Bcl 2家族蛋白是在细胞凋亡过程中起关键性作用的一类蛋白质。在线粒体上 ,Bcl 2家族蛋白通过与其他凋亡蛋白的协同作用 ,调控线粒体结构与功能的稳定性 ,发挥着细胞凋亡“主开关”的作用。Bcl 2家族包括两类蛋白质 :一类是抗凋亡蛋白 ,另一类是促凋亡蛋白。在细胞凋亡时 ,Bcl 2家族中的促凋亡蛋白成员发生蛋白质的加工修饰 ,易位到线粒体的外膜上 ,引起细胞色素c、凋亡诱导因子等其他促凋亡因子的释放 ,导致细胞凋亡 ;而平时被隔离在线粒体等细胞器内的该家族的抗凋亡蛋白成员则抑制细胞色素c和凋亡诱导因子等促凋亡因子的释放 ,具有抑制细胞凋亡的功能。但一旦这类抗凋亡蛋白成员与激活的促凋亡蛋白发生相互作用后 ,便丧失了对细胞凋亡的抑制作用 ,造成线粒体等细胞器的功能丧失和细胞器内促凋亡因子的释放 ,导致细胞凋亡。现以Bcl 2家族调控细胞凋亡的最新研究进展为基础 ,对Bcl 2家族成员及其蛋白质结构、分布和调控细胞凋亡的分子机制进行综述。  相似文献

Bcl-2蛋白质家族的抗凋亡和促凋亡成员,在线粒体水平上决定细胞的存活或死亡.在正常细胞中,这些成员呈现功能适应性的细胞内分布;抗凋亡成员主要定位于细胞内膜系特别是线粒体外膜上:但绝大多数促凋亡成员主要分布于细胞浆中.细胞接受死亡信号后,Bcl-2家族成员本身受到一系列的调节,如磷酸化、裂解、蛋白质-蛋白质相互作用等,结果之一是促凋亡成员发生细胞内定位的改变,从细胞浆转位于线粒体膜上,并引发线粒体功能异常及其内外膜间致凋亡因子的释放,最终导致细胞凋亡.  相似文献

细胞存活与凋亡之间的平衡是多细胞生物正常发育与稳态的关键。细胞凋亡是受多种因素高度调控的细胞程序性死亡过程。凋亡信号调节激酶 1 (ASK1 )是促分裂原活化蛋白激酶激酶激酶家族的成员之一 ,它分别激活SER1 JNK和MKK3 MKK6 p38途径 ,在细胞因子及应激诱导细胞凋亡过程中起着关键作用。TNF受体和Fas信号转导系统在抗凋亡与促凋亡过程中发挥重要作用 ,其中包括TNF受体Ⅰ相关死亡域蛋白 (TRADD)、Fas相关死亡域蛋白 (FADD)等多种蛋白因子。细胞色素C是线粒体依赖性死亡信号 ,受Bcl 2家族蛋白的调节。反应性氧化合物的氧化激活使硫氧还蛋白 (Trx)氧化 ,并与ASK1分离 ,从而激活ASK1造成细胞凋亡。总之 ,许多促凋亡与抗凋亡因子组成复杂的、相互拮抗的机制。在信号转导的各种不同的关卡上 ,这些因子的平衡作用最终决定细胞的生与死。  相似文献

热休克蛋白60(heat shock protein 60, HSP60)是主要存在于线粒体内的分子伴侣蛋白,对于维持线粒体蛋白的正常结构和功能不可或缺.线粒体中的HSP60可作用于凋亡相关因子而抑制线粒体凋亡通路的激活,并且能够减少线粒体产生氧自由基;胞浆中的少量HSP60亦可通过与凋亡相关因子的相互作用等途径抑制细胞凋亡.相反,在某些刺激因素作用下或者HSP60细胞定位异常时,HSP60可产生促凋亡效应.HSP60在细胞凋亡中的双重作用及其对于肿瘤等疾病诊治的意义已引起高度关注.  相似文献

细胞凋亡是机体维持组织稳态和胚胎发育的重要机制之一,受到多种信号分子的严格调控。促凋亡Bcl-2家族蛋白成员Bax和Bak蛋白在细胞凋亡中扮演着非常重要的角色。在凋亡信号的刺激下,Bax和Bak蛋白被激活并在线粒体上互相凝集成簇,使得线粒体膜的通透性增加,引起凋亡因子的释放,并最终诱导细胞的死亡。本文主要介绍Bax和Bak蛋白在细胞凋亡过程中的调控与激活机制,并详细阐述目前它们在线粒体凋亡通路中的几个激活模型,总结二者在激活线粒体凋亡通路中的作用,为进一步研究线粒体凋亡通路作一铺垫。  相似文献

Bcl-2家族蛋白在调控线粒体功能和细胞色素C释放中起重要作用。最近发现Bcl-2分子通过与其他促凋亡分子相互作用调控线粒体外膜通透性,其具体分子机制尚不完全清楚。本课题组采用化学生物学方法,在研究Bax/Bak非依赖的细胞凋亡途径中,发现了一些小分子化合物能够诱导Bim表达量急剧升高,Bim能转位到线粒体上,与Bcl-2相互作用增强,并直接促进Bcl-2构象变化。有意义的是,Bim可以诱导Bcl-2功能发生转换并能够形成大的复合体通道来介导细胞色素C释放。研究结果提示Bcl-2分子可变成促凋亡分子,参与Bax/Bak非依赖的细胞色素C释放和细胞凋亡。  相似文献

线粒体凋亡途径是细胞凋亡的主要途径之一。是目前研究凋亡的热点,各种凋亡刺激信号通过BH3(Bcl-2homology domain3)-only蛋白引起Bax(Bcl-2-asslciated proteinX)蛋白移位到线粒体外膜并多聚化,形成膜通道,刺激线粒体释放细胞色素C(CytC)和Smac(second mitochondrial-derived activator of caspase),CytC通过Apaf-1因子的多聚化与胱天蛋白酶(caspases)-9形成凋亡小体,导致下游胱天蛋白酶的级联反应,而凋亡蛋白抑制因子(IAP)和Smac通过抑制和促进胱天蛋白酶的级联反应来调控细胞凋亡。  相似文献

导言     
线粒体是细胞内具有双层膜结构和独立基因组DNA的重要细胞器,在细胞生命活动中发挥着至关重要的作用。一方面它们是真核细胞的主要能量工厂,通过有氧代谢产生ATP,为细胞生命活动提供能量;另一方面,线粒体是细胞内活性氧产生中心,同时也是细胞内主要钙库之一,调节细胞内钙信号和细胞生长活动。更为重要的是,线粒体还是细胞凋亡和衰老的调控中心。在细胞凋亡过程中,线粒体释放促凋亡因子(如细胞色素C),对细胞内凋亡信号进行整合和放大。不言而喻,线粒体在细胞生长、衰老和凋亡等生理、病理过程中扮演着重要的角色。  相似文献

心磷脂(cardiolipin, CL)是线粒体特异的一类磷脂,它不仅可以维持线粒体的结构,还对电子传递链复合体有影响。在细胞凋亡时,心磷脂会发生一些重分布,比如线粒体内膜外小叶中心磷脂含量的增加及细胞表面心磷脂的出现。同时,凋亡刺激还可以加速心磷脂的代谢循环,使心磷脂在线粒体内的含量降低。在细胞凋亡发生时,心磷脂作为一个信号整合体,对细胞色素c (cytochrome c, CytC)、胱天蛋白酶-8 (caspase-8)和促凋亡蛋白Bid发挥着重要的协调作用。本文主要就心磷脂在细胞凋亡过程中的重要作用及其在癌症中的研究进展进行综述。  相似文献

凋亡诱导因子(apoptosis-inducing factor, AIF)基因定位于X染色体上,其编码产物是一种可直接介导细胞核凋亡的效应分子. 鼠AIF前体蛋白在胞浆中合成后,通过其N端的线粒体定位信号(MLS)有效地穿入线粒体膜间隙,然后在102位甘氨酸处水解掉MLS,其余部分再与黄素腺嘌呤二核苷酸(FAD)结合,并重新折叠成为具有促凋亡潜能的成熟AIF分子,分子质量为57 ku.当凋亡信号刺激时,AIF分子从线粒体释放到胞浆,然后转位到细胞核内,引起染色体核周边凝集和DNA呈大片段断裂(~50 kb). 该作用不受广谱caspases抑制剂z-VAD.fmk的抑制,也不受Bcl-2过量表达的影响.基因剔除实验表明,AIF蛋白的促凋亡活性是胚胎小鼠形态发生过程中类胚体成腔所必需的,而且是AIF独立作用的结果,可以不依赖caspase-3的活性.因此AIF介导的细胞凋亡可能代表了独立于caspase通路之外的另一条更原始、更保守的凋亡途径.  相似文献

The Bcl-2 family of proteins consists of anti-apoptotic and pro-apoptotic members, which determine the life or death of cells by altering mitochondrial membrane permeability. Pro-apoptotic Bcl-2 family members increase mitochondrial membrane permeability, resulting in the release of mitochondrial apoptogenic factors such as cytochrome c that activates death proteases called caspases, whereas anti-apoptotic family members prevent this increase of mitochondrial membrane permeability. The release of cytochrome c is central to apoptotic signal transduction in mammals, and has been studied extensively, leading to the development of several models for cytochrome c release including rupture of the mitochondrial outer membrane and involvement of specific channels. This article describes the important role of a mitochondrial outer membrane channel, the voltage-dependent anion channel (VDAC), in apoptogenic cytochrome c release and its regulation by Bcl-2 family members, and also discusses the molecular architecture of the life - death switch in mammalian cells. Bcl-2 family proteins are central regulators of apoptosis. Various family members are located in the cytoplasm, endoplasmic reticulum, and mitochondrial outer membrane in healthy cells. However during apoptosis most of the interactions between family members that determine the fate of the cell occur at the membranes of intracellular organelles. It has become evident that interactions with membranes play an active role in the regulation of Bcl-2 family protein interactions. Here we provide an overview of various models proposed to explain how the Bcl-2 family regulates apoptosis and discuss how membrane binding affects the structure and function of each of the three categories of Bcl-2 proteins (pro-apoptotic, pore-forming, and anti-apoptotic). We also examine how the Bcl-2 family regulates other aspects of mitochondrial and ER physiology relevant to cell death.  相似文献

Mitochondria are well known as sites of electron transport and generators of cellular ATP. Mitochondria also appear to be sites of cell survival regulation. In the process of programmed cell death, mediators of apoptosis can be released from mitochondria through disruptions in the outer mitochondrial membrane; these mediators then participate in the activation of caspases and of DNA degradation. Thus the regulation of outer mitochondrial membrane integrity is an important control point for apoptosis. The Bcl-2 family is made up of outer mitochondrial membrane proteins that can regulate cell survival, but the mechanisms by which Bcl-2 family proteins act remain controversial. Most metabolites are permeant to the outer membrane through the voltage dependent anion channel (VDAC), and Bcl-2 family proteins appear to be able to regulate VDAC function. In addition, many Bcl-2 family proteins can form channels in vitro, and some pro-apoptotic members may form multimeric channels large enough to release apoptosis promoting proteins from the intermembrane space. Alternatively, Bcl-2 family proteins have been hypothesized to coordinate the permeability of both the outer and inner mitochondrial membranes through the permeability transition (PT) pore. Increasing evidence suggests that alterations in cellular metabolism can lead to pro-apoptotic changes, including changes in intracellular pH, redox potential and ion transport. By regulating mitochondrial membrane physiology, Bcl-2 proteins also affect mitochondrial energy generation, and thus influence cellular bioenergetics. Two major intracellular apoptosis signaling cascades have been characterized, the mitochondrial pathway and the death receptor pathway. The mitochondrial pathway is regulated by members of the Bcl-2 protein family. The members of this family can be subdivided into anti- and pro-apoptotic proteins. The pro-apoptotic members are further divided into two groups, the multidomain and the 'BH3 domain only' proteins. When cells are exposed to apoptotic stimulation, pro-apoptotic proteins are activated through post-translational modifications or changes in their conformation. The main site of action of the multidomain proteins are the mitochondria, where these proteins induce permeabilization of the outer membrane resulting in the release of proteins, including cytochrome c, from the intermembrane space. In the cytosol cytochrome c activates caspase cascades ultimately leading to cell death. Mounting evidence indicates that apoptosis is involved in a wide range of pathological conditions. Recent studies suggest that the mitochondrial signaling pathway is involved in several diseases. Although, so far, with the exception of C. elegans, most studies on apoptosis have been performed in mammalian systems, recently homologues to the Bcl-2 family members, including pro-apoptotic members, have been identified in Drosophila and zebrafish. Here the structure and function of the various pro-apoptotic Bcl-2 family members, their effects on mitochondria, and their involvement in diseases are discussed.  相似文献

Most anticancer therapies exert their action by triggering programmed cell death (apoptosis) in cancer cells. The mitochondrial pathway of apoptosis is initiated by mitochondrial outer membrane permeabilization, leading to the release of apoptogenic factors such as cytochrome c or Smac from the mitochondrial intermembrane space into the cytosol. Mitochondrial outer membrane permeabilization is tightly controlled, for example by pro- and anti-apoptotic proteins of the Bcl-2 family. Recent evidence indicates that inhibition of the PI3K/Akt/mTOR pathway by small-molecule PI3K inhibitors primes cancer cells to mitochondrial apoptosis by tipping the balance towards pro-apoptotic Bcl-2 proteins, resulting in increased mitochondrial outer membrane permeabilization. Thus, mitochondrial apoptotic events play an important role in PI3K inhibitor-mediated sensitization for apoptosis.  相似文献

Apoptosis is an elemental form of programmed cell death; it is fundamental to higher eukaryotes and essential to mechanisms controlling tissue homeostasis. Apoptosis is also involved in many pathologies including cancer, neurodegenerative diseases, aging, and infarcts. This cell death program is tightly regulated by Bcl-2 family proteins by controlling the formation of the mitochondrial apoptosis-induced channel or MAC. Assembly of MAC corresponds to permeabilization of the mitochondrial outer membrane, which is the so called commitment step of apoptosis. MAC provides the pathway through the mitochondrial outer membrane for the release of cytochrome c and other pro-apoptotic factors from the intermembrane space. While overexpression of anti-apoptotic Bcl-2 eliminates MAC activity, oligomers of the pro-apoptotic members Bax and/or Bak are essential structural component(s) of MAC. Assembly of MAC from Bax or Bak was monitored in real time by directly patch-clamping mitochondria with micropipettes containing the sentinel tBid, a direct activator of Bax and Bak. Herein, a variety of high affinity inhibitors of MAC (iMAC) that may prove to be crucial tools in mechanistic studies have recently been identified. This review focuses on characterization of MAC activity, its regulation by Bcl-2 family proteins, and a discussion of how MAC can be pharmacologically turned on or off depending on the pathology to be treated.  相似文献

The increase of outer mitochondrial membrane permeability is a central event in apoptotic cell death, since it releases several apoptogenic factors such as cytochrome c into the cytoplasm that activate the downstream destructive processes. The voltage-dependent anion channel (VDAC or mitochondrial porin) plays an essential role in the increase of mitochondrial membrane permeability, and it is regulated by the Bcl-2 family of proteins via direct interaction. Anti-apoptotic Bcl-2 family members close the VDAC, whereas some (but not all) pro-apoptotic members interact with the VDAC to generate a protein-conducting channel through which cytochrome c can pass. Although the VDAC is directly involved in the apoptotic increase of mitochondrial membrane permeability and is known to be a component of the permeability transition pore complex, its role in the regulation of outer membrane permeability can be separated from the occurrence of permeability transition events, such as mitochondrial swelling followed by rupture of the outer mitochondrial membrane. The VDAC not only interacts with Bcl-2 family members, but also with other proteins, and probably acts as a convergence point for a variety of life-or-death signals.  相似文献
