We have shown previously that angiotensin-II (A-II) controls proto-oncogene (c-fos, jun-B and c-jun) mRNA accumulation in bovine adrenal fasciculata cells (BAC). Since BAC contain both subtypes (AT-1 and AT-2) of the A-II receptor, we have investigated which subtype was involved in the effect of A-II on proto-oncogene mRNA by using a selective antagonist for AT-1 (DUP 753) and for AT-2 (CGP 42112A). DUP 753, but not CGP 42112A, inhibited the stimulatory effect of A-II on proto-oncogene mRNA, with ID50s of 4 x 10(-7) M, 7 x 10(-7) M and 2 x 10(-6) M for c-fos, jun-B and c-jun, respectively. Neither of the two antagonists by themselves had a direct effect on proto-oncogene mRNA. As the A-II AT-1 receptors are coupled to the phospholipase C system in BAC, we have investigated whether the A-II effects on the proto-oncogenes were mediated by protein kinase C (PKC) or by Ca2+ calmodulin. First, activation of PKC by the phorbol ester, PMA, increased the level of three proto-oncogene mRNAs, whereas calcium ionophore had no effect. Second, staurosporine, a specific inhibitor of PKC, reduced the stimulatory action of A-II on proto-oncogene mRNA by 80-90%, whereas trifluoroperazine, an inhibitor of calmodulin, had no significant effect. These results demonstrate that the effects of A-II on proto-oncogene mRNA are mediated by AT1 receptor subtypes, mainly through activation of the PKC pathway.  相似文献

Reactive oxygen species production by bovine pulmonary alveolar macrophages was evaluated by a chemiluminescence assay utilizing luminol and opsonized zymosan. Incubation with dobutamine (5 x 10(-8) and 5 x 10(-7) M) or isoproterenol (5 x 10(-8) and 5 x 10(-7) M) prior to zymosan challenge significantly (p less than 0.05) increased the time for chemiluminescence to begin, and significantly decreased the level of maximum chemiluminescence. The agonists' inhibitory effects on maximum chemiluminescence were significantly reduced by pre-incubation with the appropriate antagonist (atenolol at 1 x 10(-6) M for dobutamine; and propranolol at 1 x 10(-6) M for isoproterenol). Salbutamol at 1 x 10(-6) M significantly reduced the level of maximum chemiluminescence only, but did not increase the time for chemiluminescence to begin. This effect was significantly reduced by the presence of the beta 2-antagonist ICI 118,551 at 1 x 10(-6) M. The results reveal the presence of beta 1- and beta 2-adrenoceptors on bovine pulmonary alveolar macrophages, and suggest that these receptors are important in the regulation of reactive oxygen species production by these cells.  相似文献

Proinflammatory cytokines are important factors in the regulation of diverse aspects of skeletal muscle function; however, the muscle cytokine receptors mediating these functions are uncharacterized. Binding kinetics (dissociation constant = 39+/-4.7 x 10(-9) M, maximal binding = 3.5+/-0.23 x 10(-12) mol/mg membrane protein) of muscle tumor necrosis factor (TNF) receptors were obtained. Skeletal muscle was found to express mRNAs encoding interleukin-1 type I and II receptors, interleukin-6 receptor (IL-6R), and interferon-gamma receptor by RT-PCR, but these receptors were below limits of detection of ligand-binding assay (> or =1 fmol binding sites/mg protein). Twenty-four hours after intraperitoneal administration of endotoxin to rats, TNF receptor type II (TNFRII) and IL-6R mRNA were increased in skeletal muscle (P<0.05). In cultured L6 cells, the expression of mRNA encoding TNFRII and IL-6R receptors was induced by TNF-alpha, and all six cytokine receptor mRNA were induced by a mixture of TNF-alpha, IFN-gamma, and endotoxin (P<0.05). This suggests that the low level of cytokine receptor expression is complemented by a capacity for receptor induction, providing a clear mechanism for amplification of cytokine responses at the muscle level.  相似文献

The effects of muscarinic receptor antagonists on responses to electrical stimulation of the chorda-lingual nerve were determined in pentobarbitone-anesthetized sheep and correlated to the morphology of tissue specimens. Stimulation at 2 Hz continuously, or in bursts of 1 s at 20 Hz every 10 s, for 10 min induced similar submandibular fluid responses (19 +/- 3 vs. 21 +/- 3 microl x min(-1) x g gland(-1)), whereas vasodilatation was greater during stimulation in bursts (-52 +/- 4 vs. -43 +/- 5%; P < 0.01). Continuous stimulation at 8 Hz induced substantially greater responses (66 +/- 9 microl x min(-1) x g gland(-1) and -77 +/- 3%). While atropine (0.5 mg/kg iv) abolished the secretory response at 2 and 20 Hz (1:10 s), a small response persisted at 8 Hz (<5%). The "M1-selective" antagonist pirenzepine (40 microg/kg iv) reduced the fluid response at all frequencies tested (P < 0.05-0.01), most conspicuously at 2 Hz (reduced by 69%). Methoctramine ("M2/M4-selective"; 100 microg/kg iv; n = 5) had no effect on fluid or the vascular responses but increased the protein output at 2 (+90%, P < 0.05) and 8 Hz (+45%, P < 0.05). The immunoblotting showed distinct bands for muscarinic M1, M3, M4, and M5 receptors, and immunohistochemistry showed muscarinic M1 and M3 receptors to occur in the parenchyma. Thus muscarinic M1 receptors contribute to the secretory response to parasympathetic stimulation but have little effect on the vasodilatation in the ovine submandibular gland. Increased transmitter release caused by blockade of neuronal inhibitory receptors of the M4 subtype would explain the increase in protein output.  相似文献

The super-toxic super-complexes (SSC) of botulinic neurotoxins, types A and B, have been isolated. The preparation of type A SSC (SSC/A) consist of the proteolyzed form of type A botulinic neurotoxin (BoNT/A), 50 and 90 kD, nontoxic nonhemagglutinating protein of 140 kD (NTNH140) in the nonproteolyzed form, hemagglutinin of 17 kD (Ha17), hemagglutinin of 34 kD (Ha34) and the proteolyzed form of hemagglutinin of 70 kD (Ha70) (20 and 50 kD). The preparations of type B SSC (SSC/B) consist of the nonproteolyzed form of type B botulinic neurotoxin (BoNT/B) of 150 kD, the proteolyzed form of BoNT/B of 150 kD (45 and 105 kD), the nonproteolyzed form of NTNH140, Ha17, Ha34 and two nonidentified proteins (32 and 40 kD). As shown in this study, toxic complexes both in native toxins and in the preparations of SSC do not dissociate for several weeks at pH 8.0 and for 18 hours in 3% SDS, as well as after treatment with RNAase or 1 M NaCl. Some part of SSC/A (neurotoxin and Ha70) has been found to dissociate in 3% SDS after 1-hour incubation at 22 degrees C after the addition of 2-ME. The preparations of SSC contain nucleic acids (A260 nm/A280 nm = 2.0), supposedly ensuring the stability of the complexes. In contrast to the L-forms of Clostridium botulinum toxins, the preparations of SSC/A and SSC/B have been found to possess increased toxicity. The specific toxicity of SSC/A has proved to be 1-2 x 10(9) DLM per 1 OD280 nm and that of SSC/B, from 5 x 10(8) to 1 x 10(9) DLM per OD280 nm. One minimal lethal oral dose of these SSC preparations for mice was less than 10 DLM, introduces intraperitoneally.  相似文献

In order to examine the effects of serum proteins on the biologic activity of estrogens, we superfused uteri from ovariectomized rats with Krebs-Ringer phosphate buffer (KRP), 4% human serum albumin (HSA) in KR or charcoal-stripped human plasma (HP), alone or with estradiol (E2), estrone (E1) or estriol (E3), 5 x 10(-10), 10(-9) and 10(-8) M. Following superfusion, the uteri were homogenized and the cytosol and nuclear receptors were measured by an exchange technique. Since we could detect no significant difference in the percent of receptors in the nucleus when the time of superfusion was varied from 30-120 min, all studies were done using a 30 min superfusion at a flow rate of 0.6 ml/min. In control studies using KRP alone (n = 12) 23.8 +/- 1.8 (mean +/- SEM) of the receptors were present in the nucleus at the end of the 30 min superfusion. Addition of E1, E2 or E3 5 x 10(-10) M resulted in a significant increase compared to controls in the percent of receptors in the nucleus. The percent of nuclear receptors was significantly greater for E2 and E3 (46.5 +/- 3.2% and 43.6 +/- 1.8%) compared to E1 (34.0 +/- 0.9%). Superfusions of uteri with either E2 or E3 at 10(-9) M or 10(-8) M resulted in a significantly greater percent of nuclear receptors compared to equimolar infusions of E1. When uteri were superfused with E1 at 5 x 10(-10), 10(-9) or 10(-8) M or with E3 at 5 x 10(-10) or 10(-9) M in HSA or HP the percent of nuclear receptors was not different compared to the respective infusion of equimolar concentrations of E1 or E3 in KR. However, superfusions of E2 5 x 10(-10), 10(-9) or 10(-8) M in HSA or HP resulted in a significant decrease in the percent nuclear receptors compared to the percent after equimolar superfusions of E2 in KR. Superfusions of E2 in HSA or HP resulted in the same percent of receptors in the nucleus. The percent of receptors in the nucleus increased with increasing concentrations of E2, but at each concentration the percent of receptors was the same with HA as with HP. Using the percent of nuclear receptors as an index of biological activity, E1 has less activity than either E2 or E3. Expression of the endogenous human GH (hGH) gene in response to glucocorticoids, thyroid hormone, and insulin was studied in cultures of dispersed GH-secreting human pituitary adenomas. Results were compared to those obtained when the hGH gene was transfected into rat pituitary tumor cells (GC). In the human pituitary cells the glucocorticoid dexamethasone [(Dex) 10(-6) M] increased the release of GH and the levels of GH mRNA by 2 to 4-fold (P less than 0.05). T3 (10(-8) M) had no effect on GH mRNA but increased hGH release by 2- to 6-fold (P less than 0.01). Insulin (5 x 10(-9) M) alone had no significant effect on either hGH mRNA or protein, but blunted the effect of Dex. Among 11 of 18 GC cell clones transfected with the hGH gene with detectable hGH mRNA expression, Dex increased hGH mRNA levels in seven and T3 treatment reduced hGH mRNA levels in eight. Conversely, rat GH mRNA levels from the endogenous rat gene were increased by either Dex or T3 in all 18 clones. Insulin alone or in combination with T3 or Dex was found to increase hGH mRNA levels in some cell lines and to decrease hGH mRNA levels in others; these effects were correlated strongly (r = 0.88; P less than 0.001) with the influence of insulin on the endogenous rat GH gene, implying that individual cellular differences can simultaneously affect the insulin responsiveness of both genes.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Differentiating (3T3-L1) and nondifferentiating (3T3-C2) fibroblastic cell lines possess two classes of insulin receptors, high affinity (KD = 1.0 to 3.7 X 10(-9) M) and low affinity (KD = 2.0 to 3.6 X 10(-8) M). Confluent cultures of 3T3-L1 cells induced to differentiate by insulin (1.74 x 10(-6) M) or indomethacin (1.25 x 10(-4) M) exhibit a 3-fold increase in the number of high affinity and low affinity receptors per cell or a 1.5- to 1.8-fold increase in the number of receptors per micron2 of surface area. In contrast, nondifferentiating 3T3-C2 cells treated with insulin or indomethacin lose almost completely the high affinity insulin receptors while retaining the same levels of low affinity receptors. The loss of high affinity receptors of the 3T3-C2 cells is accompanied by the disappearance of the stimulatory effect of insulin on the production of CO2 from glucose and on the uptake of aminoisobutyrate. The levels of high affinity insulin receptors appear to be regulated by different mechanisms in the differentiating (3T3-L1) and nondifferentiating (3T3-C2) cell lines. The mode of this regulation may have a bearing on the ability of a particular cell line to differentiate.  相似文献

The relation between changes of insulin receptor and various metabolic responses were studied in adult rat hepatocytes in primary culture. In cells cultured for 3 h without insulin, the number of high affinity sites and the dissociation constant (Kd) of insulin receptor, determined from a Scatchard plot, were 1.05 x 10(5) sites/cell and 1.5 x 10(-9) M, respectively. The receptor number increased 2-fold, but the Kd value remained constant during 2-days culture in insulin-free medium (up-regulation). Addition of dexamethasone (Dex), growth hormone, glucagon or triiodothyronine did not change the number of insulin receptors or the Kd value. In contrast, 1-day culture in insulin (1 x 10(-7) M) medium decreased the receptor number by half (down-regulation) without change of the Kd value. Short-term responses of glycogenesis, amino acid transport and lipogenesis by insulin increased as the receptor number increased. In these cases, the sensitivity to insulin (Ka: half dose for the maximum response) did not change in cells with different receptor numbers, but the maximum response changed. These results show that hepatocytes, unlike adipocytes, do not have spare receptors of insulin. During down-regulation, the receptor number decreased by only half, but the insulin responses were lost almost completely. The receptor number returned to the normal level after culture in insulin-free medium for 12 h, but recovery of the responses took longer, suggesting that for the insulin response not only change of receptor number, but also other regulatory mechanisms for post-receptor processes, such as desensitization, are involved.  相似文献

The role of glucocorticoids and second messenger systems in the regulation of the vasopressin (VP) gene was studied in the human small cell lung carcinoma cell line GLC-8. Small cell lung carcinoma GLC-8 cells express VP mRNA and contain both glucocorticoid and mineralocorticoid receptors. Treatment with the synthetic glucocorticoid dexamethasone when added alone at 10(-8) M had no effect on the VP mRNA level and decreased the level by 30% at 10(-6) M. However, the effect of dexamethasone changed to positive when cells were simultaneously treated with cAMP-enhancing agents. VP mRNA levels, which were elevated by 1.5- to 2-fold by the cAMP-enhancing agents alone, increased a further 1.5- to 3-fold by dexamethasone. Thus, the combined effect of dexamethasone and cAMP stimulation was a 3- to 7.5-fold increase in VP mRNA levels. Long term treatment with the phorbol ester 12-O-tetradecanoyl-phorbol-13-acetate (TPA) reduced the VP mRNA level by 75%. The TPA-suppressed VP mRNA levels could be up-regulated about 6-fold by simultaneous treatment with 8-bromo-cAMP. Dexamethasone did not alter the TPA-suppressed VP mRNA levels. These results indicate that both cAMP and protein kinase-C pathways as well as glucocorticoid receptors are involved in the regulation of VP mRNA levels and that these factors interact. This leads to a negative or positive response of VP gene expression to glucocorticoids in a state-dependent manner. The interactions may be of significance in a physiological context and relate to the different regulation of VP-expressing systems in the brain.  相似文献

Essential oil constituents were tested for their neurophysiological effects in Periplaneta americana and Blaberus discoidalis. Eugenol depressed spontaneous and stimulus-evoked impulses recorded extracellularly in the abdominal nerve cord, with an almost complete block of spikes at 2 x 10(-3) M. Geraniol and citral had similar depressive effects but increased spontaneous firing at lower doses (threshold 2.5 x 10(-4) M). Similar effects occurred in dorsal unpaired median (DUM) neurons, recorded intracellularly in the isolated terminal abdominal ganglion of P. americana. Spontaneous firing was progressively reduced by increasing concentrations of eugenol, whereas geraniol and citral produced biphasic effects (excitation at 10(-4) M, depression at 2 x 10(-3) M). All three oils decreased excitability of silent DUM neurons that were depolarised by applied current, but eugenol (at 10(-3) M) also changed the firing pattern from single spikes to bursts driven by plateau potentials. All oils reduced spike undershoot. Low doses of citral and geraniol (threshold ca. 10(-4) M) reversibly increased the frequency of spontaneous foregut contractions and abolished them at 2 x 10(-3) M (together with response to electrical stimulation). Eugenol reversibly reduced spontaneous activity at 10(-4) M and above. Eugenol has been reported to exert its insecticidal properties via a low-dose activation of octopamine receptors. In our studies, however, octopamine was found to have opposing effects to eugenol on DUM neurons and foregut activity (excitatory in both). Furthermore, eugenol did not affect the response to octopamine in DUM neurons. These results suggest that reported effects of eugenol were on a different sub-type of octopamine receptor.  相似文献

Cell wall surface proteins of group A Streptococcus type 29 were extracted with 1 M hydroxylamine pH 6.0. The purification procedure included fractionation with ammonium sulfate and gel filtration on Sephadex G-150. SDS polyacrylamide gel electrophoresis revealed a number of proteins (approximately 20) with molecular mass of 70 kD; the difference in Mr between the proteins was 5-10 kD. Isoelectrofocusing demonstrated that the proteins are either acid (pI = 3.7) or weakly alkaline (pI = 7.7). Possible reasons for the heterogeneity of Streptococcus cell wall surface proteins are discussed.  相似文献

1. Each segmental ganglion of the leech Hirudo medicinalis contains 6 touch (T) cells, 4 pressure (P) cells and 4 nociceptive (N) cells. The receptive terminals of these cells innervate the skin in discrete areas. These cells are known to have extrasynaptic receptors. 2. We tested the effect of transmitter substances present in leech CNS on the sensitivity of T and P cells to mechanical stimuli. Substances tested included octopamine, FMRFamide, proctolin, substance P, glutamate, GABA, acetylcholine and serotonin. 3. Only acetylcholine and serotonin had consistent effects. Serotonin (1 x 10(-3) M) increased the number of action potentials of T cells elicited by a standard stimulus. Serotonin (1 x 10(-4) M) and acetylcholine (1 x 10(-3) M) increased the number and frequency of action potentials in P cells.  相似文献

Synthetic bivalent thrombin inhibitors comprise an active site blocking segment, a fibrinogen recognition exosite blocking segment, and a linker connecting these segments. Possible nonpolar interactions of the P1' and P3' residues of the linker with thrombin S1' and S3' subsites, respectively, were identified using the "Methyl Scan" method [Slon-Usakiewicz et al. (1997) Biochemistry 36, 13494-13502]. A series of inhibitors (4-tert-butylbenzenesulfonyl)-Arg-(D-pipecolic acid)-Xaa-Gly-Yaa-Gly-betaAla-Asp-Tyr-Glu-Pro-Ile-Pro-Glu-Glu-Ala- (be ta-cyclohexylalanine)-(D-Glu)-OH, in which nonpolar P1' residue Xaa or P3' residue Yaa was incorporated, were designed and improved the affinity to thrombin. Substitution of the P3' residue with D-phenylglycine or D-Phe improved the K(i) value to (9.5 +/- 0.6) x 10(-14) or 1.3 +/- 0.5 x 10(-13) M, respectively, compared to that of a reference inhibitor with Gly residues at Xaa and Yaa residues (K(i) = (2.4 +/- 0.5) x 10(-11) M). Similarly, substitution of the P1' residue with L-norleucine or L-beta-(2-thienyl)alanine lowered the K(i) values to (8.2 +/- 0.6) x 10(-14) or (5.1 +/- 0.4) x 10(-14) M, respectively. The linker Gly-Gly-Gly-betaAla of the inhibitors in the previous sentence was simplified with 12-aminododecanoic acid, resulting in further improvement of the K(i) values to (3.8 +/- 0.6) x 10(-14) or (1.7 +/- 0.4) x 10(-14) M, respectively. These K(i) values are equivalent to that of natural hirudin (2.2 x 10(-14) M), yet the size of the synthetic inhibitors (2 kD) is only one-third that of hirudin (7 kD). Two inhibitors, with L-norleucine or L-beta-(2-thienyl)alanine at the P1' residue and the improved linker of 12-aminododecanoic acid, were crystallized in complex with human alpha-thrombin. The crystal structures of these complexes were solved and refined to 2.1 A resolution. The Lys(60F) side chain of thrombin moved significantly and formed a large nonpolar S1' subsite to accommodate the bulky P1' residue.  相似文献

Isolated goat detrusor muscle exhibited spontaneous contractility with an irregular amplitude and frequency. The spontaneity of detrusor muscle exhibited a mean amplitude as 11.99 +/- 0.83 mm and frequency as 1.37 +/- 0.16/min. KATP-channel openers namely, cromakalim or pinacidil (10(-7) - 10(-4) M) added cumulatively, elicited a concentration-related inhibition of both amplitude and rate of spontaneous contractions. The mean IC50 values for both amplitude and frequency for cromakalim were 3.3 x 10(-6) M and 2.9 x 10(-6) M, respectively; and for pinacidil were 2.0 x 10(-5) M and 1.5 x 10(-5) M, respectively. Glibenclamide, a KATP-channel blocker inhibited the cromakalim-induced concentration-related relaxation of spontaneous contractions with a significant increase in its mean IC50. ACh-induced concentration-related contractile response was inhibited in the presence of either cromakalim (10(-4) M) or pinacidil (10(-4) M). The mean EC50 value of ACh, in the presence of cromakalim (2.5 x 10(-3) M) was significantly increased as compared to the control (1.2 x 10(-6) M). In the presence of glibenclamide (10(-5) M) the inhibitory effect of cromakalim was significantly reduced with consequent decrease in the EC50 value (1.9 x 10(-5) M). Application of EFS (30 V and 5 ms) on goat urinary bladder strips at 1, 2, 5, 10, 20 and 30 Hz elicited frequency-related contractile responses. Both cromakalim and pinacidil caused a rightward shift in the frequency-related contractile response curve with significant increase in the mean EF25 and EF50 values, respectively. In the presence of glibenclamide (10(-4) M), the frequency-related inhibitory response curve was shifted to left with significant (P < 0.001) increase in the mean EF25, EF50 and EF75. The present results suggest that in the goat detrusor muscle, agonist and EFS-induced contractile responses were more potently inhibited by cromakalim than pinacidil with activation of glibenclamide sensitive KATP channels.  相似文献

Possible links have been investigated between activation of protein kinase C (PKC) and endothelin (ET) production by small blood vessels. Perfusion pressures were recorded from rat isolated mesenteric artery, with or without the small intestine attached, before and after addition to the perfusate of either ET-1, ET-3 or the PKC activator 12-deoxyphorbol 13-phenylacetate (DOPPA). Rises in perfusion pressure in response to ET-1 (10(-8) M)or DOPPA (10(-6) M) were reduced significantly by pre-treatment with either the ET(A) receptor antagonist PD151242 (10(-6) M) or the PKC inhibitor Ro 31-8220 (10(-6) M). ET-3 (10(-8) M) had a significant, albeit small, effect only when the gut was still attached to the mesentery. Inthis latter preparation ET-1 and DOPPA increased the permeability of villi microvessels to colloidal carbon in the perfusate. This effect of DOPPA was reduced by pre-treatment with either PD151242 or Ro 31-8220, but the effects of ET-1 were reduced significantly only by Ro 31-8220. ET-3 (10(-8) M) was without effect. The results suggest a possible bi-directional link between ET(A) receptors and PKC in the intestinal vasculature.  相似文献

BACKGROUND: Fluorescent markers (labeled antibodies) and flow cytometry are used to enumerate the average number of receptors (antigens) on formed bodies (cells) in whole blood by using a new method that avoids the extra steps of separating bound from unbound fluorescent markers or the use of external standards. METHODS: Mean channel fluorescence intensities of equilibrated marker-cell suspension mixtures, total concentrations of marker, and targeted cell counts obtained by standard cytometry procedures are used to complete the analyses for receptors per cell. Also, flow cytometric assays using competitive binding between fluorescent marker (CD4-RD1, CD8-FITC, CD3-FITC, CD3-RD1) and unlabeled antibody (CD4, CD8, CD3, CD3-dextran) for receptors on white blood cells in whole blood are described for determination of relative and specific binding constants of unlabeled/labeled antibody for targeted receptors. RESULTS: Ranges that were obtained for receptors per cell (lymphocytes) in normal blood donors were as follows: CD4, 4.9 x 10(4)-1.5 x 10(5); CD8, 5.0 x 10(5)-2.1 x 10(6); CD3, 6.6-7.8 x 10(5). Binding constants were highest for unlabeled CD4 antibody, 2. 7 x 10(10)-2.1 x 10(12) M(-1), and then unlabeled CD3 antibody, 1.1 x 10(10)-1.9 x 10(11) M(-1). FITC- and RD1-labeled antibodies typically had binding constants that were 10-to 100-fold lower than the native antibodies. CONCLUSIONS: Values of receptors per cell and binding constants obtained by the new method from flow cytometric analyses of mixtures of whole blood with FITC- or RD1-labeled CD4, CD8, and CD3 antibodies compare well with literature values determined by other methods.  相似文献
