The time course of lectin production in culture liquid of the basidial fungus Lentinus edodes strain F-249 in different media under the conditions of submerged culture was studied. The activity of agglutinins depended on the ratio between carbon and nitrogen sources and pH of culture medium. The activity of lectin in culture medium was maximal when the fungus was grown in a medium containing L-arabinose as a source of carbon and L-asparagine as a source of nitrogen (C : N ratio, (9.5-12): 1)) on the day 15-18 of culturing at pH 8-9.  相似文献

利用本研究室已构建的重组菌Bacillus subtilis/pBSMuL3-α/β-CGTase对产B.stearothermophilus环糊精葡萄糖基转移酶的发酵产酶进行了优化,考察了培养基中重要成分:碳源、有机氮源、无机氮源、有机与无机氮源质量比、碳源与氮质量比、金属离子种类等单因素对该重组菌产α/β-CGTase的影响,并采用正交实验对发酵培养基进行优化,对优化结果分析可知,重组菌B.subtilis/pBSMuL3-α/β-CGTase发酵产α/β-CGTase的最优培养基成本为:葡萄糖5 g/L,氮源(鱼骨蛋白胨∶NH4Cl=3∶1)25 g/L,1 mmol/L Mg^2+。在最优条件下发酵培养,α/β-CGTase的酶活由原来TB发酵培养基的9.20 U/mL提高至20.32 U/mL,是优化前酶活的2.2倍,为α/β-环糊精葡萄糖基转移酶的工业应用提供了理论支持。  相似文献

利用响应面法对红曲霉M4产粉红色红曲色素的发酵培养基进行了优化。研究了不同碳源、氮源对红曲霉M4产色素的影响,利用Central composite试验设计对碳源、氮源浓度进行了试验,得到红曲色素优化培养基回归方程为Y=31.80001+12.02669X1+0.698225X2-12.33755X12+1.75X1X2-0.337496X22经分析,确定出红曲色素的最佳发酵培养基碳、氮源浓度为:红薯粉10.37%,大豆蛋白0.85%,发酵后红曲色素最高色价为每毫升41 u。  相似文献

研究了根霉12号固体发酵产生纤溶酶的工艺条件。采用单因素试验、均匀设计方法对固体发酵培养基的碳源、氮源、碳氮比、初始pH、加水量、无机盐加量进行了优化;采用正交试验对发酵时间、接种量进行了研究。结果表明,实验范围内根霉12固体发酵产纤溶酶的适宜培养基组成为:麸皮∶豆粕=1∶2,初始pH5.0,加水量0.75ml/g物料, MnSO4H2O和 (NH4)2SO4加量分别为0.25%和 1.42%(对物料)。适宜培养条件为接种量107个孢子/g物料,培养时间72h。优化条件下的纤溶酶产量平均达791.81u/g物料。  相似文献

目的对长双歧杆菌液态发酵培养基进行优化。方法以长双歧杆菌(Bifidobacteriumlongum)为发酵菌株,以MRS培养基为基础培养基,以发酵液活菌数为指标,通过单因素添加实验考察发酵培养基的碳源和氮源的种类,并验证优化后培养基的效果。结果优化后培养基的最适碳源为葡萄糖,最适氮源为酪蛋白胨、牛肉蛋白胨、水解乳蛋白,发酵液活菌数达到2.09×10^9CFU/mL,比原MRS培养基(1.22×10^9CFU/mL)提高了71.30%。结论优化后培养基优于原MRS基础培养基,可应用于长双歧杆菌的液态发酵。  相似文献

本文对枯草芽孢杆菌在不同碳源、氮源培养基的生长及破乳效能进行了研究, 并通过正交试验对枯草芽孢杆菌的发酵条件进行优化结果表明, 单一碳源葡萄糖和混合碳源葡萄糖 + 液体石蜡的培养基可提高枯草芽孢杆菌的发酵产量; 单一碳源葡萄糖、混合碳源葡萄糖 + 汽油和以硝酸铵 + 酵母膏为氮源的菌液有较高的破乳效能; 在正交试验中, 培养温度对枯草芽孢杆菌的发酵产量影响最大, 其最优组合为: 培养温度25oC, 摇床转数140 r/min, 培养pH值7.0, 接菌量6 mL, 培养时间24 h。摇床转数对枯草芽孢杆菌的发酵产物的破乳效能影响最大, 优化结果为: 培养温度25oC, 摇床转数140 r/min, pH值7.0, 培养时间20 h。  相似文献

In the present work, four strains were screened for schizophyllan production, of which Schizophyllum commune NRCM was selected for further work. The fermentation was carried out for 168 h at 28+/-2 degrees C on an orbital shaker at 180 rpm. In the first step, one factor-at-a-time method was used to investigate the effect of media constituents such as carbon and nitrogen sources on schizophyllan production. Subsequently in the second step, concentration of the medium components was optimized using Response Surface Method (RSM). The yield increased from 3.25+/-0.72 g/l in the unoptimized media to 8.03+/-1.12 g/l in the medium optimized by RSM.  相似文献

The main objective of this work was the optimization of the production of the beta-ketolase, acetopyruvate hydrolase, from Pseudomonas putida O1. Orcinol was used as an inducer for enzyme production. The growth medium was optimized in two steps. In the first step, screening for optimal glucose concentration was performed. In the second step, a central composite design was used to optimize carbon and nitrogen sources in the medium. After this optimization procedure, a medium was obtained which produced seven times more biomass than the initial medium. Acetopyruvate hydrolase enzyme production was optimized by determining the optimal time of feed and amount of orcinol, using statistical methods. In a subsequent step, the maximal orcinol-degradation rate was determined. The results obtained were used to find an optimal feeding profile for enzyme production. By using the optimized fed-batch process, acetopyruvate hydrolase activity was enhanced from 10 units l(-1)to 400 units l(-1), in comparison with previously reported fermentation experiments. Productivity could even be increased by a factor of 75, to a value of 20 units l(-1 )h(-1).  相似文献

将单因素实验结果与响应面法相结合,对高产Monacolin K的紫色红曲霉Mp-24菌株进行发酵工艺条件优化。通过摇瓶发酵对碳源、氮源、碳源含量、氮源含量、培养时间等进行单因素优化,确定Mp-24菌株摇瓶发酵适宜条件:乳糖为碳源、酵母膏为氮源、碳源含量7%、氮源含量2%、培养时间12 d,Monacolin K产量为167 mg/L。应用Box-Behnken中心组合试验设计建立数学模型,进行响应面分析优化发酵条件,结果显示最佳发酵工艺条件为:碳源(乳糖)8%,氮源(酵母膏)3%,培养时间11 d,在此条件下Monacolin K的含量达到247.8 mg/L,比优化前提高1.5倍。  相似文献

The effect of different carbon and nitrogen sources on the production of mannan-degrading enzymes, focussing on β-mannanase, by Aspergillus niger was investigated using shake flask culture. The β-mannanase activity obtained during growth of A. niger on guar gum (GG, 1495 nkat mL(-1)) was much higher than those observed on other carbon substrates, locust bean gum (1148 nkat mL(-1)), α-cellulose (10.7 nkat mL(-1)), glucose (8.8 nkat mL(-1)) and carboxymethylcellulose (4.6 nkat mL(-1)). For fermentation using GG as a carbon source, bacteriological peptone gave the highest β-mannanase activity (1744 nkat mL(-1)) followed by peptone from meat (1168 nkat mL(-1)), yeast extract (817 nkat mL(-1)), ammonium sulphate (241 nkat mL(-1)), ammonium nitrate (113 nkat mL(-1)) and ammonium chloride (99 nkat mL(-1)) when used as a nitrogen source. The composition of bacteriological peptone and initial pH of the medium were further optimized using response surface methodology (RSM). Medium consisted of 21.3 g L(-1) GG and 57 g L(-1) peptone with initial culture pH of 5.5 was optimum for β-mannanase production (2063 nkat mL(-1)) by A. niger. The β-mannanase production obtained in this study using A. niger was significantly higher than those reported in the literature.  相似文献

本实验采用摇瓶培养研究了盾壳霉(Coniothyrium minitans)产生几丁质酶的条件。改良的天然马铃薯葡萄糖培养基(mPDB)较合成培养基(SMCS)更适宜作为盾壳霉产生几丁质酶的基础培养基。添加9种不同碳源试验表明葡萄糖较适宜于盾壳霉产几丁质酶;氮源试验表明硝酸钾是产酶的较适宜氮源。盾壳霉形成几丁质酶的培养时间以15 d为佳,培养的适宜pH值为6.5。  相似文献

To enhance the production of micrococcin GO5, a bacteriocin produced by Micrococcus sp. GO5, cultivation conditions and medium composition were optimized. The optimal initial pH and temperature for bacteriocin production were 7.0-9.0 and 37 degrees C, respectively. Micrococcus sp. GO5 displayed the highest micrococcin GO5 activity when grown in modified MRS medium that contained lactose or sucrose, rather than glucose, as a carbon source. The maximum bacteriocin activity was obtained in modified MRS medium containing 0.5% tryptone and 1.0% yeast extract as nitrogen sources instead of the other nitrogen sources present in MRS medium. Bacteriocin production was greatly affected by the concentration of K(2)HPO(4); strain GO5 produced eight-fold more bacteriocin in medium containing 2.0-2.5% K(2)HPO(4) than in medium containing 0.2% K(2)HPO(4). The optimal concentration of MgSO(4).7H(2)O for bacteriocin production was 0.5%. The production of micrococcin GO5 was increased 32-fold in shake flask culture and 16-fold in a bioreactor using the optimized medium (TY medium), compared with culturing in MRS medium.  相似文献

Polysaccharides, such as barley flour, dextrin and soluble starch, were better carbon sources than monosaccharides and disaccharides, such as glucose and maltose, for cell growth of Bacillus subtilis ZJF-1A5 and beta-glucanase production. beta-Glucanase produced by B. subtilis ZJF-1A5 was associated partially with cell growth and increased significantly when cells entered stationary phase; yeast extract was the best nitrogen source, followed by soybean flour. All inorganic nitrogen sources chosen in the experiments were not favorable for cell growth and enzyme production. A fractional factorial design (2(6-2)) was applied to elucidate medium components that significantly affect beta-glucanase production. The concentration of barley flour, corn flour and soybean flour in medium were significant factors. The steepest ascent method was used to locate the optimal domain and a central composite design was used to estimate the quadratic response surface from which the factor levels for maximum production of beta-glucanase were determined. The composition of fermentation medium optimized with response surface methodology was (g/l): barley flour, 63.5; corn flour, 44.8; KH2PO4, 1.0; MgSO4 x 7H2O, 0.1; CaCl2, 0.1. beta-Glucanase activity was 251 U/ml at 48 h using optimized medium, 1.4 times higher than that in original medium.  相似文献

Cultures of Pseudomonas fluorescens DF57 were grown on different carbon and nitrogen sources. Glucose, succinate and acetate were used as carbon source and pulsed to an aerobic steady-state cultivation of P. fluorescens DF57 at D = 0.1 h(-1) with citrate as limiting carbon source. Glucose was utilised with the fastest uptake rate (19.4 C mmol l(-1) h(-1)) compared to succinate (8.8 C mmol l(-1) h(-1)) and acetate (4.3 C mmol l(-1) h(-1)). Acetate triggered an inhibition of cellular metabolism, which resulted in 2-h long growth arrest after its addition to the steady-state culture. The influence of the nitrogen source was investigated in an aerobic cultivation on a mixture of ammonium and nitrate as limiting nitrogen sources and citrate as non-limiting carbon source. When ammonia and nitrate were pulsed to the steady-state culture, they were mainly assimilated into biomass with a maximum uptake rate of 111 and 33 mg N l(-1) h(-1), respectively. Nitrate uptake was never complete as the residual concentration in the chemostat cultivation was 30 mg N l(-1) nitrate. A pulse of nitrite in the cultivation broth resulted in an inhibition of the growth but not of the primary metabolism, as nitrite was taken up at 38 mg N l(-1) h(-1), citrate was consumed and cofactors were produced continuously. In all experiments, oxygen was used as electron acceptor.  相似文献

A thermophilic isolate Bacillus coagulans BTS-3 produced an extracellular alkaline lipase, the production of which was substantially enhanced when the type of carbon source, nitrogen source, and the initial pH of culture medium were consecutively optimized. Lipase activity 1.16 U/ml of culture medium was obtained in 48 h at 55 degrees C and pH 8.5 with refined mustard oil as carbon source and a combination of peptone and yeast extract (1:1) as nitrogen sources. The enzyme was purified 40-fold to homogeneity by ammonium sulfate precipitation and DEAE-Sepharose column chromatography. Its molecular weight was 31 kDa on SDS-PAGE. The enzyme showed maximum activity at 55 degrees C and pH 8.5, and was stable between pH 8.0 and 10.5 and at temperatures up to 70 degrees C. The enzyme was found to be inhibited by Al3+, Co2+, Mn2+, and Zn2+ ions while K+, Fe3+, Hg2+, and Mg2+ ions enhanced the enzyme activity; Na+ ions have no effect on enzyme activity. The purified lipase showed a variable specificity/hydrolytic activity towards various 4-nitrophenyl esters.  相似文献

对栀子悬浮细胞合成多糖的调控因子研究表明 :B5为最适培养基 ;5~10d继代周期的细胞可以保持良好的生长状态和多糖的合成能力 ;80g L的鲜细胞的接种量有利于栀子细胞的生长和多糖的合成 ;使用单一碳源时 ,葡萄糖比蔗糖对细胞生长更有益 ,但葡萄糖成本高 ,因而混合碳源 45g L(葡萄糖 :蔗糖 =1∶1)是最佳配方 ;氮源种类对细胞生长和多糖合成没有明显的影响 ,但氮源浓度是主要因素 ,40~50mmol L是最佳浓度 ,同时运用悬浮细胞生产栀子多糖可以通过在不同时间收获的细胞来避免提取时黄色素的干扰 ,具有很好的实际意义。  相似文献

Effect of various fermentation media, carbon sources, nitrogen sources, phosphate concentration and culture requirements includes inoculum levels and age were determined on gentamicin production and biomass dry weight production for Micromonospora echinospora, a gentamicin producing strain. Of the substrates tested, starch as a sole carbon source promoted maximal gentamicin production, while maltose promoted maximal growth. Yeast extract as a sole nitrogen source promoted maximal growth, while soyabean meal for gentamicin production. Increasing phosphate concentration enhanced gentamicin production and observed optimum production at 1.2 g/1 (6% v/v) of phosphate having 72 h old inoculum in the medium. Highest gentamicin production was obtained after cultivation with shaking for 120 h in a medium containing starch 0.75% (w/v), soyabean meal 0.5%, K2HPO4 0.12%, CaCO3 0.4%, FeSO4 0.003% and CoCl2 0.0001%. The gentamicin production was 1.2-fold in this medium as compared to basal medium.  相似文献

通过单因素试验分析不同碳源、氮源、无机盐对(Sphingomonas paucimobilisFJAT-5627)产胶量的影响,确定最适碳源、氮源、无机盐,并在单因素筛选试验的基础上,利用Box-Benhnken设计和响应面分析法对碳源、氮源和无机盐进行优化,得到少动鞘脂单胞菌产生结冷肢发酵培养基最佳优化组合.实验结果表明,少动鞘脂单胞菌产胶量发酵最适碳源、氮源和无机盐分别为淀粉、豆饼粉和KH2PO4.响应面法得到产胶量(Y)与碳源淀粉(x1)、氮源豆饼粉(x2)和无机盐KH2PO4(x3)的回归方程为:Y=13.87+0.54x1+0.22x2-0.42x3-3.26x12-1.85x22-1.51x32+0.053x1x2+0.067x1x3+0.4x2x3.优化培养基组合为:淀粉浓度为30g/L,豆饼粉浓度为5 g/L,KH2PO4的浓度为0.7g/L,且此组合下少动鞘脂假单胞发酵得到结冷胶可达23.87g/L.  相似文献

The main objective of this work was the optimization of the production of the novel dioxygenase diketone cleaving enzyme (Dke1) from Acinetobacter johnsonii. Acetylacetone was used as an inducer for enzyme production. In the first step, the growth medium was optimized by using screening designs for finding the optimal carbon and nitrogen source. In the second step, a genetic algorithm was used to optimize the concentrations of all medium components. After six generations the stopping criterion was reached and a growth medium was obtained which produced sixteen times more enzyme than the starting medium. In the next step, an addition profile for the inducer acetylacetone was developed to further increase enzyme production by using a genetic algorithm. In this case, after four generations the stopping criterion was fulfilled. By using the obtained optimal addition profile Dke1 activity was enhanced from 826 to 2584Ul(-1). In comparison to the starting conditions activity could even be increased by a factor of 50.  相似文献
