A gene encoding chitinase from B. subtilis has been isolated after optimization of PCR conditions. It was cloned with two different prometers, T7 promoter of the pJET1.2/blunt cloning vector and the SP6 promoter of pGEM®-T Easy vector. After transforming E. coli DH5α, two transformants were selected, CHI-NRC-4 from the first vector and T-CHI-NRC-6 from the second vector, and used for further studies. The complete CDS sequence of chitinase gene was determined and submitted to GenBank with the accession number KX268692.1. Culture supernatants of E. coli (CHI-NRC-4) and E. coli (T-CHI-NRC-6) were investigated for their inhibitory effect on M. javanica egg hatch under laboratory conditions. Result showed up to 96% inhibition in egg hatching due to both E. coli transformants as compared to control which reflect the same expression efficiency of both used prometers. A greenhouse experiment was carried out to evaluate the nematicidal effect of culture supernatants of the two transformts E. coli (CHI-NRC-4) and E. coli (T-CHI-NRC-6) against M. javanica infected eggplant. Obtained results showed a significant reduction in nematode population in soil and roots and enhancement in eggplant growth parameters as compared to control.  相似文献

It is important to understand switchgrass (Panicum virgatum L.) productivity with relation to diverse nutrient deficiency conditions in order to optimize continuous biomass production in marginal lands. This study was conducted on a wasteland sandy soil (Aridosol) to assess biomass yield, nutrient uptake and nitrogen (N) recovery of switchgrass, and soil nitrate-N (NO3?-N) accumulation responses to N (120 kg N ha?1), phosphorus (P, 100 kg P2O5 ha?1), and potassium (K, 45 kg K2O ha?1) applications during 2015 and 2016 in Inner Mongolia, China. The experiment layout was a randomized complete block design with fertilizer mixture treatments of N, P, and K (NPK), P and K (PK), N and K (NK), N and P (NP), and a control with no fertilizer input (CK). Plant height and stem diameter remained unaffected by the different fertilizer treatments. Biomass yield with the NPK treatment in 2015 was 8.9 Mg ha?1 and in 2016 it was 7.3 Mg ha?1. In 2015, compared with the NPK treatment, a significant yield reduction of 33.7% was found with PK, 22.5% with NK, 28.1% with NP, and 40.5% with CK; however, in 2016, yield declined significantly only with CK compared to the rest of the fertilizer treatments, for which yields were statistically similar. Plant N content was reduced for the treatment PK (i.e. N omission); conversely, plant P and K content remained unaffected with P and K omission treatments. Plant nutrient uptake, particularly of N and K, was severely decreased by the nutrient omission treatments when averaged across 2 years. Apparent N recovery (ANR; quantity of N uptake per unit of N applied) was reduced for the NP and NK treatments, which led to an increase in soil NO3?-N accumulation in the top 0–20 cm layer, compared with the NPK treatment. However, ANR was the highest (37.2% in 2015) with the NPK treatment, which also reduced soil NO3?-N accumulation. A balanced N, P, and K fertilizer management approach is suggested to sustain switchgrass yield and stand persistence on semiarid, marginal, sandy wasteland.  相似文献

Mangrove sediments were collected from major mangrove stands on the Red Sea Coast of Saudi Arabia. Forty five isolates belonging to 12 genera were purified and five isolates as well as their consortium were found to be able to grow in association with petroleum oil as sole carbon source under in vitro conditions. The isolated strains were identified based on internal transcribed spacer (ITS) rDNA sequence analysis. The fungal strains with the greatest potentiality to degrade diesel oil, without developing antagonistic activity, were identified as Alternaria alternata, Aspergillus terreus, Cladosporium sphaerospermum, Eupenicillium hirayamae and Paecilomyces variotii. As compared to the controls, these fungi accumulated significantly higher biomass, produced extracellular enzymes and liberated larger volumes of CO2. These observations with GC–MS data confirm that these isolates displayed rapid diesel oil bioremoval and when used together as a consortium, there was no antagonistic activity.  相似文献

Cardiotonic glycosides, like ouabain, inhibit Na+-K+-ATPase. Recent evidence suggests that low molar concentrations of ouabain alter cell growth. Studies were conducted to examine the effect of ouabain on Akt phosphorylation and rate of cell proliferation in opossum kidney (OK) proximal tubule cells. Cells exposed to 10 nM ouabain displayed increased Akt Ser473 phosphorylation, as evidenced by an increase in phospho-Akt Ser473 band density. Ouabain-stimulated Akt Ser473 phosphorylation was inhibited by pretreatment with phosphatidylinositol 3-kinase (PI3K) inhibitors (LY294002 and wortmannin), a PLC inhibitor (edelfosine), and an Akt inhibitor. Moreover, ouabain-mediated Akt Ser473 phosphorylation was suppressed by reduction of extracellular calcium (EGTA) or when intracellular calcium was buffered by BAPTA-AM. An inhibitor of calcium store release (TMB-8) and an inhibitor of calcium entry via store-operated calcium channels (SKF96365) also suppressed ouabain-mediated Akt Ser473 phosphorylation. In fura-2 AM-loaded cells, 10 nM ouabain increased capacitative calcium entry (CCE). Ouabain at 10 nM did not significantly alter baseline cytoplasmic calcium concentration in control cells. However, treatment with 10 nM ouabain caused a significantly higher ATP-mediated calcium store release. After 24 h, 10 nM ouabain increased the rate of cell proliferation. The Akt inhibitor, BAPTA-AM, SKF96365, and cyclopiazonic acid suppressed the increase in the rate of cell proliferation caused by 10 nM ouabain. Ouabain at 10 nM caused a detectable increase in 86Rb uptake but did not significantly alter Na+-K+-ATPase (ouabain-sensitive pNPPase) activity in crude membranes or cell sodium content. Taken together, the results point to a role for CCE and Akt phosphorylation, in response to low concentrations of ouabain, that increase the rate of cell proliferation without inhibiting Na+-K+-ATPase-mediated ion transport. Annona muricata has been used in folk medicine for the treatment of cancer and tumors. This study evaluated the chemopreventive properties of an ethyl acetate extract of A. muricata leaves (EEAML) on azoxymethane-induced colonic aberrant crypt foci (ACF) in rats. Moreover, the cytotoxic compound of EEAML (Annomuricin E) was isolated, and its apoptosis-inducing effect was investigated against HT-29 colon cancer cell line using a bioassay-guided approach. This experiment was performed on five groups of rats: negative control, cancer control, EEAML (250 mg/kg), EEAML (500 mg/kg) and positive control (5-fluorouracil). Methylene blue staining of colorectal specimens showed that application of EEAML at both doses significantly reduced the colonic ACF formation compared with the cancer control group. Immunohistochemistry analysis showed the down-regulation of PCNA and Bcl-2 proteins and the up-regulation of Bax protein after administration of EEAML compared with the cancer control group. In addition, an increase in the levels of enzymatic antioxidants and a decrease in the malondialdehyde level of the colon tissue homogenates were observed, suggesting the suppression of lipid peroxidation. Annomuricin E inhibited the growth of HT-29 cells with an IC50 value of 1.62 ± 0.24 μg/ml after 48 h. The cytotoxic effect of annomuricin E was further substantiated by G1 cell cycle arrest and early apoptosis induction in HT-29 cells. Annomuricin E triggered mitochondria-initiated events, including the dissipation of the mitochondrial membrane potential and the leakage of cytochrome c from the mitochondria. Prior to these events, annomuricin E activated caspase 3/7 and caspase 9. Upstream, annomuricin E induced a time-dependent upregulation of Bax and downregulation of Bcl-2 at the mRNA and protein levels. In conclusion, these findings substantiate the usage of A. muricata leaves in ethnomedicine against cancer and highlight annomuricin E as one of the contributing compounds in the anticancer activity of A. muricata leaves.  相似文献

Plant tolerance against a combination of abiotic stresses is a complex phenomenon, which involves various mechanisms. Physiological and biochemical analyses of salinity (NaCl) and nickel (Ni) tolerance in two contrasting tomato genotypes were performed in a hydroponics experiment. The tomato genotypes selected were proved to be tolerant (Naqeeb) and sensitive (Nadir) to both salinity and Ni stress in our previous experiment. The tomato genotypes were exposed to combinations of NaCl (0, 75 and 150 mM) and Ni (0, 15, and 20 mg l−1) for 28 days. The results revealed that the tolerant and sensitive tomato genotypes showed similar response to NaCl and Ni stress; however, the level of response was significantly different in both genotypes. The tolerant tomato genotype showed less reduction in growth than the sensitive genotype against both NaCl and Ni stress. Root and shoot ionic analysis showed a decrease in Na and increase in K concentration by increasing Ni levels in the growth medium. Moreover, accumulation of Na and Ni in tissues showed a decrease in membrane stability index and an increase in malondialdehyde contents. The activity of superoxide dismutase, catalase, peroxidase and glutathione reductase under NaCl and Ni stress was significantly higher in the tolerant compared to the sensitive genotype. Enhanced activity of many antioxidant enzymes in Naqeeb under stress conditions is among the other mechanisms that enabled the genotype to better detoxify reactive oxygen species and therefore Naqeeb tolerated the stresses better than Nadir.  相似文献

Dairy production is threatened by antibiotic resistant pathogens worldwide, and alternative solutions to treat mastitis are not available. The prevalence of antibiotic resistant strains is not well known in less developed countries. The prevalence of pathogenic bacteria and their resistance to 21 commercial antibiotics were studied in milk samples taken from 122 dairy cows suffering from the symptoms of mastitis in Egypt. The bacterial species were identified with molecular methods, and antibiotic resistance was studied with disc diffusion method. The prevalence of Streptococcus aureus, Escherichia coli and Pseudomonas aeruginosa were 30%, 17% and 3.5%, respectively. Most (90%) of the S. aureus strains showed resistance to penicillin whereas only 10% of the strains were resistant to oxacillin. Nearly half (40%) of E. coli strains showed resistance to streptomycin. Six P. aeruginosa strains showed resistance to several antibiotics, including ceftriaxone, enrofloxacin and levofloxacin. This points out that despite P. aeruginosa was not common, it should be followed up carefully. Potential biocontrol agents against antibiotic resistant mastitis bacteria were searched among 30 endophytic actinobacterial strains derived from wild medicinal plants. Three plants, namely Mentha longifolia, Malva parviflora and Pulicaria undulata were chosen for a more detailed study; their endophytic actinobacteria were used to prepare metabolic extracts. The crude metabolites of the actinobacteria were extracted with ethyl acetate. All metabolic extracts inhibited the growth of S. aureus, methicillin-resistant Staphylococcus aureus (MRSA), E. coli and P. aeruginosa in vitro. The 16S rRNA sequence analysis revealed that the most efficient actinobacterial strains were two Micromonospora sp. and one Actinobacteria bacterium. We conclude that the combination of the metabolites of several endophytic actinobacteria derived from several medicinal plants would be the most efficient against pathogens. Different metabolite cocktails should be studied further in order to develop novel biocontrol agents to treat antibiotic resistant mastitis bacteria in dairy cows.  相似文献
