DNA extraction conditions fromPorphyra perforata using LiCl   总被引:1,自引:0,他引:1  
A rapid and economical method of DNA extraction from a red seaweedPorphyra perforata J. Agardh has been developed by the use of lithium chloride. This paper describes the optimization of extraction conditions. Heat treatment of tissues in a solution (0.8 M LiCl, 0.6% Sarkosyl, 10 mM EDTA, 0.2% PVPP, 5% ß-mercaptoethanol, pH 9.0) at 55 °C for 10 min extracts DNA that is of sufficient quality to be used as a template for PCR amplification. Total DNA yield was approximately 30 to 50g g–1 t of partially dried tissue. Total RNA yield was approximately 400g g–1 of partially dried tissue. Carbohydrate was contained as approximately 40 to 90 mM (expressed as glucose equivalents) from 1 g tissue, and protein contamination calculated as the O.D. 260/280 ratio was in the range of 1.4 to 1.7. The DNA was characterized by high molecular weight larger than 50 kb.  相似文献

The combined effects of water activity (aw) and temperature on mycotoxin production by Penicilium commune (cyclopiazonic acid — CPA) and Aspergillus flavus (CPA and aflatoxins — AF) were studied on maize over a 14-day period using a statistical experimental design. Analysis of variance showed a highly significant interaction (P 0.001) between these factors and mycotoxin production. The minimum aw/temperature for CPA production (2264 ng g–1 P. commune, 709 ng g–1 A. flavus) was 0.90 aw/30 °C while greatest production (7678 ng g–1 P. commune, 1876 ng g–1 A. flavus) was produced at 0.98 aw/20 °C. Least AF (411 ng g–1) was produced at 0.90 aw/20 °C and most (3096 ng g–1) at 0.98 aw/30 °C.  相似文献

A field experiment in western Kenya assessed whether the agroforestry species Tithonia diversifolia (Hemsley) A. Gray, Tephrosia vogelii Hook f., Crotalaria grahamiana Wight & Arn. and Sesbania sesban (L) Merill. had access to forms of soil P unavailable to maize, and the consequences of this for sustainable management of biomass transfer. The species were grown in rows at high planting density to ensure the soil under rows was thoroughly permeated by roots. Soil samples taken from beneath rows were compared to controls, which included a bulk soil monolith enclosed by iron sheets within the tithonia plot, continuous maize, and bare fallow plots. Three separate plant biomass samples and soil samples were taken at 6-month intervals, over a period of 18 months. The agroforestry species produced mainly leaf biomass in the first 6 months but stem growth dominated thereafter. Consequently, litterfall was greatest early in the experiment (0–6 months) and declined with continued growth. Soil pH increased by up to 1 unit (from pH 4.85) and available P increased by up to 38% (1 g P g–1) in agroforestry plots where biomass was conserved on the field. In contrast, in plots where biomass was removed, P availability decreased by up to 15%. Coincident with the declines in litterfall, pH decreased by up to 0.26 pH units, plant available P decreased by between 0.27 and 0.72 g g–1 and Po concentration decreased by between 8 and 35 g g–1 in the agroforestry plots. Declines in Po were related to phosphatase activity (R2=0.65, P<0.05), which was greater under agroforestry species (0.40–0.50 nmol MUB s–1 g–1) than maize (0.28 nmol MUB s–1 g–1) or the bare fallow (0.25 nmol MUB s–1 g–1). Management of tithonia for biomass transfer, decreased available soil P by 0.70 g g–1 and Po by 22.82 g g–1. In this study, tithonia acquired Po that was unavailable to maize. However, it is apparent that continuous cutting and removal of biomass would lead to rapid depletion of P stored in organic forms.  相似文献

The distribution of labile Cd and Zn in two contrasting soils was investigated using isotopic exchange techniques and chemical extraction procedures. A sewage sludge amended soil from Great Billings (Northampton, UK) and an unamended soil of the Countesswells Association obtained locally (Aberdeen, UK) were used. 114Cd and 67Zn isotopes were added to a water suspension of each soil and the labile metal pool (E-value) determined from the isotope dilution. Samples were obtained at 13 time points from 1h to 50 days. For the sewage sludge amended soil, 29 g Cd g–1 (86% of total) and 806 g Zn g–1 (65% of total) were labile and for the Countesswells soil the value was 8.6 g Zn g–1 (13% of total); limits of detection prevented a Cd E-value from being measured in this soil. The size of the labile metal pool was also measured by growing plants for 90 days and determining the isotopic content of the plant tissue (L-value). Thlaspi caerulescensJ. & C. Presl (alpine penny cress), a hyperaccumulator of Zn and Cd, Taraxacum officinale Weber (dandelion) and Hordeum vulgare L. (spring barley) were used. L-values were similar across species and lower than the E-values. On average the L-values were 23±0.8 g Cd g–1 and 725±14 g Zn g–1 for the Great Billings soil and 0.29±0.16 g Cd g–1 and 7.3±0.3 g Zn g–1 for the Countesswells soil. The extractable metal content of the soils was also quantified by extraction using 0.1 M NaNO3, 0.01 M CaCl2, 0.5 M NaOH, 0.43 M CH3COOH and 0.05 M EDTA at pH 7.0. Between 1.3 and 68% of the total Cd and between 1 and 50% of the total Zn in the Great Billings soil was extracted by these chemicals. For the Countesswells soil, between 6 and 83% of the total Cd and between 0.1 and 7% of the total Zn was extracted. 0.05 M EDTA and 0.43 M CH3COOH yielded the greatest concentrations for both soils but these were less than the isotopic estimates. On the whole, E-values were numerically closer to the L-values than the chemical extraction values. The use of isotopic exchange provides an alternative estimate of the labile metal pool within soils compared to existing chemical extraction procedures. No evidence was obtained that T. caerulescens is able to access metal within the soil not freely available to the other plants species. This has implications for long term remediation strategies using hyperaccumulating plant species, which are unlikely to have any impact on non-labile Cd and Zn in contaminated soil.  相似文献

The effects of medium strategies [maintenance (M), intermediary (G), and production (P) medium] on cell growth, anthraquinone (AQ) production, hydrogen peroxide (H2O2) level, lipid peroxidation, and antioxidant vitamins in Morinda elliptica cell suspension cultures were investigated. These were compared with third-stage leaf and 1-month-old callus culture. With P medium strategy, cell growth at 49 g l–1, intracellular AQ content at 42 mg g–1 DW, and H2O2 level at 9 mol g–1 FW medium were the highest as compared to the others. However, the extent of lipid peroxidation at 40.4 nmol g–1 FW and total carotenoids at 13.3 mg g–1 FW for cultures in P medium were comparable to that in the leaf, which had registered sevenfold lower AQ and 2.2-fold lower H2O2 levels. Vitamin C content at 30–120 g g–1 FW in all culture systems was almost half the leaf content. On the other hand, vitamin E content was around 400–500 g g–1 FW in 7-day-old cultures from all medium strategies and reduced to 50–150 g g–1 FW on day 14 and 21; as compared to 60 g g–1 FW in callus and 200 g g–1 FW in the leaf. This study suggests that medium strategies and cell growth phase in cell culture could influence the competition between primary and secondary metabolism, oxidative stresses and antioxidative measures. A family of 10 competing, unstructured models has been developed to model cell growth, substrate consumption, and product formation of the pyruvate producing strain Escherichia coli YYC202 ldhA::Kan strain used in fed-batch processes. The strain is completely blocked in its ability to convert pyruvate into acetyl-CoA or acetate (using glucose as the carbon source) resulting in an acetate auxotrophy during growth in glucose minimal medium. Parameter estimation was carried out using data from fed-batch fermentation performed at constant glucose feed rates of qVG=10 mL h–1. Acetate was fed according to the previously developed feeding strategy. While the model identification was realized by least-square fit, the model discrimination was based on the model selection criterion (MSC). The validation of model parameters was performed applying data from two different fed-batch experiments with glucose feed rate qVG=20 and 30 mL h–1, respectively. Certain yeast cells on solid nutrient medium produced colonies surrounded by a light zone of selenite absorption. This screening procedure resulted in the selection of 22 strains out of 200 isolates with different Se4+-absorbing capacity ranging from 16 to 98.8 g Se4+ g–1 l–1 h–1. The highest rate of Se4+ elimination from the Na2SeO3 solution was observed with an oval shaped, cream pigmented fermentative yeast, tentatively called Candida sp. strain MS4. This strain was isolated from wastewater and found to accumulate selenium oxyanions. Se4+ uptake involved both inactive and active phenomena. The amounts of selenium (initial concentration 2 mg Se4+ l–1) removed from aqueous solution by inactive and active phenomena were 667 g Se4+ g–1 l–1, and 1580 g Se4+ g–1 l–1, respectively. The aim of this work was to assess whether agroforestry species have the ability to acquire P from pools unavailable to maize. Tithonia diversifolia(Hemsley) A. Gray, Tephrosia vogelii Hook f., Zea mays and Lupinus albusL. were grown in rhizopots and pH change and depletion of inorganic and organic P pools measured in the rhizosphere. Plants were harvested at the same growth stage, after 56 days for maize and white lupin and 70 days for tithonia and tephrosia, and the rhizosphere sampled. The rhizosphere was acidified by tithonia (pH change –0.3 units to pH 4.8) and lupins (–0.2 units to 4.9), alkalinised by tephrosia (+0.4 units to pH 5.4), and remained unchanged with maize growth. Concurrent with acidification in the rhizosphere of tithonia there was a decline in resin-P (0.8 g P g–1). However, there was also a decline in NaOH extractable inorganic P (NaOH-Pi) (5.6 g P g–1 at the root surface) and organic P pools (NaOH-Po) (15.4 g P g–1 at 1.5 mm from the root), which would not be expected without specific P acquisition mechanisms. Alkalinisation of tephrosia rhizosphere was accompanied by changes in all measured pools, although the large depletion of organic P (21.6 g P g–1 at 5 mm from the root) suggests that mineralisation, as well as desorption of organic P, was stimulated. The size of changes of both pH and P pools varied with distance away from the rhizoplane. Decline of more recalcitrant P pools with the growth of the agroforestry species contrasted with the effect of maize growth, which was negligible on resin-P and NaOH-Pi, but led to an accumulation of P as NaOH-Po (14.2 g P g–1 at 5 mm from the root). Overall the depletion of recalcitrant P pools, particularly Po, suggests that the growth of tithonia and tephrosia enhance desorption and dissolution of P, while also enhancing organic P mineralisation. Both species appear to have potential for agroforestry technologies designed to enhance the availability of P to crops, at least in the short term.  相似文献

A fermentation medium based on millet (Pennisetum typhoides) flour hydrolysate and a four-phase feeding strategy for fed-batch production of baker's yeast,Saccharomyces cerevisiae, are presented. Millet flour was prepared by dry-milling and sieving of whole grain. A 25% (w/v) flour mash was liquefied with a thermostable 1,4--d-glucanohydrolase (EC 3.2.1.1) in the presence of 100 ppm Ca2+, at 80°C, pH 6.1–6.3, for 1 h. The liquefied mash was saccharified with 1,4--d-glucan glucohydrolase (EC 3.2.1.3) at 55°C, pH 5.5, for 2 h. An average of 75% of the flour was hydrolysed and about 82% of the hydrolysate was glucose. The feeding profile, which was based on a model with desired specific growth rate range of 0.18–0.23 h–1, biomass yield coefficient of 0.5 g g–1 and feed substrate concentration of 200 g L–1, was implemented manually using the millet flour hydrolysate in test experiments and glucose feed in control experiments. The fermentation off-gas was analyzed on-line by mass spectrometry for the calculation of carbon dioxide production rate, oxygen up-take rate and the respiratory quotient. Off-line determination of biomass, ethanol and glucose were done, respectively, by dry weight, gas chromatography and spectrophotometry. Cell mass concentrations of 49.9–51.9 g L–1 were achieved in all experiments within 27 h of which the last 15 h were in the fedbatch mode. The average biomass yields for the millet flour and glucose media were 0.48 and 0.49 g g–1, respectively. No significant differences were observed between the dough-leavening activities of the products of the test and the control media and a commercial preparation of instant active dry yeast. Gelidium sesquipedale is the most important raw material used for extraction of agar in Spain. Based on chemostats, a system of culture for macroalgae with a continuous flow of culture medium has been developed. A stressed morphotype from the South of Spain was cultured, and the effects of different rates of NO 3 flow on growth and internal constituents were investigated in the laboratory. Cultivation was successful after optimizing factors affecting growth, such as irradiance level, renewal rate and water movement. Mass production was dependent on N supply. With a flow of 35 mol NO3 g–1 DW d–1, optimal values of growth (2.1% d–1) and biomass yield were obtained. In these conditions, biomass yield resembled the values observed in natural populations (about 500 g DW m–2 y–1). When the flow of N was reduced to 15 mol NO 3 g–1 DW d–1, growth rate and biomass yield were reduced three-fold, and were null when N was supplied as 7 mol NO 3 g–1 DW d–1. C:N ratio was an index of the physiological status of the tissue, remaining low when N was sufficient and raised to critical values when N supply was limited. Phycobiliproteins, kept at a constant irradiance level, were affected by N supply, acting as an internal nitrogen reserve, unlike chlorophylla. An effective phycobiliprotein synthesis took place when the flow of N was sufficient. Agar yield, on dry weight basis, was similar as a function of N flow, whereas agar yield of the culture was higher when N was sufficient as a result of growth not being limited by N.This system of culture, commonly used in microalgal studies, may have an important use in macroalgae as a system to obtain biomass of high quality as well as a good tool for physiological studies in conditions of continuous and controlled flow of nutrients.  相似文献

Shoot cultures of Alyssum markgrafii O.E. Shulz, endemic nickel hyperaccumulating species of central Balkan, were established and maintained on Murashige and Skoog medium supplemented with 0.2 mg dm–3 benzyladenine (BA). Nickel in form of NiCl2 . 6 H2O was supplemented at 22 different concentrations ranging from 0.0001 to 15 mM but none of them was lethal to cultures. High Ni2+ concentrations (10 mM or more) arrested shoot growth which, upon transfer to Ni-free medium, commenced via axillary bud proliferation. Shoots that developed from axillary buds through the subculture manifested increased tolerance to Ni2+ expressed as shoot elongation. Shoot multiplication and dry biomass production decreased with increase of Ni2+ in medium. Only the accumulation of Ni2+ in tissues increased with Ni2+ content of the medium. Apart from shoot cultures, high Ni2+ accumulation was registered in undifferentiated callus cultured on medium with 0.5 mg dm–3 BA and 0.5 mg dm–3 naphthylacetic acid. Highest content of accumulated Ni was 2.37 g g–1 (d.m.) in shoots and 2.65 g g–1 (d.m.) in callus, both measured on medium with 15 mM Ni2+.  相似文献

Diurnal variation in leaf stomatal conductance (g s) of three xerophilous species (Buddleia cordata, Senecio praecox and Dodonaea viscosa) was measured over a 10-month period during the dry and wet seasons in a shrubland that is developing in a lava substratum in Mexico. Averaged stomatal conductances were 147 and 60.2 (B. cordata), 145 and 24.8 (D. viscosa) and 142.8 and 14.1 mmol m–2 s–1 (S. praecox) during the wet and dry season respectively. Leaf water potential () varied in a range of –0.6 to –1.2 (S. praecox), –0.6 to –1.8 (B. cordata) and –0.9 to –3.4 MPa (D. viscosa) during the same measurement periods. Stomata were more sensitive to changes in irradiance, air temperature and leaf–air vapour pressure difference in the rainy season than the dry season. Although stomatal responses to were difficult to distinguish in any season (dry or rainy), data for the entire period of measurement showed a positive correlation, stomata tending to open as increased, but there is strong evidence of isohydric behaviour in S. praecox and B. cordata. A multiplicative model relating g s to environmental variables and to accounted for 79%–83% of the variation of g s in three sites (pooled data); however, the performance of the model was poorer (60%–76%) for individual species from other sites not included in the pooled data.  相似文献

Photosynthetic induction responses to a sudden increase in photosynthetic photon flux density (PPFD) from lower background PPFD (0, 25, 50, and 100 mol m–2 s–1) to 1 000 mol m–2 s–1 were measured in leaves of Fagus crenata, Acer rufinerve Siebold & Zucc., and Viburnum furcatum growing in a gap and understory of a F. crenata forest in the Naeba mountains. In the gap, A. rufinerve exhibited more than 1.2-fold higher maximum net photosynthetic rate (P Nmax) than F. crenata and V. furcatum. Meanwhile, in the understory F. crenata exhibited the highest P Nmax among the three species. The photosynthetic induction period required to reach P Nmax was 3–41 min. The photosynthetic responses to increase in PPFD depended on the background PPFD before increase in PPFD. The induction period required to reach P Nmax was 2.5–6.5-fold longer when PPFD increased from darkness than when PPFD increased from 100 mol m–2 s–1. The induction period was correlated with initial P N and stomatal conductance (g s) relative to maximum values before increase in PPFD. The relationship was similar between the gap and the understory. As the background PPFD increased, the initial P N and g s increased, indicating that the degrees of biochemical and stomata limitations to dynamic photosynthetic performance decreased. Therefore, photosynthetic induction responses to increase in PPFD became faster with the increasing background PPFD. The differences in time required to reach induction between species, as well as between gap and understory, were mainly due to the varying of relative initial induction states in P N and g s at the same background PPFD.  相似文献

In Taxus cuspidata callus, vanadyl sulfate (10 mg l–1) induced a high (146 g g–1 dry wt) production of 10-deacetylbaccatin III in comparison to 7 g g–1 dry wt of the control. The content of paclitaxel in this species increased from 16 g g–1 to 74 g g–1 dry wt when 20 mg phenylalanine l–1 was used. In T. media, p-aminobenzoic acid induced the highest content of 10-deacetylbaccatin III (481 g g–1 dry wt) versus 181 g g–1 in the control. Paclitaxel increased from 89 to 139 g g–1 dry wt after adding chitosan (20 mg l–1) to the cultures.  相似文献

The induction of astaxanthin formation by reactive oxygen species in mixotrophic culture of Chlorococcum sp. was investigated. H2O2 (0.1 mM) enhanced the total astaxanthin formation from 5.8 to 6.5 mg g–1 cell dry wt. Fe2+ (0.5 mM) added to the medium with H2O2 (0.1 mM) further promoted astaxanthin formation to 7.1 mg g–1 cell dry wt. Similarly, Fe2+ (0.5 mM) together with methyl viologen (0.01 mM) promoted astaxanthin formation to 6.3 mg g–1 cell dry wt. In contrast, an addition of KI (1 mM), a specific scavenger for hydroxyl radicals (OH), together with H2O2 (0.1 mM) and Fe2+ (0.5 mM), to the medium decreased astaxanthin formation to 1.8 mg g–1 cell dry wt. KI (1 mM) also inhibited the enhancement of carotenogenesis by superoxide anion radicals (O2 ), with a decrease of astaxanthin formation to 1.7 mg g–1 cell dry wt. This suggested that O2 might be transformed to OH before promoting carotenogenesis in Chlorococcum sp.  相似文献

Specific rates of growth (Cw) of Mesocyclops leuckarti, young male and female instars varied between 0.03–0.26 and 0.03–0.17 g(w.w.)/g(w.w.)/day respectively at 15° and 22°C, whilst at IV–V copepodid stages females showed a higher Cw values. During 1969–1975 the averages of productivity and monthly P/B ratios were 44 (±23) g(w.w.) (= 5g(w.w.)/m2/month) and 3.1 respectively. P/B ratios were highly correlated (r(sup2)=0.98) with temperature changes. Metabolic parameters and P/B ratios were found to be similar to other water bodies in the world indicating an adaptation of M. leuckarti to different conditions.  相似文献

Summary Reaction kinetic analysis of the electrical properties of the electrogenic Cl pump inAcetabularia has been extended from steady-state to nonsteady-state conditions: electrical frequency responses of theAcetabularia membrane have been measured over the range from 1 Hz to 10 kHz at transmembrane potential differences across the plasmalemma (V m ) between –70 and –240 mV using voltage-clamp techniques. The results are well described by an electrical equivalent circuit with three parallel limbs: a conventional membrane capacitancec m , a steadystate conductanceg o (predominantly of the pump pathway plus a minor passive ion conductance) and a conductanceg s in series with a capacitancec p which are peculiar to the temporal behavior of the pump. The absolute values and voltage sensitivities of these four elements have been determined:c m of about 8 mF m–2 turned out to be voltage insensitive; it is considered to be normal.g o is voltage sensitive and displays a peak of about 80 S m–2 around –180 mV. Voltage sensitivity ofg s could not be documented due to large scatter ofg s (around 80 S m–2).c p behaved voltage sensitive with a notch of about 20 mF m–2 around –180 mV, a peak of about 40 mF m–2 at –120 mV and vanishing at –70 mV. When these data are compared with the predictions of nonsteady-state electrical properties of charge transport systems (U.-P. Hansen, J. Tittor, D. Gradmann, 1983,J. Membrane Biol. in press), model A (redistribution of states within the reaction cycle) consistently provides magnitude and voltage sensitivity of the elementsg o ,g s andc p of the equivalent circuit, when known kinetic parameters of the pump are used for the calculations. This analysis results in a density of pump elements in theAcetabularia plasmalemma of about 50 nmol m–2. The dominating rate constants for the redistribution of the individual states of the pump in the electric field turn out to be in the range of 500 sec–1, under normal conditions.  相似文献

Nitrate induction in spruce: an approach using compartmental analysis   总被引:6,自引:0,他引:6  
Using 13NO 3 -efflux analysis, the induction of nitrate uptake by externally supplied nitrate was monitored in roots of intact Picea glauca (Moench) Voss. seedlings over a 5-d period. In agreement with our earlier studies, efflux analysis revealed three compartments, which have been identified as surface adsorption, apparent free space, and cytoplasm. While induction of nitrate uptake was pronounced, NO 3 fluxes in induced plants were decidedly lower and the induction response was slower than in other species. Influx rose from 0.1 mol·g–1·h–1 (measured at 100 M [NO 3 o) in uninduced plants to a maximum of 0.5 mol·g–1h–1 after 3 d of exposure to 100 M [NO 3 o and declined to 0.3–0.4 mol·g–1h–1 at the end of the 5-d period. Efflux remained relatively constant around 0.02-0.04 mol·g–1h–1, but its percentage with respect to influx declined from initially high values (around 30%) to steady-state values of 4–7%. Cytoplasmic [NO 3 ] ranged from the low micromolar in uninduced plants to a maximum of 2 mM in plants fully induced at 100 M [NO 3 ]o. In-vivo root nitrate reductase activity (NRA) was measured over the same time period, and was found to follow a similar pattern of induction as influx. The maximum response in NRA slightly preceded that of influx. It increased from 25 nmol·g–1·h–1 without prior exposure to NO 3 to peak values around 150 nmol· g–1h–1 after 2 d of exposure to 100 M [NO 3 ]o. Subsequently, NRA declined by about 50%. The potential of three estuarine macroalgae (Ulvarotundata, Enteromorpa intestinalis andGracilaria gracilis) as biofilters for phosphate ineffluents of a sea bass (Dicentrarchus labrax) cultivationtank was studied. These seaweeds thrive in Cádiz Bay and were alsoselected because of their economic potential, so that environmental andeconomicadvantages may be achieved by future integrated aquaculture practices in thelocal fish farms. The study was designed to investigate the functioning of Pnutrition of the selected species. Maximum velocity of phosphate uptake (2.86mol PO4 g–1 dry wth–1) was found in U. rotundata.This species also showed the highest affinity for this nutrient. At low flowrates (< 2 volumes d–1), the three species efficientlyfiltered the phosphate dissolved in the waste water, with a minimum efficiencyof 60.7% in U. rotundata. Net phosphate uptake rate wassignificantly affected by the water flow, being greatest at the highest rateassayed (2 volumes d–1). The marked decrease in tissue P shownby the three species during a flow-through experiment suggested that growth wasP limited. However, due to the increase in biomass, total P biomass increasedinthe cultures. A significant correlation was found between growth rates and thenet P biomass gained in the cultures. A three-stage design under low water flow(0.5 volumes d–1) showed that the highest growth rates (up to0.14 d–1) and integrated phosphate uptake rates(up to 5.8 mol PO4 3– g–1dry wt d–1) were found in E.intestinalis in the first stage, with decreasing rates in thefollowing ones. As a result, phosphate become limiting and low increments oreven losses of total P biomass in these stages were found suggesting thatphosphate was excreted from the algae. The results show the potential abilityofthe three species to reduce substantially, at low water flow, the phosphateconcentration in waste waters from a D. labrax cultivationtank, and thus the quality of effluents from intensive aquaculture practices.  相似文献

The uptake of ammonium, nitrate and phosphate by laboratory-grown young sporophytes of Laminaria abyssalis was measured in a perturbed system (batch mode) at 18 °C and 35 ± 5 µE m–2 s–1 photon flux density. Uptake of all appeared to follow saturation-type nutrient uptake kinetics. The NO inf3 sup– (K s = 14.0 µM, V max = 5.0 µmol h–1 g–1 dry wt) and NH inf4 sup+ (K s = 4.6 µM, V max= 2.0 µmol h–1 g–1 dry wt) were taken up simultaneously, although NH inf4 sup+ was taken up more rapidly. Values of K 3 and V max for phosphate were, respectively, 2.21 µM and 0.83 µmol h–1 g–1 dry wt. Nitrate and phosphate were both consumed in similar rates (V max /Ks 0.37) at low concentrations. NH inf4 sup+ , thus, might be a more efficient form of N fertilizer if artificial enrichment of seawater is used.  相似文献
