The toxicity of nitrofurantoin was studied on human WI-38 fibroblasts: this chemical was lethal when added at concentrations higher than 5·10−5 M in the culture medium. The protection afforded by anitoxidants was then tested: α-tocopherol gave at 10−4 M a light protection in contrast to ascorbic acid which even became toxic at high concentrations. We also tested catalase, superoxide dismutase and glutathione peroxidase introduced intracellularly by the microinjection technique. On a molecular basis, glutathione peroxidase was 23-times more efficient than catalase and 3000-times more than superoxide dismutase. The results also showed that a similar range of enzyme concentrations was found for the protection against high oxygen pressure. This suggests that, in the case of both oxygen and nitrofurantoin toxicity, the peroxide derivatives are the most toxic intermediates of the free radical attacks.  相似文献

How to Characterize a Biological Antioxidant   总被引:15,自引:0,他引:15  
An antioxidant is a substance that, when present at low concentrations compared to those of an oxidizable substrate, significantly delays or prevents oxidation of that substrate. Many substances have been suggested to act as antioxidants in vivo, but few have been proved to do so. The present review addresses the criteria necessary to evaluate a proposed antioxidant activity. Simple methods for assessing the possibility of physiologically-feasible scavenging of important biological oxidants (superoxide, hydrogen peroxide, hydroxyl radical, hypochlorous acid, haem-associated ferryl species, radicals derived from activated phagocytes, and peroxyl radicals, both lipid-soluble and water-soluble) are presented, and the appropriate control experiments are described. Methods that may be used to gain evidence that a compound actually does function as an antioxidant in vivo are discussed. A review of the pro-oxidant and anti-oxidant properties of ascorbic acid that have been reported in the literature leads to the conclusion that this compound acts as an antioxidant in vivo under most circumstances.  相似文献

Cadmium (Cd) is a widespread heavy metal pollutant and environmental and human health hazard, which may be partially resolved using green and cost-effective phytoremediation techniques. However, the efficiency of phytoremediation is often limited by the small biomass of Cd-hyperaccumulator plants. Although cattail (Typha angustifolia L.) is tolerant of heavy metals and has a high biomass, there is little information available on its detoxification mechanisms for heavy metals, especially Cd. In the present study we investigated the tolerance of cattail to Cd and mechanisms involved in its Cd detoxification. Our results show that: (a) cattail is tolerant of Cd; (b) the root Casparian band, cell wall, vacuole, glutathione (GSH), and glutathione peroxidase (GPX) play important roles in Cd detoxification; and (c) mechanisms of Cd detoxification differ in leaf cell cytoplasm (mainly a GSH-related antioxidant defense system) and root cell cytoplasm (mainly a GSH-related chelation system). In summary, cattail possesses multiple detoxification mechanisms for Cd and is a promising species for phytoremediation of Cd-polluted environments.  相似文献

为了解根际供氮对紫青菜(Brassica campestris ssp.chinensis var.communis)生长和营养品质的影响,用不同水平的NH_4NO_3处理,对紫青菜的生理特性和营养品质进行研究。结果表明,随着供氮水平的增加,紫青菜的鲜质量、干质量、根系活力和游离氨基酸含量均呈先上升后略下降趋势;硝酸还原酶(NR)活性、叶绿素含量、NO_3~–含量和可溶性蛋白质含量均呈上升趋势;DPPH·自由基清除率、超氧化物歧化酶(SOD)活性、过氧化物酶(POD)同工酶和过氧化氢酶(CAT)同工酶活性等均提高;而花青素苷相对含量、可溶性糖含量、维生素C(Vc)含量、总酚含量和FRAP值均呈下降趋势。根际NH_4NO_3为75~300 mg L~(–1)时,紫青菜的鲜质量和干质量较低;NH_4NO_3达1200 mg L~(–1)时,其鲜质量和干质量又低于600 mg L~(–1)处理的,植株积累较多NO_3~–,导致营养品质下降,同时根际也积累NH4NO3。因此,600 mg L~(–1)的根际NH_4NO_3是适宜的氮水平,其鲜质量和干质量均最高、营养均衡、抗氧化能力强。  相似文献

In this study several investigations and tests were performed to determine the antioxidant activity and the acetylcholinesterase and tyrosinase inhibitory potential of Pulmonaria officinalis and Centarium umbellatum aqueous extracts (10% mass) and ethanolic extracts (10% mass and 70% ethanol), respectively. Moreover, for each type of the prepared extracts of P. officinalis and of C. umbellatum the content in the biologically active compounds – polyphenols, flavones and proanthocyanidins was determined. The antioxidant activity was assessed using two methods, namely the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and reducing power assay. The analyzed plant extracts showed a high acetylcholinesterase and tyrosinase inhibitory activity in the range of 72.24–94.24% (at the highest used dose – 3 mg/mL), 66.96% and 94.03% (at 3 mg/mL), respectively correlated with a high DPPH radical inhibition – 70.29–84.9% (at 3 mg/mL). These medicinal plants could provide a potential natural source of bioactive compounds and could be beneficial to the human health, especially in the neurodegenerative disorders and as sources of natural antioxidants in food industry.  相似文献

EGb761 is a well-defined mixture of active compounds extracted from Ginkgo biloba leaves. This extract is used clinically due to its neuroprotective effects, exerted probably via its potent antioxidant or free radical scavenger action. Previous studies suggest that oxidative stress, via free radical production, may play an important role in depression and animal models for depression-like behavior. Preclinical studies have suggested that antioxidants may have antidepressants properties. The aim of this study was to investigate the antidepressant-like of EGb761 due to its antioxidant role against oxidative stress induced in the forced swimming test, the most widely used preclinical model for assessing antidepressant-like behavior. Male BALB/c mice were pretreated with EGb761 (10 mg/kg, ip) daily for 17 days followed by the forced swimming test and spontaneous locomotor activity. Animals were sacrificed to evaluate lipid peroxidation, different antioxidant enzyme activities, serotonin and dopamine content in midbrain, hippocampus and prefrontal cortex. EGb761 significantly decreased the immobility time (39%) in the forced swimming test. This antidepressant-like effect of EGb761 was associated with a reduction in lipid peroxidation and superoxide radical production (indicated by a downregulation of Mn-superoxide dismutase activity), both of which are indicators of oxidative stress. The protective effect of EGb761 is not related to excitatory or inhibitory effects in locomotor activity, and was also associated with the modulation of serotonergic and dopaminergic neurotransmission. It is suggested that EGb761 produces an antidepressant-like effect, and that an antioxidant effect against oxidative stress may be partly responsible for its observed neuroprotective effects.  相似文献

Daily topical application of the aqueous ethanolic extract of the marine sea grass, Thalassia testudinum, on mice skin exposed to UVB radiation resulted in a dose-dependent recovery of the skin macroscopic alterations over a 6-day period. Maximal effect (90%) occurred at a dose of 240 μg/cm2, with no additional effects at higher doses. Bioassay-guided fractionation of the plant extract resulted in the isolation of thalassiolin B (1). Topical application of 1 (240 μg/cm2) markedly reduces skin UVB-induced damage. In addition, thalassiolin B scavenged 2,2-diphenyl-2-picrylhydrazyl radical with an EC50 = 100 μg/ml. These results suggest that thalassiolin B is responsible for the skin-regenerating effects of the crude extract of T. testudinum. Erik L. Plants are confronting a variety of environmental hazards as a result of fast climate change, which has a detrimental influence on soil, plant growth, and nutrient status. As a result, the present study aims to evaluate the influence of various fly ash concentrations (5, 10, 15, 20, 25, 30, and 35% FA) mixed with the optimum concentrations of nitrogen in the form of urea (0.5 g pot?1) on the growth, productivity and biochemical constituents of radish plants. Energy-dispersive X-ray spectroscopy (EDX) and scanning electron microscopy (SEM) were used to assess soil physical–chemical properties and FA nutrient status. Results suggested that FA added many essential plant nutrients to the growth substrate and improved some important soil characteristics such as pH, electric conductivity, porosity, and water holding capacity. Also, the results revealed that the low concentrations of FA up to 20% were found to boost radish growth, yield, chlorophyll, carotenoids, and mineral content. While the highest concentrations of FA (25–35%) decreased radish growth and yield, increased oxidative stress through increased lipid peroxidation (MDA) and caused a significant boost in ascorbic acid, proline, protein, and antioxidant enzyme activities. Furthermore, SEM of radish leaf revealed an enhancement in the stomatal pore of radish leaf under different levels of FA. In conclusion, combining 15% fly ash with 0.5 g nitrogen in the form of urea significantly enhanced radish yield by enhancing antioxidant activity such as catalase, peroxidase, ascorbate peroxidase, Guaiacol peroxidase, superoxide dismutase, nitrate reductase and reducing oxidative stress, potentially reducing fly ash accumulation and environmental pollution.  相似文献

Antioxidant enzyme (AE) activities were studied in normal hamster kidney proximal tubules and in estrogen-induced hamster kidney cancer. In vivo, kidney tumor had lower activities of manganese superoxide dismutase (MnSOD), copper, zinc superoxide dismutase, catalase, and glutathione peroxidase than kidney proximal tubules. Differences in AE activities were, in general, maintained in tissue culture, with AE activities remaining low in tumor cells compared to normal cells. Normal proximal tubular cells showed significant induction of MnSOD activity as a function of time in culture of following exposure to diethylstilbestrol, a synthetic estrogen, while MnSOD activity remained low in tumor cells under these conditions. Our results suggest that antioxidant enzymes, particularly MnSOD, are regulated differently in estrogen-induced hamster kidney tumor cells than in normal kidney proximal tubular cells, demonstrating that cancers arising from hormonal influence have similar AE profiles to those previously described in cancers arising from viral or chemical etiologies.  相似文献
