Chromosome measurements were carried out in Hordeum vulgare, H. bulbosum, and their diploid, triploid, and tetraploid hybrids. The chromosomes were classified by using relative values, and thus karyotypes were established. For comparison of these karyotypes both relative and absolute values were used. It was concluded that differential amphiplasty occurred, whereas neutral amphiplasty could not be demonstrated. In the hybrids the relative length of the parts of the chromosomes (long arm, short arm, satellite) was not changed in comparison with these lengths in the pure species. The karyotypes of both species had considerable similarities. From comparing the mean absolute genome lengths, it was, however, concluded that in the pure species, as well as in all hybrid types, the chromosomes of H. vulgare were longer than those of H. bulbosum. In the diploid and tetraploid hybrids the mean genome lengths were shorter than those in the pure species and the triploid hybrids. The differential amphiplasty was such that the secondary constriction of chromosome 6 of H. bulbosum, did not show up in the hybrids. This could be related to the suppression of nucleolar formation in the genome of H. bulbosum, because the maximum number of nucleoli in root tip cells equalled the number of satellite chromosomes. Finally it was found that the pattern of nucleolar fusion in diploid and triploid hybrids deviated from the expectation. The results were discussed in relation to chromosomal disturbances that occurred in the hybrid tissues and that resulted in elimination of chromosomes and other effects.  相似文献

Interrelationships between H. vulgare (2x=14) and H. bulbosum (2x=14; 4x=28) were estimated on the basis of the karyotypes and the pairing behaviour of the chromosomes in diploid, triploid and tetraploid hybrids obtained with the aid of embryo culture. — A comparison of the karyotypes of the two species revealed similarities as well as differences. It was concluded that at least 4 or more of the chromosomes were similar in morphology and probably closely related. — Diploid and tetraploid hybrids are rarely obtained and their chromosome numbers tend to be unstable whereas triploid hybrids (1 vulgare + 2 bulbosum genomes) were stable and relatively easy to produce. In the diploid hybrid only 40% of the meiotic cells contained 14 chromosomes while the numbers ranged from 7 to 16 in other cells. All hybrids exhibited pairing between the chromosomes of the two species. Diploid hybrids had a mean of 5.0 and a maximum of 7 bivalents per cell in those cells having 14 chromosomes. Triploid hybrids from crosses between 2x H. vulgare and 4x H. bulbosum exhibited a mean of 1.5 and a maximum of 5 trivalents per cell. In a hexaploid sector found following colchicine treatment of a triploid the mean frequencies of chromosome associations per cell were: 5.5I+8.0II+0.7III+3.7IV+0.3V+0.4VI. One unstable 27 chromosome hybrid obtained from crosses between the autotetraploid forms had a mean of 1.1 and a maximum of 4 quadrivalents per cell. The chromosome associations observed in these hybrids are consistent and are taken as evidence of homoeologous pairing between the chromosomes of the two species. Interspecific hybridization between these two species also reveals that chromosome stable hybrids are only obtained when the genomes are present in a ratio of 1 vulgare2 bulbosum. Based upon the results obtained, the possibility of transferring genetic characters from H. bulbosum into cultivated barley is discussed.  相似文献

The 140+ species of Echeveria have more than 50 gametic chromosome numbers, including every number from 12 through 34 and polyploids to n = ca. 260. With related genera, they comprise an immense comparium of 200+ species that have been interconnected in cultivation by hybrids. Some species with as many as 34 gametic chromosomes include none that can pair with each other, indicating that they are effectively diploid, but other species with fewer chromosomes test as tetraploids. Most diploid hybrids form multivalents, indicating that many translocations have rearranged segments of the chromosomes. Small, nonessential chromosomal remnants can be lost, lowering the number and suggesting that higher diploid numbers (n = 30–34) in the long dysploid series are older. These same numbers are basic to most other genera in the comparium (Pachyphytum, Graptopetalum, Sedum section Pachysedum), and many diploid intergeneric hybrids show very substantial chromosome pairing. Most polyploid hybrids here are fertile, even where the parents belong to different genera and have very different chromosome numbers. This seems possible only if corresponding chromosomes from a polyploid parent pair with each other preferentially, strong evidence for autopolyploidy. High diploid numbers here may represent old polyploids that have become diploidized by loss, mutation, or suppression of duplicate genes, but other evidence for this is lacking. Most species occur as small populations in unstable habitats in an area with a history of many rapid climatic and geological changes, presenting a model for rapid evolution.  相似文献

Sedum cremnophila and Echeveria linguifolia have generally been placed in different genera on the basis of their flowers—largely because the petals are spreading in one and erect in the other—and the genera have been placed in different subfamilies. However, they are very similar vegetatively and in their unusual inflorescence, their karyotypes are similar (n = 33), and they readily hybridize to produce fertile F1 hybrids. Study of hybrids of these two species with numerous others leads to the conclusion that each of the two is effectively diploid, with a genome consisting of 33 chromosomes that are all different and that do not pair with each other. Therefore, the good chromosome pairing and the fertility of the hybrid between them are the result of close structural and genetic homology between the corresponding chromosomes of the two species. Taxonomic revision to reflect their very close relationship is desirable. Some other species of Sedum and Echeveria also may need to be reclassified.  相似文献

Summary The chromosomes of the tetraploid wheats Triticum timopheevi (Genome AAGG) and T. araraticum (Genome AAGG) were C-banded at mitosis. The identity of the banded and unbanded chromosomes was then established by firstly making comparisons with the hexaploid species T. zhukovskyi which has the genome formula AAAAGG. Secondly, the meiotic pairing in F1 hybrids between T. timopheevi and diploid wheats was examined by means of C-banding. The results showed that the banded chromosomes belonged to the G genome, while the unbanded chromosomes belonged to the A genome. Only one of the two pairs of satellited chromosomes had strong heterochromatic bands. The relationship between the genomes of T. timopheevi and T. dicoccum (Genome AABB) was then assessed at meiosis in hybrids between these species, using the techniques of C-banding and in situ hybridisation of a cloned ribosomal RNA gene probe. It was concluded that there were differences both in the amount and distribution of heterochromatin and also translocation differences between the species.  相似文献

The karyotypes of four gerreids of the western Atlantic Ocean are documented. A diploid chromosome complement of 48 telocentric chromosomes was found in the four species (2N=48t, fundamental number FN=48). No differences were detected either in the number of chromosomes of the standard karyotype, in their karyotype size, or between the karyotypes derived from male or female specimens of any of the species. Chromosome length decreased progressively and slightly from pair 1 to pair 24. The Ag–NOR karyotypes of E. argenteus and E. harengulus were characterized by the position of the nucleolar organizer regions next to the centromere in chromosome pair 1, whereas in E. gula and E. plumieri Ag–NORs were detected in pair 4. The other 46 chromosomes showed a light staining of the centromere with no terminal or intermediate heterochromatic blocks. All Eucinostomus species showed Ag–NORs of similar size, while Eugerres plumieri showed Ag–NORs 10–20% larger than Eucinostomus species. A combination of size and position of the Ag–NORs identified E. gula, while size alone identified E. plumieri. However, the ancestral state for size and position of Ag–NORs could not be established. There was no differential staining of the chromosomes by G-banding. The karyotype of the gerreids appears similar to the hypothetical ancestral karyotype of fish. The phylogenetic relationships among these species could not be established because of the lack of chromosome G-bands. Most likely this indicates a homogeneous distribution of GC nucleotides in the chromosomes.  相似文献

This study was conducted to describe the major and the minor rDNA chromosome distribution in the spined loach Cobitis taenia (2n = 48) and the Danubian loach Cobitis elongatoides (2n = 50), and their laboratory-produced diploid reciprocal F1 hybrid progeny. It was tested by fluorescence in situ hybridisation (FISH) whether the number of 28s and 5s rDNA sites in the karyotypes of diploid hybrids corresponds to the expectations resulting from Mendelian ratio and if nucleolar organiser regions (NOR)were inherited from both parents or nucleolar dominance can be observed in the induced F1 hybrid progeny. Ten (females) or twelve (males) 28s rDNA loci were located in nine uniarm chromosomes of C. taenia. Two of such loci terminally bounded on one acrocentric chromosome were unique and indicated as specific for this species. Large 5s rDNA clusters were located on two acrocentric chromosomes. In C. elongatoides of both sexes, six NOR sites in terminal regions on six meta-submetacentric chromosomes and two 5s rDNA sites on large submetacentrics were detected. The F1 hybrid progeny (2n = 49) was characterised by the intermediate karyotype with the sites of ribosome synthesis on chromosomes inherited from both parents without showing nucleolar dominance. 5s rDNA sites were detected on large submetacentric and two acrocentric chromosomes. The observed number of both 28s and 5s rDNAs signals in F1 diploid Cobitis hybrids was disproportionally inherited from the two parental species, showing inconsistency with the Mendelian ratios. The presented rDNA patterns indicate some marker chromosomes that allow the species of the parental male and female to be recognised in hybrid progeny. The 5s rDNA was found to be a particularly effective diagnostic marker of C. elongatoides to partially discern genomic composition of diploid Cobitis hybrids and presumably allopolyploids resulting from their backcrossing with one of the parental species. Thus, the current study provides insight into the extent of rDNA heredity in Cobitis chromosomes and their cytotaxonomic character.  相似文献

The interspecific hybrids of Hordeum exhibit selective suppression of secondary constriction formation in the chromosome(s) contributed by one of the two parents. A comparison of the number of SAT (secondary constriction) chromosomes in the metaphase cells and the maximum number of nucleoli in interphase cells revealed that the chromosomes capable of organising nucleoli were not always reflected through secondary constriction formation. — The rDNA (DNA complementary to rRNA) amounts were estimated by DNA-rRNA filter hybridisation in diploid and polyploid species of Hordeum and their hybrids. While similar rDNA proportions were present in diploid and autotetraploid lines of H. bulbosum, there were up to threefold differences between H. vulgare and allohexaploids. Furthermore, differences were also apparent between species of same ploidy level. Ribosomal RNA (18S+5.8S+26S) cistron numbers in each of the five experimental hybrids exhibiting the selective suppression of secondary constriction formation revealed no selective loss of rDNA. — The presence of a higher number of nucleoli than the number of SAT chromosomes seen and the presence of expected number of rRNA cistrons suggest that the suppression of secondary constriction formation is not due to selective loss of rDNA.  相似文献

A cytogenetic analysis was performed in experimental hybrids between species of Chagas disease transmitting bugs with remarkable differences in the amount and distribution of heterochromatin. Using C-banding technique, we identified the parental species chromosomes and analysed the meiotic behaviour in the male hybrids between Triatoma platensis and T. infestans, T. platensis and T. delpontei, and T. infestans and T. rubrovaria. The two former hybrids have an entirely normal meiotic behaviour despite the extensive differences in C-banded karyotypes observed in the parental species, indicating that heterochromatin differences between homeologous chromosomes are not a barrier that influences meiotic synapsis and recombination. On the contrary, the experimental hybrids between T. infestans and T. rubrovaria show failures in pairing of homeologous chromosomes that lead to the production of abnormal spermatids and hybrid sterility. Our data suggest that karyotypic repatterning within triatomines has involved at least two different pathways. Among closely related species, chromosomal changes have largely involved addition or deletion of heterochromatic regions. In more distant species, chromosomal rearrangements (i.e. inversions and translocations) have also arisen. Hybridisation data also allow to hypothesize about the origin and divergence of this taxonomic group, as well as the mechanisms that maintain species isolation.  相似文献

Arachis batizocoi Krap. & Greg. is a suggested B genome donor to the cultivated peanut,A. hypogaea L. Until recently, only one accession of this species was available in U.S.A. germplasm collections for analyses and species variability had not been documented. The objective of this study was to determine the intraspecific variability ofA. batizocoi to better understand phylogenetic relationships in sect.Arachis. Five accessions of the species were used for morphological and cytological studies and then F1 intraspecific hybrids analyzed. Some variation was observed among accessions—for example, differences in seed size, plant height and branch length. The somatic chromosomes of accessions 9484, 30079, and 30082 were nearly identical, whereas, the karyotypes of accessions 30081 and 30097 have several distinct differences. For example, 30081 had significantly more asymmetrical chromosomes 2 and 6 and more median chromosomes 7 and 10, and 30097 had significantly more asymmetrical chromosomes 3 and 10 and more median chromosomes 1 and 5 than accessions 9484, 30079, and 30082. All F1 hybrids among accessions were highly fertile. Meiotic observations indicated that hybrids among accessions 9484, 30079, or 30082 had mostly bivalents. However, quadrivalents were observed when either 30081 or 30097 was crossed with the above three accessions and 30081 × 30097 had quadrivalents, hexavalents and octavalents. The presence of translocations is the most likely cause of multivalent formation inA. batizocoi hybrids. Summary Electrofusion was carried out between mesophyll protoplasts from the transformed diploid S. tuberosum clone 413 (2n=2x=24) which contains various genetic markers (hormone autotrophy, opine synthesis, kanamycin resistance, -glucuronidase activity) and mesophyll protoplasts of a diploid wild-type clone of N. plumbaginifolia (2n=2x=20). Hybrid calli were obtained after continuous culture on selection medium containing kanamycin. Parental chromosome numbers, determined at 2 months after fusion, revealed hybrid-specific differences between the individual calli. On the basis of these differences three categories of hybrids were distinguished. Category I hybrids contained between 8 and 24 potato chromosomes and more than 20 N. plumbaginifolia chromosomes; category II hybrids had between 1 and 20 N. plumbaginifolia chromosomes and more than 24 potato chromosomes; category III hybrids contained diploid or subdiploid numbers of chromosomes from both parents. The hybrids were evenly distributed over the three categories. After a 1-year culture of 24 representative hybrid callus lines on selection medium the karyotype of 10 hybrids remained stable, whereas 8 hybrids showed polyploidization of the genome of one parent, together with no or minor changes of the chromosome numbers of the other parent. Six hybrids showed slight changes in the hybrid karyotype. The elimination of chromosomes of a particular parent was not correlated to their metaphase location. The processes of spontaneous biparental chromosome elimination leading to the production of asymmetric hybrids of different categories are discussed.  相似文献

Although the Dendranthema zawadskii complex has been known to comprise a series of polyploids (4×, 6×, 8×), we found diploid individuals (with 2n=18) to occur in four populations of D. zawadskii var. latilobum in the southern region of Korea. Karyotypes of metaphase chromosomes were diverse because numbers of metacentric, submetacentric, and acrocentric chromosomes differ even within a population. A total of 17 karyotypes were found in 31 diploid individuals collected from the four populations. The karyotypes were also diverse in the presence or absence of chromosomes with a secondary constriction on a short or long arm and, if present, in the number of such chromosomes. They were further diverse in the presence or absence of non-homologous chromosome(s), the presence or absence of a chromosome with a satellite, and, if present, how many and where satellites are present. Almost the same pattern of diversity was found in diploid individuals (with 2n=18) of D. boreale and D. indicum as well, irrespective of whether they occur together with D. zawadskii var. latilobum or not. Structural features of chromosomes in the variously different karyotypes suggest that reciprocal translocation and the hybridization between individuals with different karyotypes had repeatedly occurred not only in D. zawadskii var. latilobum, but also in D. boreale and D. indicum. Following fusion of human diploid fibroblast-derived microcells with mouse A9 cells, we isolated seven monochromosomal hybrids containing a single human chromosome 2, 3, 5, 12, 15, 20, or 22. Cytogenetic analysis as well as PCR karyotyping (chromosome-specific banding pattern generated by Alu-PCR) was performed on all the hybrids. Karyotypes and meiotic configurations of Anthurium andraeanum and closely related taxa were analyzed. The karyotypes of A. andraeanum, A. caperatum, A. formosum, A. kamemotoanum, A. lindenianum, A. roseospadix, A. cf. sanctifidense, A. subsignatum, A. garagaranum, and an unidentified Anthurium sp. commonly consisted of four large metacentric or submetacentric chromosomes, two fairly large acrocentric chromosomes, two satellite chromosomes, and 22 smaller chromosomes. Variation in the karyotypes of A. nymphaeifolium and A. ochranthum suggested chromosomal rearrangement in the genus. All taxa showed 15 pairs of chromosomes at prometaphase I of meiosis in pollen mother cells. Four large chromosomes appeared as ring bivalents, and the rest of the chromosomes appeared as either ring or rod bivalents. Regular bivalent formation at prometaphase I of meiosis in pollen mother cells of species hybrids suggested close genomic relationships among parental taxa. On the other hand, reduction of pollen fertility estimated by pollen stainability in those hybrids suggested genetic divergence of species.  相似文献

Menzel , Margaret Y. (Florida State U., Tallahassee), and James B. Pate . Chromosomes and crossing behavior of some species of Sansevieria. Amer. Jour. Bot. 47(3) : 230—238. Illus. 1960.–Approximately 20 species (28 clones) studied were diploids, tetraploids or hexaploids of the basic numbers x = 20; about 40% of the taxa were polyploid. All species had similar karyotypes, except for chromosome number. Five of 12 combinations of diploid species gave fertile F1 hybrids; 4 studied cytologically showed 20 bivalents at metaphase I. Two triploid interspecific hybrids showed high trivalent frequencies. In contrast, multivalent formation in polyploid species was variable but rather low. Morphological relationships appeared reticulate among and between diploids and polyploids and did not coincide with barriers to crossing or to hybrid fertility. The following tentative hypothesis concerning relationships in the genus is proposed: Sansevieria is monophyletic and speciation has proceeded through genetic variation and hybridization at the diploid level and by allopolyploidy (of the segmental type) ; a low level of chromosome differentiation has accompanied speciation such that complete pairing occurs in diploid hybrids, but considerable preferential pairing occurs in allopolyploids. The occurrence of both polyploid and hybrid vigor, the fertility of hybrids between species differing greatly in morphology and physiology, and the high potential for vegetative propagation make the genus a favorable subject for breeding based on interspecific hybridization.  相似文献

Considerable karyotypic differentiation has occurred within the group of taxa comprising the eastern North American members of the genus Claytonia. Patterns of karyotypic differentiation are congruent with evolutionary groupings based on flavonoid chemistry, particularly at the diploid level. The 2n = 16 diploid chemotype found in both C. caroliniana and C. virginica possesses a karyotype composed entirely of metacentric chromosomes, while acrocentric chromosomes predominate in the karyotypes of the 2n = 12 and 2n = 14 diploid chemotypes of C. virginica. The 2n = 16 diploid also has a karyotype significantly larger than those of the other diploid cytotypes. Polyploid karyotypes of both species show varying degrees of divergence from their presumed diploid progenitors.  相似文献

Summary Meiotic chromosome behavior was studied in seven diploid species of Lotus (L. alpinus Schleich., L. japonicus (Regel) Larsen, L. filicaulis Dur., L. schoelleri Schweinf., L. krylovii Schischk. and Serg., L. tenuis Waldst. et Kit., L. corniculatus var. minor Baker) and in 51 interspecific hybrids from 16 different crosses. Meiosis in the diploid species was quite regular. In a high proportion of the PMC's of the hybrids there was close chromosome homology with a normal association of 6 II's. However, meiotic irregularities including bridges, lagging chromosomes, univalents, and quadrivalents, occurred in a small percentage of the cells. The late separation of bivalents, the presence of quadrivalents, and inversion bridges with fragments, would indicate for some hybrids that certain chromosomes were structurally differentiated. The large number of rod bivalents observed at diakinesis was also highly suggestive that genetic nonhomology in one chromosome arm could contribute to the frequency of this type of bivalent. Therefore, the maximum number of 6 II's which occurred in a high percentage of cells may be misleading in that cryptic structural differences between chromosome arms, or segments, are not revealed. Pollen fertility in the species and hybrids was not correlated with meiotic irregularities suggesting that pollen fertility is genotypically controlled.  相似文献

The relative amounts of nuclear DNA of root meristematic cells of two related diploid Avena species, A. strigosa 2x and A. pilosa, which have different karyotypes, and an autotetraploid of one, A. strigosa 4x, were measured by Feulgen microspectrophotometry. The durations of various periods of their mitotic cycles were studied by autoradiography of cells pulse-labeled with tritiated thymidine. The results show that the autotetraploid, with twice the amount of nuclear DNA of its diploid, has the same duration of S period as the diploid, while A. pilosa, with intermediate nuclear DNA content, has a longer S period. These results support the hypothesis that homologous chromosomes or genomes require similar duration for their DNA synthesis and suggest that the structures of chromosomes are involved in temporal control of the DNA synthesis in cells.  相似文献

Both diploid and tetraploid experimental interspecific hybrids betweenRanunculus silerifolius (2x) andR. chinensis (2x) exhibit normal bivalent pairing. However, microspores of diploid hybrids do not undergo mitosis and their pollen grains are highly sterile, whereas tetraploid hybrids form good pollen grains after microspore division. Evidence is forwarded for the assumption thatR. cantoniensis (4x) has originated by hybridization between these two diploid parental species and by polyploidization of the diploid hybrids. Parallelisms between the different karyotypes ofR. cantoniensis (4x) andR. silerifolius (2x) suggest that the former is a species of polyphyletic origin.  相似文献

Karyotypes of 93 individuals belonging to 18 accessions of mandarins, mandarin hybrids and two related species were analysed with the fluorochromes CMA and DAPI, to identify marker chromosomes. The karyotypes revealed highly differentiated banding patterns and could be classified in four groups (I–IV) according to the presence/absence of chromosomes with three bands (type A) or with two bands (one proximal and one terminal, type B, or both terminal, type C). The accessions of group I exhibited the simplest and homozygous karyotypes (lacking chromosome types A, B and C), represented by `Sunki' and `Cleopatra'. Group II (lacking chromosome types A and B) included three accessions of Mediterranean mandarins and `Cravo' mandarin, all of them with very similar and almost completely homozygous karyotypes. All other karyotypes of groups II and III (lacking chromosome type A) were heterozygous for one or more chromosome pairs and most of them seemed to be hybrid derivatives from non-mandarin accessions. Group IV (with chromosome types A and B) was represented only by two heterozygous hybrids (`Murcott' and `King'). The karyotype of most hybrids agrees with one of the possible combinations resulting from chromosome types segregation from their putative ancestor karyotypes, but at least `Orlando' seemed to be a more complex hybrid. Comparing with banding patterns of other Citrus species, those of group I and the Mediterranean mandarins (group II) are the best candidates to represent C. reticulata (sensu Swingle) as a true species.  相似文献
