An enzyme assay was developed to measure the initial and Mg2+–CO2activated forms of Ribulose 1,5-bisphosphate Carboxylase/Oxygenase(Rubisco) in rose leaves. The assay was verified by co-extractionof the leaflets with partially purified spinach Rubisco andthrough correlation with net photosynthetic rates of individualleaflets (r2=0.7324). Changes in activities were measured asa function of depth of leaves in the canopy for two cultivarsof greenhouse hybrid tea roses. Initial Rubisco activity declinedwith increasing canopy depth for both cultivars. The activatedform of the enzyme, however, remained constant with canopy depthfor cv. Red Success; but increased with canopy depth, then declinedafter mid-canopy in the cv. Royalty. Rubisco activities werealso measured in the cv. Red Success grown in CO2 enriched environments(100 mm3 dm–3) at three humidity levels. The activitieswere not significantly affected by humidity treatment. In this paper we report for the first time the occurrence ofan inducible weak CAM in leaves of Talinwn triangulare (Jacq.)Willd. This plant is a terrestrial perennial deciduous herbwith woody stems and succulent leaves which grows under fullexposure and in the shade in northern Venezuela. Plants grownin a greenhouse (‘sun’ plants) and a growth cabinet(‘shade’ plants) with daily irrigation showed CO2uptake only during the daytime (maximum rate, 4?0 µmolm–2 s–1) and a small acid accumulation during thenight (6?0 µmol H+g–1 FW). Twenty-four hours aftercessation of irrigation, no CO2 exchange was observed duringpart of the night. Dark fixation reached a maximum (1?0 µmolCO2 m–2 s–1, 100 µmol H+ g–1 FW) onday 9 of drought. By day 30 almost no gas exchange was observed,while acid accumulation was still 10 µmol H+ g–1FW. Rewatering reverted the pattern of CO2 exchange to thatof a C3 plant within 24 h. Daytime and night-time phosphoenolpyruvatecarboxylase activity increased up to 100% (shade) and 62% (sun)of control values after 10 and 15 d of drought, respectively.Light compensation point and saturating irradiance were similarin well-watered sun and shade plants, values being characteristicof sun plants. CAM seems to be important for the tolerance ofplants of this species to moderately prolonged (up to 2 months)periods of drought in conditions of full exposure as well asshade, and also for regaining high photosynthetic rates shortlyafter irrigation. The effect of the ‘leafless’ mutation (in whichtendrils replace leaflets and the stipules are reduced to avestigial form) upon foliage area, photosynthetic net CO2 uptakepotential, dry matter production and seed yield in Pisum sativumwas studied by comparing two near-isogenic lines of genotypeafafstst and ++++. The mutation is of potential agronomic valuein that it offers improved lodging resistance, crop drying andharvester throughput. In the conventional phenotype the total foliage area of themain axis attained a plateau (456 cm2) at day 56 from seedlingemergence, whereas corresponding values for the ‘leafless’mutant showed a total area of 208 cm2 at day 68 with no indicationof a plateau. The agronomic consequence of this is discussed.During the vegetative phase of the plant the maximum CO2 uptakepotential in the fully expanded conventional leaf was 8·5mg CO2 leaf–1 h–1 and in the ‘leafless’mutant this value was 7·0 mg CO2 leaf–1 h–1.For most ‘leaves’ of the latter phenotype this valuewas between 30 and 60 per cent less than for their conventionalcounterpart. There was a consistently higher photosyntheticpotential per unit area in tendrils of the ‘leafless’mutant than in leaflets of the conventional phenotype. The respectivemean specific values for the two phenotypes were 53 and 37 mgCO2 dm–2 h–1. The problem of obtaining a meaningfulsurface area value for tendrils is discussed and the cylindricalnature of tendrils is taken into account. The ‘leafless’ mutant consistently accumulated 50per cent less dry matter than did conventional plants in theperiod from seedling emergence to anthesis and yield of maturedry seed per plant showed a reduction of 50 per cent both inseed number and total seed weight. The implications for future breeding and selection programmesaimed at haulm reduction are discussed in relation to evaluatingthe ability of the background genotype to produce adequate tendrilsin the presence of afafstst. Physiological responses to water stress (drought) have beeninvestigated in Umbilicus rupestris (wall pennywort) by comparingcontrol (well-watered) and draughted plants with respect to(i) diurnal fluctuations in the acid content of the leaves,(ii) CO2 exchange patterns and (iii) stomatal conductance. Controlplants show no diurnal fluctuations in acid content, whereasafter 6 d of drought a clear CAM-type pattern (nocturnal acidificationfollowed by deacidification in the light) is observed. In controlplants, the CO2 exchange pattern over a 24 h period is of atypical C-3 ‘square-wave’ type, with extensive CO2uptake in the light and CO2 output in the dark. In droughtedplants the day-time CO2 uptake is confined to a morning ‘burst’,whilst night-time CO2 output is markedly reduced. There is howeverno net noctural uptake of CO2. In control plants, stomatal conductanceis high during the day (especially in the first half of theday) falling to a low level at the onset of darkness, and thenrising slowly through the remainder of the night. In droughtedplants, stomatal conductance is very low, except that thereis morning ‘burst’ of high conductance and a periodduring the night when conductance is higher than in controlplants. These results are discussed in relation to the response of U.rupestris to drought both in laboratory and in field conditions. The levels of purine and pyrimidine nucleotides in suspensioncultures of Catharanthus roseus were determined 24 h after stationary-phasecells were transferred to fresh complete (‘+Pi’)or phosphate-deficient (‘–Pi’) Murashige-Skoogmedium. The levels of ATP, GTP, UTP and CTP were from approx.3 to 5-fold greater in the cells grown in ‘+Pi’medium than in the cells grown in ‘–Pi’ medium.The levels of almost all other nucleotides were slightly higherin the cells in ‘+Pi’ medium. The rates of de novoand salvage biosynthesis of purine and pyrimidine nucleotideswere estimated from the rates of incorporation of radioactivityfrom [14C]formate, [2–14C]glycine, NaH14CO3, [6–14C]orotate,[8–14C]adenine, [8–14C]adenosine, [2–14C]uraciland [2–14C]uridine. The results indicated that the activityof both the de novo and the salvage pathway was higher in thecells in ‘+Pi’ medium than in the cells in ‘–Pi’medium. The rate of degradation estimated from the rate of releaseof 14CO2 from labelled purines and pyrimidines indicated thatdegradation of uridine was significantly reduced in the cellsin ‘+Pi’ medium, but no significant difference wasfound in the degradation of adenine, adenosine and uracil. Thepossible role of Pi in the control of the biosynthesis of nucleotidesand in the degradation of uridine is discussed. Trends in several photosynthetic parameters and their responseto changed growth light were followed for 15 d in leaves ofyoung birch saplings using a rapid-response gas exchange measuringequipment. These in vivo measurements were compared to biochemicalassays that were made from the same leaves after the gas exchangestudies. The measurements were made on leaves that were selectedprior to the study and were at that time of similar age. Forthe first 7 d the photosynthetic parameters were followed fromthe growth conditions of moderate light (200 µmol m–2s–1; referred to as controls later in the text). On day7 some of the saplings were transferred to grow either underhigh (450 µmol m–2 s–1; referred to as highlight plants) or low (75 µmol m–2 s–1; referredto as low light plants) light and the capability of the preselectedleaves for acclimation was followed for 6 d. For comparison,at the end of the experiment the measurements were made on bothcontrols and on young leaves that had developed under high andlow light. Generally the in vivo measured rate of CO2 uptake (gross photosynthesis)both at 310 ppm CO2 and 2000 ppm CO2 corresponded very wellto the biochemically determined CO2 fixation capacity in vitroafter rapid extraction (measured as the initial and total activityof Rubisco, respectively). However, if the flux of CO2 intothe chloroplasts was limited by the closure of the stomata,as was the case of the high light plants, then the in vitromeasured Rubisco activity was greater than the in vivo measuredCO2 uptake. Vmax, calculated from the mesophyll conductanceat 1% O2, exceeded the initial activity of Rubisco (assayedat saturating RuBP and CO2) constantly by 60%. The catalyticactivity of Rubisco in birch leaves was overall very low, evenwhen calculated from the total activity of Rubisco (Kcat 0.63–1.18 s–1), when compared to herbaceous C3 species. Signs of light acclimation were not observed in most of thephotosynthetic parameters and in chloroplast structure whenmature birch leaves were subjected to changes in growth lightfor 6 d. However, the change of the growth light either to highor low light caused day-to-day fluctuations in most of the measuredphotosynthetic parameters and in the case of the high lightplants signs of photoinhibition and photodestruction were alsoobserved (decrease in the amount of chlorophyll and increasein chlorophyll a/b ratio). As a result of these fluctuationsthese plants achieved a new and lower steady-state conditionbetween the light and dark reactions, as judged from the molarratio of RuBP to Rubisco binding site. The activation of ribulose–1, 5-bisphosphate carb-oxylase/oxygenase(Rubisco, EC 4.1.1.39 [EC] ) from the floating angiosperm Spirodelapolyrhiza (L.) Schleid. (giant duckweed) grown at a photon irradianceof 200 or 400 mol photons m–2 s–1 was consistentlylow, in the range of 56–62%. Similarly low values wereobserved with four other emergent aquatic species growing underfull sun irradiance. Transference of Spirodela plants for short(minutes) or long (days) periods to the higher or lower irradianceincreased or decreased, respectively, the activation by onlyabout 15%. Activation was not greatly altered by exposure ofthe plants to full sun irradiance of >2000 mol photons m–2s–1 or CO2 concentrations in air of 0 and 1170 mol mor–1but darkness caused a slow decline to 20% activation. Transientoscillations were observed following a change in irradianceor CO2 concentration indicating that Rubisco was responsiveto environmental perturbations. The low Rubisco activation wasnot due to the tight binding of inhibitors such as carboxyarabinitol-1-phosphate.It is concluded that a substantial proportion of the Rubiscoprotein in these naturally-occurring species may not be usedfor CO2-fixation at any given moment. The effects of elevated CO2 were studied on the photosyntheticgas exchange behaviour and leaf physiology of two contrastingpoplar (Populus) hybrids grown and treated in open top chambers(OTCs in Antwerp, Belgium) and in closed glasshouse cabinets(GHCs in Sussex, UK). The CO2 concentrations used in the OTCswere ambient and ambient +350 µmol mol–1 while inthe GHCs they were c. 360 µmol mol–1 versus 719µmol mol–1. Measurements of photosynthetic gas exchangewere made for euramerican and interamerican poplar hybrids incombination with measurements of dark respiration rate and Rubiscoactivity. Significant differences in the leaf anatomy and structure(leaf mass per area and chlorophyll content) were observed betweenthe leaves grown in the OTCs and those grown in the GHCs. ElevatedCO2 stimulated net photosynthesis in the poplar hybrids after1 month in the GHCs and after 4 months in the OTCs, and therewas no evidence of downward acclimation (or down-regulation)of photosynthesis when the plants in the two treatments weremeasured in their growth CO2 concentration. There was also noevidence of down-regulation of Rubisco activity and there wereeven examples of increases in Rubisco activity. Rubisco exerteda strong control over the light-saturated rate of photosynthesis,which was demonstrated by the close agreement between observednet photosynthetic rates and those that were predicted fromRubisco activities and Michaelis-Menten kinetics. After 17 monthsin elevated CO2 in the OTCs there was a significant loss ofRubisco activity for one of the hybrid clones, i.e. Beaupr,but not for clone Robusta. The effect of the CO2 measurementconcentration (i.e. the short-term treatment effect) on netphotosynthesis was always larger than the effect of the growthconcentration in both the OTCs or GHCs (i.e. the longterm growthCO2 effect), with one exception. For the interamerican hybridBeaupr dark respiration rates in the OTCs were not significantlyaffected by the elevated CO2 concentrations. The results suggestthat for rapidly growing tree species, such as poplars, thereis little evidence for downward acclimation of photosynthesiswhen plants are exposed to elevated CO2 for up to 4 months;longer term exposure reveals loss of Rubisco activity. The effects of exposure of up to 2 h with sulphur dioxide ona range of plant species was observed by measuring changes inthe rate of net photosynthesis under closely controlled environmentalconditions. Ryegrass, Lolium perenne ‘S23’ was thespecies most sensitive to SO2; significant inhibition was detectedat 200 nl l–1. Fumigations at 300 nl l–1 also inhibitedphotosynthesis in field bean (Vicia faba cv. ‘Three FoldWhite’ and ‘Blaze’) and in barley (Hordeumvulgare cv. ‘Sonja’). No effect was detected inwheat (Triticum aestivum cv. ‘Virtue’) at concentrationsup to 600 nl l–1 SO2, or in oil-seed rape (Brassica napuscv. ‘Rafal’) except at 800 nl l–1 SO2). Recoverycommenced immediately after the fumigation was terminated andwas complete within 2 h when inhibition had not exceeded 20%during the SO2 treatment. Pattern of Respiration of a Perennial Ryegrass Crop in the Field   总被引:3,自引:0,他引:3  
‘Dark’ respiratory losses of CO2 were measured ona one year old sward of S24 perennial ryegrass (Lolium perenneL.) at intervals during a 74 day reproductive growth period,between April and June, and a 21 day vegetative growth period,in July and August. Part of the sward was shaded for one weekbefore measure ments commenced. Measurements of ‘dark’respiration continued for 46 hand it was possible to distinguishtwo components which are designated ‘maintenance’and ‘synthetic’ ‘Maintenance’ respiration was taken to be the meanrate of CO2 efflux after 40–46 h darkness. When calculatedon a plant d. wt basis at 15°C it ranged between 6 to 32mgCO2 g-1 day-1 during reproductive growth and 10–14 mgCO2 g-1 day-1 during vegetative growth. During reproductivegrowth, sward protein content ranged between 7–23 percent and when maintenance respiration was recalculated on thebasis of protein content it changed relatively little throughoutthe growth period (90–140 mg CO2 g pro tein-1 day-1);the value for vegetative growth ranged between 70–100mgCO2 g protein-day-1. Total ‘synthetic’ CO2 flux was determined duringreproductive growth and a rate of ‘synthetic’ CO2flux was determined during both reproductive and vegetativegrowth. Between 15 and 35 per cent of the CO2 fixed in the previousphotoperiod was lost in ‘synthetic’ respirationof above-ground material in reproductive swards. Previous shadingincreased the proportion of ‘synthetic’ CO2 lossfrom above ground. The rate of ‘synthetic’ CO2 outputduring the first hours of darkness increased with amount ofCO2 fixed in the previous photoperiod, although it was not proportionalto it. There is some evidence that assimilate is ‘carried-over’from one photoperiod to the next.  相似文献
