losses to farmers and at the same time create negative attitudes against wildlife and conservation efforts that may result in negative interactions against wildlife and lead to human-wildlife conflicts (HWCs). For the analysis of negative interactions between humans and terrestrial wildlife species, a globally applicable scheme for monitoring was developed and applied over 6 years in study areas of two Asian (Nepal and India) and two African (Zambia and Tanzania) countries. Factors influencing crop consumption by eight different groups of herbivores were monitored and analyzed using generalized linear models. Seasonality, crop availability, type and the phenological stage of the crop seem to play an important role in the crop damaging behavior of herbivores. Crop consumers such as elephants (Loxodonta africana and Elephas maximus), zebra (Equus quagga spp.) and boars/hogs (Sus scrofa, Potamocherus larvatus and Phacochoerus africanus) show preferences for harvested and/or maturing crops. Rhinos (Rhinoceros unicornis) and antelopes/deer (Taurotragus oryx, Aepyceros melampus, Boselaphus tragocamelus and Axis axis) damage the highest numbers of fields with crops at an intermediate growth stage. The findings of this study can inform management of HWCs in areas where people and wildlife coexist. Furthermore, this study demonstrates the benefits of standardized HWC assessments in order to compare data from different continents and between different species to be able to draw generalized conclusions for the management of HWC.  相似文献

The feasibility of assessing microalgal fatty acid composition using near-infrared spectroscopy (NIRS) is described. The chlamydomonad microalga, Rhopalosolen saccatus (previously known as Characium saccatum), was isolated from the Fitzroy River, Central Queensland, Australia. R. saccatus was grown in batch culture with varying phosphorus nutrition and assessed for dry matter, total lipid and fatty acid composition using gas chromatography (GC). Transmission spectra (1100–2500 nm) were acquired of liquid culture, and reflectance spectra were acquired of wet and dry filtrates of cultures and of methyl esters. Partial least square (PLS) regression models were built on biomass, total lipid and a number of fatty acids. All sample presentation models supported PLS regression model with a cross validation correlation coefficient (R cv) >0.87 for biomass and R cv >0.68 for total lipid; however, the use of dry filtrates of culture is recommended as the sample presentation mode of choice. Models for fatty acids based on culture transmission spectra, reflectance spectra of wet and dry culture filtrates, or reflectance spectra of methyl esters in solvent were not acceptable. Dry extracts of methyl esters supported adequate models for fatty acids from C8:0 to C22:0, with the exception of capric and behenic acids, with an R cv of 0.89–0.94; however, in practice, samples processed to this stage can be easily analyzed by GC. Near-infrared spectroscopy can be a potential choice for rapid estimation of biomass (dry matter) and lipid content and composition in microalgae, with further work required to demonstrate oping robustness of the calibration model in prediction of unknown samples.  相似文献

Oreochromis mossambicus (Peters 1852) (Tilapia) is one of the most consumed fish globally. Tilapia thrives well in environments polluted by urban waste, which invariably contain antibiotic-resistant bacteria and antibiotic resistance genes (ARGs). Thus, Tilapia surviving in such polluted environments may serve as a potential source for dissemination of ARGs. To investigate this, we isolated bacterial strains from gut of Tilapia found in polluted rivers and lakes near Pune, India, and studied the prevalence of resistance genes by molecular methods. A total of 91 bacterial strains were obtained, which include fish pathogens and human pathogens such as Aeromonas hydrophila, Klebsiella pneumoniae, E. coli, Serratia marcescens, Enterobacter spp. and Shigella spp. Overall the prevalence of class 1 integrons, class 2 integrons, extended-spectrum beta-lactamases (ESBLs) bla CTX-M, bla SHV and aac(6')-Ib-cr gene was 38%, 24%, 38%, 31% and 31% respectively. Forty-two percent of the Enterobacteriaceae strains carried bla CTX-M gene, which is a common ESBL gene in clinics. The study demonstrates that tilapia found in the polluted waters can serve as reservoirs and an alternative route for human exposure to clinically important ARG-carrying bacteria. The consumption and handling of these fish may pose a potential health risk.  相似文献

Immunoglobulin G1 (IgG1) is the most abundant circulating human antibody and also the scaffold for many therapeutic monoclonal antibodies (mAbs). The destruction of IgG-coated targets by cell-mediated pathways begins with an interaction between the IgG Fc region and multiple varieties of membrane-bound Fc γ receptors (FcγRs) on the surface of leukocytes. This interaction requires the presence of an asparagine-linked (N-)glycan on the Fc, and variations in the N-glycan composition can affect the affinity of CD16A binding (an FcγR). Contemporary efforts to glycoengineer mAbs focus on increasing CD16A affinity, and thus treatment efficacy, but it is unclear how these changes affect affinity for the other FcγRs. Here, we measure binding of the extracellular Fc-binding domains for human CD16A and B, CD32A, B and C, and CD64 to 6 well-defined IgG1 Fc glycoforms that cover ~85% of the pool of human IgG1 Fc glycoforms. Core α1–6 fucosylation showed the greatest changes with CD16B (8.5-fold decrease), CD16A (3.9-fold decrease) and CD32B/C (1.8-fold decrease), but did not affect binding to CD32A. Adding galactose to the non-reducing termini of the complex-type, biantennary glycan increased affinity for all CD16s and 32s tested by 1.7-fold. Sialylation did not change the affinity of core-fucosylated Fc, but increased the affinity of afucosylated Fc slightly by an average of 1.16-fold for all CD16s and CD32s tested. The effects of fucose and galactose modification are additive, suggesting the contributions of these residues to Fc γ receptor affinity are independent.  相似文献

A recent genome-wide association study (GWAS) identified association with variants in X-linked CLDN2 and MORC4, and PRSS1-PRSS2 loci with chronic pancreatitis (CP) in North American patients of European ancestry. We selected 9 variants from the reported GWAS and replicated the association with CP in Indian patients by genotyping 1807 unrelated Indians of Indo-European ethnicity, including 519 patients with CP and 1288 controls. The etiology of CP was idiopathic in 83.62% and alcoholic in 16.38% of 519 patients. Our study confirmed a significant association of 2 variants in CLDN2 gene (rs4409525—OR 1.71, P = 1.38 x 10-09; rs12008279—OR 1.56, P = 1.53 x 10-04) and 2 variants in MORC4 gene (rs12688220—OR 1.72, P = 9.20 x 10-09; rs6622126—OR 1.75, P = 4.04x10-05) in Indian patients with CP. We also found significant association at PRSS1-PRSS2 locus (OR 0.60; P = 9.92 x 10-06) and SAMD12-TNFRSF11B (OR 0.49, 95% CI [0.31–0.78], P = 0.0027). A variant in the gene MORC4 (rs12688220) showed significant interaction with alcohol (OR for homozygous and heterozygous risk allele -14.62 and 1.51 respectively, P = 0.0068) suggesting gene-environment interaction. A combined analysis of the genes CLDN2 and MORC4 based on an effective risk allele score revealed a higher percentage of individuals homozygous for the risk allele in CP cases with 5.09 fold enhanced risk in individuals with 7 or more effective risk alleles compared with individuals with 3 or less risk alleles (P = 1.88 x 10-14). Genetic variants in CLDN2 and MORC4 genes were associated with CP in Indian patients.  相似文献

Herein, we have identified yeast Sec22p (ySec22p), a SNARE protein essential for endoplasmic reticulum to Golgi trafficking, as a suppressor of Bax-induced yeast apoptosis and corroborated published observations that ySec22p suppresses α-synuclein’s toxicity in yeast. It has been suggested that compounds which enhance expression, in neurons, of human homologues of ySec22p (Sec22Bp/Sec22p/Sec22A) would prevent synucleinopathies, such as Parkinson’s disease. With the aim of finding a small molecule that would mimic ySec22p, a library of natural products consisting of 394-compounds was screened using yeast cells that express either human α-synuclein or human Bax. The antioxidant aegeline, an alkaloid-amide occurring in the leaves of the plant Aegle marmelos Correa, was the only molecule that overcame apoptosis induced by both α-synuclein and Bax in yeast. Besides, aegeline also prevented growth block in cells expressing the more toxic A53T α-synuclein mutant. Restoration of cell growth occurred through inhibition of increased ROS levels, mitochondrial membrane potential loss and nuclear DNA fragmentation, characteristics of apoptosis manifested in α-synuclein or Bax-expressing cells. These results highlight the importance of yeast systems to identify rapidly molecules that may prevent the onset of apoptosis that occurs in Parkinson’s disease.  相似文献
