Ecological features and conservation requirements of populations at the latitudinal limits of a species’ geographical range frequently differ from those in other parts of the range. Identifying such differences is key to implementing effective conservation strategies for threatened range‐edge populations especially, in the context of rapid global warming, at the lower‐latitude range edge. We studied habitat selection and diet of the endangered Cantabrian Capercaillie Tetrao urogallus cantabricus in a recently discovered population at the southernmost edge of the sub‐species’ range. This is the only Western Capercaillie population in the Mediterranean biogeographical region. We combined non‐systematic surveys based on questionnaires, reports and field sampling with data from radiotracking to assess habitat selection. Diet was surveyed by micro‐histological methods from droppings collected in the new population, which inhabits Pyrenean Oak Quercus pyrenaica forests and Scots Pine Pinus sylvestris plantations, and in two Cantabrian populations inhabiting Eurosiberian forests. Capercaillie preferred large (> 500 ha) and medium‐sized (100–500 ha) Pyrenean Oak forest fragments and large Scots Pine plantations. Forest fragments smaller than 100 ha and non‐forested habitats were always avoided. Diet differed markedly between Mediterranean and Eurosiberian populations. Bilberry Vaccinium myrtillus is common in the diet of most Capercaillie populations but was scarce in the study area and so was rare in the diet of the new population. Instead, Rockrose Halimium lasianthum was described for the first time as a major food resource for the Capercaillie and was consumed in autumn and winter. Pine needles were also heavily consumed in winter. We document for the first time the strong preference of Capercaillie for Pyrenean Oak forests and a moderately high consumption of the leaves, buds and acorns of this tree species throughout the year. Habitat selection and diet of this Mediterranean population differ from those of the core Cantabrian and other populations. Our results suggest a wider environmental tolerance (phenotypic plasticity) in the species than previously recognized. We advocate specific protection for this unique range‐edge Capercaillie population and its Pyrenean Oak forest habitat. 相似文献

Between 2000 and 2009 we studied the diet and breeding success of Egyptian Vultures Neophron percnopterus in southern Spain. Wild species accounted for 74.9% of prey items (n = 1071) with a predominance of mammals (62.3%), followed by birds (20.8%) and reptiles (13.1%). Spatially, the diet was highly varied and not restricted to carcasses of livestock; wild Rabbits Oryctolagus cuniculus accounted for 54% of the overall remains. The spatial variability may reflect regional and local disparity in the availability of main prey. The temporal relationship between variation in trophic diversity and Vulture nesting productivity (both values showing a long‐term decrease) might suggest a causal link between variation in diet and reproductive output. We hypothesize that high turnover rates could explain productivity variation as a consequence of the recruitment of less experienced individuals to the breeding population. This could in turn generate covariation between diet and reproductive output. 相似文献

The disruption of the circadian system in humans has been associated with the development of chronic illnesses and the worsening of pre-existing pathologies. Therefore, the assessment of human circadian system function under free living conditions using non-invasive techniques needs further research. Traditionally, overt rhythms such as activity and body temperature have been analyzed separately; however, a comprehensive index could reduce individual recording artifacts. Thus, a new variable (TAP), based on the integrated analysis of three simultaneous recordings: skin wrist temperature (T), motor activity (A) and body position (P) has been developed. Furthermore, we also tested the reliability of a single numerical index, the Circadian Function Index (CFI), to determine the circadian robustness. An actimeter and a temperature sensor were placed on the arm and wrist of the non-dominant hand, respectively, of 49 healthy young volunteers for a period of one week. T, A and P values were normalized for each subject. A non-parametric analysis was applied to both TAP and the separate variables to calculate their interdaily stability, intradaily variability and relative amplitude, and these values were then used for the CFI calculation. Modeling analyses were performed in order to determine TAP and CFI reliability. Each variable (T, A, P or TAP) was independently correlated with rest-activity logs kept by the volunteers. The highest correlation (r = −0.993, p<0.0001), along with highest specificity (0.870), sensitivity (0.740) and accuracy (0.904), were obtained when rest-activity records were compared to TAP. Furthermore, the CFI proved to be very sensitive to changes in circadian robustness. Our results demonstrate that the integrated TAP variable and the CFI calculation are powerful methods to assess circadian system status, improving sensitivity, specificity and accuracy in differentiating activity from rest over the analysis of wrist temperature, body position or activity alone. 相似文献

Obesity is the result of a positive balance between total energy intake and its catabolism. Although many factors are involved in the regulation of energy metabolism, the discovery of leptin led to energy homeostasis being investigated in greater depth. Since its identification, leptin has been considered important in the development of obesity, given its anorexigenic effect and influence on food intake and energy expenditure. Leptin is involved in diverse physiological processes such as energy balance, appetite and body weight control, fat and carbohydrate metabolism, and reproduction. However, to be able to function, this hormone has many specific receptors both centrally (hypothalamus) and peripherally in the skeletal muscle, lungs and kidneys. Measurements of the prostate-specific antigen (PSA) levels in blood are widely used as diagnostic, predictive and prognostic marker of prostate disease. The selective detection of molecular forms of PSA can contribute clinically to meaningful enhancements of the conventional PSA-test. As it is plausible that an in-depth search for structural variants of PSA gene products may increase our ability to discriminate distinct patho-biological basis and stages of prostate diseases, we have developed a multi-step protocol comprising gel-based methods followed by mass spectrometric identification.Our current aim was to provide a comprehensive identification of PSA variants occurring in seminal fluid. We provide a proof-of-principle for this multiple step analytical approach to identify multiple PSA variants from complex biological samples that revealed distinct molecular characteristics. In addition, sequence-annotated protein bands in SDS–PAGE gels were compared to those detected by Western blots, and by monitoring the enzymatic activity in zymogram gels, using gelatin as a substrate. The high accuracy annotations were obtained by fast turnaround MALDI-Orbitrap analysis from excised and digested gel bands. Simple Sequence Repeats (SSRs) are known to be scattered and present in high number in eukaryotic genomes. We demonstrate
that dye-labeled oligodeoxyribonucleotides with repeated mono-, di-, tri, or tetranucleotide motifs (15-20 nucleotides in
length) have an unexpected ability to recognize SSR target sequences in non-denatured chromosomes. The results show that all
these probes are able to invade chromosomes, independent of the size of the repeat motif, their nucleotide sequence, or their
ability to form alternative B-DNA structures such as triplex DNA. This novel and remarkable property of binding SSR oligonucleotides
to duplex DNA targets permitted the development of a non-denaturing fluorescence in situ hybridization method that quickly
and efficiently detects SSR-enriched chromosome regions in mitotic, meiotic, and polytene chromosome spreads of different
model organisms. These results have implications for genome analysis and for investigating the roles of SSRs in chromosome

The naturally occurring steroid dehydroepiandrosterone (DHEA) is reported to reduce glial fibrillary acidic protein (GFAP) overexpression in a model of reactive gliosis due to its conversion to estradiol by the enzyme aromatase. In the present study we examined the biological effect of a new epimerized derivative of DHEA, 16α-iodomethyl-13α-dehydroepiandrosterone derivative (16α-iodomethyl-13α-DHEAd, 16α-iodomethyl-13α-androst-5-en-3β,17β-diol), using the same model system, and compared the 3D structure of this molecule with that of DHEA and two steroidal type aromatase inhibitors, formestane and exemestane. The synthetic compound, in contrast to the reported effect of DHEA, was able to reduce GFAP overexpression only if the enzyme aromatase was inhibited. Data obtained from computational calculations fortified by X-ray crystallography revealed that contrary to the nearly planar sterane framework of DHEA, the synthetic derivative 16α-iodomethyl-13α-DHEAd has a bent sterane skeleton, resulting in a 3D structure that is similar to that of formestane or exemestane. Moreover, 16α-iodomethyl-13α-DHEAd resulted to be metabolically more stable than DHEA.The results suggest that epimerization of the sterane skeleton of DHEA inclines the plane of the D ring, leading to a significantly altered biological activity. The synthetic molecule has a DHEA-like effect on GFAP overexpression when the enzyme aromatase is inhibited and the naturally occurring DHEA is ineffective in this respect. On the other hand, based on their structural similarity it can be hypothesized that 16α-iodomethyl-13α-DHEAd applied alone might have a biological effect similar to that of formestane or exemestane. 相似文献
