Reactivation and Dedifferentiation of Differentiated Murine Erythroleukemic Cell Nuclei

A murine erythroleukemic cell line, 745 A4-TG, deficient in hypoxanthine-guanine-phosphoribosyl transferase, can be induced with 3 mM hexamethylene bisacetamide to yield at least 50% of cells undergoing irreversible erythroid differentiation and finally losing capacity for cell divisions. The effects of such induced differentiation of 745 A4-TG on its ability to form viable and proliferating hybrids when fused with 3T3 1T22 fibroblasts were investigated. We found that when the induced 745 A4-TG cells were used, more continuously proliferating hybrids were obtained than could be accounted for by the residual uninduced cells which remained in these induced preparations. This suggests that some of the induced 745 A4-TG cells, when fused with 3T3 1T22 reverted from the induced phenotype of a limited capacity for cell proliferation to an uninduced state of continuous proliferation. This observation was further confirmed with the use of fully differentiated 745 A4-TG cells, which were obtained after selection with a bromodeoxyuridine suicide treatment to eliminate the uninduced and the partially differentiated cells in the preparations. When these selected, fully differentiated cells, as characterized by their lack of proliferation capacity and thymidine kinase activity, were fused with 3T3 1T22 (also deficient in thymidine kinase), it was found that not only were viable hybrid colonies obtained in a selection medium, which precluded the proliferation of either parental cells, but these hybrids continued to proliferate for more than two months in selection medium. These data thus confirmed that some fully differentiated erythroleukemic nucleus components in the hybrids were reactivated to regain capacity for cell proliferation and to dedifferentiate to synthesize thymidine kinase for survival in the selection medium. The lack of hemoglobin synthesis by these hybrids also indicates dedifferention of these murine erythroleukemic components in the hybrids.     

Volatile components of developing tomato fruit grown under field and greenhouse conditions

Volatile components of field- and glass greenhouse-grown tomatoes(Lycopersicon esculentum MILL., variety, V. R. Moscow) of differentmaturities were studied by gas-liquid and thin-layer chromatography.The following components were separated and identified: isobutylalcohol, isopentyl alcohol, hexyl alcohol, 2-methyl-3-hexanol,isovaleraldehyde, caproaldehyde, benzaldehyde, furfural, isopentylacetate, isopentyl butyrate, isopentyl isovalerate, butyl hexanoate,hexyl hexanoate, methyl salicylate and -pinene. Chromatogramswere consistent for both field- and greenhouse-grown tomatoesat all stages of maturity. Except for isovaleraldehyde and hexylalcohol, the biosynthesis of volatile components increased alongwith the growth of fruit. Isovaleraldehyde and hexyl alcohol,presumed to give the "green leafy" aroma of tomatoes, were foundto be in their highest concentrations at the breaker and thelarge-green stages, respectively. ¹This research was supported by a research grant—UI 00449—fromthe National Center for Urban and Industrial Health, U. 九种罕见的人类染色体异常核型报告

人工饲养蒙新河狸的发情交配行为

人工饲养的蒙新河猪其动情周期可划分为４个阶段：动情前期、动情期（发情交配期）、动情后期和停滞期（动情间隔期）。动情期间雌雄河狸均表现出活跃好动,浮躁不安,食量减小;发情交配期雌雄河狸均发出低沉嘶哑的叫声,雄性叫声频率比性略高,爬跨交配是动情期发情的高峰,动情周期各阶段的行为特征,在雌性与阴道涂片细胞学及外生殖器形态、色泽、阴道分泌物周期性变化特征相吻合,在雄性则与睾丸及阴茎形态特征吻合,在人工饲养     

神经生长因子结构与功能研究进展

神经生长因子(NGF)是神经营养因子家族的典型代表, 它控制着脊椎动物周围和中枢神经系统中部分神经元的发育和存活.NGF的三维结构是以“胱氨酸结”和β折叠为基础,它以二聚体的形式结合细胞表面的受体从而发生生物学效应.参与这些反应的氨基酸残基已通过化学修饰和定点突变法加以确定,这有助于更进一步理解其结构与功能的关系.     

利用染色体涂染方法建立人和白眉长臂猿(Hylobates hoolock)的比较染色体图谱

除人Y染色体外,本文采用生物素标记的人全部整条染色体特异探针与白眉长臂猿(Hylobates hoolock)有丝分裂中期分裂相进行染色体原位杂交即染色体涂染法以研究人和白眉长臂猿染色体之间的同源性。在白眉长臂猿18对常染色体上检测出了与人22对常染色体同源的59对染色体片段,确定了人和白眉长臂猿之间的精度较高的染色体连锁群。结果表明:自人与白眉长臂猿的祖先分歧以来,大量的染色体间重排(至少发生了39次易位)和染色体内的重排导致了二者核型的差异。根据杂交结果绘制了首份人和白眉长臂猿比较染色体图谱,并结合已有的人和白掌长臂猿(Hylobates lar)(2n=44)和合趾长臂猿(Hylobates syndactylus)(2n=50)的比较染色体图谱对长臂猿属的染色体进化作了初步的探讨。     

杉梢小卷蛾新种记述(鳞翅目:卷蛾科)

近年来由于杉梢小卷蛾在福建、江西、浙江、江苏、湖南、湖北、安徽、广东、广西等省(区)的为害,严重地影响了杉木的成长。现经研究鉴定为一新种,属鳞翅目、卷蛾科(Tortricidae)、小卷蛾亚科(Olethreutinae)、新小卷蛾族(Olethreutini)、Polychrosis属,订名为 Polychrosis cunninghamiacola。     

青海湖裸鲤自然产卵场的生境特征及无人机遥测判别——以泉吉河为例

自然产卵场是维持物种延续最关键的栖息地,青海湖裸鲤（Gymnocypris przewalskii）的自然繁殖栖息地状况和生境特征尚缺乏详细的定量研究。以青海湖入湖河流泉吉河为例,在平水期采用现场调查和无人机遥测的方法对青海湖裸鲤的自然产卵场分布及生境状况进行调查,确定其产卵场生境特征参数,建立基于无人机遥测识别产卵场的方法并进行复核验证。结果表明：泉吉河河道形态可分为弯曲型、分汊型和顺直型等三种典型河型,青海湖裸鲤自然产卵场主要分布在弯曲型和分汊型河道中,分汊型河道几乎100%都有产卵场分布,弯曲型河道有70%为产卵场;产卵场河道平均长度（135.13±61.13） m,平均宽度（30.01±17.51） m,平均曲折度1.09±0.07;平均面积（4586.6±4201.61） m²;产卵场常位于缓水浅滩处,平均水深（0.19±0.10） m （范围：0.03-0.44m）,平均流速（0.24±0.20） m/s （范围：0.01-0.81m/s）,河床质为含有沙粒的卵石（粒径：163-256mm）底质。河道形态、沙洲分布、水深特征等特征参数可作为无人机遥测识别产卵场的判断条件,并实现验证成功。研究结果可为开展整个流域的青海湖裸鲤自然产卵场现状评估及保护对策制定提供技术支撑。
