The crustacean genus Artemia naturally inhabits various saline and hypersaline environments and is the most frequently laboratory-hatched animal for live feed in mari- and aquaculture. Because of its high economic importance, Artemia-bacteria interactions were so far studied mostly in laboratory strains. In this study, we focused our attention on the Artemia-associated microbiota in its natural environment in the solar salterns of Eilat, Israel. We applied a culture-independent method (clone libraries) to investigate the bacterial community structure associated with Artemia in five evaporation ponds with salinities from slightly above seawater (5%) to the point of saturation (32%), in two different developmental stages: in nauplii and in the intestine of adult animals. Bacteria found in naupliar and adult stages were classified within the Proteobacteria, Bacteroidetes, Firmicutes, Actinobacteria and Cyanobacteria. The halophilic proteobacterial genera Halomonas spp. and Salinivibrio spp. dominated the Artemia microbiota in both stages in all ponds. We also analysed a clone library of entire adult animals, revealing a novel bacterial phylogenetic lineage. Though our knowledge on microbial degradation of organic pollutants at high salt concentrations is still limited, the list of toxic compounds shown to be degraded or transformed in media of high salinity is growing. Compounds transformed aerobically include saturated and aromatic hydrocarbons (by certain archaeobacteria), certain aromatic compounds, organophosphorus compounds, and formaldehyde (by halotolerant eubacteria). Anaerobic microbial transformations of toxic compounds occurring at high salt concentrations include reduction of nitroaromatic compounds, and possibly transformation of chlorinated aromatic compounds. 相似文献

The drought-resistant cyanobacteria Phormidium autumnale, strain LPP4, and a Chroococcidiopsis sp. accumulated trehalose, sucrose, and both trehalose and sucrose, respectively, in response to matric water stress. Accumulated sugar concentrations reached values of up to 6.2 μg of trehalose per μg of chlorophyll in P. autumnale, 6.9 μg of sucrose per μg of chlorophyll in LPP4, and 4.1 μg of sucrose and 3.2 μg of trehalose per μg of chlorophyll in the Chroococcidiopsis sp. The same sugars were accumulated by these cyanobacteria in similar concentrations under osmotic water stress. Cyanobacteria that did not show drought resistance (Plectonema boryanum and Synechococcus strain PCC 7942) did not accumulate significant amounts of sugars when matric water stress was applied. 相似文献

The red palm weevil (Rhynchophorus ferrugineus; Olivier, 1790) (Coleoptera: Curculionidae) has recently become the most severe palm pest in the Mediterranean basin. Its dispersal was initially supported mainly by the acquisition of infested trees, but was further facilitated by the weevils’ flight. Therefore, knowledge of weevils’ flight capacity is a key element in evaluating their dispersal capability and setting preventive actions in advance. We tested the weevils’ flight ability in repeated flights that were 7–10 days apart by computer-monitored flight mill with a seesaw design. Tested flight parameters were: flight distance, duration, and velocity, number of flights, and cumulative flight distance, of virgin and mated weevils of both sexes. Our tests found no differences in flight distance between virgin and mated individuals or between sexes. Weevils showed flight capability between the ages of 2 and 97 days, and covered up to 315 km of cumulative distance during this time. In addition, we tested the effect of age of flight initiation and found that old starters perform fewer flights than young starters, and are thus assumed to possess inferior dispersal capabilities. 相似文献

Abstract: Microbial crusts are present on surfaces of soils throughout the world. A key feature of these crusts in arid zones is the abundance of filamentous sheath-forming and polysaccharide-excreting cyanobacteria. Several isolates of cyanobacteria were prepared from crust samples (Nizzana sand dunes, north-western Negev Desert, Israel). Optimal growth conditions for two such isolates of Microcoleus sp. were defined, and the role of the excreted polysaccharides in affecting the hydrological properties of crust-covered sand dunes was studied. After a period of more than ten years in which bacterial and algal community sizes were extremely small, a dense bloom of halophilic archaea developed in the upper 5–10 m of the Dead Sea water column in the summer of 1992. The development of this bloom followed a dilution of the upper water layer by winter rainfloods, which enabled the development of a short-lived dense bloom of the unicellular green alga Dunaliella parva. The dense archaeal community (up to 3.5 × 107 cells m1–1 in June 1992) imparted a red coloration to the Dead Sea, due to its high content of bacterioruberin. Bacteriorhodopsin was not detected. High levels of potential heterotrophic activity were associated with the bloom, as measured by the incorporation of labeled organic substrates. After the decline of the algal bloom, archaeal numbers in the lake decreased only little, and most of the community was still present at the end of 1993. The amount of carotenoid pigment per cell, however, decreased 2–3-fold between June 1992 and August 1993. No new algal and archaeal blooms developed after the winter floods of 1992–1993, in spite of the fact that salinity values in the surface layer were sufficiently low to support a new algal bloom. A remnant of the 1992 Dunaliella bloom maintained itself at the lower end of the pycnocline at depths between 7 and 13 m (September 1992–August 1993). Abstract: Acetate uptake and turnover rates were determined for the heterotrophic community in hypersaline environments (saltern crystallizer ponds, the Dead Sea) dominated by halpphilic Archaea. Acetate was formed from glycerol, which is potentially the major available carbon source for natural communities of halophilic Archaea. Values of [ K t + S n ] (the sum of the substrate affinity and the substrate concentration present in situ) for acetate measured in saltern crystallizer ponds were around 4.5–11.5 μM, while in the Dead Sea during a Dunaliella bloom values up to 12.8 μM were found. Maximal theoretical rates ( V max ) of acetate uptake in saltern crystallizer ponds were 12–56 nmol l−1 h−1 , with estimated turnover times for acetate ( T t ) between 127–730 h at 35°C. V max values measured in the Dead Sea were between 0.8 and 12.8 nmol l−1 h−1 , with turnover times in the range of 320–2190 h. V max values for acetate were much lower than those for glycerol. Comparisons with pure cultures of halophilic Archaea grown under different conditions showed that the natural communities were not adapted for preferential use of acetate. Both in natural brines and in pure cultures of halophilic Archaea, acetate incorporation rates rapidly decreased above the optimum pH value, probably since acetate enters the cell only in its unionized form. The low affinity for acetate, together with low potential utilization rates result in the long acetate turnover times, which explains the accumulation of acetate observed when low concentrations of glycerol are supplied as a nutrient to natural communities of halophilic Archaea. 相似文献

Framebuilders of Cenozoic coral reefs are limited by their photic requirements to the contemporaneous sea-level, and therefore shallow water reef facies are reliable paleo sea-level indicators. Sea-level lowstands leave no record on coral reefs in areas subject to tectonic uplift, such as the Huon Peninsula, New Guinea, but are recorded by coral reefs in areas subject to tectonic subsidence. A eustatic sea-level fall which exceeds the rate of subsidence subaeriallyexposes the upper section of the reef complex, creating a meteoric ground water system whose diagenetic imprint on the reef carbonates offers a good indicator of a sea-level stillstand. Cenozoic reef platforms thus may contain records of sea-level fluctuations, whether eustatic and global, or tectonic and local. Those reef platforms which developed on seamounts formed in mid-oceanic plate settings are particularly useful for the study of eustatic sea-level changes because their subsidence history is relatively simple, and the tectonic factor can be accounted for when estimating the eustatic sea-level component. Conventional petrographic and biostratigraphic methods used to delineate erosional unconformities in Cenozoic carbonate sections are often deficient. We demonstrate here that stable oxygen and carbon isotopes of the carbonates can reveal the location of both the exposure surface and the paleo water table with greater confidence on account of the specific imprint of meteoric diagenesis. In addition, the87Sr/86Sr isotope technique offers a promising dating tool of disconformities linked to sea-level lowstands with a resolution superior to the conventional biostratigraphic techniques. Although oxygen, carbon, and strontium isotopes monitor different aspects of global sea-level changes, when used in conjunction they provide deeper insights into the past than either one could achieve alone. Examples from previous and ongoing studies of Pacific mid-oceanic carbonate platforms illustrate the potential of the isotope techniques to unravel sea-level changes. At Midway Atoll, stable carbon and oxygen isotopes along with lithologic and biostratigraphic data suggest a sharp eustatic sealevel fall during the Early Miocene and a series of rapid, brief eustatic fluctuations during the Pliocene-Quaternary. The frequency and timing of the latter is supported by sea-level data from Enewetak Atoll obtained on the basis of detailed strontium isotopes and lithology. The Enewetak data also indicate a series of rapid, brief eustatic fluctuations around the Early-Middle Miocene boundary. At Niue, a carbonate platform about 500 km south of Samoa, oxygen, carbon, and strontium isotope records cover the critical interval of the Miocene-Pliocene boundary and show two distinct disconformities. The mid-oceanic carbonate platforms offer a testing ground of Vail-Haq type eustatic sea-level curves derived primarily from sections along passive continental margins and continental interiors. We show that Neogene sea-level data obtained from Midway, Enewetak, and Niue differ from Vail and Hardenbol's contemporaneous sea-level curve and support Haq et al.'s version. 相似文献

Deep wastewater reservoirs are used throughout Israel to store domestic wastewater effluents for summer irrigation. These effluents contain high concentrations of ammonia (≤5 mM) that are frequently toxic to photosynthetic microorganisms and that lead to development of anoxic conditions. Population dynamics of nitrifying bacteria and rates of nitrification were studied in two wastewater reservoirs that differed in organic load and degree of oxygenation and in the laboratory under controlled conditions, both by serial dilutions in mineral medium and microscopically with fluorescein isothiocyanate-conjugated antibodies prepared against local isolates. The difference in counts by the two methods was within 1 order of magnitude. In the laboratory, an O2 concentration of 0.2 mg liter−1 was close to optimal with respect to growth of NH3 oxidizers on domestic wastewater, while O2 concentrations of 0.05 mg liter−1 supported significant rates of nitrification. It was found that even hypertrophic anaerobic environments such as the anaerobic hypolimnion of the wastewater reservoir or the anaerobic settling ponds are capable of sustaining a viable, although not actively nitrifying, population of Nitrosomonas spp. and Nitrobacter spp., in contrast to their rapid decline when maintained anaerobically in mineral medium in the laboratory. Nitrification rates of NH3 in effluents during storage in the reservoirs were slower by 1 to 2 orders of magnitude compared with corresponding rates in water samples brought to the laboratory. The tolerance of halophilic archaebacteria towards bromide was tested in view of the fact that bromide occurs in natural brines in concentrations of up to 66 mM. It was found that, while concentrations of up to 0.8–1M are tolerated well by all halobacterial types examined, great differences exist between species with respect to bromide tolerance. WhileHalobacterium (H. salinarium, H. halobium, andH. sodomense) andNatronobacterium species are only moderately tolerant,Haloarcula (H. vallismortis, H. marismortui), andHaloferax species (H. mediterranei, H. gibbonsii) tolerate higher concentrations.Haloferax volcanii proved extremely tolerant and showed growth in bromide media at very low chloride concentrations (below 50 mM). No correlation was found between bromide tolerance and the bromide concentration in the habitat from which the strains were isolated. Iodide proved much more toxic than bromide.
