A cytochemical study of intracellular thiol distribution inAcetabularia mediterranea intact cells was performed using thefluorescent thiol-labelling agent monobromobimane (mBBr). Differentdevelopmental stages were examined during the vegetative phaseand generative phase of the algal life cycle up to cyst maturation.Important changes in thiol localization have been found to coincidewith turning-points ofAcetabularia development. During the rapid growth phase, overall thiol content steadilyincreased along the stalk, being maximal shortly before capdifferentiation. At this stage, the thiol distribution patternparalleled that of cap morphogenesis essential processes: thiolsbeing accumulated at the apex where morphogenesis is going tobe expressed. High thiol contents were also present in the rhizoidal partof the alga throughout the vegetative phase. At the onset ofthe generative phase, important alterations in rhizoid thioldistribution coincided with the presumptive time of nucleardivision. Overall thiol content strongly decreased and thiolsbecame highly concentrated in definite zones localized in thecentral area of the rhizoid. Later on, during the sequence of morphogenetic events leadingto cyst differentiation, changes in thiol localization and relativecontent were observed in the cap rays. Positioning of secondarynuclei into the cap coincided with a high increase in thiolcontent in the entire cap. During the process of cyst formation,thiol content slightly decreased and thiols were localized incyst domains. Thiol distribution was also studied during regenerative processesafter merotomy. The microscopic anatomy of the extraocular eye muscles of the shark Scyliorhinus canicula is described. In contrast to swimming muscles, the fibres are not differentiated into distinct fibre type groups. The sarcoplasmic reticulum has narrow terminal cisternae in the Z band region, longitudinal tubules and a fenestrated cisterna in the H band region. Triads or dyads are mostly located at the Z discs. A number of variations from this general pattern is described, with main emphasis on triad location, orientation and structure. Since triads in vertebrates usually are located either at the Z disc or at the A-I junction, and with a fixed structure, these variations in shark muscles are of phylogenetic interest, as they may provide basis for an explanation of development to the two different triad locations.  相似文献

The accessibility of new genomic resources, high‐throughput molecular technologies and analytical approaches such as genome scans have made finding genes contributing to fitness variation in natural populations an increasingly feasible task. Once candidate genes are identified, we argue that it is necessary to take a mechanistic approach and work up through the levels of biological organization to fully understand the impacts of genetic variation at these candidate genes. We demonstrate how this approach provides testable hypotheses about the causal links among levels of biological organization, and assists in designing relevant experiments to test the effects of genetic variation on phenotype, whole‐organism performance capabilities and fitness. We review some of the research programs that have incorporated mechanistic approaches when examining naturally occurring genetic and phenotypic variation and use these examples to highlight the value of developing a comprehensive understanding of the relationship between genotype and fitness. We give suggestions to guide future research aimed at uncovering and understanding the genetic basis of adaptation and argue that further integration of mechanistic approaches will help molecular ecologists better understand the evolution of natural populations.  相似文献

Mimics can have both accurate mimicry and phenotypic variation if deception operates in multiple sensory modes. Australian Tongue orchids (Cryptostylis species) attract their sole pollinator, male Lissopimpla excelsa wasps (Ichneumonidae), by accurately mimicking the scent and colour of female L. excelsa wasps. To test for shape mimicry of female wasps, both traditional and geometric morphometric comparisons were performed with allopatric Cryptostylis ovata and the often sympatric Cryptostylis erecta, Cryptostylis leptochila, and Cryptostylis subulata. Although some floral parts accurately mimicked the female wasp, the overall floral shape differed dramatically among orchid species. The function (if any) of this interspecific shape variation is unknown, although it does not cause character displacement of pollen attachment locations to reduce interspecific pollen transfer. Analyses showed that floral parts involved in pollinia transfer were similarly shaped for three of the four Cryptostylis species and all attach their pollinia to the same location on the pollinator's abdomen. Shape may interact with pollinator behaviour: in the field, pollination rates doubled when two Cryptostylis species were present, regardless of orchid abundances. Perhaps variation in shape hinders pollinator recognition and the avoidance of orchids, similar to scent and colour variation in other sexually deceptive orchid systems. © 2012 The Linnean Society of London, Biological Journal of the Linnean Society, 2012, 106 , 469–481.  相似文献

During studies on the purification of cucumber mosaic virus (strain W) it was found that preparations were most infective and stable when made from tobacco leaves (10–12 days after inoculation) homogenized in phosphate buffer containing EDTA and thioglycollic acid and clarified with diethyl ether. The preparations were further purified by centrifugation in sucrose density gradients containing EDTA at pH 9.0 and were then stable at 2 °C for > 100 days. When mounted in neutralized ammonium molybdate they were shown to consist of predominantly intact particles. In tube and ring precipitin tests and in agar gel-diffusion tests, specific precipitation with homologous antiserum occurred only in media containing alkaline adjusted solutions (ammonium molybdate and dipotassium hydrogen phosphate).  相似文献

1. A litter‐bag experiment was undertaken in a pond on the margins of a large temperate floodplain in south‐western France to assess the potential influence of the replacement of native by exotic riparian species on organic matter degradation. We determined initial litter chemical composition, breakdown rates and the invertebrate assemblages associated with the litter for five pairs of native dominant and exotic invasive species co‐occurring at different stages along a successional gradient. 2. Litter chemical composition, breakdown rates and abundance and diversity of detritivorous invertebrates were similar for the exotic and native species overall. No overall changes in organic matter degradation can thus be predicted from the replacement of dominant natives by exotic invasives. Breakdown rates were primarily driven by the C/N ratio. 3. One invasive species (Buddleja davidii) showed significantly higher breakdown rates than its native counterpart (Populus nigra), resulting in the disappearance of leaf litter 6 months prior to the next litterfall. In some cases, therefore, invasion by exotic species may result in discontinuity of resource supply for decomposers.  相似文献

Plants of the apple rootstock M.VII were grown for a singleseason by spraying their roots continuously with nutrient solutions:(a) complete, (b) low iron, (c) low magnesium, and (d) low ironand low magnesium. Detailed records were taken throughout theseason of fresh weight, shoot length, diameter, and leaf area,while the fresh and dry weights of component parts were determinedon harvested samples. The leaves of the iron-deficient plants were chlorotic and hada lower dry weight and higher water content per unit area thanthose of the control plants. Growth and net assimilation ratewere both reduced but the distribution of assimilates was similarto that in the controls. Total growth was also markedly reduced by the low-magnesiumtreatment, but the leaf symptoms were different and the plantswere morphologically distinct from the control plants. Verylittle chlorosis occurred, but necrosis and severe defoliation,progressing up the shoot, reduced the leaf area ratio and probablylargely accounted for the low net assimilation rate and relativegrowth rate. The diameter of the shoot was affected much morethan its length and the shoot/root ratio was greatly increased. Plants deficient in both iron and magnesium showed less defoliationthan when magnesium alone was low. More dry matter accumulatedin the old stem and less in the new shoot than would have occurredwith a simple combination of the single deficiencies.  相似文献

SIGNIFICANT changes in RNA metabolism have been described during early sea urchin development. Until recently the only detectable class of RNA synthesized during cleavage stages was that with a low G + C base composition and heterogeneous sedimentation properties (DNA-Jike RNA)1. The genes for nucleolar ribosomal RNA (26S and 18S) were believed to become active only following gastrulation2–4 and the products of nuclear transfer RNA (4S) genes were first detected at the mesenchyme blastula stage5. Any label in the 4S region of sucrose gradients of RNA from the cleavage stages of embryogenesis was interpreted as reflecting the turnover of the pCpCpA region of pre-existing transfer RNA (tRNA) molecules.  相似文献
