Background aims
Although mesenchymal stromal cells (MSCs) have shown therapeutic potential in intestinal tissue repair, controversy concerning their short survival and poor biodistribution in recipient tissues still remains. Therefore, we investigated the paracrine role of MSC in three-dimensional culture of colon with experimental colitis.Methods
Colitis was induced in mice by oral administration of dextran sulfate sodium (DSS) for 7 days. Inflammatory responses were assessed on the basis of clinical signs, morphological, and histopathological parameters. On days 2 and 5, colonic explants were removed, and a three-dimensional culture was performed. The structural integrity of the intestinal mucosa was tested by treating the cultures with MSC or conditioned medium (CM) for 24 h, and then the colons were analyzed for histology/immunohistochemistry and interleukin (IL)-6 production.Results
Histological analysis demonstrated that both MSC and CM treatment reduced colon damage in organ culture. An increase in cell proliferation (Ki-67 staining) was observed after CM treatment. Additionally, MSC treatment was able to reduce CD3+ cells. The therapeutic effect of MSC and CM was mediated by the downregulation of IL-6.Discussion
The intestinal in vitro model has shown to be potentially useful for studying cellular interactions in a three-dimensional cell arrangement. Moreover, our results provide strong evidence that both MSC and CM treatments can alleviate colonic damage in organ culture. Importantly, these results suggest that MSC-secreted factors are able to protect the colon from inflammation caused by DSS-induced colitis independent of cell transplantation. 相似文献52.

We isolated one trinucleotide and seven tetranucleotide microsatellite loci for the Eurasian woodcock (Scolopax rusticola). We describe polymerase chain reaction conditions and primers for the successful amplification of these loci and report the results obtained from their use in 42 specimens from two populations in Europe. The number of alleles per locus ranged from three to 15, observed heterozygosity was comprised between 0.11 and 1.00 and expected heterozygosity ranged between 0.10 and 0.91. Cross‐specific amplification experiments highlighted the potential usefulness of these molecular markers for the study of three related scolopacid waders. 相似文献

Molecular (partial mitochondrial 12S ribosomal DNA sequences), morphological and meristic analysis of Lepadogaster lepadogaster lepadogaster , L. l. purpurea and L. zebrina were performed to investigate the relationships between these taxa. On the western shore of mainland Portugal, where the two subspecies of L. lepadogaster occur sympatrically, they differ in microhabitat preferences and their breeding seasons are largely out of phase. This information, combined with data on distribution patterns, led to the following conclusions: Lepadogaster l. purpurea is considered to be a valid species, L. purpurea ( Bonnaterre, 1788 ), different from L. l. lepadogaster , now designated L. lepadogaster ( Bonnaterre, 1788 ). L. zebrina was found to be a synonym of L. lepadogaster . The two newly defined species were found to be in sympatry at Madeira and the Canary islands, the Atlantic coast of the Iberian Peninsula, and the Mediterranean at least as far as Genoa (Italy). Abstract. 1. Fields such as ecology, macroecology, and conservation biology rely on accurate and comparable data. This is especially important for mostly unknown and megadiverse taxa such as spiders and regions such as the Mediterranean. Short-term sampling programmes are increasingly seen as the best option for sampling spiders. Comparability of results, however, demands standard procedures both in methodology and in sampling period. Cost-efficiency dictates that this period should be the most species rich.
2. Pitfall trapping was conducted in 23 sites from north to south Portugal, comprising three large-scale environmental zones and many different habitat types, during 10 months in each site. The annual richness pattern, differences in this pattern between areas and habitats, the complementarity between sampling periods and possible environmental correlates of richness were studied.
3. May and June present the optimal time for collecting spiders in Mediterranean areas. Northern areas have a later peak in richness and dense tree-cover sites offer more flexibility for sampling, with a higher proportion of species present at each period throughout the year.
4. Day length is the environmental factor most correlated with species richness. Maximum daily temperature may reduce richness, especially in southernmost areas, where summer temperatures can be extremely harsh.
5. It is recommended that short-term sampling programmes, intended to give a reasonable picture of spider communities in Portugal and in the Iberian Peninsula (and possibly extending to all the Mediterranean), should be conducted during May or June, with variable flexibility according to area and habitat. The proposed suggestions should appeal to everyone working in the field, given the cost-efficiency and comparability of results by adopting a common standardised approach. 相似文献
