1. Whether Drosophila larvae and pupae naturally experience temperatures that can cause heat damage or death is poorly understood, but bears directly on numerous investigations of the thermal biology and heat-shock response in Drosophila . Accordingly, the temperatures of necrotic fruit, which Drosophila larvae and pupae inhabit, the temperatures of larvae and pupae outside the laboratory, and the levels of the heat-shock protein hsp 70 expressed by larvae in nature were examined.
2. When necrotic fruit was sunlit, internal temperatures rose to levels that can harm indwelling insects. Fruit size and evaporative water loss affected these temperatures. Temperatures of larvae and pupae in the field commonly exceeded 35 °C, with living larvae recorded at >44°C and pupae at >41°C. Natural mortality was evident, presumably because of heat.
3. In the laboratory, these temperatures kill larvae rapidly, with LT50s (time taken for half the sample to be killed) of 30 min at 39 °C, 15 min at 40 °C and 8·5 min at 41 °C. Gradual transfer from 25°C to these temperatures resulted in no lesser mortality than did direct transfer.
4. Hsp 70 levels in lysates of whole larvae were measured by ELISA (enzyme-link immunosorbent assay) with an hsp 70-specific antibody. For larvae within necrotic apples experimentally transferred from shade to sun and within necrotic fruit in situ , hsp 70 levels equalled or exceeded levels detected in parallel laboratory studies of whole larvae or cells in culture.
5. These data provide an ecological context for studies of thermal stress and the heat-shock response in Drosophila that has heretofore been lacking.  相似文献

Thermal tolerance of a northern population of striped bass Morone saxatilis   总被引:1,自引:0,他引:1  
Thermal tolerance of age 0+ year Shubenacadie River (Nova Scotia, Canada) striped bass Morone saxatilis juveniles (mean ± s . e . fork length, L F, 19·2 ± 0·2 cm) acclimated in fresh water to six temperatures from 5 to 30° C was measured by both the incipient lethal technique (72 h assay), and the critical thermal method ( C m). The lower incipient lethal temperature ranged from 2·4 to 11·3° C, and the upper incipient lethal temperature ( I U) from 24·4 to 33·9° C. The area of thermal tolerance was 618° C2. In a separate experiment, the I U of large age 2+ year fish (34·4 ± 0·5 cm L F) was 1·2 and 0·6° C lower ( P < 0·01) than smaller age 1+ year fish (21·8 ± 0·5 cm L F) at acclimation temperatures of 16 and 23° C. Using the C m, loss of equilibrium occurred at 27·4–37·7° C, loss of righting response at 28·1–38·4° C and onset of spasms at 28·5–38·8° C, depending on acclimation temperature. The linear regression slopes for these three responses were statistically similar (0·41; P > 0·05), but the intercepts differed (25·3, 26·0 and 26·5° C; P < 0·01). The thermal tolerance of this northern population appears to be broader than southern populations.  相似文献

Various haematological parameters have been measured in a sample of wild African elephants from two game parks in Uganda. The results are compared with those of other authors for the African and Indian elephant and other mammals. No intersex differences were found to occur but calves (1–5 years) differed from the population as a whole in several respects. Some values varied with season and this may be attributed to the effect of dehydration during the dry season. A number of elephants from one of the parks had abnormally high red cell numbers and the possibility is considered that this may reflect an incidence of habitat induced cardiovascular disease.  相似文献

Oxygen consumption, rectal temperature, thermal conductance, and evaporative water loss (EWL) were determined in resting captive Tatera leucogaster at ambient temperatures of between 14 and 38 °C. Basal metabolic rate (BMR) was 0.86 ml O2. min−1 (S.D.=0.15, n = 6), 45% of that expected for a rodent of the same body mass (106.2 g). Minimum wet thermal conductance was 0.21 ml O2. min−1, °C−1 (S.D. = 0.01, n = 6), 113% of that expected for a mammal of the same body mass. Wet thermal conductance increased exponentially at temperatures greater than 32 °C. Mean rectal temperature was 35.3 °C below 35 °C (S.D. = 0.5, n = 6) and 39.3 (S.D. = 0.6, n = 5) at 38 °C. Mean resting EWL was 1.43 mg. min−1 (S.D. = 0.14, n = 6) between 15 and 32 °C and increased dramatically at temperatures above 32 °C. Combining our data with data from the literature suggests that gerbils (Family Muridae; subfamily Gerbillinae) have, on average, low BMR and average minimum wet thermal conductance when compared to other rodents and other mammals, respectively, of the same body mass. Similarly, rodents (including gerbils) from arid habitats have, on average, lower rates of EWL when at rest below thermoneutrality than do other rodents of the same body mass from mesic habitats.  相似文献

The digestive tract of elephants is surprisingly short compared to other herbivorous mammals. However, measurements relating the length of the intestine to the body mass of the respective individual are rare. In this study, we report such data for an African elephant and an Asian elephant. Our data support the hypothesis that Asian elephants have a longer intestinal tract than their African counterparts. These findings are in accord with the observation of longer retention times and higher digestion coefficients in Asian as compared to African elephants. This difference between the species could be the reflection of slightly different ecological niches, with Asian elephants adapted to a natural diet with a higher proportion of grass.  相似文献

Survival, recoverability and sublethal injury of two strains of Listeria monocytogenes , Scott A and an environmental strain KM, on exposure to sea water at 12·8 or 20·8 °C was determined using in situ diffusion chambers. Plate counts were used to assess recoverability and injury while 5-cyano-2,3-ditolyl tetrazolium chloride (CTC) reduction was used to determine respiratory activity. T90 values (times for 10-fold decreases in numbers of recoverable cells) on non-selective medium (trypticase soya agar with 0·6% yeast extract) at 12·8 and 20·8 °C were 61·7 and 69·2 h for L. monocytogenes Scott A, and 103·0 and 67·0 h for L. monocytogenes KM, respectively. On selective medium (Oxford agar), T90 values at 12·8 and 20·8 °C were 60·6 and 56·9 h for L. monocytogenes Scott A, and 83·0 and 65·9 h for L. monocytogenes KM, respectively. With Scott A, the percentage of sublethally injured cells at 12·8 and 20·8 °C was 1·7 and 17·7%, respectively, while for KM the values were 19·0 and 1·6%, respectively. The fraction of cells reducing CTC but which were not recoverable on plating progressively increased on exposure to sea water. Listeria monocytogenes KM challenged at 58 °C showed an apparent increase in heat resistance after exposure to sea water at 20·8 °C for 7 d ( D 58= 2·64 min) compared with before exposure ( D 58= 1·24). This increase in thermal resistance was not apparent at temperatures greater than 63 °C, and analysis of the best-fit regression lines fitted to the thermal data obtained from the two cell populations indicated that their thermal resistance was not significantly different ( P > 0·05) over the temperature range tested (58–62 °C).  相似文献

Most African elephant (Loxodonta africana africana) populations are isolated and thus threatened by a loss of genetic diversity. As a consequence, genetic analysis of African elephant populations will play an increasing role in their conservation, and microsatellite loci will be an important tool in these analyses. Previously published sets of polymorphic microsatellites developed for African elephants are all dinucleotide repeats, which are prone to typing error. Here, we characterize 11 tetranucleotide microsatellite loci in the African elephant. All loci were polymorphic in 32 faecal samples and two tissue samples from 33 individual African savannah elephants.  相似文献

Among mammals, non-offspring nursing is the most extreme form of communal parenting. This is because lactation is the most energetically costly part of parental investment (Clutton-Brock, 1991; Packer, Lewis & Pusey, 1992). Non-offspring nursing is most common in species characterized by large litters and small kin groups (Packer et al ., 1992; e.g. lions Panthera leo : Pusey & Packer, 1994). Although non-offspring nursing has also been reported in monotocous species (e.g. water buffalo Bubalus bubalus , Murphey et al ., 1995; African elephant Loxodonta africana : Dublin, 1983; Lee, 1987; Indian elephant Elaphus maximus : MacKay, 1973; Rapaport & Haight, 1987; fallow deer Cervus dama : San José & Braza, 1993) it is almost always associated with reproductive errors (Riedman, 1982) such as milk theft or exclusive adoption (Packer et al ., 1992). However, simultaneous non-offspring nursing in monotocous species has been reported in some bat species (e.g. McCracken, 1984; Eales, Bullock & Slater, 1988), African elephants (Lee, 1987), and captive Indian elephants (Rapaport & Haight, 1987). Recent research, however, suggests that nutritive non-offspring nursing in African elephants is rarer than previously thought as most reported instances were probably non-lactating juveniles allowing infants to suckle (Lee & Moss, 1986; Lee, 1987, 1989).  相似文献

We provide here unique data on elephant skeletal ontogeny. We focus on the sequence of cranial and post-cranial ossification events during growth in the African elephant (Loxodonta africana). Previous analyses on ossification sequences in mammals have focused on monotremes, marsupials, boreoeutherian and xenarthran placentals. Here, we add data on ossification sequences in an afrotherian. We use two different methods to quantify sequence heterochrony: the sequence method and event-paring/Parsimov. Compared with other placentals, elephants show late ossifications of the basicranium, manual and pedal phalanges, and early ossifications of the ischium and metacarpals. Moreover, ossification in elephants starts very early and progresses rapidly. Specifically, the elephant exhibits the same percentage of bones showing an ossification centre at the end of the first third of its gestation period as the mouse and hamster have close to birth. Elephants show a number of features of their ossification patterns that differ from those of other placental mammals. The pattern of the initiation of the ossification evident in the African elephant underscores a possible correlation between the timing of ossification onset and gestation time throughout mammals.  相似文献

The chief objective was to determine the critical thermal limits for alevins, fry and parr of Arctic charr, Salvelinus alpinus , (L.) from four races living in Windermere (northwest England). The experimental fish were reared in a hatchery but were the progeny of wild parents. As comparisons between tethal temperatures at four acclimation temperatures (5, 10, 15, 20° C) revealed few significant racial differences, the data were pooled to estimate the lethal values for survival over 7 days (incipient lethal temperature) and over only 10 min (ultimate lethal temperature) for each life stage. Upper lethal values increased with acclimation temperatures for alevins but this effect was negligible for fry and parr, Alevins were generally less tolerant than fry and parr at lower, but not higher, acclimation temperatures; e.g. after acclimation at 5° C, mean upper ultimate values were 23·3, 25·1 and 25·7° C and mean upper incipient values were 18·7, 21·5 and 21·5° C for alevins, fry and parr respectively; after acclimation at 20° C, mean upper ultimate and incipient values were 26·2, 26·1 and 26·6° C and 20·8, 20·8 and 21·6° C for alevins, fry and parr respectively. The area of the temperature tolerance polygon (expressed as ° C2) for juvenile Arctic charr is amongst the lowest recorded for salmonids; being 409, 439 and 461° C2 for alevins, fry and parr respectively. These low values are due to lower upper tolerance limits, not high lower tolerance limits; the latter being close to 0° C (<1°C for parr and fry, <0·3° C for alevins) at all acclimation temperatures. Arctic charr are therefore amongst the least resistant of salmonids to high temperatures but probably the most resistant to low temperatures.  相似文献

The effect of temperature on thermoregulation, metabolism, evaporative water loss and thermal conductance was studied in Gerbillus pusillus . Its resting body temperature (TB) was 34·6°C, approximately 5°C higher than the mean ambient temperature (TA) encountered in its burrow. As TA increased above 34°C, its ability to lose heat to the environment decreased. It overcame this problem by tolerating increases in TB to a non-lethal maximum of 41°C, whilst also eliminating increasing quantities of obligate heat by pulmocutaneous evaporation and conduction.
Metabolic rate was 41% lower than that predicted from Kleiber's (1975) allometric equation. This confers a considerable saving in energy in an environment where food is often scarce, whilst simultaneously reducing heat production and the degree of gaseous exchange in the already oxygen-poor and carbon dioxide-rich environment encountered in the plugged burrows of its natural milieu.
Gerbillus pusillus , therefore, does not maintain strict homeothermy and utilizes a labile TB and reduced metabolic rate as an adaptive mechanism for survival in the arid zones of tropical Africa.  相似文献

Rate of metabolism, body temperature, wet thermal conductance, and evaporative water loss were measured at different ambient temperatures in four lesser gymnures Hylomys suillus. Gymnures responded as typical endothermic homoiotherms to changes in ambient temperature. Below the lower critical temperature of 32°C, they maintained a body temperature of 37.3± 0.3°C by an increased rate of metabolism. Minimum wet thermal conductance was 111% of that expected on the basis of body mass. Average basal rate of metabolism was 1.04 ml O2 g−1 h−1, which represents 106% of the expected value. Within and above the thermoneutral zone, heat loss by evaporation did not account for more than 30% of the heat produced. As a consequence, the body temperature of gymnures was maintained 4°C above ambient temperature. These metabolic and thermoregulatory patterns differ strikingly from those of other members of the family Erinaceidac and can be interpreted as a result of physiological adaptation to a different ecology. Being smaller than hedgehogs and inhabiting montane tropical rainforests, lesser gymnures lack the physiological traits which enable many hedgehogs to invade hot, arid and/or strongly seasonal environments.  相似文献

We describe scaling of morphological variables that influence total insulation in eight species of marine mammals ranging in average size from 35 to 30000 kg. We also calculate total heat loss and the partitioning of heat loss through the body surface and appendages. For the eight species investigated, heat loss in 0°C water is appreciably higher than the predicted basal metabolic rates for small species such as the ringed seal. Rorquals, on the other hand, will probably not need to raise their metabolic rates to keep warm. At rest, 10–30% of the heat production of a resting animal is lost through flippers, fins and flukes. This amount can increase to 70–80% during moderate exercise. Whole-body conductance scales with body size in the same way in marine as in terrestrial mammals, although conductance is higher for a given body size in a marine mammal.  相似文献

Aim Large, charismatic and wide‐ranging animals are often employed as focal species for prioritizing landscape linkages in threatened ecosystems (i.e. ‘connectivity conservation’), but there have been few efforts to assess empirically whether focal species co‐occur with other species of conservation interest within potential linkages. We evaluated whether the African elephant (Loxodonta africana), a world‐recognized flagship species, would serve as an appropriate focal species for other large mammals in a potential linkage between two major protected area complexes. Location A 15,400 km2 area between the Ruaha and Selous ecosystems in central Tanzania, East Africa. Methods We used walking transects to assess habitat, human activity and co‐occurrence of elephants and 48 other large mammal species (> 1 kg) at 63 sites using animal sign and direct sightings. We repeated a subset of transects to estimate species detectability using occupancy modelling. We used logistic regression and AIC model selection to characterize patterns of elephant occurrence and assessed correlation of elephant presence with richness of large mammals and subgroups. We considered other possible focal species, compared habitat‐based linear regression models of large mammal richness and used circuit theory to examine potential connectivity spatially. Results Elephants were detected in many locations across the potential linkage. Elephant presence was highly positively correlated with the richness of large mammals, as well as ungulates, carnivores, large carnivores and species > 45 kg in body mass (‘megafauna’). Outside of protected areas, both mammal richness and elephant presence were negatively correlated with human population density and distance from water. Only one other potential focal species was more strongly correlated with species richness than elephants, but detectability was highest for elephants. Main conclusions Although African elephants have dispersal abilities that exceed most other terrestrial mammals, conserving elephant movement corridors may effectively preserve habitat and potential landscape linkages for other large mammal species among Tanzanian reserves.  相似文献

To determine whether American martens Martes americana behaviourally thermoregulate in winter, and to estimate the energetic cost of resting in various forest microhabltats, we studied microenvironments used by martens in a coniferous forest in the Medicine Bow Mountains, Wyoming We hypothesized that martens would rest in sites that minimized energetic costs, and that the relative thermal efficiency of sites would vary as a function of snow depth and weather By measuring heat loss from a heated model and calculating operative temperature, we found that equivalent marten metabolic rates were inversely related to operative temperatures Operative temperatures were generally higher outside resting cavities than inside, except when snow was > 15 cm. outside air temperature was ≤−5°C. and wind was ≥0.3 m s−1 For the models lying on a snowy substrate, total body conductance was 0 32 W °C−1 23% higher than with the models lying on dry wood chips The microenvironments of resting sites and the preferences of martens generally are consistent with our hypothesis of behavioural thermoregulation Conductance contributed strongly to total thermal losses and to explaining selection of resting sites We propose a decision tree to predict where American martens should rest under a range of environmental conditions  相似文献

Disease susceptibility and resistance are important factors for the conservation of endangered species, including elephants. We analyzed pathology data from 26 zoos and report that Asian elephants have increased neoplasia and malignancy prevalence compared with African bush elephants. This is consistent with observed higher susceptibility to tuberculosis and elephant endotheliotropic herpesvirus (EEHV) in Asian elephants. To investigate genetic mechanisms underlying disease resistance, including differential responses between species, among other elephant traits, we sequenced multiple elephant genomes. We report a draft assembly for an Asian elephant, and defined 862 and 1,017 conserved potential regulatory elements in Asian and African bush elephants, respectively. In the genomes of both elephant species, conserved elements were significantly enriched with genes differentially expressed between the species. In Asian elephants, these putative regulatory regions were involved in immunity pathways including tumor-necrosis factor, which plays an important role in EEHV response. Genomic sequences of African bush, forest, and Asian elephant genomes revealed extensive sequence conservation at TP53 retrogene loci across three species, which may be related to TP53 functionality in elephant cancer resistance. Positive selection scans revealed outlier genes related to additional elephant traits. Our study suggests that gene regulation plays an important role in the differential inflammatory response of Asian and African elephants, leading to increased infectious disease and cancer susceptibility in Asian elephants. These genomic discoveries can inform future functional and translational studies aimed at identifying effective treatment approaches for ill elephants, which may improve conservation.  相似文献

Pinfish Lagodon rhomboides acclimation rates were determined by modelling changes in critical thermal minimum ( T crit min, ° C) estimates at set intervals following a temperature decrease of 3–4° C. The results showed that pinfish gained a total of 3·7° C of cold tolerance over a range of acclimation temperatures ( T acc, ° C) from (23–12° C), that cold tolerance increased with exposure time to the reduced temperature at all T acc, but that the rate of cold tolerance accruement (mean 0·14° C day−1) was independent of T acc. A highly significant ( P < 0·001) multivariate predictive model was generated that described the acclimation rates and thermal tolerance of pinfish exposed to reduction in water temperature: log10 T crit min= 0·41597 − 0·01704 T acc+ 0·04320 T plunge− 0·08376[log10 ( t + 1)], where T plunge is plunge temperature (° C) and t is the time (days). A comparison of the present data, with acclimation rate data for other species, suggests that factors such as latitude or geographic range may play a more important role than ambient temperature in determining cold acclimation rates in fishes.  相似文献

Spores of Bacillus stearothermophilus CNCH 5781 were suspended in distilled water or nutrient medium. A 28 μl aliquot of each was inoculated into haematocrit capillaries and subjected at different time intervals to sublethal temperatures of 63° or 100°C as heat activation for germination. This was followed by heat treatment at 121·1°C and the heat parameter D 121·1 was measured. Contrary to standard observations, heat resistance was observed to increase following activation, a phenomenon which we named 'heat-induced resistance'.  相似文献

Listeria monocytogenes strains 1151 and Scott A were grown in broth at 30 °C and transferred to half cream, double cream and butter stored at 5 °C to determine the influence of dairy product composition on heat resistance at 52, 56, 60, 64 and 68 °C. Strain 1151 showed a higher heat resistance than strain Scott A. The heat resistance of both strains was higher in the dairy products than in broth, particularly at lower temperatures. A significant difference was observed between log 10 of the D -values in the different dairy products. The D -values obtained for both strains resuspended in all the dairy products would result in efficient elimination of the pathogen at 72·7 °C for 15 s. The highest D -value was 11·30 s at 68 °C and by using a z -value of 6·71 °C it can be determined that at 72·7 °C the D -value would be 1·5 s. The 15 s process would therefore achieve 10 log reductions. The effect of growth conditions on the heat resistance at 60 °C of L. monocytogenes Scott A was also investigated. When the cells were grown in the dairy products themselves, and particularly butter, the heat resistance of Scott A was enhanced; for example, the D -values were 7·15 times higher than in broth. Further studies are required to investigate if this protection against heating exists at higher temperatures, in which case the efficiency of pasteurization treatments or other heat treatments would be considerably lowered.  相似文献
