In this study, micromorphological features of the leaf epidermal cells of 17 species of Iranian Cotoneaster were determined. The studied species were compared with each other and with phylogenetic relationships established by previous studies. Three populations of C. integerrimus and C. multiflorus were studied as representatives of C. subgen. Cotoneaster and Chaenopetalum, respectively. To evaluate quantitative and qualitative characters, multiple correspondence analysis (MCA) and factor analysis of mixed data (FAMD) were performed. Finally, relationships among the taxa were illustrated by hierarchical clustering analysis. Three stomatal types were identified, i.e. anomocytic, stephanocytic and actino‐stephanocytic. The wax and cuticle density was higher in species from semi‐moist habitats than in those from dry and semi‐dry habitats. The patterns formed by the anticlinal walls of the epidermal cells were sinuous, repand and straight‐curved. Interestingly, there was a significant relationship between the length of leaves, the density of the indumentum and the pattern formed by the anticlinal cell walls. The leaves of species with large leaves also have lower hair density and more curved anticlinal cell walls. It appears probable that climatic conditions affected the evolution of leaf micro‐morphological characters during the early diversification of the genus, but that these characters have subsequently been permanently fixed by phylogenetic constraints and thus appear diagnostic for extant species.  相似文献

Abstract

Biochar is considered a novel soil amendment for cadmium (Cd) stabilization in contaminated soils. A pot experiment was conducted to examine the efficiency of wheat straw and sugarcane bagasse induced biochar on Cd mobility in soil and its bioavailability to spinach in contaminated soil. Soil pH, Cd contents in plant tissues and microbial biomass were examined. Results showed that Cd was significantly decreased by 30.95% and 20.83% with wheat straw and sugarcane bagasse biochar at 2% application rate respectively, relative to the control. Similarly, Cd contents were decreased in plants shoots by 15.41 and 14.33%, while in roots by 48.3 and 35.54%, when wheat straw and sugarcane biochar were added at 2% application rate respectively. Moreover, soil microbial biomass was significantly increased with the application of all biochar types and their applications rates. Finally, wheat straw biochar at 2% application rate can be considered as an effective approach for Cd stabilization in contaminated soils.  相似文献

An electrochemical biosensor was developed for Hg2+ determination based on DNA hybridization. In the presence of Hg2+, the target and probe DNAs with thymine–thymine (T–T) mismatches could hybridize by forming T–Hg2+–T complex. This induced DNA hybridization led to the decrease in reduction peak currents of ethyl green (EG) as electroactive label, which could be used for determination of Hg2+. The difference in the value of the peak currents of EG before and after DNA hybridization (ΔI) was linear with the concentration of Hg2+ in the range of 9.0 × 10−11–1.0 × 10−9 M. The detection limit was 3.08 × 10−11 M.  相似文献

Rab3A is expressed predominantly in brain and synaptic vesicles. Rab3A is involved specifically in tethering and docking of synaptic vesicles prior to fusion which is a critical step in regulated release of neurotransmitters. The precise function of Rab3A is still not known. However, up-regulation of Rab3A has been reported in malignant neuroendocrine and breast cancer cells. In the present study, the structure of Rab3A protein was generated using MODELLER 9v8 software. The modeled protein structure was validated and subjected to molecular docking analyses. Docking with GTP was carried out on the binding site of Rab3A using GOLD software. The Rab3A-GTP complex has best GOLD fitness value of 77.73. Ligplot shows hydrogen bondings (S16, S17, V18, G19, K20, T21, S22, S31, T33, A35, S38, T39 and G65) and hydrophobic interacting residues (F25, F32, P34, F36, V37, D62 and A64) with the GTP ligands in the binding site of Rab3A protein. Here, the ligand molecules of NCI diversity set II from the ZINC database against the active site of the Rab3A protein were screened. For this purpose, the incremental construction algorithm of GLIDE and the genetic algorithm of GOLD were used. Docking results were analyzed for top ranking compounds using a consensus scoring function of X-Score to calculate the binding affinity and Ligplot was used to measure protein–ligand interactions. Five compounds which possess good inhibitory activity and may act as potential high affinity inhibitors against Rab3A active site were identified. The top ranking molecule (ZINC13152284) has a Glide score of ?6.65 kcal/mol, X-Score of ?3.02 kcal/mol and GOLD score of 64.54 with 03 hydrogen bonds and 09 hydrophobic contacts. This compound is thus a good starting point for further development of strong inhibitors.  相似文献

Background

Current study has been designed to evaluate the chemical composition of essential and fixed oils from stem and leaves of Perovskia abrotanoides and antioxidant and antimicrobial activities of these oils.

Results

GC-MS analysis of essential oil identified 19 compounds with (E)-9-dodecenal being the major component in stem and hexadecanoic acid in leaves. In contrast, GC-MS analysis of fixed oil showed 40 constituents with α-amyrin the major component in stem and α-copaene in leaves. The antioxidant activity showed the highest value of 76.7% in essential oil from leaves in comparison with fixed oil from stem (45.9%) through inhibition of peroxidation in linoleic acid system. The antimicrobial assay tested on different microorganisms (e.g. E. coli, S. aureus, B. cereus, Nitrospira, S. epidermis, A. niger, A. flavus and C. albicans) showed the higher inhibition zone at essential oil from leaves (15.2 mm on B. cereus) as compared to fixed oil from stem (8.34 mm on S. aureus) and leaves (11.2 mm on S. aureus).

Conclusions

The present study revealed the fact that essential oil analyzed from Perovskia abrotanoides stem and leaves could be a promising source of natural products with potential antioxidant and antimicrobial activities, as compared to fixed oil.  相似文献

Doxorubicin‐induced renal toxicity overshadows its anticancer effectiveness. This study is aimed at assessing the possible modulator effects of meloxicam, a cyclooxigenase‐2 inhibitor, on doxorubicin‐induced nephrotoxicity in mice and exploring some of the modulator mechanisms. Forty male mice were divided for treatment, for 2 weeks, with saline, meloxicam (daily), doxorubicin (twice/week), or both meloxicam and doxorubicin. Doxorubicin induced a significant increase in relative kidney weight to body weight, kidney lipid perooxidation, plasma levels of interleukin‐6 and tumor necrosis factor‐α, kidney caspase‐3 activity, and kidney prostaglandin E2 (PGE2) content. Doxorubicin disturbed kidney histology, abrogated renal function tests (serum creatinine, uric acid, and blood urea nitrogen), induced a significant decrease in antioxidant enzyme activities (superoxide dismutase, glutathione peroxidase, and catalase) and reduced glutathione (GSH) content. The administration of meloxicam with doxorubicin mitigated all doxorubicin‐disturbed parameters. Meloxicam ameliorated doxorubicin‐induced renal injury via inhibition of inflammatory PGE2, inflammatory cytokines, caspase‐3 activity, antioxidant effect, and free radical scavenging activity.  相似文献

Cytochrome c oxidase (COX) deficiency, one of the most common respiratory-chain defects in humans, has been associated with mutations in either mitochondrial DNA genes or nucleus-encoded proteins that are not part in but promote the biogenesis of COX. Mutations of nucleus-encoded structural subunits were sought for but never found in COX-defective patients, leading to the conjecture that they may be incompatible with extra-uterine survival. We report a disease-associated mutation in one such subunit, COX6B1. Nuclear-encoded COX genes should be reconsidered and included in the diagnostic mutational screening of human disorders related to COX deficiency.  相似文献

Pseudomonas aeruginosa is an opportunistic bacterium known for causing chronic infections in cystic fibrosis and chronic obstructive pulmonary disease (COPD) patients. Recently, several drug targets in Pseudomonas aeruginosa PAO1 have been reported using network biology approaches on the basis of essentiality and topology and further ranked on network measures viz. degree and centrality. Till date no drug/ligand molecule has been reported against this targets.In our work we have identified the ligand /drug molecules, through Orthologous gene mapping against Bacillus subtilis subsp. subtilis str. 168 and performed modelling and docking analysis. From the predicted drug targets in PA PAO1, we selected those drug targets which show statistically significant orthology with a model organism and whose orthologs are present in all the selected drug targets of PA PAO1.Modeling of their structure has been done using I-Tasser web server. Orthologous gene mapping has been performed using Cluster of Orthologs (COGs) and based on orthology; drugs available for Bacillus sp. have been docked with PA PAO1 protein drug targets using MoleGro virtual docker version 4.0.2.Orthologous gene for PA3168 gyrA is BS gyrAfound in Bacillus subtilis subsp. subtilis str. 168. The drugs cited for Bacillus sp. have been docked with PA genes and energy analyses have been made. Based on Orthologous gene mapping andin-silico studies, Nalidixic acid is reported as an effective drug against PA3168 gyrA for the treatment of CF and COPD.  相似文献

NAD biosynthesis is of substantial interest because of its important roles in regulating various biological processes. Nicotinamide mononucleotide adenylyltransferase 3 (Nmnat3) is considered a mitochondria-localized NAD synthesis enzyme involved in de novo and salvage pathways. Although the biochemical properties of Nmnat3 are well documented, its physiological function in vivo remains unclear. In this study, we demonstrated that Nmnat3 was localized in the cytoplasm of mature erythrocytes and critically regulated their NAD pool. Deficiency of Nmnat3 in mice caused splenomegaly and hemolytic anemia, which was associated with the findings that Nmnat3-deficient erythrocytes had markedly lower ATP levels and shortened lifespans. However, the NAD level in other tissues were not apparently affected by the deficiency of Nmnat3. LC-MS/MS-based metabolomics revealed that the glycolysis pathway in Nmnat3-deficient erythrocytes was blocked at a glyceraldehyde 3-phosphate dehydrogenase (GAPDH) step because of the shortage of the coenzyme NAD. Stable isotope tracer analysis further demonstrated that deficiency of Nmnat3 resulted in glycolysis stall and a shift to the pentose phosphate pathway. Our findings indicate the critical roles of Nmnat3 in maintenance of the NAD pool in mature erythrocytes and the physiological impacts at its absence in mice.  相似文献

To assess comparative losses of Trianthema portulacastrum (HP) relative to other weeds, the experiment was set during consecutive summer seasons 2018 and 2019 at the Research Farm MNS-University of Agriculture, Multan, Pakistan. Experiment consisted three replications which were laid out under randomized complete block design. Experiment consisted of ten treatments viz: weeds free (whole season), HP free till 20 Days after emergence (DAE), HP free till 40 DAE, HP free till 60 DAE, all weeds free 20 DAE, all weeds free 40 DAE, all weeds free 60 DAE, weedy check (all weeds), weedy check except HP and weedy check containing only HP. During 2018 in all weeds weedy check, maximum HP relative density (33.33%) was observed while in 2019, plot where weeds were controlled from growing till 20 DAE showed (80%) relative density at 30 DAE. HP maximum frequency (66.67%, 77.78%) and relative frequency (66%, 100%) was recorded at 45 DAE in plots where HP was kept controlled till 20 DAE and all weeds kept controlled till 20 DAE, respectively. Maximum number of grains per cob (738, 700.68), 1000 grain weight (306.66, 271.51 g) and grain yield (6150, 8015 kg hec-1) were recorded in plots which were kept all weed free till 60 DAE. As the competition period of weeds increased over 40 DAE, it substantially reduced yield of maize. Keeping the plots HP free till 40 DAE in the maize fields with HP as the major dominating weed, likely increase in maize grain yield is up to 30% compared to the fields where HP left un attended throughout the growing season. However, if maize field is infested with a mix of weeds with more than one dominating weeds including HP, compared to weedy situation the whole season, 30% higher grain yield can be obtained if all weeds are kept controlled till 40 DAE. Hence it can be concluded that whether the farmers face heavy HP infestation only or the mix of weeds as dominating weeds, in either case farmer should control weeds within first 40 days in maize field for better grain yield.  相似文献
