The apparent activity coefficients for sodium (gamma'Na) in the blood of six insect species have been calculated from measurements made with sodium-selective electrodes and a flame photometer. In every case gamma Na was significantly lower than that for this cation in free solution (gammaNa). In Periplaneta americana gammaNa varied considerably, during a period of 90 days, so that a relatively constant sodium activity (aNa) was maintained in the blood in the face of large variations in the total sodium content measured by flame photometry. Despite the relative constancy of aNa (of around 0.088M) appreciable fluctuations were observed in the sodium and potassium content of nervous connective over a period of 140 days. The values of aNa and aK were used to devise a satisfactory cockroach saline for use in experiments with isolated nerve cords.  相似文献

The role played by molecules of the strigolactone family in stimulating the germination of seeds of parasitic weeds of the genera Striga, Orobanche and Alectra has never been clearly elucidated. The biogenesis of these unusual terpenoid lactones, originally identified in minute quantities in the root exudates of a small number of host plants and two or three "false hosts", also remains obscure. These lactones, as the chemical signals which initiate the life cycle of Striga, are consequently at the forefront of the Striga research effort. This paper reviews recent key discoveries relating to the biosynthesis and mode of action of strigolactones, and summarises the evidence suggesting that these molecules may be far more widely distributed and have a greater physiological significance than has hitherto been appreciated.  相似文献

Human cysteine protease cathepsin L was inactivated at acid pH by a first-order process. The inactivation rate decreased with increasing concentrations of a small synthetic substrate, suggesting that substrates stabilize the active conformation. The substrate-independent inactivation rate constant increased with organic solvent content of the buffer, consistent with internal hydrophobic interactions, disrupted by the organic solvent, also stabilizing the enzyme. Circular dichroism showed that the inactivation is accompanied by large structural changes, a decrease in alpha-helix content being especially pronounced. The high activation energy of the reaction at pH 3.0 (200 kJ.mol-1) supported such a major conformational change occurring. The acid inactivation of cathepsin L was irreversible, consistent with the propeptide being needed for proper folding of the enzyme. Aspartic protease cathepsin D was shown to cleave denatured, but not active cathepsin L, suggesting a potential mechanism for in-vivo regulation and turnover of cathepsin L inside lysosomes.  相似文献

A pectic polysaccharide, lemnan, was isolated from freshly collected duckweed Lemna minor L. Its sugar chain was shown to be mainly composed of the residues of D-galacturonic acid (64%), galactose, arabinose, xylose, and D-apiose, a branched chain sugar. The high content of D-apiose (25%) indicated that lemnan is an apiogalacturonan type pectin similar to zosteran, a pectic polysaccharide from a sea phanerogam of the Zosteraceae family. The results of partial acidic hydrolysis, pectinase digestion, and NMR studies of lemnan demonstrated that its macromolecule contains regions of the linear alpha-1,4-D-galacturonan and branched apiogalacturonan. The side chains of apiogalacturonan were found to be formed of single and 1,5-linked residues of D-apiofuranose attached to 2- and 3-positions of the D-galacturonic acid residues of the apiogalacturonan backbone. Lemnan was shown to exhibit an immunomodulatory effect by activating the system of phagocytosis.  相似文献

This paper attempts to combine (and thereby briefly review) various sets of physiological data in order to outline a qualitative model of the different states of stochastic neural activity underlying different forms of physiological tremor. Particular emphasis in put on spatial distributions of the properties of neural elements and their interconnexions, and on discharge characteristics of motor units and muscle spindle afferents including so-called early discharges and nonlinearities. It is argued that the wide variety of internal anatomical and functional structures of skeletal muscles and of their reflex organization must be considered when dealing with stability problems. Computer simulations of stochastic population models of the involved neuromuscular elements are advocated as means to investigate the relative importance of the many factors possibly contributing to stabilizing or de-stabilizing neuromuscular systems.  相似文献

Bacillus cereus sphingomyelinase activity was assayed on large unilamellar vesicles composed of sphingomyelin (SM)/cholesterol (Ch) mixtures at varying proportions. Natural (egg) SM was used with a gel–fluid transition temperature at ca. 40 °C. When the enzyme was assayed at 37 °C, the activity on pure SM was exceedingly low, but a small increase was observed as soon as some Ch was added, and a large enhancement of activity occurred with Ch proportions above 25 mol%. The data were interpreted in terms of sphingomyelinase activity being higher in the cholesterol-induced liquid-ordered phase than in the gel phase. The abrupt increase in activity above 25 mol% Ch would occur as a result of a change in domain connectivity, when the Ch-rich liquid-ordered domains coalesced. In equimolar SM/Ch mixtures, that were in the liquid-ordered state in a wide range of temperatures, sphingomyelinase activity was virtually constant in the 30–70 °C range. The results demonstrate that at the mammalian and bird physiological temperatures Ch modulates sphingomyelinase activity, and that this can occur precisely because most SM have a gel–fluid transition temperature above the physiological temperature range. In addition, Ch activation of sphingomyelinase and the strong affinity of Ch for SM allow the rapid, localised and self-contained production of the metabolic signal ceramide in specific microdomains (rafts).  相似文献

An assay method for glyceraldehyde-3-phosphate dehydrogenase in which none of the primary products accumulate and which gives linear kinetics under physiological conditions has been developed. It is based on the use of the 1,3-diphosphoglycerate produced by the enzyme for the formation of NADPH, while the NADH produced is recycled with an auxiliary system. Revised Km values at pH 7.4 for the muscle (rabbit and rat) enzyme are: glyceraldehyde-3-P, 50 μM; NAD, 100 μM; Pi, 10 mM. The rat erythrocyte enzyme gave similar values except for glyceraldehyde-3-P which was 300 μM. Cooperativity for NAD+ tends to be positive but is a variable parameter.  相似文献

We investigate the relationship between the extensor electromyogram (EMG) and tremor time series in physiological hand tremor by cross-spectral analysis. Special attention is directed to the phase spectrum and the effects of observational noise. We calculate the theoretical phase spectrum for a second-order linear stochastic process and compare the results to measured tremor data recorded from subjects who did not show a synchronized EMG activity in the corresponding extensor muscle. The results show that physiological tremor is well described by the proposed model and that the measured EMG represents a Newtonian force by which the muscle acts on the hand. Electroreduction studies were performed on several cross-conjugated mesomeric betaines containing the fused pyrazolium (2) and fused imidazolium (3) ring systems. Studies at acidic pH were of principal interest. Substituent effects for 2 were in line with prior findings, and reduction potentials were comparatively negative (-0.96 to -1.34 V). Reduction potentials fit the modified Hammett equation. Compound 3 was more readily reduced (-0.88 V). The related psi-oxatriazoles (6) gave values in the range of -0.85 to -1.22 V. The electrochemical characteristics are compared with those of the mesoionic sydnones (4) and sydnoneimines (5). These mesoionic compounds were generally reduced at more positive potentials than 2 and 3. A relationship between electroreduction and physiological activity is proposed. The overall results are in keeping with the hypothesis of widespread participation of iminium-type species in biological systems.  相似文献
