Fluorescence of the marine alga Thalassiosira weissflogii (Grunow) Fryxell et Hasle with open (F o ) and closed (F m ) reaction centers of photosystem 2 (PS 2) and its relative variable fluorescence (F v/F m ) were measured at various levels of inorganic nitrogen. A significant heterogeneity of the population in terms of these parameters was revealed. Some cells within the population were more sensitive to nitrogen deficiency, and their photosynthetic apparatus was disrupted to a greater extent. The cells within a population also differed in terms of their ability to recover after incubation at low nitrogen levels. Enhancement of nitrogen deficiency resulted in an increase in the variability of the F o and F v/F m values of the cells. Fluorescence variability decreased at a less pronounced deficiency. Fluorescence variability should be taken into consideration in the studies concerning responses of algae to changes in nutrient contents.  相似文献

The effects of exogenous applied proline (Pro), on photosystem II (PSII) photochemistry of drought stressed (DS) 4-week old Arabidopsis thaliana plants, was studied by using chlorophyll (chl) fluorescence imaging. The maximum quantum yield of PSII photochemistry (F v /F m) in DS plants decreased significantly to 77% of that of the control value, suggesting that DS plants could not maintain PSII function, possibly due to accelerated photoinhibition of PSII. Free Pro and total soluble sugars (SS) increased, in response to DS. Exogenous foliar application of Pro by spraying, led to a remarkable increase in the accumulation of Pro and surprisingly also of SS. Both of them served to scavenge reactive oxygen species (ROS), as it was evident by the decreased lipid peroxidation level measured as malondialdehyde (MDA). DS plants sprayed with Pro showed a tolerance to photoinhibition, this indicated by F v/F m being close to values typical of healthy leaves by maintaining more than 98% of PSII function. Also the higher quantum efficiency of PSII photochemistry (Φ PSΙΙ ) and the decreased excitation pressure (1 ? q p ) recorded for stressed leaves with Pro, lead us to conclude that Pro appears to be involved in the protection of chloroplast structures by quenching ROS. The enhanced dissipation of excess light energy of PSII, in part accounts for the observed increased resistance to DS in A. thaliana leaves with Pro. Our data pointed out that Pro signalling interacts with SS signaling pathway and provided a new insight in Pro metabolism.  相似文献

The native alpine plant Saussurea superba is widely distributed in Qinghai–Tibetan Plateau regions. The leaves of S. superba grow in whorled rosettes, and are horizontally oriented to maximize sunlight exposure. Experiments were conducted in an alpine Kobresia humilis meadow near Haibei Alpine Meadow Ecosystem Research Station (37°29′–37°45′N, 101°12′–101°33′E; alt. 3200 m). Leaf growth, photosynthetic pigments and chlorophyll fluorescence parameters were measured in expanding leaves of S. superba. The results indicate that leaf area increased progressively from inner younger leaves to outside fully expanded ones, and then slightly decreased in nearly senescent leaves, due to early unfavorable environmental conditions, deviating from the ordinary growth pattern. The specific leaf area decreased before leaves were fully expanded, and the leaf thickness was largest in mature leaves. There were no significant changes in the content of chlorophylls (Chl) and carotenoids (Car), but the ratios of Chl a/b and Car/Chl declined after full expansion of the leaves. The variation of Chl a/b coincided well with changes in photochemical quenching (q P) and the fraction of open PSII reaction centers (q L). The maximum quantum efficiency of PSII photochemistry after 5 min dark relaxation (F (v)/F (m)) continuously increased from younger leaves to fully mature leaves, suggesting that mature leaves could recover more quickly from photoinhibition than younger leaves. The light-harvesting capacity was relatively steady during leaf expansion, as indicated by the maximum quantum efficiency of open PSII centers (\(F_{\text{v}}^{{\prime }}\)/\(F_{\text{m}}^{{\prime }}\)). UV-absorbing compounds could effectively screen harmful solar radiation, and are a main protection way on the photosynthetic apparatus. The decline of q P and q L during maturation, together with limitation of quantum efficiency of PSII reaction centers (L (PFD)), shows a decrease of oxidation state of QA in PSII reaction centers under natural sunlight. Furthermore, light-induced (Φ NPQ) and non-light-induced quenching (Φ NO) were consistent with variation of L (PFD). It is concluded that the leaves of S. superba could be classified into four functional groups: young, fully expanded, mature, and senescent. Quick recovery from photoinhibition was correlated with protection by screening pigments, and high level of light energy trapping was correlated with preservation of photosynthetic pigments. Increasing of Φ NPQ and Φ NO during leaves maturation indicates that both thermal dissipation of excessive excitation energy in safety and potential threat to photosynthetic apparatus were strengthened due to the declination of q P and q L, and enhancement of L (PFD).  相似文献

The impact that the parasitic plant field dodder (Cuscuta campestris Yunk.) has on chlorophyll fluorescence and chlorophyll content of infested alfalfa (Medicago sativa L.) and sugar beet (Beta vulgaris L.) was examined under controlled conditions. Several parameters of chlorophyll fluorescence were measured in infested and non-infested alfalfa and sugar beet plants over a period of twenty days, beginning with the day of infestation. Chlorophyll contents (total, relative and ratio of chlorophyll a to b) were determined 1, 7, 14 and 20 days after infestation (DAI). Field dodder was found to affect both the total and relative chlorophyll contents in infested alfalfa and sugar beet, causing significant reduction in chlorophyll content in both host plants. This parasitic plant also affects a number of parameters of chlorophyll fluorescence (Fo, Fv/Fm, ΦPSII, Fv and IF), showing that these parameters may be considered sensitive indicators of the impact that field dodder has on its host plants.  相似文献

Using chlorophyll (Chl) a fluorescence many aspects of the photosynthetic apparatus can be studied, both in vitro and, noninvasively, in vivo. Complementary techniques can help to interpret changes in the Chl a fluorescence kinetics. Kalaji et al. (Photosynth Res 122:121–158, 2014a) addressed several questions about instruments, methods and applications based on Chl a fluorescence. Here, additional Chl a fluorescence-related topics are discussed again in a question and answer format. Examples are the effect of connectivity on photochemical quenching, the correction of F V /F M values for PSI fluorescence, the energy partitioning concept, the interpretation of the complementary area, probing the donor side of PSII, the assignment of bands of 77 K fluorescence emission spectra to fluorescence emitters, the relationship between prompt and delayed fluorescence, potential problems when sampling tree canopies, the use of fluorescence parameters in QTL studies, the use of Chl a fluorescence in biosensor applications and the application of neural network approaches for the analysis of fluorescence measurements. The cadmium (Cd), being a widespread soils pollutant and one of the most toxic heavy metals in the environment, adversely affects sustainable crop production and food safety. Pot experiment was conducted to quantify and simulate the response of purslane (Portulaca oleracea L.) plants to Cd toxicity. The purslane germinated seeds were cultivated in twelve Cd concentrations (from 0 to 300 mg/kg of Cd in soil) for six weeks and then some growth characteristics, photosynthesis pigments, and chlorophyll a fluorescence parameters were measured. The influence of Cd gradients in the soil on all growth parameters, photosynthesis pigments and chlorophyll a fluorescence parameters (except Fm and carotenoid content) were described by a segmented model. Furthermore, Fm and carotenoid contents were fitted to a linear model. The growth characteristics, chlorophyll content, photosynthetic pigments and some parameters of chlorophyll a fluorescence such as Fv, Fv/Fm, Y(II) and ETR decreased when Cd concentration increased. In contrast, F0, Y(NPQ) and Y(NO) increased and Fm was not significantly affected. In general, most variations in the studied parameters were recorded with low concentrations of cadmium, which ranged from 0 to 125 mg/kg. Also, the growth characteristics (especially stem, leaf, and shoot dry weights) were more sensitive to Cd contamination than other parameters. Moreover, among chlorophyll fluorescence parameters, Y(NPQ) was the most sensitive to Cd concentration gradients in the soil that can be due to disturbances of antennae complex of PSII.  相似文献

The effects of the exogenous application of nitric oxide (NO, in the form of sodium nitroprusside, SNP) on the diurnal variation in photosynthesis, chlorophyll content, chlorophyll fluorescence, light response curve and the net assimilation of CO2 against intercellular CO2 concentration (A-Ci) curve parameters were investigated in the leaves of bamboo (Indocalamus barbatus McClure) exposed to simulated acid rain (SAR, pH 3.0) stress. According to the results of the diurnal variation in photosynthesis, foliar applications of 100–400 mg/L SNP effectively inhibited the decrease in net photosynthetic rate (Pn) as a result of non-stomatal factors, and mitigated midday depression under acid rain stress. The mitigating effect was most pronounced at 400 mg/L SNP. However, at higher concentrations of SNP (700 and 1000 mg/L), the mitigating effect became weak and even counterproductive. The results of the chlorophyll content, light response and A-Ci curve parameters suggested that the regulating role of NO against acid rain in the photosynthetic processes occurs through improving not only the efficiency of the light-harvesting and the activity of photosynthetic apparatus, but also the absorption of CO2 and the availability of CO2 for photosynthesis. The results of the chlorophyll fluorescence investigation further indicated that NO protected PSII activity from the damage of acid rain toxicity by enhancing the electron transport activity and photochemical efficiency, especially concerning the increase in the proportion of PSII open reaction centers. Furthermore, NO induced an increase in photorespiration (Rp), rather than an increase in non-photochemical quenching (NPQ), to dissipate the excessive excitation energy, which provided some protection to the photosynthetic apparatus under acid rain stress.  相似文献

The chlorophyll (Chl) fluorescence imaging technique was applied to cashew seedlings inoculated with the fungus Lasiodiplodia theobromae to assess any disturbances in the photosynthetic apparatus of the plants before the onset of visual symptoms. Two-month-old cashew plants were inoculated with mycelium of L. theobromae isolate Lt19 or Lt32. Dark-adapted and light-acclimated whole plants or previously labelled, single, mature leaf from each plant were evaluated weekly for Chl fluorescence parameters. From 21 to 28 days, inoculation with both isolates resulted in the significantly lower maximal photochemical quantum yield of PSII (Fv/Fm) than those for control samples, decreasing from values of 0.78 to 0.62. In contrast, the time response of the measured fluorescence transient curve from dark-acclimated plants increased in both whole plants and single mature leaves in inoculated plants compared with controls. The Fv/Fm images clearly exhibited photosynthetic perturbations 14 days after inoculation before any visual symptoms appeared. Additionally, decays in the effective quantum yield of PSII photochemistry and photochemical quenching coefficient were also observed over time. However, nonphotochemical quenching increased during the evaluation period. We conclude that Fv/Fm images are the effective way of detecting early metabolic perturbations in the photosynthetic apparatus of cashew seedlings caused by gummosis in both whole plants and single leaves and could be potentially employed in larger-scale screening systems.  相似文献

The physiological state of the leaves of the small-leaved linden (Tilia cordata), silver birch (Betula pendula), and northern white cedar (Thuja occidentalis) under urban conditions was assessed via recording the kinetics of chlorophyll under fluorescence induction. Different sensitivities of the plants to adverse growing conditions were revealed. The most sensitive parameters of the fluorescence JIP test, viz., PI ABS , F V/F 0, and F V/F M, were identified as indicators of the physiological state of the urban phytocoenosis. In order to investigate the effect of day/night temperature difference (DIF) on photosynthetic characteristics of tomato plants (Solanum lycopersicum, cv. Jinguan 5) at fruit stage, an experiment was carried out in climate chambers. Five day/night temperature regimes (16/34, 19/31, 25/25, 31/19, and 34/16°C) with respective DIFs of -18, -12, 0, +12, and +18 were used and measured at mean daily temperature of 25°C. The results showed that chlorophyll (Chl) a, Chl b, net photosynthetic rate (PN), stomatal conductance (gs), maximum quantum yield of PSII photochemistry (Fv/Fm), effective quantum yield of PSII photochemistry (?PSII), and photochemical quenching (qp) significantly increased under positive DIF, while they decreased with negative DIF. In contrast, the Chl a/b ratio and nonphotochemical quenching (NPQ) decreased under positive DIF, while increased with negative DIF. Chl a, Chl b, PN, gs, Fv/Fm, ?PSII, and qp were larger under +12 DIF than those at +18 DIF, while Chl a/b and NPQ showed an opposite trend.  相似文献

Changes of chlorophyll (Chl) a fluorescence and photosynthetic pigment contents were analysed in galled leaves (visibly damaged and undamaged parts) and intact leaves. The values of minimal fluorescence of the dark-adapted state, maximal quantum yield of PSII photochemistry, effective quantum yield of PSII photochemical conversion, and photochemical quenching coefficient decreased in Ulmus pumila L. leaves galled by Tetraneura ulmi (L.) and in U. glabra Huds. galled by Eriosoma ulmi (L.). Colopha compressa (Koch.) feeding affected these parameters only in damaged parts of U. laevis Pall. galled leaves. The increasing number of T. ulmi galls progressively decreased photosynthetic performance. In gall tissues of all analysed aphid species, the lowest photosynthetic pigment content was found, indicating that the photosynthetic capacity must have been low in galls. Significant reduction of Chl and carotenoid contents were observed in damaged and undamaged portions of galled leaves only in the case of T. ulmi feeding.  相似文献

Pigment mutants CC-124y-1, СС-124у-2, and СС-124р-2 of the green microalga Chlamydomonas reinhardtii Dangeard were obtained by means of UV-induced mutagenesis. Morphological properties and photosynthetic activity of these mutants were examined. The mutants displayed a lowered content of chlorophyll b and carotenoids. Analysis of fluorescence induction curves revealed the decreased rates of electron transport in photosystem II (PSII), the increase in the fraction of QB-nonreducing centers, and the enhancement of nonphotochemical fluorescence quenching. The mutations had no direct impact on oxidation of pigment P700 in PSI and on the decline of delayed fluorescence. The parameters of induction curves of prompt and delayed fluorescence are proposed for use in early diagnostics of UV-induced mutagenesis. The pigment mutants obtained can be applied in biomonitoring studies.  相似文献

Pistachio (Pistacia vera L.) has a high tolerance to drought and soil salinity. Although adult pistachio trees are well known to be drought tolerant, the studies on physiological adaptation of pistachio cultivars to drought are limited. Therefore, three pistachio cultivars, i.e., Akbari, Kaleghochi, and Ohadi were subjected to three osmotic drought stress treatments: control (?0.1 MPa), moderate (?0.75 MPa) and severe drought (?1.5 MPa) stress using PEG 6000 for a 14-day period. All drought stress treatments decreased net photosynthesis (P n), stomatal conductance (g s), intercellular CO2 concentration (C i), and transpiration rate (E), but Ohadi maintained better its photosynthetic capacity compared to Akbari and Kaleghochi. Maximum quantum yield of PSII photochemistry (F v /F m), effective PSII quantum yield (ΦPSII) and photochemical quenching (qP) were also reduced. The chlorophyll fluorescence parameters indicated that Akbari was more susceptible to the applied drought stress. Drought stress levels decreased chlorophyll pigments, fresh weight, stem elongation, leaf nitrogen content (N), leaf water potential and increased water use efficiency (WUE). Proline increased strongly under drought stress for Akbari. After 2 weeks of stress a recovery of 2 weeks was applied. This period was insufficient to fully restore the negative effects of the applied stress on the studied cultivars. Based on the reduction of photosynthesis and the increase of the proline content Akbari seems more sensitive to the applied drought stress.  相似文献

Bioenergetics of the aerobic bacteriochlorophyll a-containing (BCl a) bacterium (ABC bacterium) Roseinatronobacter thiooxidans is a combination of photosynthesis, oxygen respiration, and oxidation of sulfur compounds under alkaliphilic conditions. The photosynthetic activity of Rna. thiooxidans cells was established by the photoinhibition of cell respiration and reversible photobleaching discoloration of the BCl a of reaction centers (RC), connected by the chain of electron transfer with cytochrome c 551 oxidation. The species under study, like many purple bacteria and some of the known ABC bacteria, possesses a light-harvesting pigment-protein (LHI) complex with the average number of 30 molecules of antenna BCl a per one photosynthetic RC. Under microaerobic growth conditions, the cells contained bc 1 complex and two terminal oxidases: cbb 3-cytochrome oxidase and the alternative cytochrome oxidase of the a 3 type. Besides, Rna. thiooxidans was shown to have several different soluble low- and high-potential cytochromes c, probably associated with the ability of utilizing sulfur compounds as additional electron donors.  相似文献

The OJDIP rise in chlorophyll fluorescence during induction at different light intensities was mathematically modeled using 24 master equations describing electron transport through photosystem II (PSII) plus ordinary differential equations for electron budgets in plastoquinone, cytochrome f, plastocyanin, photosystem I, and ferredoxin. A novel feature of the model is consideration of electron in- and outflow budgets resulting in changes in redox states of Tyrosine Z, P680, and QA as sole bases for changes in fluorescence yield during the transient. Ad hoc contributions by transmembrane electric fields, protein conformational changes, or other putative quenching species were unnecessary to account for primary features of the phenomenon, except a peculiar slowdown of intra-PSII electron transport during induction at low light intensities. The lower than F m post-flash fluorescence yield F f was related to oxidized tyrosine Z. The transient J peak was associated with equal rates of electron arrival to and departure from QA and requires that electron transfer from QA ? to QB be slower than that from QA ? to QB ?. Strong quenching by oxidized P680 caused the dip D. Reduced plastoquinone, a competitive product inhibitor of PSII, blocked electron transport proportionally with its concentration. Electron transport rate indicated by fluorescence quenching was faster than the rate indicated by O2 evolution, because oxidized donor side carriers quench fluorescence but do not transport electrons. The thermal phase of the fluorescence rise beyond the J phase was caused by a progressive increase in the fraction of PSII with reduced QA and reduced donor side.  相似文献

The methods of chlorophyll fluorescence induction and HPLC were used to study the influence of autumnal temperature decrease on photochemical electron-transport activity of photosystem II (PSII), nonphotochemical quenching of excessive excitation energy, and the composition of pigments in the firstyear needles of Pinus sуlvestris L. trees grown naturally in Central Yakutia. In the period from the beginning of September to October 10, the chlorophyll content was reduced by half, while the Chl a/b ratio increased from 2.9 to 4.3–4.5, indicating the degradation of peripheral antenna complexes. The decrease in average daily temperature to 4.9–6.4°C led to a transient increase in the quantum yield of nonphotochemical quenching (ΔpH-dependent parameter φNPQ). These changes were accompanied by a slow accumulation of unregulated zeaxanthin fraction insensitive to illumination conditions. The further decrease in average daily temperature to near-zero levels was paralleled by a sharp increase in zeaxanthin content, while the pH-dependent quenching was replaced with the constitutive quenching (parameter φf,D) because of the supposed structural reorganization of PSII. These processes were accompanied by a fast decrease in PSII functional activity, which was mostly due to the impairment of plastoquinone photochemical reduction. Freezing temperatures (from–3.6 to–12.1) destructed the oxygen-evolving complex in PSII and completely inactivated the PSII reaction centers. It is concluded that the largest changes in the condition of photosynthetic apparatus occur at a near-zero temperature range and proceed until complete inactivation of PSII under the action of freezing temperatures.  相似文献

The objective of this investigation was to evaluate the simultaneous action of light stress and salinity. Pulse amplitude modulated chlorophyll fluorescence, P700 redox state, and pigment analysis were used to assess the impact of high light intensity on Paulownia tomentosa × fortunei and Paulownia elongata × elongata grown on soils with different salinity. It was found that light stress reduced the amount of pigments and the efficiency of photochemical energy conversion, inhibited the maximum and the effective quantum yields of PSII photochemistry, decreased photochemical quenching and photosynthetic rate. Data also showed influence on the primary quinone acceptor (QA) reoxidation, which led to the restriction of the electron flow from QA to plastoquinone and stimulation of the cyclic electron flow. The possible reasons for the increased effects of the light stress under conditions of high salt concentration in soil for Paulownia tomentosa × fortunei are discussed.  相似文献

The purpose of the present study was to investigate the mechanism of DA-6 in alleviating the salinity inhibition of Cassia obtusifolia L. seeds and seedlings. NaCl (100 mM) was used to mimic salinity stress in a series of experiments. Varying combinations of DA-6 were added to seeds and seedlings under salinity stress. Seed germination indices and seedling parameters were investigated. Seed germination and seedling growth were significantly inhibited under salinity stress. NaCl-induced inhibitory effects on seed germination and seedling growth were ameliorated by DA-6 with different concentrations. Addition of DA-6 to seeds (50 µM) and seedlings (100 µM) significantly alleviated damage to the plant cells under salinity stress. DA-6 (regardless of the presence or absence of NaCl) enhanced chlorophyll concentration, total soluble sugars, free proline, and soluble protein, and improved photosystem II (PSII) photochemical efficiency levels (F v/F m), PSII actual photochemical efficiency (ΦPSII), and the photochemical quench coefficient. In contrast, the initial fluorescence (F o) and the non-photochemical quenching coefficient decreased. Application of DA-6 also enhanced the activities of superoxide dismutase (SOD; EC 1.15.1.1), peroxidase (POD; EC 1.11.1.7), catalase (CAT; EC 1.11.1.6), ascorbate peroxidase (APX; EC 1.11.1.11), and glutathione reductase (GR; EC 1.6.4.2), thus alleviating oxidative damage, as indicated by decreases in thiobarbituric acid-reactive substances, hydrogen peroxide concentration (H2O2), relative conductivity, and lipoxygenase activity (LOX; EC 1.13.11.12). Based on the experimental results, we conclude that DA-6 induces advantageous effects on the attenuation of salt-stress inhibition of C. obtusifolia seeds and seedlings and alleviates oxidative damage by conferring beneficial cytoprotection and activating antioxidant enzymes. DA-6 can be used as an effective plant growth regulator to alleviate salinity stress.  相似文献

Seasonal changes in the characteristics of chlorophyll fluorescence were studied in the bark of several species of trees originating in various climatic zones: Siberian cedar (Pinus sibirica), larch (Larix sibirica), eastern arborvitae (Thuja occidentalis), pendent white birch (Betula pendula), wild black cherry (Padus virginiana), horse chestnut (Aesculus hippocastanum), red oak (Quercus rubra), Manchurian catalpa (Catalpa bungei), linden (Tilia cordata), goat willow (Salix caprea), Amur cherry (Padus maackii), and apple Korichnaya (Malus domestrica B.). Tree bark has a sufficient amount of chlorophyll for measuring the parameters of chlorophyll fluorescence throughout the year. The relative yield of the variable fluorescence of chlorophyll (F v/F m) can be used to assess seasonal changes in the physiological state of various trees.  相似文献

Xantha-702 mutant of cotton (Gossypium hirsutum L.) proved to have blocked synthesis of 5-aminolevulinic acid in the light. Accordingly, mutant leaves accumulated 2–5% chlorophyll of baseline. Mutant plants demonstrated disturbed production of pigment-protein complexes of photosystems I (PSI) and II (PSII) and generation of the chloroplast membrane system blocked at the early stages, largely, at the stages of vesicles and single short thylakoid. The functional activity of the PSI and PSII reaction centers was close to zero. Only the chlorophyll a/b light-harvesting complexes of PSI and PSII with the chlorophyll fluorescence peaks at 728 and 681 nm, respectively, were produced in the xantha-702 mutant. We propose that the genetic block of 5-aminolevunilic acid biosynthesis in the light in the xantha-702 mutant disturbs the formation and activity of the complexes of the reaction centers of PS-I and PS-II and inhibits the development of the whole membrane system of chloroplasts.  相似文献
