为探讨光强及光质对高山红景天(Rhodiola sachalinensis)生物量和红景天甙含量的影响。于2000年4月至6月在东北林业大学温室内以移栽于大兴安岭加格达奇圃地人工种植生长3a的高山红景天为材料,通过纱布遮荫及遮以不同颜色的滤光膜分别进行了光强、光质控制实验(处理45d)。随着光强的降低,高山红景天全株生物量、根生物量、根的红景天甙含量和产量以及叶中叶绿素a、叶绿素b、总叶绿素的含量均有降低的趋势,但叶绿素含量变化很小,不同光强及对照之间的差异均未达到显著水平。相对光强为67.75%和44.71%的两种处理下的全株生物量、根生物量、根的红景天甙含量和产量差异不显著,它们的全株生物量和红景天甙含量与对照(全光照)的差异也不显著,但根生物量和红景天甙产量与对照的差异显著。当相对光强减弱至31.96%,全株生物量、根生物量、根的红景天甙含量和产量均大幅度下降,根冠比显著增加。4种滤光膜处理均使高山红景天的全株生物量及根生物量显著降低,蓝膜和绿膜处理的降低幅度大于红膜和黄膜处理的。红膜处理的红景天甙的含量和产量均高于对照,但黄膜、蓝膜和绿膜处理的红景天甙含量和产量则低于对照。通过计算去除4种滤光膜的光强因素,仅从光质的作用看。4种滤光膜处理仍是减小了全株生物量和根生物量,红膜和绿膜处理提高了红景天甙的含量和产量,而黄膜处理降低了红景天甙的含量和产量,蓝膜处理几乎没有效果。4种滤光膜处理均使叶绿素含量增加,但只有蓝膜处理的与对照差异显著。红膜处理不仅显著提高根中红景天甙的含量(为对照的3.42倍),而且对根生物量的影响较小(为对照的90.24%)。因而提高了高山红景天根的红景天甙产量,这意味着在生产上可能会有一定的实践意义。  相似文献

高山红景天细胞悬浮培养过程中,3mg·L916-BA、0.3mg·L-1NAA、60mmol·L-1氮源(其中NH_4~:NO_3~- 为1:1)、0.5~1.25mmol·L-1KH2PO4和200mg·L-1蛋白胨,较适合于细胞生长和红景天苷的积累。在适宜的接种量(3.2g·L-1)下细胞培养24d后,生物量达14.04g·L-1,干细胞中红景天苷含量为5.66mg·g-1。  相似文献

采用纱布遮荫(覆以透明膜)和红色滤光膜处理人工种植生长3年和4年的高山红景天,观察了根生物量和红景天甙含量的季节变化结果表明,纱布遮荫处理(相对光强为全光照的51.8%)下根的生物量显著下降;红景天甙含量略有增加,红景天甙产量(红景天甙含量与根生物量乘积)则有所降低,但差异均不显著,红色滤光膜处理(相对光强为全光照的51.8%)与纱布遮荫处理相比,根生物量减少但不显著,红景天甙的含量和产量则显著增加,生长季末前者的红景天甙含量为后者的1.63倍(生长3年)和1.55倍(生长4年),红景天甙产量为后者的1.44倍(生长3年)和1.45倍(生长4年)。  相似文献

为探讨野外条件下光强及光质对高山红景天 (Rhodiola sachalinensis)生物量和红景天甙含量的影响 ,于 2 0 0 1年 5月 8日至 9月 16日在大兴安岭加格达奇的高山红景天种植圃地 ,利用纱布及红色、蓝色和绿色的滤光膜遮光处理 ,对生长 3a和 4 a的高山红景天进行了光强、光质控制实验。与温室实验类似 ,遮荫显著抑制高山红景天根的生长 ,并使红景天甙的含量略有提高。红膜处理使光强大约降低一半 ,但仅从光质的角度而言 ,红膜处理对根的生长影响不大 ,却显著增加了根中的红景天甙含量和产量 ,不过效果不如温室实验明显。绿膜处理未表现出对红景天甙积累的促进作用 ,这与温室实验结果不同。红膜处理不同天数的结果表明 ,处理时间对红景天甙含量提高的程度影响很小。这意味着在野外种植的情况下 ,可以在临近收获的最后一段时间用红膜对高山红景天进行处理 ,这样既可避免红膜处理对高山红景天根生长的抑制 (由于减弱了光照 ) ,又可显著提高根的红景天甙含量 ,从而达到大幅度提高红景天甙产量的目的。  相似文献

建立了一种高效液相色谱法测定红景天根中红景天甙的含量。色谱柱为Hypersil—C18(5μm,150mm×4.6mm),甲醇-水为流动相,梯度洗脱,柱温25℃,在250nm波长处检测。研究结果表明:红景天甙的检测限为50μg/L,线性范围为4—40mg/L,回归方程为Y=5.6348X-4.0931,r=0.9996,加样回收率为98.05%。方法操作简便、快速、准确。  相似文献

在黑龙江省大海林地区海拔1400-1620m的高山上,依据高山红景天的生境于岳桦林内、高山草甸、苔原上选择了6个样地,测定了高山红景天根部的红景天甙含量。生境不同的高山红景天,根部红景天甙的含量有明显差异,红景天甙含量范围内0.101%-0.684%。从测定结果看,光照条件好、土壤肥沃的生境下高山红景天根部的红景天甙含量较高。  相似文献

对1998年8月和1999年7月采自吉林省长白山自然保护区内共14个天然居群和人工栽培于黑龙江省大兴安岭的高山红景年龄和根部红景天甙含量的关系进行了研究。高山红景天的年龄根据其极茎上芽磷痕的数量判断,结果表明,人工栽培的高山红景天的年龄与其根部红景天甙的含量呈显著正相关,而长白山自然保护区内自然生境下的高山红景天,其根部的红景天甙含量与年龄的正相关性则较弱。  相似文献

Some closely related members of the monocotyledonous familiesAlismataceae, Liliaceae, Juncaceae, Cyperaceae, Poaceae andAraceae with variable modes of pollination (insect- and wind-pollination) were studied in relation to the ultrastructure of pollenkitt and exine (amount, consistency and distribution of pollenkitt on the surface of pollen grains). The character syndromes of pollen cementing in entomophilous, anemophilous and intermediate (ambophilous or amphiphilous) monocotyledons are the same in principal as in dicotyledons. Comparing present with former results one can summarize: 1) The pollenkitt is always produced in the same manner by the anther tapetum in all angiosperm sub-classes. 2) The variable stickiness of entomophilous and anemophilous pollen always depends on the particular distribution and consistency of the pollenkitt, but not its amount on the pollen surface. 3) The mostly dry and powdery pollen of anemophilous plants always contains a variable amount of inactive pollenkitt in its exine cavities. 4) A step-by step change of the pollen cementing syndrome can be observed from entomophily towards anemophily. 5) From the omnipresence of pollenkitt in all wind-pollinated angiosperms studied one can conclude that the ancestors of anemophilous angiosperms probably have been zoophilous (i.e. entomophilous) throughout.

Cyclophilin A (CypA) is a peptidyl-prolyl cis/trans isomerase that interacts with the matrix protein (M1) of influenza A virus (IAV) and restricts virus replication by regulating the ubiquitin–proteasome-mediated degradation of M1. However,the mechanism by which CypA regulates M1 ubiquitination remains unknown. In this study, we reported that E3 ubiquitin ligase AIP4 promoted K48-linked ubiquitination of M1 at K102 and K104, and accelerated ubiquitin–proteasome-mediated degradation of M1. The recombinant IAV with mutant M1 (K102 R/K104 R) could not be rescued, suggesting that the ubiquitination of M1 at K102/K104 was essential for IAV replication. Furthermore, CypA inhibited AIP4-mediated M1 ubiquitination by impairing the interaction between AIP4 and M1. More importantly, both the mutations of M1 (K102 R/K104 R) and CypA inhibited the nuclear export of M1, indicating that CypA regulates the cellular localization of M1 via inhibition of AIP4-mediated M1 ubiquitination at K102 and K104, which results in the reduced replication of IAV.Collectively, our findings reveal a novel ubiquitination-based mechanism by which CypA regulates the replication of IAV.  相似文献
