Leaf age-dependent changes in structure, nitrogen content, internal mesophyll diffusion conductance (gm), the capacity for photosynthetic electron transport (Jmax) and the maximum carboxylase activity of Rubisco (Vcmax) were investigated in mature non-senescent leaves of Laurus nobilis L., Olea europea L. and Quercus ilex L. to test the hypothesis that the relative significance of biochemical and diffusion limitations of photosynthesis changes with leaf age. The leaf life-span was up to 3 years in L. nobilis and O. europea and 6 years in Q. ilex. Increases in leaf age resulted in enhanced leaf dry mass per unit area (MA), larger leaf dry to fresh mass ratio, and lower nitrogen contents per dry mass (NM) in all species, and lower nitrogen contents per area (NA) in L. nobilis and Q. ilex. Older leaves had lower gm, Jmax and Vcmax. Due to the age-dependent increase in MA, mass-based gm, Jmax and Vcmax declined more strongly (7- to 10-fold) with age than area-based (5- to 7-fold) characteristics. Diffusion conductance was positively associated with foliage photosynthetic potentials. However, this correlation was curvilinear, leading to lower ratio of chloroplastic to internal CO2 concentration (Cc/Ci) and larger drawdown of CO2 from leaf internal air space to chloroplasts (ΔC) in older leaves with lower gm. Overall the age-dependent decreases in photosynthetic potentials were associated with decreases in NM and in the fraction of N in photosynthetic proteins, whereas decreases in gm were associated with increases in MA and the fraction of cell walls. These age-dependent modifications altered the functional scaling of foliage photosynthetic potentials with MA, NM, and NA. The species primarily differed in the rate of age-dependent modifications in foliage structural and functional characteristics, but also in the degree of age-dependent changes in various variables. Stomatal openness was weakly associated with leaf age, but due to species differences in stomatal openness, the distribution of total diffusion limitation between stomata and mesophyll varied among species. These data collectively demonstrate that in Mediterranean evergreens, structural limitations of photosynthesis strongly interact with biochemical limitations. Age-dependent changes in gm and photosynthetic capacities do not occur in a co-ordinated manner in these species such that mesophyll diffusion constraints curb photosynthesis more in older than in younger leaves.  相似文献

The characterization of 11 Leptinotarsa decemlineata microsatellite loci isolated using the fast isolation by AFLP of sequences containing repeats (FIASCO) protocol is reported. Three to 15 alleles per locus were detected in 54 beetles collected from four populations of L. decemlineata. The mean number of alleles was 7.4 ± 3.2 and the level of expected heterozygosity ranged from 0.451 to 0.798. The total exclusionary probabilities using these loci for the first and the second parent were 0.982 and 0.999, respectively. These are the first microsatellite loci characterized from the Colorado potato beetles that can be used for estimating genetic diversity, population structure and parentage analysis.  相似文献

1. Multiple male copulations can have detrimental effects on female fitness due to sperm limitation. 2. Monandrous Naryciinae females are immobile while the males are short‐lived and do not feed. Multiple male mating is therefore expected to lead to sperm limitation in females. Sperm limitation and male limitation are hypothesised as causes of the repeated evolution of parthenogenetic reproduction in the Psychidae. 3. In this study, the effects of multiple male mating on female reproduction are investigated in several species of Naryciinae by allowing males multiple copulations. The results for two species, Siederia listerella and Dahlica lichenella, are compared. The sex ratios of 53 natural populations are examined for indications of male limitation. 4. Previous copulations by the male increased the female's risk of remaining unfertilised. However, contrary to expectations, those unfertilised females were capable of successful re‐mating. 5. In S. listerella, the number of previous copulations of males negatively influenced female fitness. Females produced 30% fewer offspring if they mated with a previously mated male. In D. lichenella, the older the male and the lower its number of total lifetime copulations, the higher the female's reproductive success. 6. Only a fraction of the investigated populations had a female‐skewed sex ratio, but differences in development time between males and females could lead to reproductive asynchrony. 7. In conclusion, male mating history did not lead to strong sperm limitation in Naryciinae as had been suggested by their life history.  相似文献

In Stenotaenia, a strongly conserved overall morphology is matched by an unusual interspecific variation in both the body size of fully grown specimens (from 1.7 cm in S. romana to 7.7 cm in S. sturanyi) and the number of leg‐bearing segments (from 43 in male S. romana to 115 in female S. sturanyi). In this work the modulation of the regio- and stereo-selective hydroxylation of testosterone by vinclozolin was studied in evaluating cocarcinogenic properties. Changes of cytochrome P450-(CYP)-catalysed drug metabolism was investigated in liver, kidney and lung microsomes of Swiss Albino CD1 mice of both sexes after single (625 or 1250 mg kg-1 b.w.) or repeated (daily 750 mg kg-1 b.w. for 3 days) i.p. administrations. Treatment of mice with a single dose of vinclozolin caused in a dose-dependent fashion from 2 1 to 14 1-fold increase in the 7-, 6- and 2-hydroxylations of testosterone in liver. Lower increase in extrahepatic tissues ranging from 2 3 to 8 1-fold for testosterone 6-, 16 -, 2- and 2- hydroxylase activity in the kidney or from 2 2 to 5 1-fold for 6-, 16 -, 16 - and 2- hydroxylase activity in the lung were observed. Repeated treatment with this fungicide did not substantially modify the extent and pattern of induction, the liver being the only tissue responsive (up to 7 6-fold increase, 7-hydroxylation) in both male and female. In the kidney (7-, 6-, 16 -, 2-, 7-hydroxylations) and lung (6-, 7-, 6-, 16 -, 16 - and 2- hydroxylations), a typical sex-dependent induction (up to 9 0-fold, 16 -hydroxylation in the lung, female) was achieved. In general, however, vinclozolin has a complex pattern of induction and suppression of CYP-dependent enzymes, as exemplified from the reduced expression of some hydroxylations depending upon dose, sex and organ considered. For example, after a single administration, 16 -hydroxylation was suppressed in liver (up to 78% loss in male, higher dose), whereas 16 -hydroxylation was reduced in kidney up to 50% in both sexes (at the higher dose). Glutathione S-transferase activity, measured as index of post-oxidative reactions, was markedly increased by vinclozolin in the liver (up to 5 2-fold, female) and kidney (up to 3 9-fold, female) but not in the lung. Because both phase I and phase II reactions were enhanced by vinclozolin treatment in liver and kidney, the ratio between activation/detoxification mechanisms was slightly affected. Conversely, this ratio was shifted toward activating mechanisms in the lung, sustaining, in part, the expression of certain type of tumours tissue-dependent. Taken together, these findings seem to indicate the cotoxic, cocarcinogenic and promoting potential of this fungicide.  相似文献

Measured profiles of photosynthetic capacity in plant crowns typically do not match those of average irradiance: the ratio of capacity to irradiance decreases as irradiance increases. This differs from optimal profiles inferred from simple models. To determine whether this could be explained by omission of physiological or physical details from such models, we performed a series of thought experiments using a new model that included more realism than previous models. We used ray‐tracing to simulate irradiance for 8000 leaves in a horizontally uniform canopy. For a subsample of 500 leaves, we simultaneously optimized both nitrogen allocation (among pools representing carboxylation, electron transport and light capture) and stomatal conductance using a transdermally explicit photosynthesis model. Few model features caused the capacity/irradiance ratio to vary systematically with irradiance. However, when leaf absorptance varied as needed to optimize distribution of light‐capture N, the capacity/irradiance ratio increased up through the crown – that is, opposite to the observed pattern. This tendency was counteracted by constraints on stomatal or mesophyll conductance, which caused chloroplastic CO2 concentration to decline systematically with increasing irradiance. Our results suggest that height‐related constraints on stomatal conductance can help to reconcile observations with the hypothesis that photosynthetic N is allocated optimally.  相似文献

The uptake and release of 3H-labelled fusicoccin (FC) fed topea internode segments and the transport of [3H]FC in wholeplants and in excised parts of plants of pea, maize, and pumpkinhave been investigated. FC uptake showed three phases apparentlycorresponding to the penetration of FC: (1) into the free space;(2) into the cytoplasm; (3) into the vacuole. The first phase is a little thermosensitive and apparently includesadsorption binding to some free space component (cell surface?).The second and third phases are highly temperaturesensitiveand in part energy-dependent. However, no accumulation of FCagainst a concentration gradient is observed at high externalFC concentration. Accumulation at low concentration seems dueto the formation of a poorly exchangeable complex with somecell structure. The efflux of the FC penetrated within the cells is limitedby some highly temperature-sensitive process. A small fractionof the accumulated FC tends to be retained in the tissue, eitherbecause sequestered in some compartment or because bound tosome cellular component. FC transport in whole plants as well as in isolated plant partsseems to depend mainly on simple diffusion and on transportby mass flow in the xylem. Long distance transport of the toxinin pumpkin plants mainly occurs in the xylem, while the slowmovement in non-vascular tissues seems to depend on diffusioncombined with mass flow in the free space, and does not seemheavily influenced by metabolic factors.  相似文献

ABSTRACT. We observed different low salinity tolerances between two groups of populations of Euplotes crassus. After mating analysis, the less tolerant populations could be assigned to three mating groups. Two of them were separated by preconjugative barriers but both conjugated with a third one. A breeding test showed evidence of (potential) introgression mediated by this "bridging" group. On the other hand, the more tolerant populations formed a fourth mating group unable to conjugate with the others and characterized by distinct zymogram patterns. We conclude that natural introgression among the studied populations is not complete and that the species structure of E. crassus comprises intrafertile sets of populations among which gene flow may be difficult or virtually absent.  相似文献

Bois Noir is an emergent disease of grapevine that has been associated to a phytoplasma belonging to the XII‐A stolbur group. In plants, phytoplasmas have been found mainly in phloem sieve elements, from where they spread moving through the pores of plates, accumulating especially in source leaves. To examine the expression of grapevine genes involved in sucrose transport and metabolism, phloem tissue, including sieve element/companion cell complexes and some parenchyma cells, was isolated from healthy and infected leaves by means of laser microdissection pressure catapulting (LMPC). Site‐specific expression analysis dramatically increased sensitivity, allowing us to identify specific process components almost completely masked in whole‐leaf analysis. Our findings showed decreased phloem loading through inhibition of sucrose transport and increased sucrose cleavage activity, which are metabolic changes strongly suggesting the establishment of a phytoplasma‐induced switch from carbohydrate source to sink. The analysis focused at the infection site also showed a differential regulation and specificity of two pathogenesis‐related thaumatin‐like genes (TL4 and TL5) of the PR‐5 family.  相似文献
