Myostatin (GDF-8, MSTN) is a member of trans- forming growth factors (TGF-β) superfamily, which was first described by McPherron et al. in 1997[1]. Myostatin appears to act as a negative regulator of muscle development and controls not only fibre size but also fibre number[2,3]. 本实验旨在构建雪莲类PEBP基因原核表达载体,在大肠杆菌中表达并纯化类PEBP基因所编码的蛋白,为进一步研究奠定基础.将雪莲类PEBP基因开放阅读框序列克隆到原核表达载体pET30( )上,转化感受态表达菌株BL21(DE3),低浓度IPTG低温诱导融合蛋白的表达,纯化产物,Western blotting鉴定目的蛋白.IFTG低诱导PEBP,经SDS-PAGE分析,其相对分子量约为28 kD,与预期相符,表达量约占菌体蛋白的26.8%,并且通过亲和层析纯化了重组融合蛋白,Western blotting鉴定为阳性.成功构建了原核表达载体pET-PEBP,获得了高效表达产物,并为进一步研究雪莲类PEBP基因的抗冻功能打下基础.  相似文献

猪是研究糖尿病最理想的模型动物,研究胰岛素和胰岛素抵抗是研究糖尿病的重要环节。为明确SOCS-3在胰岛素抵抗中的作用,分别用100nmol/L的胰岛素,300nmol/L的地基米松处理原代培养的猪脂肪细胞诱导胰岛素抵抗;利用半定量RT-PCR技术分别检测SOCS-3、OB、GLUT4和PPARγ基因表达变化。结果发现,胰岛素增加了GLUT4、SOCS-3和PPARγ基因的表达,对OB基因表达变化没有显著性影响;地塞米松诱导的胰岛素抵抗状态下OB和SOCS-3基因表达水平升高,而GLUT4和PPARγ基因表达水平显著下调。研究结果表明,GLUT4基因表达量水平的升高可能是由于PPARγ的高表达引起,SOCS-3基因的不同表达水平对胰岛素信号的抑制效果不同。地塞米松诱导的胰岛素抵抗不仅表现在对葡萄糖转运的抑制,也反映在抑制了胰岛素信号;而SOCS-3基因可能是消除胰岛素抵抗的一个有效靶基因。  相似文献

在一个继代周期中,培养基中添加10-50 g.L-1的蔗糖、葡萄糖和果糖后,白桦愈伤组织的鲜重均上升,浓度为20-30 g.L-1的三种糖,其鲜重积累量均较高.培养基中添加不同浓度葡萄糖和果糖的白桦愈伤组织中三萜物质含量先上升后下降,而添加蔗糖的则呈直线上升.其中,葡萄糖,果糖和蔗糖浓度为30 g.L-1的三萜类物质积累量最高.  相似文献
