Lactobacillus casei Zhang是一株分离自传统酸马奶中的益生菌。本文研究了不同碳源、氮源、碳氮比例、微量元素及缓冲盐对Lactobacillus casei Zhang增殖培养的效果, 并采用响应面法对优选的碳源、氮源和缓冲盐类的组成含量进行优化, 得到Lactobacillus casei Zhang的增殖培养基为:葡萄糖 20.9 g/L、大豆蛋白胨10.45 g/L、酵母粉10.45 g/L、K2HPO4 3.5 g/L、醋酸钠14.6 g/L、柠檬酸钠2.3 g/L、MgSO4?7H2O 1 g/L、MnSO4?5H2O 54 mg/L、CuSO4?5H2O 10 mg/L、吐温80为1 g/L。Lactobacillus casei Zhang在此增殖培养基中经37℃18 h培养活菌数可达到4.78×109 CFU/mL, 比在MRS中(4.8×108 CFU/mL)提高近10倍。  相似文献

本文重点在于提高酪酸梭状芽孢杆菌活菌数量,降低发酵培养基成本.通过对发酵培养基中不同碳源、氮源、生长因子等进行单因素研究,得到最佳培养基组成:可溶性淀粉10 g/L,豆粕(中性蛋白酶水解3 h)20 g/L,玉米浆3g/L.用此培养基在37℃培养24h,采用高层半固体琼脂试管法对酪酸梭状芽孢杆菌进行活菌计数,活菌数可达8.2×108 cfu/mL.培养基中添加K2HPO4 5g/L、MgSO4·7H2O 0.2 g/L、MnSO4·H2O 0.2 g/L培养32 h时,酪酸梭状芽孢杆菌芽孢转化率可达95%.  相似文献

为提高一株具有藻毒素清除能力的干酪乳杆菌Lactobacillus casei BBE10-212单位体积的活菌数,针对其营养需求,研究了不同碳源、氮源、缓冲盐、微量元素及生长因子对该菌株生长情况及发酵特性的影响.通过响应面法对碳源、氮源、生长因子等进行优化,获得最佳培养基配方为:α-乳糖43.8 g/L,酵母膏79.5 g/L,无水乙酸钠13.12 g/L,冰醋酸9.17 mL/L,MnSO4 ·H2O 190mg/L,吐温-80 5.15 mL/L.经37℃培养18 h,菌体干重达到4.97 g/L,比在普通MRS培养基中(1.32 g/L)提高近4倍.基于乳酸菌发酵过程中的产酸特性,通过外源添加5 g/L谷氨酸,促使菌体浓度进一步提高15%,并提前1.2h进入生长稳定期.上述研究结果为食品行业重要生产菌株干酪乳杆菌的高密度培养技术提供了可借鉴的研究思路.  相似文献

研究不同碳源、氮源和无机盐对毕赤酵母AX181菌株产木聚糖酶的影响.实验表明,分别采用葡萄糖和玉米浆干粉为碳源和氮源可以明显提高木聚糖酶的产量.无机盐单因子优化实验显示添加适量的(NH4)2SO4、KH2 PO4、MnSO4·H2O、FeSO4·7H2O也可以部分提高木聚糖酶产量.在此基础上利用响应面法优化毕赤酵母产木聚糖酶培养基,利用12次实验的Plackett - Burman设计实验筛选出影响产木聚糖酶的3个主要因素,即玉米浆干粉、MrSO4 ·H2O和FeSO4·7H2O.并进一步通过最陡爬坡路径逼近最大响应区域,采用中心组合实验设计确定最佳条件.优化后的产木聚糖酶培养基组分为(g/L):葡萄糖40.00,玉米浆干粉80.84,(NH4)2SO4 6.25,KH2PO4 1.25、MnSO4·H2O 0.35,FeSO4 ·7H2O 1.31.培养基优化后,实际产酶2 883.86 U/mL,是优化前YPD培养基产酶的2.51倍.  相似文献

为实现L. casei Zhang的高密度培养,在之前优化增殖培养基的基础上进一步寻求适宜该菌的培养条件。研究了不同中和剂、缓冲盐浓度、葡萄糖浓度、pH值控制、通气条件和补料分批培养对菌体在恒pH条件下发酵的影响,根据不同条件下菌体的比生长速率、菌体密度和活菌数情况,确定L. casei Zhang较适宜的高密度培养条件为:培养基葡萄糖浓度为80 g/L~100 g/L,以氨水为中和剂使pH保持5.9,采用间歇通氮气的方法保持环境厌氧,分批培养方式下37°C保温发酵10 h~12 h后,L. casei Zhang细胞干重达到7 g/L,活菌数3.5×1010 CFU/mL,比优化前提高7倍以上,能够满足益生菌制品生产要求的高菌体密度。  相似文献

以淡色生赤壳菌(Bionectria ochroleuca)Bo-1菌株发酵液乙酸乙酯粗提物对水稻白叶枯病菌(Xanthomonas oryzaepv.oryzae,Xoo)抑菌活性为检测指标,采用单因素试验优化Bo-1菌株产生抗菌物质培养所需的碳源、氮源、无机盐;通过正交试验优化培养基配方和摇瓶发酵条件。研究结果表明,Bo-1菌株产生抗菌物质适宜的碳源、氮源和无机盐分别为淀粉、蛋白胨、MgSO4·7H2O;优化的培养基配方为:淀粉30 g/L,蛋白胨2 g/L,MgSO4·7H2O 0.5 g/L;适宜的发酵条件为:温度30℃,转速150 r/min,装液量80 mL/250 mL,pH 6.5。  相似文献

利用一株分离自传统发酵酸马奶中的益生干酪乳杆菌(Lactobacillus casei Zhang)进行固态发酵(Solid State Fermentation,SSF)。以发酵物中的活菌数为主要指标,采用九因素四水平(L32(4^9))的正交试验优化固态发酵培养基,并在优化的培养基基础上研究不同的初始含水量及培养时间对Lactobacillus casei Zhang活菌数的影响。实验结果表明,在固态发酵培养基组成为4g豆粕、5g麸皮、0.6g乳清粉、0.3g葡萄糖、0.3g碳酸钙、0.02g硫酸铵、0.01g硫酸镁,初始含水量为55%的优化条件下,37℃发酵60h,发酵物中Lactobacillus casei Zhang活菌数可达到4.08×10^10CFU/g。  相似文献

【目的】对一株具有抗肿瘤活性的银杏内生真菌Aspergillus oryzae YX-5的发酵培养基进行优化。【方法】以发酵后的菌体干重、粗提物质量和粗提物抗肿瘤活性为指标,通过单因素实验选出合适的碳源和氮源,再以选出的碳源、氮源以及K2HPO4、MgSO4·7H2O、KCl共5个因素进行正交实验,确定出最适宜的发酵培养基配方。【结果】YX-5的最佳发酵培养基配方为葡萄糖45 g/L,蛋白胨8 g/L,K2HPO4 0.5 g/L,MgSO4·7H2O 0.2 g/L,KCl 1 g/L,FeSO40.01 g/L。优化后菌体干重、代谢物的产量和抗肿瘤活性分别提高了41.88%、226.52%和19.31%。【结论】通过对米曲霉菌(Aspergillus oryzae)YX-5的发酵培养基进行优化,明显提高了粗提物产量和抗肿瘤活性,这对后期规模化发酵中减小发酵规模和降低工作量十分有利。  相似文献

通过对6种藓类植物,即褶叶青藓(Brachythecium salebrosum(Web.et Mohr.)B.S.G.)、湿地匐灯藓(Plagiomnium acutum(Lindb.)Kop.)、侧枝匐灯藓(Plagiomnium maximoviczii(Lindb.)Kop.)、大凤尾藓(Fissidensnobilis Griff.)、大羽藓(Thuidium cymbifolium(Doz.et Molk.)B.S.G.)和大灰藓(Hypnum plumaeforme Wils.)嫩茎和老茎的石蜡切片和显微观察发现,同一藓类植株的嫩茎和老茎,茎结构稳定,不同种藓类植物茎横切面具有不同特征.植物体茎横切面形状、表层细胞的层数、细胞大小和细胞壁厚薄、皮层细胞大小和形状、中轴的有无以及比例等特征可以作为藓类植物的分科分类依据之一.  相似文献

Seed germination of an aurea mutant of tomato ( Lycopersicon esculentum Mill.) is promoted by continuous irradiation with red, far-red or long-wavelength far-red (758 nm) light as well as by cyclic irradiations (5 min red or 5 min far-red/25 min darkness). Far-red light applied immediately after each red does not change the germination behaviour. Seed germination of the isogenic wild-type, cv. UC-105, is promoted by continuous and cyclic red light while it is inhibited by continuous and cyclic far-red light and by continious 758 nm irradiation. Far-red irradiation reverses almost completely the promoting effect of red light. The promoting effect (in the aurea mutant) and the inhibitory effect (in the wild-type) of continuous far-red light do not show photon fluence rate dependency above 20 nmol m−2 s−1. It is concluded that phytochrome controls tomato seed germination throgh low energy responses in both the wild type and the au mutant. The promoting effect of continuous and cyclic far-red light in the au mutant can be attributed to a greater sensitivity to Pfr.  相似文献

A complex analysis of seasonal fluctuations of the mean group parameters of the system of regulation of lipid peroxidation has been performed in liver of Balb/c mice. Association of lipid characteristics and morphophysiological parameters is studied in the Balb/c mouse liver. An inter-connection is revealed between the liver index and the amount of lysoforms of phospholipids, the scale and character of the interconnection differing essentially depending on proportion of phos-phatidylcholine in mouse liver phospholipids.  相似文献

Seven strains of aerobic carbon monoxide-oxidizing bacteria (carboxydebacteria) when growing on CO as sole source of carbon and energy had doubling times which ranged from 12–42 h. The activity profiles obtained after discontinuous sucrose density gradient centrifugation indicated that the CO-oxidizing enzymes are soluble and the hydrogenases are membrane-bound in all strains examined. The CO-oxidizing enzymes of Pseudomonas carboxydohydrogena, Pseudomonas carboxydoflava, Comamonas compransoris, and the so far unidentified strains OM2, OM3, and OM4 had a molecular weight of 230,000; that of Achromobacter carboxydus amounted to 170,000. The molecular weights of the CO-oxidizing and H2-oxidizing enzymes turned out to be identical. The cell sonicates were shown to catalyze the oxidation of both CO and H2 with methylene blue, thionine, phenazine methosulfate, toluylene blue, dichlorophenolindophenol, cytochrome c or ferricyanide as electron acceptors. Methyl viologen, benzyl viologen, FAD+, FMN+, and NAD(P)+ were not reduced. The spectrum of electron acceptors was identical for all strains tested. Neither free formate, hydrogen nor oxygen gas were involved in the CO-oxidation reaction. Methylene blue was reduced by CO at a 1:1 molar ratio. The results indicate that CO-oxidation by carboxydobacteria is catalyzed by identical or similar enzymes and that the reaction obeys the equation CO+H2OCO2+2H++2e- as previously shown for Pseudomonas carboxydovorans.Dedicated to Otto Kandler remembering almost three decades of enjoyable cooperation  相似文献

Since the introduction of the concepts of allostery about four decades ago, much advancement has been made in elucidating the structure-function correlation in allostery. However, there are still a number of issues that remain unresolved. In this review we used mammalian pyruvate kinase (PK) as a model system to understand the role of protein dynamics in modulating cooperativity. PK has a triosephosphate isomerase (TIM)(α/β)8 barrel structural motif. PK is an ideal system to address basic questions regarding regulatory mechanisms about this common (α/β)8 structural motif. The simplest model accounting for all of the solution thermodynamic and kinetic data on ligand-enzyme interactions involves two conformational states, inactive ET and active ER. These conformational states are represented by domain movements. Further studies provide the first evidence for a differential effect of ligand binding on the dynamics of the structural elements, not major secondary structural changes. These data are consistent with our model that allosteric regulation of PK is the consequence of perturbation of the distribution of an ensemble of states in which the inactive ET and active ER represent the two extreme end states. Sequence differences and ligands can modulate the distribution of states leading to alterations of functions. The future work includes: defining the network of functionally connected residues; elucidating the chemical principles governing the sequence differences which affect functions; and probing the nature of mutations on the stability of the secondary structural elements, which in turn modulate allostery.  相似文献

Summary Embryogenic tissues of sugi (Cryptomeria japonica) were induced on a modified Campbell and Durzan (CD) medium containing 1 μM 2,4-dichlorophenoxyacetic acid (2,4-D) and 600 mg l−1 glutamine, and subcultured in the medium of the same composition for over 1 yr. This resulted in a mixed culture of embryogenic and non-embryogenic cells. When embryogenic cells were isolated and cultured independently, their capacity to form embryogenic aggregates was lost. Thus, the non-embryogenic cells present within a mixed culture system were essential to the formation of embryogenic aggregates. When embryogenic tissues were isolated and cultured independently on a high glutamine-containing (2400 mg l−1) medium, dry weights and endogenous levels of glutamine increased, and the tissue could generate a large number of embryogenic aggregates. Amino acid analysis of embryogenic and non-embryogenic cells from the maintenance culture indicated a higher level of glutamine was present in the latter. The high endogenous level of glutamine in the non-embryogenic portion of mixed cell masses may be the supplier of glutamine for maintaining the embryogenic property of the tissues.  相似文献
