通过对5株蛹虫草的菌丝形态和生长速率、液体培养生物量和胞外多糖、人工栽培和子实体中活性物质虫草多糖和虫草素的对比研究,筛选出优良的蛹虫草菌株。试验结果表明:蛹虫草6号菌株的菌丝的生长速率比其他菌株快;液体培养生物量和胞外多糖含量明显高于其他菌株;人工栽培的蛹虫草子实体头部大,子囊壳丰富,颜色橘黄,出草整齐均匀,出草率高,子实体中虫草多糖和虫草素的含量均高于其他菌株,表明6号菌株具有较高的经济价值,是值得开发和推广的好品种。  相似文献

利用场强为0.1、0.25、0.4T恒定磁场,以流速为1 m·s-1,分别对普通水进行9次处理,串联(SC)磁场处理3次的磁处理水,进而用于栽培蛹虫草,并研究了生物磁效应对蛹虫草虫草素、虫草酸、多糖含量及生物学转化效率的影响。结果表明,0.4T处理有利于蛹虫草虫草酸含量及生物学效率的提高,相比对照组分别提高了7.29%和15.37%;0.25T处理组有利于虫草素的积累,比对照组提高了11.31%;0.1T处理有利于多糖的积累,相比对照组提高了14.80%。方差分析表明,经不同场强处理的磁化水对蛹虫草子实体生长和活性物的代谢影响差异显著。但磁处理水对于不同物质含量的影响规律存在一定的差异性。  相似文献

蛹虫草胞外多糖具有增强免疫力、抗疲劳等药理活性,有极高的保健价值。为高效地获取蛹虫草胞外多糖,本研究通过向发酵培养基中添加适量的扁桃斑鸠菊叶粉末,来提高蛹虫草发酵液中胞外多糖的产量,并对优化得到的胞外多糖红外吸收光谱和化学抗氧化活性进行了研究。实验结果表明,液体发酵最优条件为:扁桃斑鸠菊叶粉末添加量8 g/L、发酵时间9 d、pH 6.5、接种量5.0 mL,在此条件下,蛹虫草胞外多糖的产量可达(5.24±0.28) mg/mL,与未添加扁桃斑鸠菊叶的空白组相比,胞外多糖产量提高了约205.20%;红外分析与抗氧化活性实验结果显示,扁桃斑鸠菊叶对蛹虫草生产的胞外多糖结构和活性影响较小。该研究结果表明扁桃斑鸠菊叶能够有效地提高蛹虫草胞外多糖的产量,为蛹虫草胞外多糖的高效生产提供了新思路。  相似文献

本文探讨了蛹虫草多糖对急性酒精性肝损伤小鼠模型的影响,并对蛹虫草多糖相对分子质量和单糖组分进行了分析。以12.86m L/kg BW的剂量一次性灌胃50%乙醇诱导小鼠产生急性酒精性肝损伤模型,灌胃蛹虫草多糖,结果表明蛹虫草多糖可以显著降低急性酒精性肝损伤后小鼠肝匀浆中的过氧化脂质降解产物丙二醛(MDA)、甘油三酯(TG)的含量(P0.01);提高肝匀浆中的还原型谷胱甘肽(GSH)的含量(P0.01);脂肪变性程度均低于阳性对照组(P0.01)。采用凝胶过滤法测定蛹虫草多糖的相对分子质量为117k Da;高效气相色谱结果分析表明蛹虫草多糖是由鼠李糖、木糖、甘露糖、葡萄糖和半乳糖组成的,其组成比例分别为鼠李糖:木糖:甘露糖:葡萄糖:半乳糖=1:2.39:5.40:30.67:13.37。  相似文献

报道了蛹虫草(Gordyceps militaris)多糖发酵工程中的关键因子对菌丝体生物量和多糖产量影响的研究结果.对蛹虫草液体发酵培养基中的碳源、氮源和温度进行了正交试验,考察了三因子对蛹虫草生物量以及多糖的综合效应.实验结果表明:各因素不同水平间的组合对蛹虫草生物量、胞外多糖、胞内多糖以及总多糖的影响不同.其中.在6%的碳源和1%的氮源以及25℃的条件下.能获得蛹虫草的最大生物量以及总多糖和胞外多糖的最大产量;碳源为6%、氮源为1%、温度为22℃时.胞内多糖的产量达到最高.  相似文献

本研究以细脚棒束孢、蛹虫草、蝉棒束孢和球孢白僵菌的菌丝体粗多糖为对象,分析4种虫草相关真菌菌丝体粗多糖含量与生物量的相关性,并进一步对其抗氧化能力和抗肿瘤活力进行评价。液体发酵结果表明,蝉棒束孢MF12、MF13和蛹虫草MF27、MF1的菌丝体粗多糖含量(>40mg/g)显著高于其他菌株,蝉棒束孢MF11、MF13和蛹虫草MF27菌丝体生物量(>12g/L)显著高于其他菌株,但相关性分析表明,4种虫草相关真菌10个菌株菌丝体的多糖含量与生物量之间没有显著相关性;抗氧化活性表明,蛹虫草MF27、MF1和球孢白僵菌MF10菌丝体粗多糖具有良好的体外抗氧化活性,其EC50均小于0.9mg/mL;抗肿瘤活性表明,蛹虫草MF1、MF28、MF27和球孢白僵菌MF10菌丝体粗多糖在体外能有效抑制HepG-2细胞增殖,其IC50均小于1.5mg/mL。综上,蛹虫草MF27、MF1和球孢白僵菌MF10虫草菌株具有良好的开发和应用潜力。  相似文献

忍冬桑黄和蛹虫草两种药用真菌均可产活性多糖,共发酵模式是产生新化合物或提高化合物含量或药效的潜在方式。本研究尝试用忍冬桑黄和蛹虫草共发酵联产真菌多糖,并对其共发酵所得的菌丝体多糖开展抗氧化活性试验。结果表明,当忍冬桑黄和蛹虫草预先分别发酵3d和1d后再同时接种共发酵,两菌可以较好地进行共生长,菌体总量和总多糖得率显著高于两菌单独进行发酵时的相应量。进一步对适宜两菌共发酵的培养基进行了优化,获得适宜两菌共发酵高产多糖的培养基组成为:可溶性淀粉30g/L、牛肉粉12g/L、磷酸氢二钾和硫酸镁各1.5g/L。共发酵菌丝体中多糖和黄酮含量均高于两菌单个菌体中的相应含量,但三萜含量和桑黄菌体中的三萜含量没有显著差异。抗氧化活性试验表明,共发酵菌体多糖、忍冬桑黄菌体多糖和蛹虫草菌体多糖对DPPH·自由基、羟基自由基、超氧阴离子自由基均具有清除作用,其中共发酵菌体多糖对3种自由基的清除作用显著强于两菌单个菌体多糖的清除效果。本研究表明忍冬桑黄和蛹虫草共发酵联产真菌多糖具有可行性。  相似文献

柞蚕蛹虫草中含有虫草素、腺苷、多糖等多种活性成分,具有提高免疫力、抗疲劳、保护心脑血管、抗癌等方面的作用,是冬虫夏草的良好替代品。以柞蚕蛹虫草的继代培育为基础,分别检测蛹虫草菌在不同传代次数时柞蚕蛹虫草子实体生长状态、蛹虫草中腺苷、虫草素、虫草多糖含量及蛹虫草菌菌丝、分生孢子中活性氧的分布。第1代蛹虫草菌接种后柞蚕蛹出现腐烂现象;第2、3代培育的子实体生长量、腺苷及虫草素含量均较高;第4代子实体生长量、腺苷及虫草素含量均出现大幅下降的现象。柞蚕蛹虫草中虫草多糖含量在传代过程中也逐渐降低,但降低幅度较腺苷和虫草素缓慢。第2代培育的柞蚕蛹虫草子实体生长状态优于第3代,故第2代蛹虫草菌更适合应用于批量生产。蛹虫草菌退化后含有活性氧的分生孢子比例增大,这可能是发生退化的表象之一。  相似文献

对功能性蛹虫草、野生蛹虫草、柞蚕蛹虫草及冬虫夏草的代谢组学进行非靶向成分比较分析,并对功能性蛹虫草的常见生物活性成分进行定量分析。结果表明,功能性蛹虫草共检测出代谢产物成分2 213种,占成分总量的72.67%,其特有成分497种,与冬虫夏草、柞蚕蛹虫草和野生蛹虫草相比具有显著的成分优势。除目前已经报导的部分成分外,还含有丰富的特殊性代谢产物,如灵芝酸、茯苓新酸、人参皂苷、仙茅皂苷、苦玄参苷、黄芪皂苷、金铁锁环肽、王不留行环肽、甾醇、丹酚酸等。常规成分定量分析表明功能性蛹虫草子实体中虫草素和喷司他丁含量均较高,麦角甾醇、SOD酶、维生素E含量均显著高于对照组。大量药物活性物质在蛹虫草代谢产物成分体系中得到表达,对于揭示蛹虫草保健价值的物质基础具有重要意义。  相似文献

以蛹虫草为材料.采用超声波水提法、超声波醇提法、水热回流法和醇热回流法4种提取方法选择虫草素和虫草多糖的最优提取办法.并用正交试验方法考察水热回流法中提取温度、提取次数、提取时间、料液比4个因素对虫草素、虫草多精综合提取效率的影响.以确定从蛹虫草中综合提取虫草素、虫草多糖最佳工艺。结果表明.水热回流法是提取虫草多糖和虫草素的最优提取方法.其最佳工艺条件为:用10倍量的水于80℃热回流提取3次.每次90min。  相似文献

通过室内瓶栽试验对蛹虫草(Cordyceps militaris)9个菌株的主要子实体形成情况进行了分析,并采用DNS法和HPLC法测定了不同菌株胞内糖和虫草素含量。综合上述3个方面的试验结果,9个供试菌株中,8号菌株表现最优,子实体粗壮且生长整齐,虫草素及胞内糖含量显著高于其他菌株。  相似文献

采用定量检测和代谢组学方法对柞蚕蛹虫草的成分和参与的代谢途径及通路进行了分析,并开展了其对嘌呤代谢障碍引起的高尿酸血症作用研究。结果表明,柞蚕蛹虫草共含有代谢产物233种,其中不仅含有定量检测已经报道的虫草素、虫草酸等成分,还含有皮质固醇、肾上腺素及谷胱甘肽等多种特殊代谢成分,这些成分主要隶属于羧酸及衍生物、有机氧化合物及脂肪酰基类等10个不同类别,KEGG分析其显著性富集在ABC转运蛋白、氨基酸生物合成及嘌呤代谢等19种代谢途径;动物试验结果表明,柞蚕蛹虫草中、高剂量组与模型组比较,血尿酸水平和黄嘌呤氧化酶活性均显著降低(P<0.05),对防治高尿酸血症具有明显的作用,并对其作用机制进行了分析探讨。本研究为柞蚕蛹虫草在高尿酸血症作用及其相关药理学研究和开发应用提供了理论依据。  相似文献

Proteomic analyses of fruits are confronted with a series of specific obstacles: a general low protein content in plant tissues, allergen extraction from highly complex matrices and protein determination in the presence of interfering compounds. Different methods are currently being introduced to achieve higher protein yields and a simultaneous removal of interfering substances, such as polyphenols and polysaccharides. However, no universal protocol suitable for protein purification from any given plant species is available. Protein profiling by 2DE-western blotting offers a powerful tool for the detection and characterization of known and novel plant allergens. Moreover, the detection of IgE-reactive proteins from fruits is improved by combining western blot and alternative visualization techniques. The recent developments in bioinformatics and databases facilitate the interpretation of profiling studies with regard to novel potential fruit allergens.  相似文献

Cordyceps militaris SU5-08 was derived from an initial strain (C. militaris SU5) by ultraviolet mutagenesis of protoplasts, and the extraction parameters for C. militaris SU5-08 exopolysaccharide (EPS) produced during submerged culture were optimized. The extraction rate of EPS was 1919.16±165.27 mg/l, which was 120.38±11.36% higher than that of C. militaris SU5. The in vitro scavenging effects of EPS of C. militaris SU5-08 on hydroxyl, superoxide anion and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals at a dosage of 5 g/l were 63.64±3.52%, 75.27±5.16%, and 6.46±5.03%, respectively. The reducing power of EPS of C. militaris SU5-08 was 0.21±0.01. The results suggest that the EPS of C. militaris SU5-08 can be used as a potential antioxidant which enhances adaptive immune responses.  相似文献

The active components of most meningococcal vaccines are four antigenic serogroup capsular polysaccharides (A, C, Y, W135). The vaccines, monovalent or multivalent mixtures of either free polysaccharides or polysaccharides conjugated to antigenic carrier proteins, may be in liquid or lyophilised formulations, with or without excipients. Acid hydrolysis and chromatographic methods for serogroup quantitation, which were previously optimised and qualified using polysaccharide-based standards and a narrow range of real vaccines, are here challenged with multiple lots of a broad assortment of additional multivalent polysaccharide-based meningococcal vaccine products. Centrifugal filtration successfully removed all interfering lactose excipient without loss of polysaccharides to allow for the determination of Y and W135 serogroups. Replicate operations by three different analysts indicated high method reproducibility. Results indicated some lot-to-lot and product-to-product variations. However, all vaccines were within general specifications for each serogroup polysaccharide, with the exception of all lots of one polysaccharide vaccine – which by these methods were found to be deficient in the serogroup A component only. These robust techniques are very useful for the evaluation of antigen content and consistency of manufacture. The deformulation, hydrolysis and chromatographic methods may be adaptable for the evaluation of other types of polysaccharide-based vaccines.  相似文献

光作为一种重要的环境因子,可影响真菌的生长发育、生理周期、形态变化及次级代谢产物的产生,对于蛹虫草而言,光照还是其子实体生长发育的必要条件。选择2株不同来源的蛹虫草菌株,研究了自然光照和蓝光光照条件下,其子实体生长、抗氧化酶活性和主要活性成分的变化。结果表明蓝光光照对于蛹虫草子实体产量没有明显的促进和抑制作用。自然光照条件下成熟子实体的超氧化物歧化酶(superoxide dismutase,SOD)活性显著高于蓝光光照,过氧化氢酶(catalase,CAT)活性没有明显差别,而过氧化物酶(peroxidase,POD)活性则表现出菌株差异。子实体主要活性成分方面,腺苷、甘露醇含量不受蓝光影响,类胡萝卜素在蓝光光照条件下含量极显著高于自然光光照,蓝光对子实体、虫草素、粗多糖含量的影响则存在菌株差异。研究为蛹虫草子实体的栽培条件优化,提高蛹虫草子实体质量提供依据。  相似文献

本文从研究铜绿微囊藻表型转换机制的需要出发,综述了非生物因素对藻类胞外多聚糖含量的影响.很多藻类的胞外多聚糖分泌量与环境中主要营养盐比例之间存在着一定的响应关系,在高碳氮比或高碳磷比条件下,即在氮不足或磷不足时,藻类光合作用固定的有机物质主要以不含氮磷的碳水化合物形式存在,胞内碳水化合物的过量累积导致其逐步向胞外转移释放,使得胞外多聚糖含量显著升高.在碳氮代谢不平衡的情况下,胞外多聚糖充当了接收过剩固定碳的汇.对于一些藻类,不同光谱、光强和光周期均可影响其胞外多聚糖的合成与分泌.温度对藻类胞外多聚糖的产生也有一定影响.由于胞外多聚糖在藻细胞相互粘结形成群体上发挥着重要作用,通过调控相关非生物因素使得铜绿微囊藻胞外多聚糖产量增加可能有助于在室内模拟铜绿微囊藻的群体形成.  相似文献

In this study, biodegradation of natural organic matter (NOM) in a biological aerated filter (BAF) as pretreatment of UF treating river water was investigated. Photometric measurement, three-dimensional excitation–emission matrix (EEM) fluorescence spectroscopy and liquid chromatography with online organic carbon detector (LC-OCD) were used to investigate the fate of NOM fractions in the BAF + UF process. Results showed that the BAF process could effectively remove particles and parts of dissolved organic matter, which led to a lower NOM loading in the UF system, but different NOM fractions showed different biodegradation potentials. Further biodegradation batch experiments confirmed this observation and identified that polysaccharides and proteins (quantified using photometric methods) contained a large proportion of readily biodegradable matter while humic substances were mainly composed of inert organic substances. According to EEM measurements, it is evident that protein-like substances were more readily eliminated by microorganisms than humic-like substances. LC-OCD data also supported the phenomena that the polysaccharides and large-size proteins were more degradable than humic substances.  相似文献
