The building sector is one of the most relevant sectors in terms of environmental impact. Different functional units (FUs) can be used in life cycle assessment (LCA) studies for a variety of purposes. This paper aimed to present different FUs used in the LCA of buildings and evaluate the influence of FU choice and setting in comparative studies.
Methods
As an example, we compared the “cradle to grave” environmental performance of four typical Brazilian residential buildings with different construction typologies, i.e., multi-dwelling and single dwelling, each with high and basic standards. We chose three types of FU for comparison: a dwelling with defined lifetime and occupancy parameters, an area of 1 m2 of dwelling over a year period, and the accommodation of an occupant person of the dwelling over a day.
Results and discussion
The FU choice was found to bias the results considerably. As expected, the largest global warming indicator (GWi) values per dwelling unit and occupant were identified for the high standard dwellings. However, when measured per square meter, lower standard dwellings presented the largest GWi values. This was caused by the greater concentration of people per square meter in smaller area dwellings, resulting in larger water and energy consumption per square meter. The sensitivity analysis of FU variables such as lifetime and occupancy showed the GWi contribution of the infrastructure more relevant compared with the operation in high and basic standard dwellings. The definition of lifetime and occupancy parameters is key to avoid bias and to reduce uncertainty of the results when performing a comparison of dwelling environmental performances.
Conclusions
This paper highlights the need for adequate choice and setting of FU to support intended decision-making in LCA studies of the building sector. The use of at least two FUs presented a broader picture of building performance, helping to guide effective environmental optimization efforts from different approaches and levels of analysis. Information regarding space, time, and service dimensions should be either included in the FU setting or provided in the building LCA study to allow adjustment of the results for subsequent comparison.

Mechanobiology is focused on how the physical forces and mechanical properties of proteins, cells, and tissues contribute to physiology and disease. Although the response of proteins and cells to mechanical stimuli is critical for function, the tools to probe these activities are typically restricted to single-molecule manipulations. Here, we have developed a novel microplate reader assay to encompass mechanical measurements with ensemble biochemical and cellular assays, using a microplate lid modified with magnets. This configuration enables multiple static magnetic tweezers to function simultaneously across the microplate, thereby greatly increasing throughput. We demonstrate the broad applicability and versatility through in vitro and in cellulo approaches. Overall, our methodology allows, for the first time (to our knowledge), ensemble biochemical and cell-based assays to be performed under force in high-throughput format. This approach substantially increases the availability of mechanobiology measurements. 相似文献
Abstract This study proposes a bioclimatic characterization and a new biogeographic division for the Antarctic territories up to the province level following the criteria and models of Rivas-Martínez et al. The Antarctic Kingdom comprises the continent of Antarctica, the surrounding ice-covered Antarctic islands, and the associated cold oceanic islands and archipelagos. It has two biogeographic regions: the Antarctic Region and the Subantarctic Insular Region. The Antarctic Region includes the entire pergelid Antarctic continent and the surrounding islands and archipelagos, and is characterized by upper suprapolar hyperoceanic and oceanic or Polar pergelid bioclimates on the coasts. The region has been divided into three pr6ovinces: Maritime Antarctica, West Antarctica and East Antarctica. The Subantarctic Insular Region comprises the circumantarctic islands and archipelagos that are widespread at the southern tip of the planet’s most important oceans, mostly in the subtemperate latitudinal zone inside or not far from the Antarctic Convergence. Bioclimatically, all insular subantarctic territories (excluding the South-American Tierra de Fuego, Terra Magellanica and large islands) are characterized by thermo-suprapolar and semipolar antarctic hyperoceanic bioclimates on the coasts. Four provinces – Falklandian-South Georgian, Kerguelenian, Macquarian and Aucklandian-Campbellian – have been recognized in this region. A derivative of fuculose-1-phosphate aldolase, immobilized with high loading on glyoxal–agarose gels, has been characterized and evaluated as a biocatalyst for an aldol addition reaction. The reaction of the solid biocatalyst was diffusion-controlled for conversion of its natural substrate. Nevertheless, when catalyzing the synthesis of a biologically active aminopolyol, the lower reaction rate with non-natural substrates led to a process controlled by the intrinsic enzyme kinetics. The resulting biocatalyst has high synthetic specific activity and has been successfully used in batch synthesis reactions with high conversion. In addition, the immobilized aldolase has been employed in fed-batch synthesis, increasing the selectivity of the reaction and obtaining high conversion (88%). 相似文献
Bcause oxidative stress is one of the main sources of severe cellular damage, cells have different defense weapons against reactive oxygen species. Ubiquitous plasma membrane redox systems play a role in defense against oxidative stress damage. On the other hand, a tightly controlled and localized production of reactive oxygen species by a plasma membrane NADPH oxidase can be used as a potent microbicidal weapon. Small biopsy samples are used increasingly to assess the biomarker expression for prognostic information and for monitoring therapeutic responses prior to and during neoadjuvant therapy. The issue of intratumor heterogeneity of expression of biomarkers, however, has raised questions about the validity of the assessment of biomarker expression based on limited tissue samples. We examined immunohistochemically the expression of HER-2neu (p185erbB-2), epidermal growth factor receptor (EGFR), Bcl-2, p53, and proliferating cell nuclear antigen (PCNA) in 30 breast carcinomas using archived, paraffin embedded tissue and determined the extent of intratumor heterogeneity. Each section was divided into four randomly oriented discrete regions, each containing a portion of the infiltrating carcinoma. For each tumor, the entire lesion and four regions were analyzed for the expression of these markers. Scores of both membrane and cytoplasmic staining of HER-2neu and EGFR, scores of cytoplasmic staining of Bcl-2, and scores of nuclear staining of both p53 and PCNA were recorded. The intensity of staining and the proportion of immunostained cells were determined. A semiquantitative immunoscore was calculated by determining the sum of the products of the intensity and corresponding proportion of stained tumor cells. We analyzed both invasive (IDC) and in situ (DCIS) carcinomas. The Wilcoxon signed-rank test was used for paired comparisons between overall and regional immunoscores and between overall and regional percentages of stained cells. Spearman's correlation coefficients were used to assess the level of agreement of overall biomarker expression with each of the regions. Generalized linear models were used to assess overall and pair-wise differences in the absolute values of percent changes between overall and regional expression of biomarkers. For IDCs, there were no statistically significant differences in the expression of the biomarkers in terms of either the percentage of cells staining or the immunoscores when comparing the entire tumor with each region except for the lower EGFR expression of arbitrarily selected region 1 and lower p53 expression of region 1 compared to that of the entire tumor section. For DCIS, there were no statistically significant differences in the expression of the biomarkers between the entire tumor and each region except in PCNA of region 2 compared to that of entire tumor section. Positive correlation of immunoscores was observed between the entire tumor and each region as well as across all four regions for IDC. Similar observations were noted with DCIS except for HER-2neu and PCNA. No statistically significant differences were observed in the absolute values of percent changes of biomarker expression between overall and the four regions for both DCIS and IDC. Therefore, no significant intratumor heterogeneity in the expression of HER-2neu, Bcl-2, and PCNA was observed in IDC. Minor regional variations were observed for EGFR and p53 in IDC. Similarly, no significant regional variation in the expression of markers was observed in DCIS except for PCNA. 相似文献
