Bark beetles oxidize the defensive monoterpenes of their host trees both to detoxify them and convert them into components of their pheromone system. This oxidation is catalyzed by cytochrome P450 enzymes and occurs in different tissues of the insect, including the gut (i.e., the site where the beetle's pheromones are produced and accumulated) and the antennae (i.e., the olfactory organs used for perception of airborne defensive monoterpenes as well as other host-associated compounds and pheromones). We identified ten new CYP genes in the pine beetle Dendroctonus rhizophagus in either antennae or gut tissue after stimulation with the vapors of major host monoterpenes α-pinene, β-pinene and 3-carene. Five genes belong to the CYP4 family, four to the CYP6 family and one to the CYP9 family. Differential expression of almost all of the CYP genes was observed between sexes, and within these significant differences among time, stimuli, anatomical region, and their interactions were found upon exposure to host monoterpenes. Increased expression of cytochrome P450 genes suggests that they play a role in the detoxification of monoterpenes released by this insect's host trees.  相似文献

Constitutive and inducible terpene production is involved in conifer resistance against bark beetles and their associated fungi. In this study 72 Norway spruce (Picea abies) were randomly assigned to methyl jasmonate (MJ) application, inoculation with the bluestain fungus Ceratocystis polonica, or no-treatment control. We investigated terpene levels in the stem bark of the trees before treatment, 30 days and one year after treatment using GC–MS and two-dimensional GC (2D-GC) with a chiral column, and monitored landing and attack rates of the spruce bark beetle, Ips typographus, on the trees by sticky traps and visual inspection. Thirty days after fungal inoculation the absolute amount and relative proportion of (+)-3-carene, sabinene, and terpinolene increased and (+)-α-pinene decreased. Spraying the stems with MJ tended to generally increase the concentration of most major terpenes with minor alteration to their relative proportions, but significant increases were only observed for (?)-β-pinene and (?)-limonene. Fungal inoculation significantly increased the enantiomeric ratio of (?)-α-pinene and (?)-limonene 1 month after treatment, whereas MJ only increased that of (?)-limonene. One year after treatment, both MJ and fungal inoculation increased the concentration of most terpenes relative to undisturbed control trees, with significant changes in (?)-β-pinene, (?)-β-phellandrene and some other compounds. Terpene levels did not change in untreated stem sections after treatment, and chemical induction by MJ and C. polonica thus seemed to be restricted to the treated stem section. The enantiomeric ratio of (?)-α-pinene was significantly higher and the relative proportions of (?)-limonene were significantly lower in trees that were attractive to bark beetles compared to unattractive trees. One month after fungal inoculation, the total amount of diterpenes was significantly higher in putative resistant trees with shorter lesion lengths than in putative susceptible trees with longer lesions. Thus, terpene composition in the stem bark may be related to resistance of Norway spruce against I. typographus and C. polonica.  相似文献

Ips pini bark beetles use myrcene hydroxylases to produce the aggregation pheromone component, ipsdienol, from myrcene. The enantiomeric ratio of pheromonal ipsdienol is an important prezygotic mating isolation mechanism of I. pini and differs among geographically distinct populations. We explored the substrate and product ranges of myrcene hydroxylases (CYP9T2 and CYP9T3) from reproductively-isolated western and eastern I. pini. The two cytochromes P450 share 94% amino acid identity. CYP9T2 mRNA levels were not induced in adults exposed to myrcene-saturated atmosphere. Functional assays of recombinant enzymes showed both hydroxylated myrcene, (+)- and (?)-α-pinene, 3-carene, and R-(+)-limonene, but not α-phellandrene, (?)-β-pinene, γ-terpinene, or terpinolene, with evidence that CYP9T2 strongly preferred myrcene over other substrates. They differed in the enantiomeric ratios of ipsdienol produced from myrcene, and in the products resulting from different α-pinene enantiomers. These data provide new information regarding bark beetle pheromone evolution and factors affecting cytochrome P450 structure–function relationships.  相似文献

Spruce bark beetle (Ips typographus L.) is the most destructive insect pest of spruce forests in Eurasia. However, contact toxicity, in vivo metabolism, and ecological functions of host monoterpenes are poorly understood at the spruce tree–bark beetle–predator tritrophic level. Spruce monoterpenes including S-(–)-α-pinene, R-(+)-α-pinene, and myrcene showed contact toxicity to I. typographus, with LD50 values ranging from 22–32 μg/mg. When topically treated with S-(–)-α-pinene or R-(+)-α-pinene, the amounts of volatile metabolites, including 4S-(–)-cis-verbenol, 4S-(+)-/4R-(–)-trans-verbenol, R-(+)-/S-(–)-verbenone and 1R-(–)-/1S-(+)-myrtenol, in the hindgut extracts of I. typographus varied significantly between sexes, and their quality (enantiomeric composition) varied significantly with the chirality of α-pinene. More importantly, S-(–)-α-pinene induced male adults to produce large amounts of 4S-(–)-cis-verbenol and S-(–)-verbenone. When topically treated with myrcene, the expected semiochemicals such as E-myrcenol, ipsenol and ipsdienol were not detected in the beetle hindguts, indicating that the pheromone biosynthetic system of I. typographus does not participate in the metabolism of host myrcene. In trap tests, S-(–)-α-pinene and R-(+)-α-pinene increased the catches of I. typographus and its predator Thanasimus substriatus in pheromone-baited traps, whereas myrcene exhibited a strong repellent (or inhibitory) effect on I. typographus but not on its predator. I. typographus seems to adopt different ecological strategies (e.g. avoidance to myrcene and preference for α-pinene) to adapt to and tolerate different host monoterpenes. Extensive investigation of these monoterpenes will help us understand their roles in manipulating the arms race between host trees and bark beetles, and potentially improve the efficacy of controlling I. typographus via the push-pull strategy using host kairomones.  相似文献

Six monoterpenes, ((?)-citronellal, p-cymene, (?)-menthone, α-pinene, α-terpinene and (?)-terpinen-4-ol) and two phenylpropenes, (trans-cinnamaldehyde and eugenol) were tested for their insecticidal activity against the rice weevil, Sitophilus oryzae under laboratory conditions. The bioassays were carried out on wheat at the concentrations of 0.5, 1.0 and 5.0?g/kg. Adult mortality was assessed after 14?days of exposure. After this interval, the treated wheat was retained for an additional period of 90?days, in order to evaluate progeny production and wheat loss. At the concentrations of 0.5 and 1.0?g/kg, trans-cinnamaldehyde and eugenol achieved highest adult mortality. At 5.0?g/kg, all compounds except for p-cymene and α-pinene caused complete (100%) adult mortality. Generally, trans-cinnamaldehyde was the most potent compound causing complete inhibition of progeny at the three concentrations. Similarly, no progeny were observed in the wheat treated with (?)-citronellal, eugenol, p-cymene, (?)-menthone, and (?)-terpinen-4-ol at 5.0?g/kg after 45 and 90?days. Similar trends were noted for wheat weight loss and damage as the treatment with monoterpenes and phenylpropenes at the highest rate preserved the wheat intact and free from damage by S. oryzae for 90?days. Our findings suggest the tested compounds except p-cymene and α-pinene could be recommended for use as part of an integrated pest management program for S. oryzae control in stored wheat.  相似文献

Fungal volatile compounds can mediate fungal-insect interactions. Whether fungi can emit insect pheromones and how volatile chemicals change in response to chemicals the fungi naturally encounter is poorly understood. We analyzed volatiles emitted by Grosmannia clavigera (symbiont of the mountain pine beetle) and Ophiostoma ips (symbiont of the pine engraver beetle) growing in liquid media amended with compounds that the fungi naturally encounter: (−)-α-pinene, (+)-α-pinene, (−)-trans-verbenol, verbenone, or ipsdienol. Nine volatile compounds were identified among the fungal and amendment treatments. Volatiles qualitatively and quantitatively differed between fungal species and among amendment treatments. The bark beetle anti-aggregation pheromone (−)-verbenone was detected from both fungi growing in (−)-trans-verbenol-amended media. G. clavigera and O. ips can emit beetle pheromones and other beetle semiochemicals, suggesting that ophiostomatoid fungi could contribute to the chemical ecology of bark beetles. However, such contributions could be modulated by the presence of other environmental chemicals.  相似文献

C-3 of (+) and (?)-α-pinene and of (?)-β-pinene biosynthesized in several Pinus species was derived from C-2 of mevalonate; and the hydrogen at C-5 in all the isomers was derived from that at C-6 in nerol. This pattern is consistent with two routes for bicyclization of the acyclic biosynthetic precursor: one leads to (?)-β-pinene and the other to (+)-α-pinene of opposite absolute configuration. (?)-α-Pinene probably results from subsequent isomerisation of the (?)-β-isomer, and (very small) amounts of (+)-β-pinene result from similar (unfavoured thermodynamically) isomerisation of the (+)-α-isomer.  相似文献

2002年,利用寄主挥发性物质(+)-α-蒎烯、(-)-β-蒎烯、3-蒈烯和(-)柠檬烯按不同配比制成诱芯,采用自行研制的挥发物释放装置,在山西省关帝山森林经营局西葫芦林场对危害油松Pinus tabulaeformis Carr.的重大外来入侵林业害虫红脂大小蠹Dendroctomus valens LeConte进行了引诱试验。结果表明,3蒈烯对红脂大小蠹的引诱作用最强,明显优于单独使用(+)-α-蒎烯和(-)-β-蒎烯的诱虫效果,也比北美应用的标准诱芯[(+)- α蒎烯(-)-β-蒎烯与3-蒈烯的比例为1∶1∶1]效果好;在标准诱芯中加入油松另一主要成分-(-)柠檬烯会明显降低诱虫效果。红脂大小蠹引诱剂最佳释放量为150 mg/d。该研究不仅对我国红脂大小蠹的监控具有实际应用意义,也从理论上探讨了红脂大小蠹与寄主油松协同进化的化学生态学过程。  相似文献

Analyses of volatiles in hindguts of Ips typgraphus males from different spruce trees and attack phases are reviewed. The composition of monoterpenes, and the chirality of α-pinene, have been determined in phloem samples. Relationships between compounds emanating from spruce trees and bark beetles, respectively, have been studied. Male beetles depend on their host tree for the production of pinene alcohols. The ratio between the pinene alcohols is almost constant in males boring in the same tree but can vary widely between males from different spruce trees. Very good correlations were found between some host tree monoterpene hydrocarbons and bark beetle produced pinene alcohols. The production of the essential pheromone component 2-methyl-3-buten-2-ol, was not correlated with the monoterpene content in the host trees, while the production of the other essential pheromone component, cis-verbenol, depends on the amount of the precursor, (−)-α-pinene in the phloem. Male beetles boring in a resistant spruce tree will continue to produce the pinene alcohols, including cis -verbenol, as long as the tree defends itself with resin.  相似文献

In the search for compounds that contribute to the host or habitat discrimination, antennae of Ips typographus were screened for sensitivity to volatiles released by spruce trap-trees using gas chromatography linked to electroantennography. The antennally active compounds were determined using comprehensive two-dimensional gas chromatography with time-of-flight mass spectrometric detection. Data show that I. typographus antennae respond to compounds emitted by the host. In total, 18 of antennally active compounds were detected: α-pinene, camphene, sabinene, β-pinene, myrcene, Δ-3-carene, p-cymene, limonene, β-phellandrene, 1,8-cineole, γ-terpinene, terpinolene, nonanal, camphor, trans-pinocamphone, cis-pinocamphone, terpinen-4-ol, and verbenone. Unequivocal identification of all active minor compounds is provided and confirmed using synthetic standards. Compounds in minor quantities like 1,8-cineole, β-phellandrene, camphor, cis-pinocamphone, and trans-pinocamphone were more active than major spruce monoterpenes. We hypothesize that the minor spruce compounds may play so far unrecognized role in conveying information about host suitability for I. typographus.  相似文献

In the present study, six monoterpenes [(?)-citronellal, p-cymene, (?)-menthone, α-pinene, α-terpinene, and (?)-terpinen-4-ol] and two phenylpropenes [trans-cinnamaldehyde and eugenol] were evaluated for their contact and fumigant toxicities against Sitophilus oryzae adults. The effects of these compounds on the mortality of S. oryzae adults in stored wheat and their inhibitory effects on acetylcholinesterase (AChE) and adenosine triphosphatases (ATPases) were examined. The tested compounds showed varying degrees of contact toxicity, with trans-cinnamaldehyde (LC50 = 0.01 mg/cm2) being the most potent compound, followed by (?)-menthone (LC50 = 0.013 mg/cm2) and eugenol (LC50 = 0.015 mg/cm2). In a fumigant toxicity assay, the monoterpenes α-terpinene, p-cymene, and (?)-menthone showed the highest toxicities (LC50 = 50.79, 52.37, and 54.08 μl/L air, respectively). Trans-cinnamaldehyde, (?)-citronellal, and eugenol were the least toxic (LC50 > 100 μl/L air). In general, the oxygenated compounds exhibited high contact toxicities while the hydrocarbon compounds exhibited high fumigant toxicities. When tested for their insecticidal activities against S. oryzae in stored wheat, trans-cinnamaldehyde was found to be the most potent compound, with 73.9% mortality at an application rate of 0.5 g/kg and complete mortality (100%) at 1 and 5 g/kg after 1 week of treatment. All of the tested compounds showed AChE inhibition, although (?)-citronellal and trans-cinnamaldehyde presented the strongest enzyme inhibition, with IC50 values of 18.40 and 18.93 mM, respectively. On the other hand, (?)-terpinene-4-ol exhibited the highest inhibition of ATPases, followed by α-pinene and α-terpinene.  相似文献

In response to infection by shoot infecting pathogens, Scots pine releases cortical resin into affected tissues. The resin contains a mixture of monoterpene compounds (α-pinene, β-pinene, 3-carene, β-myrcene, limonene and β-phellandrene) that retard the growth of a range of pathogens. The proportion of each monoterpene in the resin shows substantial variation among trees within a population. Thus pathogens on different trees encounter quite different monoterpene environments. To investigate the evolutionary response of pathogens to the chemically heterogeneous environment provided by Scots pine, isolates of the ascomycete canker pathogen Crumenulopsis sororia were collected from trees within a range of natural Scots pine populations. Growth rates of these isolates were measured in the presence and absence of five host monoterpenes. Substantial heritable variation for growth rate in the presence and absence of monoterpenes, and for monoterpene tolerance was recorded, suggesting the potential for the evolution of chemically specialized pathogen subpopulations on different trees within a wood. However, genetic correlations between growth rates in different monoterpene environments and between tolerance of different monoterpenes were either positive or non-significant, and there was no evidence of “tradeoffs” in performance under different monoterpene regimes. The results suggest that, on its own, the presence of monoterpene variability within Scots pine will not lead to disruptive selection on the C. sororia population. The relationship between defensive chemical diversity and pest resistance is discussed in the light of these results.  相似文献

The pinewood nematode (PWN) Bursaphelenchus xylophilus, vectored primarily by the sawyer beetle, Monochamus alternatus, is an important invasive pest and causal agent of pine wilt disease of Chinese Masson pine, Pinus massoniana. Previous work demonstrated that the ratios and concentrations of α-pinene∶β-pinene differed between healthy trees and those trees containing blue-stain fungus (and M. alternatus pupae). However, the potential influence of the altered monoterpene ratios and concentrations on PWN and associated fungi remained unknown. Our current results show that low concentrations of the monoterpenes within petri dishes reduced PWN propagation, whereas the highest concentration of the monoterpenes increased PWN propagation. The propagation rate of PWN treated with the monoterpene ratio representative of blue-stain infected pine (α-pinene∶β-pinene = 1∶0.8, 137.6 mg/ml) was significantly higher than that (α-pinene∶β-pinene = 1∶0.1, 137.6 mg/ml) representative of healthy pines or those damaged by M. alternatus feeding, but without blue stain. Furthermore, inhibition of mycelial growth of associated fungi increased with the concentration of the monoterpenes α-pinene and β-pinene. Additionally, higher levels of β-pinene (α-pinene∶β-pinene = 1∶0.8) resulted in greater inhibition of the growth of the associated fungi Sporothrix sp.2 and Ophiostoma ips strains, but had no significant effects on the growth of Sporothrix sp.1, which is the best food resource for PWN. These results suggest that host monoterpenes generally reduce the reproduction of PWN. However, PWN utilizes high monoterpene concentrations and native blue-stain fungus Sporothrix sp.1 to improve its own propagation and overcome host resistance, which may provide clues to understanding the ecological mechanisms of PWN''s successful invasion.  相似文献

Pheromones and metabolites of host (ponderosa pine) compounds were found in association with the hindgut of both naturally fed and of non-fed, host vapour-exposed bark beetles, Ips paraconfusus and Dendroctonus brevicomis. Much smaller amounts were found in the corresponding heads and mid guts. Sex-specific differences in content of pheromones were observed as in earlier studies. Exposure of I. paraconfusus to vapours of a pheromone component, ipsenol and other monoterpene alcohols resulted in their accumulation in the hindgut but relatively very low amounts in the head. The possible sites of pheromone biosynthesis are discussed. Exposure of male I. paraconfusus to vapours of host compounds, myrcene and α-pinene, revealed that immature adults do not produce the pheromone components, ipsenol and ipsdienol, as mature adults do while both immature and mature sexes produced another pheromone component, cis-verbenol, as well as trans-verbenol and myrtenol. Immature D. brevicomis adults did not contain pheromones until their exposure to vapours of (?)-α-pinene which caused production of trans-verbenol but only about 10% that of mature adults treated similarly. Verbenone, a male-produced inhibitory pheromone of D. brevicomis, apparently was not synthesized from (?)-α-pinene in females nor was its synthesis in males enhanced by exposure to this host compound.  相似文献

Bark beetle infestation and associated fungal infection are a serious disease problem in conifer species. Conifers have evolved elaborate, constitutive and inducible, terpene-based defense mechanisms to deter insect pests and their symbiotic fungal pathogens. This process involves the secretion of oleoresin, a complex mixture of monoterpenes, sesquiterpenes and diterpenoid acids. Induced oleoresinosis in grand fir (Abies grandis) provides a model system for studying the regulation of defensive terpene biosynthesis and for identifying relevant genes. The ecological relationships between conifers, beetle pests, beetle predators and fungal pathogens present several possible avenues for manipulating oleoresin composition to improve tree resistance. Possible examples include chemically disguising the host, adding toxins and altering the levels of pheromone precursors, attractants for predators or hormone mimics to disrupt insect development. Strategies and prospects for generating transgenic conifers with increased defense capability are discussed.  相似文献

The alien red turpentine beetle (RTB), Dendroctonus valens LeConte, is one of the most economically destructive forest pests in China, having killed more than 6 million pines in recent years. There is a need to understand the basic biology and ecology of the beetle in order to develop an effective monitoring and management strategy. In this study, the effects of hillside exposure (south- and north-facing), host-tree locations according to relief (valley, mid-slope, and ridge-top) and tree diameters on RTB colonization were investigated in one valley (3 sites). The results showed that (i) RTB clearly preferred colonizing pines growing on south-facing hillsides, especially in the valley; (ii) RTB preferred to colonize the pines growing at the valley rather than pines growing at mid-slope or on ridge-top; (iii) RTB preferred to colonize trees with large diameter over small and medium-sized pines; (iv) the attack density of RTBs (measured by pitch tubes/pine) was obviously higher on larger trees standing in the valley than other trees standing at other places. We conclude from RTB colonization patterns, that RTB prefers to attack large trees in the valley, which may be useful in developing a pest-management strategy.  相似文献

The volatile composition of six Hypericum species has been studied. The essential oils were obtained by steam distillation in 500 mL H2O for 2 h in a modified Clevenger apparatus with a water-cooled oil receiver to reduce hydrodistillation over-heating artifacts, and their analyses were performed by GC and GC–MS. Identification of the substances was made by comparison of mass spectra and retention indices with literature records. A total of 100 different compounds were identified. The main constituents of the investigated populations of each taxon have been revealed as follows: Hypericum alpinum: (−)-β-pinene, γ-terpinene, (−)-(E)-caryophyllene; Hypericum barbatum: (−)-α-pinene, (−)-β-pinene, (−)-limonene, (−)-(E)-caryophyllene, (−)-caryophyllene oxide; Hypericum rumeliacum: (−)-α-pinene, (−)-β-pinene, (−)-limonene, Hypericum hirsutum: nonane, undecane, (−)-(E)-caryophyllene, (−)-caryophyllene oxide; Hypericum maculatum: spathulenol, globulol; Hypericum perforatum: (−)-α-pinene, (Z)-β-farnesene, germacrene D; Monoterpene hydrocarbons were shown to be the main group of the taxa belonging to the section Drosocarpium, while the taxa of section Hypericum were more rich in sesquiterpene hydrocarbons.  相似文献

Selected monoterpenes inhibited methane oxidation by methanotrophs (Methylosinus trichosporium OB3b, Methylobacter luteus), denitrification by environmental isolates, and aerobic metabolism by several heterotrophic pure cultures. Inhibition occurred to various extents and was transient. Complete inhibition of methane oxidation by Methylosinus trichosporium OB3b with 1.1 mM (−)-α-pinene lasted for more than 2 days with a culture of optical density of 0.05 before activity resumed. Inhibition was greater under conditions under which particulate methane monooxygenase was expressed. No apparent consumption or conversion of monoterpenes by methanotrophs was detected by gas chromatography, and the reason that transient inhibition occurs is not clear. Aerobic metabolism by several heterotrophs was much less sensitive than methanotrophy was; Escherichia coli (optical density, 0.01), for example, was not affected by up to 7.3 mM (−)-α-pinene. The degree of inhibition was monoterpene and species dependent. Denitrification by isolates from a polluted sediment was not inhibited by 3.7 mM (−)-α-pinene, γ-terpinene, or β-myrcene, whereas 50 to 100% inhibition was observed for isolates from a temperate swamp soil. The inhibitory effect of monoterpenes on methane oxidation was greatest with unsaturated, cyclic hydrocarbon forms [e.g., (−)-α-pinene, (S)-(−)-limonene, (R)-(+)-limonene, and γ-terpinene]. Lower levels of inhibition occurred with oxide and alcohol derivatives [(R)-(+)-limonene oxide, α-pinene oxide, linalool, α-terpineol] and a noncyclic hydrocarbon (β-myrcene). Isomers of pinene inhibited activity to different extents. Given their natural sources, monoterpenes may be significant factors affecting bacterial activities in nature.  相似文献
