Hyalesthes obsoletus Signoret (Hemiptera: Auchenorrhyncha: Cixiidae) is a univoltine, polyphagous planthopper that completes its life cycle, including the subterranean nymph cryptic stage, on herbaceous weeds. In vineyards, it can transmit ‘Candidatus Phytoplasma solani’, an obligate parasitic bacterium associated with bois noir (BN) disease of grapevine, from its host plants to grapevine when occasionally feeding on the latter. The main disease management strategies are based on vector(s) control. Insecticide treatments on grapevine canopy are completely inefficient on H. obsoletus, due to its life cycle. Consequently, control of this planthopper focuses on the nymphs living on the roots of their host plants. Such practices, based on herbicide application and/or weed management, can reduce vector density in the vineyard but can impact the environment or may not be applicable, highlighting the necessity for alternative strategies. In this study, the efficacy of entomopathogenic nematodes (EPNs; Steinernema carpocapsae, S. feltiae, Heterorhabditis bacteriophora) and fungi (EPFs; Beauveria bassiana, Metarhizium anisopliae, Isaria fumosorosea, Lecanicillium muscarium) against H. obsoletus nymphs (EPNs) and adults (EPNs and EPFs) was assessed under laboratory and greenhouse conditions. The majority of examined EPNs and EPFs were able to kill H. obsoletus exhibiting a range of effectiveness. S. carpocapsae (among EPNs) and I. fumosorosea (among EPFs) were found to be the most effective biocontrol agents in all trials carried out. Advantages and limitations of such promising biocontrol agents were discussed. Ecological competency and conditions that can impede or enhance the EPNs and EPFs performance should be investigated to optimize their performance under field conditions.
Flow Cytometric (FCM) DNA content analysis of paraffin-embedded tissues has become a widely accepted procedure in assessing the biologic course in some tumors from archival material. Difficulty in interpreting histograms is frequently due to high levels of debris, and wide coefficients of variation (CV). These may lead to underestimating near-diploid abnormality. Although the clinical significance of low-degree aneuploidy has yet to be established, the procedure reported here improved our endeavor to detect DNA Indices (DI) of at least 1.1%. Experience has shown that careful technique can result in overall improvement of DNA histograms by lowering levels of debris and % CV, dramatically so in some cases. Archival histograms can be generated that rival in quality fresh tissue results. Archival material is currently employed for diagnostic and research purposes. 相似文献
AbstractThe development of new treatments for malignant melanoma, which has the worst prognosis among skin neoplasms, remains a challenge. The tumor microenvironment aids tumor cells to grow and resist to chemotherapeutic treatment. One way to mimic and study the tumor microenvironment is by using three-dimensional (3D) co-culture models (spheroids). In this study, a melanoma heterospheroid model composed of cancer cells, fibroblasts, and macrophages was produced by liquid-overlay technique using the agarose gel. The size, growth, viability, morphology, cancer stem-like cells population and inflammatory profile of tumor heterospheroids and monospheroids were analyzed to evaluate the influence of stromal cells on these parameters. Furthermore, dacarbazine cytotoxicity was evaluated using spheroids and two-dimensional (2D) melanoma model. After finishing the experiments, it was observed the M2 macrophages induced an anti-inflammatory microenvironment in heterospheroids; fibroblasts cells support the formation of the extracellular matrix, and a higher percentage of melanoma CD271 was observed in this model. Additionally, melanoma spheroids responded differently to the dacarbazine than the 2D melanoma culture as a result of their cellular heterogeneity and 3D structure. The 3D model was shown to be a fast and reliable tool for drug screening, which can mimic the in vivo tumor microenvironment regarding interactions and complexity.Graphic abstract相似文献
In livestock production, antibiotics are used to promote animal growth, control infections and thereby increase profitability. This practice has led to the emergence of multiresistant bacteria such as Salmonella, of which some serovars are disseminated in the environment. The objective of this study is to evaluate microcin J25 as an inhibitor of Salmonella enterica serovars of various origins including human, livestock and food. Among the 116 isolates tested, 37 (31.8%) were found resistant to at least one antibiotic, and 28 were multiresistant with 19 expressing the penta-resistant phenotype ACSSuT. Microcin J25 inhibited all isolates, with minimal inhibitory concentration values ranging from 0.06 μg/ml (28.4 nM) to 400 μg/ml (189 μM). Interestingly, no cross-resistance was found between microcin J25 and antibiotics. Multiple sequence alignments of genes encoding for the different proteins involved in the recognition and transport of microcin J25 showed that only ferric-hydroxamate uptake is an essential determinant for susceptibility of S. enterica to microcin J25. Examination of Salmonella strains exposed to microcin J25 by transmission electronic microscopy showed for the first-time involvement of a pore formation mechanism. Microcin J25 was a strong inhibitor of several multiresistant isolates of Salmonella and may have a great potential as an alternative to antibiotics. 相似文献
An extracellular amylase (AmyKS) produced by a newly isolated Bacillus subtilis strain US572 was purified and characterized. AmyKS showed maximal activity at pH 6 and 60°C with a half-life of 10 min at 70°C. It is a Ca2+ independent enzyme and able to hydrolyze soluble starch into oligosaccharides consisting mainly of maltose and maltotriose. When compared to the studied α-amylases, AmyKS presents a high affinity toward soluble starch with a Km value of 0.252 mg ml−1. Coupled with the size-exclusion chromatography data, MALDI–TOF/MS analysis indicated that the purified amylase is a dimer with a molecular mass of 136,938.18 Da. It is an unusual feature of a non-maltogenic α-amylase. A 3D model and a dimeric model of AmyKS were generated showing the presence of an additional domain suspected to be involved in the dimerization process. This dimer arrangement could explain the high substrate affinity and catalytic efficiency of this enzyme. 相似文献

Decomposition of the seagrass Halophila stipulacea (Forsk.) Aschers. followed in situ over 340 days showed that the plant lost 40.8% of its initial weight during the first 85 days and only 21.2% over the remaining 255 days of the incubation period. The observed overall rate of decay of 0.18% day?1 indicates the high resistance of Halophila tissue to decomposition.An exponential model showing a regularly decreasing decay coefficient was fitted to the data of weight loss. The correlation between observed and calculated weight remaining was highly significant (r = 0.9881, P < 0.0001).The samples of decomposing leaf litter harboured 2.2 × 1011 bacteria g?1 dry wt. on average and consumed 0.25?1.07 mg O2?1 dry wt. h?1. The early colonizing bacteria of the detritus were mostly non-sporogenous forms. The most prominent genera involved in decomposition were Actinomyces, Arthrobacter, Bacillus and Pseudomonas, all of which are capable of degrading structural carbohydrates. 相似文献
