Secoiridoid glycosides are naturally occurring phytochemicals of great importance for the food and pharmaceutical industry because of their various biological activities. Certain Gentiana and Centaurium species, which are recognized as the most important sources of these compounds, have become critically endangered due to overexploitation. In this study we describe a laboratory-scale approach for further implementation in large-scale production of secoiridoid glycosides, using a hairy root culture system of Centaurium maritimum L. Fritch, an underutilized and phytochemically unexplored species. Hairy roots were induced by Agrobacterium rhizogenes strain A40M70GUS and grown in Erlenmeyer flasks, as well as in RITA® temporary immersion bioreactors (TIBs). About 2–4 times higher biomass production rate and up to 8 times higher total secoiridoid glycosides production rate were achieved in RITA® bioreactors. Among the selected hairy root lines, line HR3 cultured in RITA® TIBs proved to be the most efficient considering both biomass and secoiridoid glycosides production rate.  相似文献

In this study, we investigated how neighbors (i.e., competitors) altered resistance phenotypes, namely plant size and levels of secondary compounds (iridoid glycosides), of individual plants and specifically tested whether neighbor identity mattered. We conducted a greenhouse experiment with Plantago lanceolata and Plantago major (Plantaginaceae) in which each species served as focal plants as well as neighbors in a factorial design. In addition, we harvested plants six and nine weeks after transplantation to test whether effects changed as plants grew. In both species, competition reduced plant size, and this effect increased over time. Plantago lanceolata neighbors suppressed growth of both focal plant species more than P. major neighbors. Effects of competition on levels of secondary compounds were more complex. Concentrations of iridoid glycosides were increased by competition in both species at harvest one. By the second harvest, an effect of competition on iridoid glycosides was found only in P. major. Neighbor identity influenced levels of iridoid glycosides in P. lanceolata at harvest one; concentrations were higher in plants grown with P. lanceolata neighbors than in plants grown with P. major neighbors. We also tested whether there was a trade-off between growth (biomass) and defense (levels of iridoid glycosides). Biomass and iridoid glycoside content were significantly correlated only in plants grown with competition and harvested at nine weeks, and this relationship was positive in both species, indicating that there was no trade-off between growth and defense. This study suggests that neighbor identity could play an important role in interspecific interactions, including the interactions of plants with other trophic levels.  相似文献

Eight species of Mentzelia (Loasaceae) have been investigated for iridoid glycosides. In addition to the known glucosides deutzioside, decaloside, mongolioside, loganin and sweroside, several novel compounds have been isolated and characterized by chemical and spectroscopic means. 6′-O-Acetyl deutzioside was found in a single species, while the diglycosidic compounds glucosyl-decaloside, allosyl-decaloside and quinovosyl-decaloside were each isolated from one or more species. In addition, a novel compound, epoxydecaloside (= 11-hydroxy-deutzioside), together with glucosyl-epoxydecaloside, allosyl-epoxydecaloside and mentzelosyl-epoxydecaloside are described. The last compound contains a 4-deoxy-α-l-erythro-pentopyranosyl moiety, whose parent sugar, named mentzelose, has not been encountered so far in nature. A non-glycosidic iridoid, mentzetriol, has been characterized solely by spectroscopic means and a structure is proposed. The secoiridoid secoxyloganin has been found for the first time in a plant source, and the coumarin glucoside scopolin has been isolated from two species of Mentzelia. 13C and 1H NMR spectra of several iridoid compounds are presented. The biosynthesis of the compounds is considered and the systematic position of Loasaceae discussed concluding in a possible derivation from Cornalean ancestors.  相似文献

The Tibetan folk medicine Qinjiao is traditionally used to treat various conditions, and its main active constituents comprise four iridoid glycosides, i.e., loganic acid, swertiamarin, gentiopicroside, and sweroside. The traditional crude medicine Qinjiao is derived from the dried roots of three species belonging to Gentiana sect. Cruciata (Gentianaceae) growing in the Qinghai‐Tibetan Plateau (QTP). In this study, we determined by HPLC the contents of the four main active constituents in the dried roots collected from 83 localities at different altitudes across the QTP. The material was classified under the seven taxonomic species G. straminea, G. dahurica, G. crassicaulis, G. waltonii, G. officinalis, G. ihassica, and G. macrophylla. Our results suggested that the four constituents were present in the roots of all seven species for all localities, but their concentrations varied greatly within and between species. The level of gentiopicroside revealed to be the most dominant for all examined localities (2.1–12.4 mg/g), and G. macrophylla Pall . contained the highest concentration of all the four constituents at the species level. Except for loganic acid in G. officinalis, there was no significant correlation between the contents of these constituents and the altitude of the sampling localities. These results suggest that all species of all origins can be used as reliable resource for the crude medicine Qinjiao. However, a few species contain higher concentrations of the main active constituents, irrespective of their origin.  相似文献

Gentiana crassicaulis Duthie ex Burk . and Gentiana macrophylla Pall . are two main sources of Radix Gentianae Macrophyllae (Qinjiao) available in markets, which has a wide range of anti‐inflammatory effects and has been extensively used for fighting rheumatoid arthritis. However, they vary in terms of chemical compositions, pharmacological activities, and biomass. In this study, a combined chemical and genetic (HPLC and DNA barcoding) approach was used to compare these two plants. Four predominant bioactive compounds, namely, gentiopicroside, loganic acid, swertiamarin, and sweroside, were used to assess the chemical variations. Based on chemical variations, 15 samples were clustered into two groups through PCA analyses. DNA barcoding utilizing the variable nuclear ITS2 regions were sequenced, aligned, and compared. Together with 61 sequences collected from GenBank, 76 batches of Qinjiao were clustered in two groups according to species origin. The genetic relationships indicated by the ITS2‐based NJ tree were consistent with the chemical variations. Thus, the chemical profiles determined by HPLC and DNA profiles obtained from ITS2 region could be applied for the quality control of Qinjiao.  相似文献

DNA barcoding of plants poses particular challenges, especially in differentiating, recently diverged taxa. The genus Gentiana (Gentianaceae) is a species-rich plant group which rapidly radiated in the Himalaya-Hengduan Mountains in China. In this study, we tested the core plant barcode (rbcL + matK) and three promising complementary barcodes (trnH-psbA, ITS and ITS2) in 30 Gentiana species across 6 sections using three methods (the genetic distance-based method, Best Close Match and tree-based method). rbcL had the highest PCR efficiency and sequencing success (100%), while the lowest sequence recoverability was from ITS (68.35%). The presence of indels and inversions in trnH-psbA in Gentiana led to difficulties in sequence alignment. When using a single region for analysis, ITS exhibited the highest discriminatory power (60%-74.42%). Of the combinations, matK + ITS provided the highest discrimination success (71.43%-88.24%) and is recommended as the DNA barcode for the genus Gentiana. DNA barcoding proved effective in assigning most species to sections, though it performed poorly in some closely related species in sect. Cruciata because of hybridization events. Our analysis suggests that the status of G. pseudosquarrosa needs to be studied further. The utility of DNA barcoding was also verified in authenticating ‘Qin-Jiao’ Gentiana medicinal plants (G. macrophylla, G. crassicaulis, G. straminea, and G. dahurica), which can help ensure safe and correct usage of these well-known Chinese traditional medicinal herbs.  相似文献

The essential oils (EOs) extracted from four species of the genus Guatteria, G. australis, G. ferruginea, G. latifolia, and G. sellowiana were analyzed. A total of 24, 22, 25, and 19 constituents of the oils from four species, respectively, were identified by GC/MS. These oils showed qualitative and quantitative differences. All the oils contained the oxygenated sesquiterpenes spathulenol (11.04 – 40.29%) and caryophyllene oxide (7.74 – 40.13%) as predominant constituents. Evaluation of antiproliferative activity of the EOs showed strong selectivity (1.1 – 4.1 μg/ml) against the tumor cell line OVCAR‐03 (ovarian cancer), i.e., more active than the positive control doxorubicin (11.7 μg/ml). All EOs showed strong antibacterial activity (minimum inhibitory concentrations of 0.062 – 0.25 mg/ml) against strains of Rhodococcus equi.  相似文献

Iridoid glycosides are plant defence compounds that are deterrent and/or toxic for unadapted herbivores but are readily sequestered by dietary specialists of different insect orders. Hydrolysis of iridoid glycosides by β‐glucosidase leads to protein denaturation. Insect digestive β‐glucosidases thus have the potential to mediate plant–insect interactions. In the present study, mechanisms associated with iridoid glycoside tolerance are investigated in two closely‐related leaf beetle species (Coleoptera: Chrysomelidae) that feed on iridoid glycoside containing host plants. The polyphagous Longitarsus luridus Scopoli does not sequester iridoid glycosides, whereas the specialist Longitarsus tabidus Fabricius sequesters these compounds from its host plants. To study whether the biochemical properties of their β‐glucosidases correspond to the differences in feeding specialization, the number of β‐glucosidase isoforms and their kinetic properties are compared between the two beetle species. To examine the impact of iridoid glycosides on the β‐glucosidase activity of the generalist, L. luridus beetles are kept on host plants with or without iridoid glycosides. Furthermore, β‐glucosidase activities of both species are examined using an artificial β‐glucosidase substrate and the iridoid glycoside aucubin present in their host plants. Both species have one or two β‐glucosidases with different substrate affinities. Interestingly, host plant use does not influence the specific β‐glucosidase activities of the generalist. Both species hydrolyse aucubin with a much lower affinity than the standard substrate. The neutral pH reduces the β‐glucosidase activity of the specialist beetles by approximately 60% relative to its pH optimum. These low rates of aucubin hydrolysis suggest that the ability to sequester iridoid glycosides has evolved as a key to potentially preventing iridoid glycoside hydrolysis by plant‐derived β‐glucosidases.  相似文献

Introduction – The popular use of black cohosh products (Actaea racemosa L., syn. Cimicifuga racemosa L.) is growing as the demand for alternatives to estrogen therapy has increased. Critical to safe use is the assurance of unadulterated, high‐quality products. Questions have been raised about the safety of black cohosh due to cases of liver toxicity in patients who reported taking it; subsequent evaluation found some products to be adulterated with other related herbal species. Correct plant species identification is a key first step for good manufacturing practices of safe black cohosh products. Objectives – To develop analytical methods which distinguish black cohosh from other species (American and Asian) of Actaea increasingly found as adulterants in commercially available black cohosh products. Material and methods – Fifteen species of Actaea were collected from North America and Asia, and the phytochemical fingerprints of these samples were established using HPLC‐PDA and LC‐MS techniques. Results – The HPLC and LC‐MS fingerprints for polyphenols and triterpene glycosides revealed distinct patterns that make black cohosh clearly distinguishable from most other species of Actaea. Two marker compounds, cimifugin and cimiracemoside F, were found to be important to distinguish black cohosh from most Asian species of Actaea. Formononetin was not found from either Asian or American species of Actaea. Conclusions – Phytochemical fingerprinting is a practical, reliable method for authenticating black cohosh and distinguishing it from other species of Actaea increasingly found as adulterants in commercially available black cohosh products. Introduction – Seeds of wild Peganum harmala Linn., P. multisectum (Maxim) Bobr., P. nigellastrum Bunge and a probable indeterminate species, herein referred to as P. variety, are commonly used in Chinese medicine. These seeds cannot be differentiated based on morphology. Objective – Seeds of P. harmala Linn., P. multisectum (Maxim) Bobr., P. nigellastrum Bunge and P. variety were collected in different provinces in China and their HPLC profiles were recorded for statistical analysis and pattern recognition. Methodology – HPLC chromatograms of seed extracts were recorded under the same conditions. Individual HPLC chromatograms for each species were evaluated against the mean chromatogram for the same species generated using a similarity evaluation computer program. Data from chromatographic fingerprints were also processed using principal component analysis (PCA), hierarchical cluster analysis (HCA) and linear discriminant analysis (LDA). Results – The Peganum sp. seed extracts had similar HPLC fingerprints but with some inter‐specific differences. The chromatographic fingerprints combined with PCA, HCA and LDA could distinguish the seeds of the different species of Peganum investigated. Conclusion – HPLC fingerprints can be used to authenticate and differentiate the seeds of three different species of genus Peganum indigenous to China. G. macrophylla pall, G. dahurica Fisch, G. straminea Maxim, and G. crassicaulis Duthie ex Burk are the major cultivated Gentiana species in Gansu Province, China. The present study has collected 107 wild samples from 19 natural populations and focused on the analysis of their medicinal ingredients, genetic diversity, and relationship, concerning four species’s worrying situation and solutions. High performance liquid chromatography (HPLC) results show that their gentiopicroside contents are 9.75 ± 2.0%, 9.29 ± 2.6%, 8.18 ± 3.2%, and 7.96 ± 3.2% respectively and higher than the criterion standard for Radix Gentianae in Chinese Pharmacopoeia. Low genetic polymorphism was found among these four species with HE (expected heterozygosity) = 0.147, GST (interspecies coefficient of gene differentiation) = 0.3207, and Nm (gene flow) = 1.0593. The unweighted pair group method with arithmetic mean (UPGMA) analysis was performed with POPGENE 1.31 software and Nei’s method but did not clearly differentiate these four species. Their medicinal ingredients did not correlate with the genetic variants (r = ?0.179, P = 0.586). The results indicate that there is no significant difference among four species in regard to the traits of medicinal qualities. All four Gentiana species can be widely cultivated to not only meet the demand of domestic and international market for Chinese traditional medicine, but also help the local conservation of the wild Gentiana resources in China.  相似文献

Five new saccharide fatty acid esters, named nonioside P ( 3 ), nonioside Q ( 4 ), nonioside R ( 8 ), nonioside S ( 10 ), and nonioside T ( 14 ), and one new succinic acid ester, butyl 2‐hydroxysuccinate (=4‐butoxy‐3‐hydroxy‐4‐oxobutanoic acid) ( 31 ), were isolated, along with 26 known compounds, including eight saccharide fatty acid esters, 1, 2, 5, 6, 7, 9, 12 , and 13 , three hemiterpene glycosides, 15, 17 , and 18 , six iridoid glycosides, 21 – 25 , and 27 , and nine other compounds, 20, 28, 29 , and 32 – 37 , from a MeOH extract of the fruit of Morinda citrifolia (noni). Upon evaluation of these and five other glycosidic compounds, 11, 16, 19, 26 , and 30 , from M. citrifolia fruit extract for their inhibitory activities against melanogenesis in B16 melanoma cells induced with α‐melanocyte‐stimulating hormone (α‐MSH), most of the saccharide fatty acid esters, hemiterpene glycosides, and iridoid glycosides showed inhibitory effects with no or almost no toxicity to the cells. These compounds were further evaluated with respect to their cytotoxic activities against two human cancer cell lines (HL‐60 and AZ521) and their inhibitory effects on Epstein? Barr virus early antigen (EBV‐EA) activation induced with 12‐O‐tetradecanoylphorbol‐13‐acetate (TPA) in Raji cells.  相似文献

A rapid analytical method has been developed to determine xanthone and secoiridoid glycoside in in vitro and in vivo Swertia chirayita extracts. Ultra performance liquid chromatography–electrospray ionization mass spectrometry (LC-ESI/MS) was applied and validated for the analysis of xanthone and secoiridoid glycoside a potential active component isolated from methanolic extracts of in vitro and in vivo Swertia chirayita plantlets. Chromatographic separation was achieved on a RP-C18 column using gradient elution. Mangiferin (Xanthone), Amarogentin and Swertiamarin (Secoiridoid glycosides) were identified in both the extracts. In the LC/ESI-MS spectra, major [M + H] + and [M + Na] + ions were observed in positive ion mode and provided molecular mass information. An ultra-performance liquid-chromatography in combination with electrospray ionization tandem mass spectrometry involving metal cationisation was successfully utilized for the rapid identification of xanthone and secoiridoid glycosides. This method is suitable for the routine analysis, as well as for the separation and identification of known and novel secoiridoid glycoside and xanthone.  相似文献

Five novel iridoid glycosides, avellanedaesides A‐E ( 1  –  5 ) were isolated from the H2O extract of Tabebuia avellanedae. Their structures were determined on the basis of NMR and MS analysis. Isolated compounds suppressed inflammatory cytokine, tumor‐necrosis factor‐α and interleukin‐1β production in cultured human myeloma THP‐1 cells co‐stimulated with lipopolysaccharide (LPS). In addition, the study revealed iridoid glycosides inhibited the activity of cytochrome CYP3A4 enzyme.  相似文献

  • 2 We analysed the iridoid glycoside content of E.gillettii adults from two different populations, Warm Lake, Idaho, and Granite Creek, Wyoming, that have different patterns of hostplant use.
  • 3 Gas chromatographic analysis of thirty butterflies from the Wyoming population showed that they contained a mean of 1.27 (±0.19 SE) % dry weight iridoid glycosides. Notably, 20% of these butterflies contained no detectable iridoid glycosides.
  • 4 In contrast, nineteen butterflies from the Idaho population contained a mean of 3.89 (±0.38 SE) % dry weight iridoid glycosides, and all butterflies contained iridoid glycosides.
  • 5 These results illustrate how the chemical defence of herbivorous insects varies according to differential use of potential hostplants.

In addition to two well-known glucosides, secologanin and morroniside, two rare secoiridoids, secoxyloganin and secologanoside, have been isolated and characterized by chemical and spectroscopic means. Iridoid glycosides are plant defence compounds with potentially detrimental effects on non-adapted herbivores. Some plant species possess β-glucosidases that hydrolyse iridoid glycosides and thereby release protein-denaturing aglycones. To test the hypothesis that iridoid glycosides and plant β-glucosidases form a dual defence system, we used Plantago lanceolata and a polyphagous caterpillar species. To analyse the impact of leaf-age dependent differences in iridoid glycoside concentrations and β-glucosidase activities on insect performance, old or young leaves were freeze-dried and incorporated into artificial diets or were provided freshly to the larvae. We determined larval consumption rates and the amounts of assimilated nitrogen. Furthermore, we quantified β-glucosidase activities in artificial diets and fresh leaves and the amount of iridoid glycosides that larvae feeding on fresh leaves ingested and excreted. Compared to fresh leaves, caterpillars grew faster on artificial diets, on which larval weight gain correlated positively to the absorbed amount of nitrogen. When feeding fresh young leaves, larvae even lost weight and excreted only minute proportions of the ingested iridoid glycosides intact with the faeces, indicating that the hydrolysis of these compounds might have interfered with nitrogen assimilation and impaired larval growth. To disentangle physiological effects from deterrent effects of iridoid glycosides, we performed dual choice feeding assays. Young leaves, their methanolic extracts and pure catalpol reduced larval feeding in comparison to the respective controls, while aucubin had no effect on larval consumption. We conclude that the dual defence system of P. lanceolata consisting of iridoid glycosides and β-glucosidases interferes with the nutrient utilisation via the hydrolysis of iridoid glycosides and also mediates larval feeding behaviour in a concentration- and substance-specific manner.  相似文献

  • 1 The simultaneous effects of daytime temperature (20°C versus 30°C) and leaf age (new versus intermediate-aged) on a generalist insect herbivore were examined. Fourth-instar Spilosoma congrua caterpillars were tested on plantain (Plantago lanceolata), one of this lepidopteran species’host plants, for which the major defensive chemicals, iridoid glycosides (aucubin and catalpol), could be quantified.
  • 2 Cool temperature depressed amount of food eaten, amount of frass, and consumption and growth rates, and increased the proportion of time spent in the non-feeding period (from head-capsule slippage to ecdysis).
  • 3 Average iridoid glycoside concentration was 2.8% dry weight (d.w.) in new leaves and 1.6% d.w. in intermediate-aged leaves. When fed new leaves, the caterpillars had a higher efficiency of conversion of ingested food to biomass and a higher growth rate than those fed intermediate-aged leaves. Furthermore, the proportion of time spent in the non-feeding period was prolonged by a diet of intermediate-aged leaves.
  • 4 A second experiment showed that the percentage dry weight of aucubin, catalpol and total iridoid glycosides increased over 24 h in incubated, excised leaves, which meant that the caterpillars in the first experiment experienced somewhat higher iridoid glycoside concentrations than the levels in fresh leaves.
  • 5 Overall, these results indicate that this generalist should prefer new plantain leaves over older leaves even though new leaves contain higher concentrations of iridoid glycosides.
