Aim

Accurately documenting and predicting declines and shifts in species’ distributions is fundamental for implementing effective conservation strategies and directing future research; species distribution models (SDM) have become a powerful tool for such work. Nevertheless, much of the data used to create these models are opportunistic and often violate some of their basic assumptions. We use amphibian declines and extinctions linked to the fungus Batrachochytrium dendrobatidis (Bd) to examine how sampling biases in data collection can affect what we know of this disease and its effect on amphibians in the wild.

Location

Queensland, Australia.

Methods

We developed a distribution model for Bd incorporating known locality records for Bd and a subset of climatic variables that should correctly characterize its distribution. We tested this (original) model with additional surveys, recorded new Bd observations in novel environments and reran the distribution model. We then investigated the difference between the original and new models, and used frog abundance and infection status data from two of these new localities to look at the susceptibility of the torrent frog Litoria nannotis to chytridiomycosis.

Results

While largely correct, the original SDM underestimated the distribution of Bd; sampling in ‘unsuitable’ drier environments discovered abundant populations of susceptible frogs with pathogen prevalences of up to 100%. The validation surveys further uncovered a new population of the frog Litoria lorica coexisting with the pathogen; this species was previously believed to be an extinct rain forest endemic.

Main conclusion

Our results indicate that SDMs constructed using opportunistically collected data can be biased if species are not at equilibrium with their environment or because environmental gradients have not been adequately sampled. For disease ecology, the better estimations of pathogen distribution may lead to the discovery of new populations persisting at the edge of their range, which has important implications for the conservation of species threatened by chytridiomycosis.

This study determined if a spatial hierarchy existed with respect to reach-scale habitat, subwatershed-scale, and watershed-scale geomorphology and land use stressors on fish assemblages in southeastern U.S. coastal plain streams. During May–October 2009–2012, fishes were sampled by seine, and habitat was assessed at 50 reaches in the Lake Pontchartrain Basin (USA). Using partial redundancy analysis (pRDA) a variance decomposition procedure was used to partial out influences of confounding covariables at each spatial scale. Reach-scale habitat had the strongest association with the assemblage. Stream width, depth, aquatic vegetation and human debris cover, rapid habitat assessment score, and large woody debris volume were the most important variables. At subwatershed and watershed scales, natural and anthropogenic characteristics were important, including elevation, gradient, watershed area, wetland cover, stream density, road, dam and oil/gas well densities. Six species were associated most strongly with the watershed variables, compared to reach- and subwatershed-scale variables. These species had more “r-selected” life-history strategies (e.g., smaller eggs, shorter life spans, multiple broods, longer spawning season, and trophic generalists). In contrast, most species that were associated strongest with reach-scale variables exhibited more “k-selected” life-history traits (e.g., larger eggs, longer life spans, shorter spawning season, single brood, and trophic specialists).  相似文献

Walking speeds were calculated for nine clones of the peach potato aphid Myzus persicae collected from three countries along a latitudinal cline of its European distribution from Sweden to Spain (Sweden, UK and Spain), and the effects of collection origin and intra and intergenerational acclimation were investigated. Walking speeds declined with decreasing temperature, with maximum performance at temperatures closest to acclimation temperature (fastest median walking speed of 5.8 cm min(-1) was recorded for clone UK 3, collected from the UK, at 25°C after acclimating to 25°C for one generation). Following acclimation at both 20°C and 25°C, walking ceased (as indicated by median walking speeds of 0.0 cm min(-1)) at temperatures as high as 7.5°C and 12.5°C. However, acclimation at 10°C enabled mobility to occur to temperatures as low as 0°C. There was no relationship between mobility and latitude of collection, suggesting that large scale mixing of aphids may occur across Europe. However, clonal variation was suggested, with clone UK 3 outperforming the majority of other clones across all temperatures at which mobility was maintained following acclimation at 10°C for one and three generations and at 25°C for one generation. The Scandinavian clones consistently outperformed their temperate and Mediterranean counterparts at the majority of temperatures following acclimation for three generations at 25°C.  相似文献

Many parasites and pathogens suppress host immunity to maintain infection or initiate disease. On the skin of many amphibians, defensive peptides are active against the fungus Batrachochytrium dendrobatidis (Bd), the causative agent of the emerging infectious disease chytridiomycosis. We tested the hypothesis that infection with the fungus may be linked to lower levels of defensive peptides. We sampled both ambient (or constitutive) skin peptides on the ventral surface immediately upon capture, and stored skin peptides induced from granular glands by norepinephrine administration of Australian green-eyed treefrogs, Litoria serrata. Upon capture, uninfected frogs expressed an array of antimicrobial peptides on their ventral surface, whereas infected frogs had reduced skin peptide expression. Expression of ambient skin peptides differed with infection status, and antimicrobial peptides maculatin 1.1 and 2.1 were on average three times lower on infected frogs. However, the repertoire of skin peptides stored in granular glands did not differ with infection status; on average equal quantities were recovered from infected and from uninfected frogs. Our results could have at least two causes: (1) frogs with reduced peptide expression are more likely to become infected; (2) Bd infection interferes with defence peptides by inhibiting release or causing selective degradation of peptides on the skin surface. Immune evasion therefore may contribute to the pathogenesis of chytridiomycosis and a mechanistic understanding of this fungal strategy may lead to improved methods of disease control.  相似文献

The cyclic peptide ANP 4-23 and the linear peptide analogue AP-811 have been shown to be selective ANP-CR antagonists. Via alanine scanning and truncation studies we sought to determine which residues in these molecules were important in their binding to the clearance receptor and the relationship between these two molecules. These studies show that several modifications to these compounds are possible which improve physical properties of these molecules while retaining high affinity for the ANP-CR.  相似文献

The second osmotic virial coefficient (B) is a measure of solution nonideality that is useful for predicting conditions favorable for protein crystallization and for inhibition of aggregation. Static light scattering is the technique most commonly used to determine B values, typically using protein concentrations less than 5 mg/mL. During static light scattering experiments at low protein concentrations, frequently the protein is assumed to exist either as a single nonassociating species or as a combination of assembly states independent of protein concentration. In the work described here, we examined the limit for ignoring weak reversible dimerization (Kd > or =1 mM) by comparing B values calculated with and without accounting for self-association. Light scattering effects for equilibrium dimer systems with Kd <20 mM and Kd <1 mM will significantly affect apparent B values measured for 20 and 150-kDa proteins, respectively. To interpret correctly light scattering data for monomer-dimer equilibrium systems, we use an expanded coefficient model to account for separate monomer-monomer (B(22)), monomer-dimer (B(23)), and dimer-dimer (B(33)) interactions.  相似文献
