从线粒体基因探讨中国大头蛙群的分类及其属内地位 总被引:6,自引:0,他引:6
Partial sequences of the mitochondrial 12S rRNA and 16S rRNA gene were determined for 8 populations of three species of Chinese Limnonectes, and aligned with the published sequences of Limnonectes from other parts of the world. When Nanorana parker, Paa boulengeri, Fejervarya limnocharis and Hoplobatrachus rugulosus was used as outgroup taxa (Accession Nos. AY158705, AY313685, AF206111, AF206491, AY322311). The sequences of the 12S rRNA and 16S rRNA genes totaled 950 nueleotide positions with gaps including 510 variable sites. We reconstructed phylogenetie trees using Clustal X 1.8, Mega 2.1 and PHYLIP 3.5e software, and using the maximum parsimony and maximum likelihood methods, respectively. Our analyses suggest that these fanged frogs from China are another monophyletie group in addition to the four monophyletie groups identified by previous studies. The Chinese Limnonectes were grouped into three elades (BCL 55% ). The first elade contains one species (BCL 100% ), from a population of Limnoneetes fragilis from Hainan Province. The second contains four individuals (BCL 100% ), i. e. two populations of Limnonectes kuhlii from Yunnan Province. The third contains one species (BCL 100% ), i. e. five populations of Limnonectes fujianensis from Fujian Province and 1 from Taiwan Province. The resulted phylogenetie trees indicate L. fragilis is basal to L. kuhlii L. fujianensis 。 相似文献

泛希姬蝽Himacerus apterus(Fabricius)是半翅目姬蝽科中重要的天敌种类,本文旨在分析泛希姬蝽不同地理种群的遗传多样性和遗传分化情况。利用COI作为标记基因,使用DnaSP、Arlequin、MEGA等软件分析了中国4个省12个种群33个体的单倍型多样性、遗传结构分化、系统发育等。共发现19个单倍型,所有单倍型中仅H1为共享单倍型,为宁夏和陕西共有,且出现频率最高。总体单倍型多样性指数Hd=0.913,核苷酸多样性指数π=0.006,平均核苷酸差异数k=4.083。Tajima's D=-0.853,P>0.100,表明泛希姬蝽未经过种群扩张。AMOVA分析表明种群内的分子变异大于种群间分子变异,变异比例分别为45.393%和36.594%,遗传分化指数均大于0.250,差异水平极显著。泛希姬蝽不同地理种群遗传多样性较高,宁夏与内蒙古、山西、陕西省分组间存在极度的遗传分化。种群间遗传距离和地理距离无相关性,表明地理距离不是影响种群间遗传距离的重要因素。通过比较4个省采集点的环境特点,认为地区间基因流受限和气温、猎物的差异可能是影响遗传分化的重要因素。 相似文献

到目前为止,多数学者认为小熊猫种内已分指名亚种A.f.fulgens和川西亚种A.f.styani。然而,由于其个体毛色变异较大,一些学者对其亚种分化问题提出质疑。本文采用DNA指纹方法,对小熊猫的种内遗传和亚种分化进行了研究。结果表明,川西亚种所有个体在分子量约为8.4kb处均有一条指名亚种不具有的共有谱带,指名亚种所有个体则在分子量约为1.8kb处具有另外一条川西亚种不具有的共有谱带,且这两条谱带可通过双亲遗传给子代,说明此共有谱带可分别作为区分小熊猫川西亚种和指名亚种的特征带。另外,种内的遗传分化研究表明,川西亚种基因组的多态性强于指名亚种,且川西亚种内各个体之间的遗传变异高于指各亚种,说明小熊猫种内已形成遗传分化。因此,笔者认为以上研究结果为基因水平上进一步证明小熊猫种内已产生遗传分化,并形成两独立亚种,目前的亚种地位成立。 相似文献

利用COI基因序列分析长江与澜沧江水系日本沼虾群体的遗传结构 总被引:3,自引:0,他引:3
测定了我国长江水系和澜沧江水系的日本沼虾9个群体,共79个个体的线粒体COI基因序列片段(约450bp),结果发现有89个变异位点,共计有46个单倍型。其中云南昆明(KM)群体具有较丰富的遗传多样性(h=1.000,π=0.028),而重庆(CQ)群体的遗传多样性最小(h=0.700,π=0.008)。AMOVA分析表明,群体间的遗传变异占总遗传变异的9.66%,而90.34%的遗传变异源于群体内。采用邻接法(NJ)构建的分子系统树显示,46个单倍型明显地聚为长江中下游和长江上游与澜沧江两个族群。其结果可以为合理开发和利用日本沼虾自然野生资源,以及建立和保护日本沼虾种质资源库及基因库提供必要的参考。 相似文献

为深入了解罗氏沼虾(Macrobrachium rosenbergii)不同种质资源的遗传背景,研究采用微卫星标记和线粒体基因相结合,对罗氏沼虾4个育种群体,即选育3代的数丰核心群体(SF)、引进的正大群体(ZD)、正大和数丰杂交的群体(ZDS)、正大子代和数丰杂交的群体(ZD2S)的遗传多样性进行了研究。150个个体的微卫星分析结果显示, 7个微卫星位点均表现出高度多态性(PIC>0.5), 4个群体平均的等位基因数(Na)、期望杂合度(He)和多态信息含量(PIC)分别为19.43、0.8980和0.8867。SF群体的平均He(0.874)和PIC(0.854)最高,ZD2S的He (0.863)和PIC (0.834)其次, ZDS群体的He (0.798)和PIC (0.761)最低。4个群体间的遗传分化指数(FST)为0.04166—0.10438,处于中低水平的遗传分化,其中, ZD和ZDS的遗传分化最大, FST... 相似文献

采用RAPD和mtDNA 16S rRNA基因序列分析方法对海南和马来西亚斑节对虾(Penaeus monodon)种群进行了遗传多样性和遗传分化研究.结果表明:15个RAPD随机引物共检测到82个位点,海南和马来西亚种群的多态位点比例分别为75.90%和76.83%,杂合度分别为0.199和0.218,遗传多样性指数分别为0.276和0.288,种群间的遗传距离为0.015;16S rRNA基因检测的种内遗传变异较低,马来西亚和海南种群的核苷酸多样性分别为0.011和0,种群之间的遗传距离为0.008;马来西亚种群比海南种群的遗传变异水平要高得多,且海南种群可能起源于马来西亚种群;进行遗传选育时可考虑引进马来西亚亲虾作为奠基群体. 相似文献

测定了12个不同地理种群的茶树害虫假眼小绿叶蝉Empoasca vitis(Gothe)和1个外群共103个个体的线粒体DNA 16S rRNA基因部分序列,比较其同源性,计算核苷酸组成,并构建单倍型进化关系图。结果表明:在获得的假眼小绿叶蝉536 bp的序列中A+T含量占76.8%,其中56个核苷酸位点存在变异;100个个体共检测出56种单倍型,单倍型多样性指数(H)为0.971,核苷酸多样性指数(Pi)0.006;12个种群间的平均基因流(Nm)为1.64。总群体的固定系数Fst为0.026;AMOVA分子方差分析结果表明假眼小绿叶蝉的遗传分化主要来自于种群内部(97.4%)。各地理种群的遗传距离与地理分布不具有直接对应的关系。根据构建的单倍型进化关系网,各地理种群的单倍型呈现一种混杂的分布格局,未显示出与地理分布的一致性。 相似文献

2012年和2013年在湖南省桑植县天平山采集到6号臭蛙类标本,经形态比较和分子系统学分析,鉴定为宜章臭蛙(Odorrana yizhangensis),表明天平山为该物种的新分布点。这些标本与宜章臭蛙的主要鉴别特征相符:体型较小;体背面棕色斑大而密,形状不规则;腹面褐色,其上斑纹稀少;股后部色浅,深色斑明显。雄性头体长(52.45 ± 2.54)mm(48.96 ~ 54.92 mm,n = 3),雌性头体长(73.11 ± 1.43)mm(71.26 ~ 74.74 mm,n = 3),均比模式标本群体的头体长略大。基于线粒体16S rRNA基因片段(502 bp)的系统关系分析结果表明,分析的天平山臭蛙标本与宜章臭蛙已知种群聚为一支系,天平山种群与宜章臭蛙已知种群间的遗传距离为0.002 0 ~ 0.008 1,远小于宜章臭蛙与其近缘种龙胜臭蛙(O. lungshengensis)之间的遗传距离(0.035 2 ~ 0.043 9);这表明采自天平山的臭蛙样本属于宜章臭蛙。本文为宜章臭蛙物种的地理分布与系统学提供了新的信息。 相似文献

利用16S rRNA RFLP,16S rRNA序列分析和16S-23S IGS PCR RFLP技术对43株花生根瘤菌和来自其他种属的15个参比菌株进行了群体遗传多样性和系统分析。16S rRNA PCR RFLP分析结果表明,所有供试花生根瘤菌均属于慢生根瘤菌属,在系统发育上与B.japonicum的亲缘关系最近,具有相同的16S rRNA RFLP基因型,而与B.elkanii相对较远。16S rRNA 序列分析结果表明,供试花生根瘤菌在系统发育上更接近于B.liaoningense,序列间差异小于1%,而B.liaoningense在系统发育上与B.japonicum相距很近,其序列间差异小于1%,16S-23S rRNA IGS RFLP分析结果表明,尽管花生根瘤菌与B.japonicum和B.elkanii的亲缘关系很近,但在71%的相似性水平上供试花生根瘤菌仍各自聚为一群,并可进一步分为A、B、C和D4个亚群,该分群还明显反映了地理因素对群体遗传多样性和系统发育的影响。 相似文献

对中国已发表的125个人群Rh血型系统表型分布频率数据进行了收集和整理。用累积计算法(Counting method)重新计算单倍型频率,通过各单倍型的分布差异分析中国不同地区不同种族、人群的遗传差异,分组比较了不同地区汉族和少数民族人群的基因多样度和基因分化度,结果表明南方少数民族和北方少数民族民族亚群体基因多样度有明显差异,用不同方法计算中小组遗传距离并进行聚类分析。最后,计算了除维吾尔族和哈萨克族外的68个人群间的遗传距离并聚类比较,结果表明Rh基因单倍型的分布与地理分布基本相符。 相似文献

目的建立PCR-CE-RFLP方法直接检测女性生殖道感染标本中的细菌,为临床提供快速、准确的诊断依据。方法利用细菌16SrRNA集保守性和变异性于一体的特点,合成1对通用引物,其上游5’端用5'-FAM标记。将8株已知菌和38例临床诊断为生殖道炎症的标本及其培养物进行PCR扩增,扩增产物经限制性内切酶HaeⅢ消化,经毛细管电泳(CE)通过限制性片断长度多态性分析(RFLP)来鉴定不同病原菌。结果(1)已知菌株PCR扩增阳性率为75%(6/8)、生殖道感染标本的PCR扩增阳性率为79%(30/38),毛细管电泳阳性率两者均为100%。(2)已知菌株PER扩增产物的5’端HaeⅢ完全酶切片段的毛细管电泳结果,与电子克隆产物的酶切片段基本一致。(3)用传统培养检测法在生殖道能检出的常见致病菌,实验用PCR-CE-RFLP法同样也能检出。(4)实验还发现数种传统培养法未检出的细菌。结论PCR-CE-RFLP法比传统的细菌培养法具有快速、准确、灵敏的优点,比传统方法缩短20h,可用于临床标本的直接鉴定。 相似文献

对中国主要淡水湖泊(鄱阳湖、洞庭湖、太湖、洪泽湖及巢湖)三角帆蚌5个野生群体的线粒体DNA 16S rRNA基因片段进行了扩增和测序,得到473bp的碱基序列,没有发现插入/缺失突变的核苷酸位点。检测到了32个多态性核苷酸位点,共7种单倍型。鄱阳湖群体的222(C→G)和325(A→G)位点,太湖群体的233(A→G)位点,巢湖群体的40(A→G)、138(A→T)和294(C→T)位点,洪泽湖群体的241(A→C)位点的变异可以作为区分群体分子遗传标记位点。洞庭湖群体未发现特异位点。在5个群体间鄱阳湖群体多态性位点、核苷酸多态性、单倍型多态性和单倍型数量4个指标都最高,表明鄱阳湖群体具有最为丰富的遗传结构,遗传变异最大,可作为三角帆蚌选育的基础群体。 相似文献

AIMS: In order to understand the genetic diversity of Acacia tortilis ssp. raddiana-rhizobia in Tunisia, isolates from nine geographical locations were obtained and analysed. METHODS AND RESULTS: Characterization using restriction fragment length polymorphism analysis (RFLP) of PCR-amplified 16S rRNA gene and the intergenic spacer (IGS) between the 16S and 23S rRNA genes was undertaken. Symbiotic efficiency of the strains was also estimated. Analysis of the 16S rRNA by PCR-RFLP showed that the isolates were phylogenetically related to Ensifer ssp., Rhizobium tropicii-IIA, and Rhizobium tumefaciens species. Analysis of 16S-23S spacer by PCR-RFLP showed a high diversity of these rhizobia and revealed eleven additional groups, which indicates that these strains are genetically very diverse. Full 16S rRNA gene-sequencing showed that the majority of strains form a new subdivion inside the genera Ensifer, with Ensifer meliloti being its nearest neighbour. Nodulation test performed on the plant host demonstrated differences in the infectivity among the strains. CONCLUSION: Rhizobial populations that nodulate specifically and efficiently Acacia tortilis ssp. raddiana in representative soils of Tunisia is dominated by E. meliloti-like genomospecies. SIGNIFICANCE AND IMPACT OF THE STUDY: This paper provides the first clear characterization and symbiotic efficiency data of rhizobia strains nodulating A. tortilis in Tunisia. 相似文献

Diversity of bacterial community in water layer of Woopo wetland was investigated. Cultivable bacterial strains were isolated
by the standard dilution plating technique and culture-independent 16S rRNA gene clones were obtained directly from DNA extracts
of a water sample. Amplified rDNA restriction analysis (ARDRA) was applied onto both of the isolates and 16S rRNA gene clones.
Rarefaction curves, coverage rate and diversity indices of ARDRA patterns were calculated. Representative isolates and clones
of all the single isolate/clone phylotype were partially sequenced and analyzed phylogenetically. Sixty-four and 125 phylotypes
were obtained from 203 bacterial isolates and 235 culture-independent 16S rRNA gene clones, respectively. Bacterial isolates
were composed of 4 phyla, of which Firmicutes (49.8%) and Actinobacteria (32.0%) were predominant. Isolates were affiliated with 58 species. Culture-independent 16S rRNA gene clones were composed
of 8 phyla, of which Proteobacteria (62.2%), Actinobacteria (15.5%), and Bacteroidetes (13.7%) were predominant. Diversity of 16S rRNA gene clones originated from cultivation-independent DNA extracts was higher

Bacteria have been actively regulating cycles of various elements in the environment. To explore the potential bacterial role in gold biogeochemical cycling, this study analyzed the bacterial diversity of mine rock (MR) and surface soil (SS) samples from Linglong gold mine using 16S rRNA gene clone library analysis and cultivation method. From MR, 24 operational taxonomic units (OTUs) were identified from MR, covering 3 phyla and 18 genera. Meanwhile, 24 OTUs were identified from SS, including 4 phyla and 18 genera. Compared with 16S rRNA gene clone library analysis, 28 aerobic and 34 anaerobic isolates were obtained, whereas 26 aerobic and 71 anaerobic strains were isolated from SS. The cultivable bacteria were affiliated with Firmicutes, Proteobacteria and Actinobacteria phyla, and dominated by Firmicutes. These results underscore the high level of bacterial diversity in the gold mine. Our study provides information on the microbial diversity in Linglong gold mine and sheds light on the existence and potential function of bacteria in the gold biogeochemical cycling. 相似文献
