Summary -Galactosidase specific production fromK. marxianus was shown to be related with k a at low rates. A power type curve fitted very well when specific activity was plotted vs. k a. Summary Examination of the relationship between the rate of oxygen transfer and the rate of polymer production revealed an unexpectedly high requirement for oxygen. At a cell density of about 3 g (dry wt)/L, the threshhold value for OTR for optimal synthesis of polymer is about 50 mmoles O2/L.hr. Whereas Rushton turbines are effecient at transfering oxygen to solution, their use reduces the quality of the recovered polymer. Although better quality polymer can be produced in a reactor employing an agitator which causes less shear stress, the productivity can be compromised due to the inefficiency in OTR. The present study describes operating conditions for the provision of sufficient OTR in a system compatible with the production of high-quality polymer whereby turbine impellers were replaced with a marine-type propeller and mass transfer was assisted by means of a gas dispersion device.  相似文献

The Salton Sea is a shallow (mean depth = 8 m; maximum depth = 15 m), saline (41–45 g l–1), intermittently mixing, 57 km long, 980 km2 lake located in the arid southwestern United States. The Sea is a wind driven system, with predominant winds paralleling the long axis of the lake, being strongest in spring and weakest in summer and fall. The Sea mixed daily or nearly daily between September and January. During this cooling period, moderate to high levels of dissolved oxygen (3–11 mg l–1) were found throughout the water column. Mean water column temperature ranged from a minimum of 13–14 °C in early January to a maximum of 30–34 °C in July–September. During most of this warming period, the Sea was thermally stratified but subject to periodic wind driven mixing events. Winds were stronger in spring 1998 than in 1997 or 1999, causing more rapid heating of the lake that year and also delaying onset of anoxic conditions in bottom waters. During summer months, mid-lake surface waters were sometimes supersatured with oxygen, and bottom waters were hypoxic or anoxic with sulfide concentrations > 5 mg l–1. Oxic conditions (> 1 mg O2 l–1) often extended a few meters deeper nearshore than they did well offshore as a consequence of greater mixing nearshore. Mixing events in late summer deoxygenated the entire water column for a period of days. Consumption of oxygen by sulfide oxidation likely was the principal mechanism for these deoxygenation events. Sulfide concentrations in surface waters were 0.5–1 mg l–1 approximately 3 days after one mixing event in mid-August 1999. These mixing events were associated with population crashes of phytoplankters and zooplankters and with large fish kills. In the southern basin, freshwater inflows tended to move out over the surface of the Sea mixing with saline lake water as a function of wind conditions. Salinity gradients often contributed more to water column stability than did thermal gradients in the southeasternmost portion of the lake.  相似文献

Three potential routes to generation of reactive oxygen species (ROS) from α-tocopherolquinone (α-TQ) have been identified. The quinone of the water-soluble vitamin E analogue Trolox C (Trol-Q) is reduced by hydrated electron and isopropanol α-hydroxyalkyl radical, and the resulting semiquinone reacts with molecular oxygen to form superoxide with a second order rate constant of 1.3 × 108 dm3/mol/s, illustrating the potential for redox cycling. Illumination (UV-A, 355 nm) of the quinone of 2,2,5,7,8-pentamethyl-6-hydroxychromanol (PMHC-Q) leads to a reactive short-lived (ca. 10? 6 s) triplet state, able to oxidise tryptophan with a second order rate constant greater than 109 dm3/mol/s. The triplet states of these quinones sensitize singlet oxygen formation with quantum yields of about 0.8. Such potentially damaging reactions of α-TQ may in part account for the recent findings that high levels of dietary vitamin E supplementation lack any beneficial effect and may lead to slightly enhanced levels of overall mortality.  相似文献

1. Magnesium-protoporphyrin chelatase activity, previously shown in whole cells of Rhodopseudomonas spheroides, could not be demonstrated in cell-free extracts prepared in different ways, although spheroplasts retained moderate activity. Slight activity was detected also in whole cells of Rhodospirillum rubrum. 2. The effects on the activity of the enzyme of inhibitors of electron and energy transfer were studied in whole cells of Rps. spheroides. Amytal, rotenone, azide and cyanide inhibited at low pO(2) in the dark but not under anaerobic conditions in the light. Antimycin A and 2-heptyl-4-hydroxyquinoline N-oxide, as well as uncouplers and oligomycin, inhibited under all environmental conditions. 3. The effects on magnesium chelatase activity of intermediates of the tricarboxylic acid cycle, of thenoyltrifluoroacetone, of a number of artificial electron donors or acceptors, of various quinones and of the oxidation-reduction indicator dyes Benzyl Viologen and Methyl Viologen are described. 4. It was concluded that electron transport between a b-type and a c-type cytochrome as well as associated energy conservation and transformation reactions were essential for activity. There was also a specific requirement for ATP. 5. Exogenous protoporphyrin and magnesium protoporphyrin monomethyl ester were incorporated into bacteriochlorophyll or late precursors by whole cells. 6. Evidence is presented that the insertion of magnesium was the only step inhibited by oxygen in the biosynthetic pathway between protoporphyrin and bacteriochlorophyll.  相似文献

The resting oxygen consumption     , postprandial and post-exercise peak oxygen consumption     of 137 juvenile southern catfish Silurus meridionalis , weighing 18·5 ± 0·8 g (mean ± s . d .), were measured at 25° C to determine whether     is positively related to postprandial and post-exercise     in sedentary S. meridionalis . In addition, postprandial metabolic response [ i.e. the specific dynamic action (SDA)] after a satiating meal and the growth performance as a consequence of a 3 week feeding-growth trail were measured in 40 S. meridionalis , weighing 14·3 ± 0·2 g, at 25° C to determine whether postprandial     is positively related to growth rate. Postprandial     was positively correlated with     , while post-exercise     was not. Both postprandial     and post-exercise     were positively correlated with factorial and absolute scope. There was no significant correlation between the growth rate and postprandial     in S. meridionalis . It suggested that as a sit-and-wait forager with low     , low post-exercise     and high postprandial     , the expenditure of energy for maintenance in S. meridionalis may be more closely related to digestive processes than locomotor activities.  相似文献

Worm density of a deposit feeding oligochaete (Lumbriculus variegatus) did not affect egestion whereas both temperature and sediment type had a significant influence. The worms egested less actively at the lowest temperature (6 °C). The egestion rate, expressed as mg dry feces produced, was highest in the sandy sediment and lowest in the sediment derived almost exclusively from decaying plant material. The amount of dry material and the volume of wet sediment passing through the worms varied between the test sediments; the results are probably dependent on the chosen unit of measure. Reproduction was significantly decreased in sandy sediment with low organic carbon content. Reproduction was also dependent on the worm size, larger worms reproducing more frequently. Most worms lost weight during the tests, but the loss was lowest in the sediment with the highest organic carbon content. The cyclic nature of walking can lead to repetitive stress and associated complications due to the rate of loading (ROL) experienced by the body at the initial contact of the foot with the ground. An individual's gait kinematics at initial contact has been suggested to give rise to the ROL, and a repetitive, high ROL may lead to several disorders, including osteoarthritis. Additionally, proprioception, the feedback signaling of limb position and movement, may play a role in how the foot strikes the ground and thus, the ROL. Our goal was to explore the relationship between proprioception, gait kinematics and ROL. Thirty-eight women were recruited for gait analysis, and the gait characteristics 50 ms prior to and at initial contact were examined. Two proprioception tests, joint angle reproduction and threshold to detect passive motion were used to examine the subject's proprioceptive acuity. Our results indicate that individuals with a larger knee angle (i.e., greater extension) 50 ms prior to initial contact (IC) experience a higher ROL during gait and have poorer proprioceptive scores. However, it remains unclear whether poor proprioception causes a high ROL or if a high ROL damages the mechanoreceptors involved in proprioception, but the apparent relationship is significant and warrants further investigation.  相似文献

In this article, we attempt to estimate the contemporary phytoplankton species pool of a particular lake, by assessing the rate of floral change over a period of 15 years. Phytoplankton time series data from Lake Stechlin, an oligo-mesotrophic lake in the Baltic Lake District (Germany) were used. Of the 254 algal species recorded during the 15-year of studies with roughly biweekly sampling, 212 species were planktonic. In the individual plankton years, the recorded total number of species changed between 97 and 122, of which the number of dominants (>1% contribution to the annual average of total biomass) was only 10–19. The 15-year cumulative number of species exhibited an almost linear increase after an initial saturation phase. This increase was attributed to two reasons: increase of sample size and immigration of species new to the flora. Based on a probabilistic model developed in this study, we estimated the number of co-existing planktonic species of the lake as some 180, and the rate of floral change as 1–2 species per year. Of these co-existing species, only few maintain the matter–energy processing ecosystem functions in any particular plankton year. Selection of these dominants is probably driven by mesoclimatic cycles, coupled with human-induced forcing, like eutrophication. All others are hiding as an ecological memory, in the sense of the capacity or experiences of past states to influence present or future responses of the community. Data analyses suggest that selection of the ‘memory species’ that show temporary abundance increases over shorter (several years) periods are largely dependent upon the dominants. These results show that interspecific interactions and the particular autecological features of the dominants, together with their effects on the whole ecosystem, act as a major organizing force. Some phytoplankton species, like Planktothrix rubescens, are efficient ecosystem engineers with cascading effects of both a top-down and bottom-up nature. Historical scientific data on Planktothrix blooms in Lake Stechlin suggest cyclic patterns in long-term development of phytoplankton which, as the legend of the Red Cock suggests, dates back much further than scientific archives.  相似文献

Seventeen different strains of filamentous fungi were grown in batch cultures to compare their abilities for the production of β-fructofuranosidase. Three of them, Aspergillus oryzae IPT-301, Aspergillus niger ATCC 20611 and strain IPT-615, showed high production with total fructosyltransferase activity higher than 12,500 units l−1. In addition, the β-fructofuranosidases of those strains have a high fructosyltransferase activity-to-hydrolytic activity ratio. The temperature and pH effects on the sucrose-β-fructofuranosidase reaction rate were studied using a 22 factorial experimental design. The comparative analysis of the tested variable coefficients shows that the variable pH contributes mostly to the changes in the fructosyltransferase and hydrolytic rates and in the V t/V h ratio. At 40 and 50°C, there were no significant differences between the fructosyltransferase and hydrolytic velocities of these enzymes.  相似文献

In vivo investigations have confirmed the beneficial effects of hydrophilic, cationic Mn(III) porphyrin-based catalytic antioxidants in different models of oxidative stress. Using a cell culture model of rat mixed neuronal/glial cells, this study investigated the effect of MnTnOct-2-PyP5+ on oxygen and glucose deprivation (OGD)-induced cell death as compared to the effects of widely studied hydrophilic analogues MnTE-2-PyP5+ and MnTDE-2-ImP5+ and a standard compound, dizocilpine (MK-801). It was hypothesized that the octylpyridylporphyrin, MnTnOct-2-PyP5+, a lipophilic but equally potent antioxidant as the other two porphyrins, would be more efficacious in reducing OGD-induced cell death due to its higher bioavailability. Cell death was evaluated at 24 h using lactate dehydrogenase (LDH) release and propidium iodide staining. At concentrations from 3–100 µm, all three porphyrins reduced cell death as compared to cultures exposed to OGD alone, the effects depending upon the concentrations and type of treatment. To assess the effect of lipophilicity the additional experiments were performed using submicromolar concentrations of MnTnOct-2-PyP5+ in an organotypic hippocampal slice model of OGD with propidium iodide and Sytox staining. When compared to oxygen and glucose deprivation alone, concentrations of MnTnOct-2-PyP5+ as low as 0.01 µm significantly (p<0.001; power 1.0) reduced neuronal cells similar to control. This is the first in vitro study on the mammalian cells which indicates that MnTnOct-2-PyP5+ is up to 3000-fold more efficacious than equally potent hydrophilic analogues, due entirely to its increased bioavailability. Such remarkable increase in efficacy parallels 5.7-orders of magnitude increase in lipophilicity of MnTnOct-2-PyP5+ (log P=?0.77) when compared to MnTE-2-PyP5+ (log Pow=?6.43), Pow being partition coefficient between n-octanol and water.  相似文献

The tropical coastal dunes in central Gulf of Mexico have been stabilizing over the last decades resulting in reduced substrate mobility, and promoting primary succession. We describe changes in species richness and diversity in dune vegetation during 20?years. Our questions: (a) Do species richness and diversity increase over time as predicted by models of ecological succession or do they show a hump-backed manner similar to the observations in temperate coastal dunes?, (b) What is the interaction between vegetation cover and diversity and species richness?, (c) Is there a relationship between species diversity and succession rate and does succession rate change over time?, and (d) How do plant functional types change during succession? In order to answer these questions, we set 140 4?×?4?m permanent plots in a mobile dune area and monitored vegetation cover and species richness from 1991 to 2011. In time, diversity increased in a logistic manner toward an asymptotic value once vegetation cover surpassed 60?%. Species richness increased in a humped-back shape, also reaching a maximum peak at 60?% vegetation cover. The succession rate of diversity was measured by the Euclidean distance, and showed a significant humped-back relation, meaning that it was slower in early and late successional stages. The study supports the intermediate disturbance theory. The conservation of coastal dunes vegetation should focus on all, species-poor and species-rich habitats that help to maintain the ecological integrity of these ecosystems. The understanding of community dynamics and diversity patterns becomes an essential component of coastal dune management and conservation.  相似文献

Genomes contain evidence for the history of life and furthermore contain evidence for lateral gene transfer, which was an important part of that history. The geological record also contains evidence for the history of life, and newer findings indicates that the Earth's oceans were largely anoxic and highly sulfidic up until about 0.6 billion years ago. Eukaryotes, which fossil data indicate to have been in existence for at least 1.5 billion years, must have therefore spent much of their evolutionary history in oxygen-poor and sulfide-rich environments. Many eukaryotes still inhabit such environments today. Among eukaryotes, organelles also contain evidence for the history of life and have preserved abundant traces of their anaerobic past in the form of energy metabolic pathways. New views of eukaryote phylogeny suggest that fungi may be among the earliest-branching eukaryotes. From the standpoint of the fungal feeding habit (osmotrophy rather than phagotrophy) and from the standpoint of the diversity in their ATP-producing pathways, a eukaryotic tree with fungi first would make sense. Because of lateral gene transfer and endosymbiosis, branches in the tree of genomes intermingle and occasionally fuse, but the overall contours of cell history nonetheless seem sketchable and roughly correlateable with geological time.  相似文献

Nitrogen (N) rate and plant density (PD) are important factors for sustainable cotton production. The objective of this study is to examine the effects of nitrogen rate and plant density on plant growth, source–sink relationship, and cotton yield. A split-plot arrangement was used in the field experiment with the main plots assigned to N rate (120 and 180 kg/ha), and the sub-plots assigned to plant density (8, 10, and 12 plants/m2). Results showed significant N and PD interaction on plant growth, leaf gas exchange, and yield. Higher plant growth and cotton yield were noted under low nitrogen rate and high planting density than other treatment combinations. Leaf photosynthesis, stomatal conductance, intercellular CO2, transpiration rate, and water use efficiency were considerably influenced by planting density and nitrogen rate. Maximum values of these traits were obtained under low nitrogen rate with high planting density or high nitrogen rate with medium planting density, while the least values were under low nitrogen rate with low planting density. Correlation analysis revealed highly significant and positive relation between leaf gas exchange and cotton yield.  相似文献

Insecticides cause a range of sub‐lethal effects on targeted insects, which are frequently detrimental to them. However, targeted insects are able to cope with insecticides within sub‐lethal ranges, which vary with their susceptibility. Here we assessed the response of three strains of the maize weevil Sitophilus zeamais Motschulsky (Coleoptera: Curculionidae) to sub‐lethal exposure to the pyrethoid insecticide cypermethrin. We expected enzyme induction associated with cypermethrin resistance since it would aid the resistant insects in surviving such exposure. Lower respiration rate and lower activity were also expected in insecticide‐resistant insects since these traits are also likely to favor survivorship under insecticide exposure. Curiously though, cypermethrin did not affect activity of digestive and energy metabolism enzymes, and even reduced the activity of some enzymes (particularly for cellulase and cysteine‐proteinase activity in this case). There was strain variation in response, which may be (partially) related to insecticide resistance in some strains. Sub‐lethal exposure to cypermethrin depressed proteolytic and mainly cellulolytic activity in the exposed insects, which is likely to impair their fitness. However, such exposure did not affect respiration rate and walking behavior of the insects (except for the susceptible strain where walking activity was reduced). Walking activity varies with strain and may minimize insecticide exposure, which should be a concern, particularly if associated with (physiological) insecticide resistance.  相似文献

Herbaceous plants grown with free access to nutrients exhibit inherent differences in maximum relative growth rate (RGR) and rate of nutrient uptake. Measured rates of root respiration are higher in fast-growing species than in slow-growing ones. Fast-growing herbaceous species, however, exhibit lower rates of respiration than would be expected from their high rates of growth and nitrate uptake. We investigated why the difference in root O2 uptake between fast- and slow-growing species is relatively small. Inhibition of respiration by the build-up of CO2 in closed cuvettes, diurnal variation in respiration rates or an increasing ratio of respiratory CO2 release to O2 uptake (RQ) with increasing RGR failed to explain the relatively low root respiration rates in fast-growing grasses. Furthermore, differences in alternative pathway activity can at most only partly explain why the difference in root respiration between fast- and slow-growing grasses is relatively small. Although specific respiratory costs for maintenance of biomass are slightly higher in the fast-growing Dactylis glomerata L. than those in the slow-growing Festuca ovina L., they account for 50% of total root respiration in both species. The specific respiratory costs for ion uptake in the fast-growing grass are one-third of those in the slow-growing grass [0·41 versus 1·22 mol O2 mol (NO3)–1]. We conclude that this is the major cause of the relatively low rates of root respiration in fast-growing grasses.  相似文献
