为了解濒危兰科植物小叶兜兰(Paphiopedilum barbigerum Tang et Wang)胚珠和雌配子体的发育过程,采用常规石蜡切片技术对其果实的生长动态进行了研究。结果表明,授粉后60~75 d的蒴果内种子数量迅速增加,到授粉后120 d时种子充满整个蒴果。授粉后40 d的胎座上分化形成多数由1层表皮细胞包被1列细胞的胚珠原基;授粉后60 d时位于胎座指状结构末端处紧靠表皮细胞下方的孢原细胞分化为大孢子母细胞。之后,大孢子母细胞经过减数分裂和有丝分裂最终形成成熟胚囊;授粉后135 d胚囊发育成熟,附着在胎座上的种子个体分化明显。小叶兜兰胚囊的发育类型为双孢子葱型,胚珠为倒生胚珠,薄珠心,单珠被,成熟胚囊为8核。这为小叶兜兰的生殖生物学及繁殖体系的建立提供理论依据。  相似文献

应用改进的整体染色透明激光扫描共聚焦显微术(WCLSM),对同源四倍体水稻PDER-2B-4x胚囊的形成与发育过程进行观察。发现其胚囊的形成发育过程与二倍体的一致,可以清楚地划分为8个发育时期,即孢原细胞形成期、大孢子母细胞形成期、大孢子母细胞减数分裂期、功能大孢子形成期、单核胚囊形成期、胚囊有丝分裂期、八核胚囊发育期和成熟胚囊期。除正常发育的过程外,大孢子发育的各个过程均出现一些异常现象,包括:细胞退化、核位置异常、核数目异常和细胞分化异常等。这些异常可能最终导致多种结构异常成熟胚囊的形成。  相似文献

五唇兰(Doritispulcherrima Lindl.)的胚珠属于倒生型,具薄珠心,两层珠被.胚囊发育类型为双孢子葱型,授粉后约45 d形成七细胞八核的成熟胚囊.五唇兰未受精胚珠在离体培养初期对外源激素的依赖性很小,在没有外源激素的培养基上,大孢子母细胞也能经过减数分裂发育为二核胚囊.在培养后期,外源激素对胚囊发育的影响很大.在培养基无外源激素或仅含生长素或细胞分裂素时,雌配子体的发生过程不能顺利完成;在改良VW培养基上添加0.5 mg/L BA和0.1 mg/LNAA时,形成成熟胚囊.  相似文献

采用常规石蜡切片法,对车桑子大孢子的发生和雌配子体的发育进行观察,探讨车桑子自然结籽率低的原因和明确其胚胎发育特征。结果表明:(1)车桑子花柱有花柱道,子房3室,中轴胎座,横生胚珠,每心室两枚胚珠,双珠被,厚珠心,无承珠盘。(2)位于珠心表皮细胞下的孢原细胞经平周分裂产生造孢细胞,造孢细胞发育为大孢子母细胞,大孢子母细胞经减数分裂形成线性四分体,靠近珠孔端3个大孢子退化消失,靠合点端大孢子发育为功能大孢子,大孢子发生类型为单孢子发生型。(3)单核胚囊经3次有丝分裂形成7细胞8核的成熟胚囊,胚囊发育类型为蓼型。(4)花器官形态的变化和大孢子发育过程有一定联系,可根据雌花形态特征大致判断大孢子发育时期。研究认为,车桑子雌配子体发育过程中出现的胚囊不中空、游离核不进一步细胞化等异常现象,可能是导致车桑子自然结籽率低的原因之一。  相似文献

本试验结果表明 ,雌雄配子体形成过程和程序与前人结论相一致 ,但发现 :1 )尽管雌雄蕊同时形成 ,但在雌雄配子形成不同步的情况下 ,花粉和胚囊中的大量淀粉却能同时充实 ,并且同时成熟等待受精。 2 )大孢子母细胞形成比小孢子母细胞迟 0 .7~ 0 .8片展叶 ;雌配子体形成比雄配子体迟 0 .5~ 0 .6片展叶。 3)雌雄两性细胞发育特点不同 ,雄性是小孢子形成快而花粉粒发育慢 ,雌性是大孢子形成慢而胚囊发育快 ,为此在所用时间上 ,花粉粒发育约是小孢子形成的 1 .6倍 ,是胚囊发育的 2 .3倍 ,而大孢子形成约是胚囊发育的 2 .2倍 ,约是小孢子形成的 1 .4倍。  相似文献

以20年生白桦(Betula platyphylla)的雌花序为材料,研究了白桦的大孢子发生、雌配子体形成和胚胎发育过程.结果表明:1)大孢子母细胞经过减数分裂形成直线排列的4个大孢子,其中珠孔端的3个大孢子退化,合点端的大孢子进行3次有丝分裂,形成八核胚囊,8个核迅速细胞化,最终发育为成熟胚囊.胚囊发育类型为单孢子蓼型胚囊;2)胚的发育经过原胚、球形胚、心形胚和鱼雷胚等阶段发育为成熟胚;胚柄在球形胚时期最发达,但胚柄短小,只由3个扁平细胞组成,之后逐渐退化.本文同时对雌花的结构和发育特点在桦木科各属中的异同、白桦胚胎发生类型及胚胎发育进程的同步性进行了讨论.  相似文献

应用电子显微镜对鹤顶兰(Phaius tankervilliae(Aiton)Bl.)珠心细胞进行了观察,结果发现,珠心细胞程序死亡(programmed cell death,PCD)过程中伴随着液泡破裂、染色质凝聚、细胞质解体等明显特征。在鹤顶兰功能大孢子形成之前,大孢子母细胞的侧细胞壁存在明显的内突。随着胚囊体积的逐渐增大,衰退珠心细胞残留的细胞壁叠合在一起,从而使胚囊壁不断加厚。胚囊成熟前,合点端珠心细胞与胚囊之间有胞间连丝相连。合点端珠心细胞的细胞质状态,特别是液泡形态与大孢子母细胞、功能大孢子、成熟胚囊时期的细胞状态高度相似。结果表明,衰退的珠心细胞不仅为胚囊的扩大提供空间,同时也为胚囊的发育提供营养,合点端珠心细胞对胚囊发育内环境的稳定性起着重要的屏障作用。  相似文献

采用半薄切片技术和组织化学染色法对宁夏枸杞大孢子发生和雌配子体发育过程中的细胞结构变化及营养物质积累特征进行了观察。结果表明,(1)宁夏枸杞为中轴胎座,多室子房,倒生胚珠,单珠被,薄珠心类型。(2)位于珠心表皮下的孢原细胞可直接发育为大孢子母细胞,减数分裂后形成直线型大孢子四分体,合点端第一个大孢子发育为功能大孢子,胚囊发育类型为蓼型,具有珠被绒毡层。(3)初形成的胚囊外周组织中没有营养物质积累,成熟胚囊时期出现了大量的淀粉粒且呈珠孔端明显多于合点端的极性分布特征。(4)助细胞的珠孔端具有明显的丝状器结构,呈PAS正反应表现出多糖性质,成熟胚囊具有承珠盘结构。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

A complex analysis of seasonal fluctuations of the mean group parameters of the system of regulation of lipid peroxidation has been performed in liver of Balb/c mice. Association of lipid characteristics and morphophysiological parameters is studied in the Balb/c mouse liver. An inter-connection is revealed between the liver index and the amount of lysoforms of phospholipids, the scale and character of the interconnection differing essentially depending on proportion of phos-phatidylcholine in mouse liver phospholipids.  相似文献

Seven strains of aerobic carbon monoxide-oxidizing bacteria (carboxydebacteria) when growing on CO as sole source of carbon and energy had doubling times which ranged from 12–42 h. The activity profiles obtained after discontinuous sucrose density gradient centrifugation indicated that the CO-oxidizing enzymes are soluble and the hydrogenases are membrane-bound in all strains examined. The CO-oxidizing enzymes of Pseudomonas carboxydohydrogena, Pseudomonas carboxydoflava, Comamonas compransoris, and the so far unidentified strains OM2, OM3, and OM4 had a molecular weight of 230,000; that of Achromobacter carboxydus amounted to 170,000. The molecular weights of the CO-oxidizing and H2-oxidizing enzymes turned out to be identical. The cell sonicates were shown to catalyze the oxidation of both CO and H2 with methylene blue, thionine, phenazine methosulfate, toluylene blue, dichlorophenolindophenol, cytochrome c or ferricyanide as electron acceptors. Methyl viologen, benzyl viologen, FAD+, FMN+, and NAD(P)+ were not reduced. The spectrum of electron acceptors was identical for all strains tested. Neither free formate, hydrogen nor oxygen gas were involved in the CO-oxidation reaction. Methylene blue was reduced by CO at a 1:1 molar ratio. The results indicate that CO-oxidation by carboxydobacteria is catalyzed by identical or similar enzymes and that the reaction obeys the equation CO+H2OCO2+2H++2e- as previously shown for Pseudomonas carboxydovorans.Dedicated to Otto Kandler remembering almost three decades of enjoyable cooperation  相似文献

Since the introduction of the concepts of allostery about four decades ago, much advancement has been made in elucidating the structure-function correlation in allostery. However, there are still a number of issues that remain unresolved. In this review we used mammalian pyruvate kinase (PK) as a model system to understand the role of protein dynamics in modulating cooperativity. PK has a triosephosphate isomerase (TIM)(α/β)8 barrel structural motif. PK is an ideal system to address basic questions regarding regulatory mechanisms about this common (α/β)8 structural motif. The simplest model accounting for all of the solution thermodynamic and kinetic data on ligand-enzyme interactions involves two conformational states, inactive ET and active ER. These conformational states are represented by domain movements. Further studies provide the first evidence for a differential effect of ligand binding on the dynamics of the structural elements, not major secondary structural changes. These data are consistent with our model that allosteric regulation of PK is the consequence of perturbation of the distribution of an ensemble of states in which the inactive ET and active ER represent the two extreme end states. Sequence differences and ligands can modulate the distribution of states leading to alterations of functions. The future work includes: defining the network of functionally connected residues; elucidating the chemical principles governing the sequence differences which affect functions; and probing the nature of mutations on the stability of the secondary structural elements, which in turn modulate allostery.  相似文献

对我国花型Hemibarbuspattern鱼类作了骨骼系统比较,结果表明,此类型鱼类脑颅较长,副蝶骨平直或稍弯曲,眶蝶骨腹纵嵴发达(铜鱼Coreius septentrionalis例外),下颞窝和咽突中等大,基枕骨后突发达;脑颅中的上筛骨的后突、侧筛骨的外筛突,蝶耳骨的外突、上耳骨的后突、围眶骨和后颞窝等均有明显的差异;咽颅中的舌颌骨、尾舌骨、鳃盖骨和下咽齿的列数等又有显著的区别;附肢骨骼中的腰带骨、脊椎骨中的复合神经骨和第4椎骨腹侧的悬器等也有不同之处。据此,这些差异和区别可作为属间或种间的分类依据。  相似文献
