In the framework of a willow rust research project, it was necessary to include vegetatively propagated plant material of selected sallow trees (Salix caprea L.) into biotests for identification of pathotypes. Since it was not possible to root sufficient clonal plants by conventional cutting propagation, the applicability of tissue culture methods was tested. From 10 selected donor trees of Salix caprea newly sprouted shoots were harvested and transferred to nutrient media after surface disinfection. The cultures were grown at 20--22 °C, illuminated with warm-white fluorescent tubes. The majority of shoot tips and nodal segments died during the first month, but only with nursery-grown plants this was caused by bacteria contamination. Two clones could be established easily on hormone-free medium. Five clones could be initiated only after repeated subcultures on various media variants. Three clones failed completely. Different basic media compositions were tested and Woody Plant Medium, supplemented with 0.1% activated charcoal, proved to be best for most of the sallow clones. Well developed rooted plantlets were used in vitro for microcutting propagation. The resulting plants were transferred to soil and could be included in the rust screening program after acclimatising. The applicability of micropropagation for selected Salix caprea donor trees was strongly depending on the genotype. But the comparison of results from microcuttings with conventional cutting propagation showed that these methods were successful for different clones each. 相似文献

Abstract 1. Most ants are more like plants and marine invertebrates than other insects because the adults are sessile and the immature stages are motile. 2. This paper reported the results from a field experiment that increased food levels on the fungus gardening ant, Trachymyrmex septentrionalis, which has served as a model system for understanding the complexities of these obligate mutualisms. 3. Food supplementation increased the growth of the symbiotic fungus and this led to an increase in the amount of fungal biomass. The amount of fungal biomass was generally more important in determining the amount of ant offspring than colony size. 4. Background: Understanding the role of livestock grazing on plant diversity can be improved by an accurate measurement of diversity at all hierarchical scales due to the changeability of diversity components in space.Aims: We evaluated the effects of grazing on plant species diversity at different scales of all common and rare species in two regions that have different climatic conditions (arid vs. semi-arid).Methods: In each region, we collected abundant data of plant species from a nested sampling design that consisted of local (80 plots) and regional (16 sites) scales. We partitioned total species diversity (γ) into within plots (αl), among plots (βl) and among sites (β2) using the additive partitioning.Results: Diversity among sites contributed the most to total diversity for all and rare plant species in both regions. In addition, α1 and β1 diversities in ungrazed areas were greater than those in grazed areas for all and common species in both climates.Conclusion: Abandonment of grazing after 10 years resulted in significant regeneration of common species at the local scale, with no change in rare species. We conclude that low grazing intensity is likely to be an important tool for conservation of plant diversity in which all scales should be considered. 相似文献

A number of homeobox genes have been found to be expressed in skin and its appendages, such as scale and feather, and appear to be candidates for the regulation of the development of these tissues. We report that the proline-rich divergent homeobox gene Hex is expressed during development of chick embryonic skin and its appendages (scale and feather). In situ hybridization analysis revealed that, during development of the skin, a transient expression of the Hex gene was observed. While the expression of Hex in the dermis was closely correlated with proliferation activity of epidermal basal cells, that in the epidermis was related to a suppression of epidermal differentiation. When dermal fibroblasts were transfected with Hex, stimulation of both DNA synthesis and proliferation of the epidermal cells followed by two-fold scale ridge elongation and increase in epidermal area was observed during culture of the skin, whereas epidemal keratinization was not affected. This is the first study to demonstrate that Hex is expressed during development of the skin and its appendages and that its expression in the dermal cells regulates epidermal cell proliferation through epithelial mesenchymal interaction. 相似文献

Three high-resolution climatic reconstructions, based on diatom analyses from lake sediment cores from the Canadian prairies, show that shifts in drought conditions have prevailed on centennial to millennial time scales for at least the past six millennia. These shifts in mean aridity exhibit broad regional synchrony, with particularly pronounced shifts at all sites between ∼1700–2000 cal. yr bp and ∼3600–3900 cal. yr bp , as well as at ∼5400–5500 cal. yr bp for the two sites which extend back to at least 6000 cal. yr bp . The two Saskatchewan lakes exhibited significant coherence in both the timing and direction of these shifts, whereas inferred changes at the westernmost site in Alberta were significantly correlated to the Saskatchewan sites, but opposite in sign, and exhibited more high-frequency variation on the scale of centuries. The mechanisms behind these abrupt shifts in aridity are poorly understood, but may be linked to changes in oceanic–atmospheric interactions that influence the mean position of the jetstream and the associated storm tracks. Natural shifts in mean climatic conditions may accelerate with increasing carbon dioxide levels intensifying the likelihood of extreme droughts in North American prairies. 相似文献

Knowledge of the spatial scales of diversity is necessary to evaluate the mechanisms driving biodiversity and biogeography in the vast but poorly understood deep sea. The community structure of kinetoplastids, an important group of microbial eukaryotes belonging to the Euglenozoa, from all abyssal plains of the South Atlantic and two areas of the eastern Mediterranean was studied using partial small subunit ribosomal DNA gene clone libraries. A total of 1364 clones from 10 different regions were retrieved. The analysis revealed statistically not distinguishable communities from both the South-East Atlantic (Angola and Guinea Basin) and the South-West Atlantic (Angola and Brazil Basin) at spatial scales of 1000–3000 km, whereas all other communities were significantly differentiated from one another. It seems likely that multiple processes operate at the same time to shape communities of deep-sea kinetoplastids. Nevertheless, constant and homogenous environmental conditions over large spatial scales at abyssal depths, together with high dispersal capabilities of microbial eukaryotes, maintain best the results of statistically indistinguishable communities at larger spatial scales. 相似文献

We compared the results of two biologging techniques used to study the foraging behaviour of a colony of small inshore predators, little penguins (Eudyptula minor). The first technique involved the use of satellite transmitters and diving loggers deployed on separate individuals, which has been the conventional method of tracking the movements and behaviour of this species for > 10 years. The second technique combined a diving logger and a global positioning system (GPS) logger deployed on the same individual, which is similar to the biologging methods presently being developed and used for many other species. We then considered the value of each technique as a conservation tool operating at the small scale (foraging area < 5000 ha and duration < 1 day).We found that the separately deployed satellite transmitters significantly underestimated the penguins' foraging area size. However, the size of the foraging area and other foraging parameters, such as total distance travelled, were influenced by the degree of GPS location sub-sampling. Furthermore, only the combined diving and GPS loggers could confidently describe the diving behaviour of the penguins in relation to the sea floor and identify that they were using small areas of conservation interest (shipping channel) inside their foraging area.Hence, the method employed to assess habitat use at fine scales can influence conservation measures that rely upon the data collected. We suggest that researchers fast-track their adoption of high resolution multi-loggers for increased data confidence when tracking animals at a fine scale, but also consider the potential effect of sampling rate on the calculation of parameters of interest. 相似文献
