Thymidylate synthetase has been titrated in mouse liver extracts with [3H]fluorodeoxy-uridine monophosphate using centrifugally eluted gel filtration columns. The method is quick, efficient, and yields a linear dependence upon added extracted protein. The stability of the complex showed a pH maximum near 7.5. Specific binding of the tritiated nucleotide derivative to thymidylate synthetase was shown by denaturation of the complex in sodium dodecyl sulfate and gel electrophoresis of the denatured complex. Centrifugal elution of the sodium dodecyl sulfate-quenched complex from gel filtration columns was used to estimate the kinetics of complex formation at 0 and 12°C. 相似文献

DNA “fingerprinting” and the genetic management of a captive chimpanzee population (Pan troglodytes)
DNA fingerprinting probes are cloned sequences which simultaneously detect a large number of similar hypervariable loci in the target DNA. The resulting highly polymorphic pattern visualized on an autoradiograph allows resolution of questions concerning individual identification and parentage. M13 bacteriophage has been used as a DNA fingerprinting probe for paternity ascertainment among captive chimpanzees housed in multi-male groups as part of the National Chimpanzee Breeding and Research Program. In 31 cases of unknown paternity where DNA samples for mother, offspring, and all potential sires were available, DNA fingerprinting with M13 resulted in the unambiguous assignment of paternity for all 31 infants. Knowledge of pedigrees among the captive-born animals is used to address several issues important in the genetic management of captive breeding colonies, including estimation of effective population size and of the rate of decline in genetic variability, variance in male and female reproduction, and the effect of social dominance on male reproductive success. Our analysis demonstrates the beneficial effects of genetic management by comparing the managed dedicated cohort to the Bastrop colony as a whole. 相似文献

Tachykinin activation of muscarinic inhibition in canine small intestine is SPP in nature 总被引:1,自引:0,他引:1
Substance P when injected intraarterially into the small intestine of the anaesthetized dog during phasic activity produces three concentration dependent responses of the circular muscle. At lowest doses (approximately 10(-12) moles) inhibition occurs via release of acetylcholine to a muscarinic auto-receptor. At slightly higher doses (10(-10) moles) inhibition is preceeded by excitation via release of acetylcholine to muscarinic receptors on the smooth muscle. At still higher doses (10(-9) moles) substance P excites the smooth muscle directly. The present study demonstrates that other members of the tachykinin family also produce inhibition in vivo. The potency sequence was found to be physalaemin greater than or equal to substance P = neuromedin K greater than kassinin greater than alpha neurokinin = eledoisin. Such a sequence suggests that substance P is a natural stimulant of this pathway and that the receptor is SPP-like. The C-terminal fragment, substance P8-11, was a weak agonist at this receptor, while substance P1-9 was ineffective. 相似文献

A model that continuously predicts the concentration of microorganisms in complex medium fermentations is suggested. The model uses carbon dioxide evolution as its primary input and assumes that respiration activity can be differentiated into growth-related and maintenance-related functions. This model can be programmed on computer-coupled vessels and used to standardize on a physiological fermentation inoculum transfer time. The cell concentration estimate can also be used to calculate specific growth rate and can be combined with additional monitored information to calculate other important fermentation parameters such as specific oxygen uptake. 相似文献

Chimpanzees (Pan troglodytes) are endangered in the wild and may no longer be imported into the United States. Of those animals presently in captivity, candidates for breeding programs must be identified to insure a self-sustaining captive population. Some have suggested that poor reproductive performance might be linked to the performance of abnormal behaviors. In Study 1, three institutions housing breeding colonies of chimpanzees (86 males, 103 females) surveyed their animals for abnormal behaviors, copulatory performance, and, for females, maternal competence. In neither sex was there a positive association between absence of copulation and the presence of any of 18 forms of abnormal behavior. No one abnormal behavior was positively associated with inadequate maternal performance. Contrary to expectations, in both sexes (significantly for females), copulators exhibited more forms of abnormal behaviors than did noncopulators. In contrast, good mothers did show slightly fewer different forms of abnormal behaviors than did inadequate mothers. No specific combination of abnormal behaviors was associated with lack of copulatory performance in either sex or with inadequacy of maternal behavior. Significant sex differences occurred only in 2 of the 18 abnormal behaviors (coprophagy and self-clinging), both with females showing the higher prevalence. In Study 2, the rate of rocking in 5 male and 14 female chimpanzees at the Primate Foundation of Arizona was found to be relatively high among some reproductively competent subjects. Some rocking had forms and contexts indicative of aggressive displays. We conclude that chimpanzees should not be disregarded as potential breeders or as candidates for resocialization and breeding programs solely because they exhibit abnormal behaviors. Anyone involved in assessing well-being needs to be aware of individual differences among animals in the stimuli that may elicit rocking behavior. • Premise of study: A survey of the root-nodule symbiosis in Astragalus and its interaction with selenium (Se) has not been conducted before. Such studies can provide insight into how edaphic conditions modify symbiotic interactions and influence partner coevolution. In this paper plant-organ Se concentration ([Se]) was investigated to assess potential Se exposure to endophytes. • Methods: Selenium distribution and molecular speciation of root nodules from Se-hyperaccumulators Astragalus bisulcatus, A. praelongus, and A. racemosus was determined by Se K-edge x-ray absorption spectroscopy. A series of greenhouse experiments were conducted to characterize the response of root-nodule symbiosis in Se-hyperaccumulators and nonhyperaccumulators. • Key results: Nodules in three Se-hyperaccumulators (Astragalus crotalariae, A. praelongus, and A. preussii) are reported for the first time. Leaves, flowers, and fruits from Se-hyperaccumulators were routinely above the hyperaccumulator threshold (1,000 µg Se g−1 DW), but root samples rarely contained that amount, and nodules never exceeded 110 µg Se g−1 DW. Nodules from A. bisulcatus, A. praelongus, and A. racemosus had Se throughout, with a majority stored in C-Se-C form. Finally, an evaluation of nodulation in Se-hyperaccumulators and nonhyperaccumulators indicated that there was no nodulation inhibition because of plant Se tolerance. Rather, we found that in Se-hyperaccumulators higher levels of Se treatment (up to 100 µM Se) corresponded with higher nodule counts, indicating a potential role for dinitrogen fixation in Se-hyperaccumulation. The effect was not found in nonhyperaccumulators. • Conclusions: As the evolution of Se hyperaccumulation in Astragalus developed, root-nodule symbiosis may have played an integral role. 相似文献
